Sugar beet leaves as new source of hydroperoxide lyase in a bioprocess producing green-note aldehydes.

Hydroperoxide lyase activity was found in sugar beet leaves. Its optimum pH and temperature were, respectively, 6.7 and 22 degrees C. Under these conditions, conversion of linolenic acid 13-hydroperoxide to cis-3-hexenal with a maximum yield of 80% was reached after only 2 min. The stability of cis-3-hexenal was improved by acidifying the reaction medium. Based on these studies, a bioprocess producing green-note aldehydes in a laboratory-scale was achieved.

Development of a biocatalytic process for the production of c6-aldehydes from vegetable oils by soybean lipoxygenase and recombinant hydroperoxide lyase.

Volatile C6- and C9-aldehydes and alcohols are widely used as food flavors to reconstitute the "fresh green" odor of fruits and vegetables lost during processing. To meet the high demand for natural flavors, an efficient, cheap, and versatile biocatalytic process was developed to produce C6-aldehydes on a large scale. Vegetable oils were converted by soybean lipoxygenase and recombinant hydroperoxide lyase into hexanal and (2E)- or (3Z)-hexenal. In contrast to plant extracts, generally used as enzyme sources, high molar conversions were obtained with recombinant hydroperoxide lyase (50% for he

HCN production and hydroxynitrile lyase: a natural activity in plants and a renewed biotechnological interest.

Over 3,000 plant species are cyanogenic. Cyanogenesis is defined as the hydroxynitrile lyase catalysed release of a cyanide group in the form of HCN and the corresponding aldehyde or ketone. When a plant is attacked, HCN released is a self defence mechanism. A special characteristic of enzymatic reactions is that all enzymatic reactions are reversible-hydroxynitrile lyases can also be used for the synthesis of enantiomerically pure cyanohydrins which are of great importance in industry. This article presents a comprehensive review of the role of hydroxynitrile lyases, both in nature and indust

Epigenetic regulation of pro-inflammatory cytokine secretion by sphingosine 1-phosphate (S1P) in acute lung injury: Role of S1P lyase.

Cellular level of sphingosine-1-phosphate (S1P), the simplest bioactive sphingolipid, is tightly regulated by its synthesis catalyzed by sphingosine kinases (SphKs) 1 & 2 and degradation mediated by S1P phosphatases, lipid phosphate phosphatases, and S1P lyase. The pleotropic actions of S1P are attributed to its unique inside-out (extracellular) signaling via G-protein-coupled S1P1-5 receptors, and intracellular receptor independent signaling. Additionally, S1P generated in the nucleus by nuclear SphK2 modulates HDAC1/2 activity, regulates histone acetylation, and transcription of pro-inflamma

Mammalian 2-hydroxyacyl-CoA lyases: Discovery and assays.

2-Hydroxyacyl-CoA-lyases (HACLs) catalyze a carbon-carbon cleavage of long chain 2-hydroxyacyl-CoAs, generating formyl-CoA and a one-carbon shortened long chain aldehyde as primary products. Two enzymes were characterized in mammals, one being a peroxisomal matrix enzyme (HACL1), the other associated with the endoplasmic reticulum (HACL2). They are the sole known thiamine pyrophosphate depending enzymes involved in the breakdown of fatty acids, and are crucial for the α-oxidation of phytanic acid and long chain 2-hydroxy fatty acids.

Synthesis of ortho-Formylphenylphosphonic Acids as Covalent Probes of Active Site Lysines.

During the course of an investigation of targeted inhibition of DNA polymerase beta (pol β) lyase activity using small molecules, we observed the formation of an aldimine between (2-formyl)phenylphosphonic acid (2FPP) and butylamine under basic aqueous conditions; complete deprotonation of the phosphonate group was required to stabilize the imine product. Results of computational docking studies suggested that the reaction of Lys-72 on the lyase active site with an aldehyde group could be facilitated by a proximal phosphonate, not only because of the phosphonate's ability to mimic phosphate in

A lipid-hydrolysing activity involved in hexenal formation.

Short-chain aldehydes such as (3Z)-hexenal and n-hexanal are formed from lipids through sequential actions of lipid-hydrolysing, lipoxygenase and fatty acid hydroperoxide lyase activities. The aldehydes are formed upon wounding of plant tissues, and are reported to have bactericidal and fungicidal activities. Furthermore, it has been reported that the aldehydes can induce expression of a subset of genes involved in disease resistance and that they are involved in a defence response against insect herbivores. Although several genes encoding lipoxygenases and the lyases have been isolated, and c

Hydroxynitrile lyases from prunus seeds in the preparation of cyanohydrins.

The hydroxynitrile lyase (HNL) activity of nine defatted Prunus seeds was compared for catalyzing the addition of HCN to aromatic, heteroaromatic and α,β-unsaturated aldehydes. Although the conversion and enantiomeric excess (ee) of the corresponding cyanohydrins were both influenced by the HNL source and the chemical structure of the aldehyde, Prunus HNLs were all suitable for the enantioselective preparation of cyanohydrins.

Structural and functional analysis of hydroxynitrile lyase from Baliospermum montanum with crystal structure, molecular dynamics and enzyme kinetics.

Hydroxynitrile lyases (HNLs) catalyze degradation of cyanohydrins to hydrogen cyanide and the corresponding ketone or aldehyde. HNLs can also catalyze the reverse reaction, i.e., synthesis of cyanohydrins. Although several crystal structures of S-selective hydroxynitrile lyases (S-HNLs) have been reported, it remains unknown whether and how dynamics at the active site of S-HNLs influence their broad substrate specificity and affinity. In this study, we analyzed the structure, dynamics and function of S-HNL from Baliospermum montanum (BmHNL), which has an α/β hydrolase fold. Two crystal structu

Glyoxalase enzyme system in human muscular dystrophy.

Glyoxalase I and glyoxalase II activities were determined in skeletal muscle of control subjects and of patients with Duchenne dystrophy, other major forms of muscular dystrophies and certain neuromuscular disorders. The glyoxalase I activity was normal in all diseases examined except in Duchenne and limb girdle types of muscular dystrophy, where it showed a significant moderate decrease. The glyoxalase II activity in normal human muscle was very low, and the activity was unaltered in muscle of patients with Duchenne and other major forms of muscular dystrophies and spinal muscular atrophy. DERA-Catalyzed Chemoenzymatic Access to Nucleobase-Substituted Candidate Statin Precursors.

Aldolases are powerful biocatalysts for the stereoselective formation of carbon-carbon bonds and are widely used in the synthesis of chiral intermediates for pharmaceutical applications. Among them, 2-deoxyribose-5-phosphate aldolase (DERA) has been extensively exploited for the preparation of the conserved side chain of statins. In this work, we report a novel chemoenzymatic approach for the synthesis of nucleobase-substituted lactol products as potential precursors of new statin analogues. A C49M variant of DERA from Pectobacterium atrosepticum (PaDERA C49M) was employed to catalyze sequenti

Formaldehyde aldolases: Key enzymes in C-C bond formation and C1 assimilation.

The conversion of C1 compounds such as CO2, formic acid, formaldehyde (FALD), and methanol into longer-chain molecules depends on efficient C-C bond formation. Enzymatic strategies offer sustainable routes for such transformations, but their application remains limited by insufficient stability, specificity, and/or activity. A critical step here is the enzymatic condensation of FALD, which is promising building block due to its reactivity. This review discusses the catalytic mechanism, application, and limitation of three classes of aldolases that catalyze C-C bond formation using formaldehyde

Asymmetric synthesis of an (R)-cyanohydrin using enzymes entrapped in lens-shaped gels.

[structure: see text] A novel synthesis of (R)-cyanohydrins is described which is based on the use of cross-linked and subsequently poly(vinyl alcohol)-entrapped (R)-oxynitrilases. These immobilized lens-shaped biocatalysts have a well-defined macroscopic size in the mm range, show no catalyst leaching, and can be recycled efficiently. Furthermore, this immobilization method is cheap and the entrapped (R)-oxynitrilases gave similar good results compared with those of free enzymes. The (R)-cyanohydrin was obtained in good yields and with high enantioselectivities of up to >99% ee.

Modeling-Assisted Design of Thermostable Benzaldehyde Lyases from Rhodococcus erythropolis for Continuous Production of α-Hydroxy Ketones.

Enantiopure α-hydroxy ketones are important building blocks of active pharmaceutical ingredients (APIs), which can be produced by thiamine-diphosphate-dependent lyases, such as benzaldehyde lyase. Here we report the discovery of a novel thermostable benzaldehyde lyase from Rhodococcus erythropolis R138 (ReBAL). While the overall sequence identity to the only experimentally confirmed benzaldehyde lyase from Pseudomonas fluorescens Biovar I (PfBAL) was only 65 %, comparison of a structural model of ReBAL with the crystal structure of PfBAL revealed only four divergent amino acids in the substrat

Studies on the kinetics of cyanohydrin synthesis and cleavage by the the flavoenzyme oxynitrilase.

Almond oxynitrilase (D-alpha-hydroxynitrile lyase, EC 4.1.2.10) catalyzes the reversible condensation of HCN with aldehydes to form D-alpha-hydroxynitriles. Steady-state kinetic parameters for cleavage and synthesis of mandelonitrile and vanillin cyanohydrin were determined at pH 5.5 which is near the pH optimum of the enzyme. Benzaldehyde and vanillin act as competitive inhibitors of cyanohydrin cleavage while noncompetitive inhibition was observed for HCN. The results are consistent with an ordered uni bi mechanism in which aldehyde is the first substrate bound. Competitive inhibition of cya

Asymmetric Conjugate Hydrocyanation of α,β-Unsaturated Aldehydes Catalyzed by Engineered 2-Deoxy-D-ribose-5-phosphate Aldolase.

The enantioselective conjugate hydrocyanation of α,β-unsaturated aldehydes remains a long-standing challenge in synthetic chemistry. Here, we report the redesign of 2-deoxy-D-ribose-5-phosphate aldolase (DERA) into an efficient biocatalyst capable of promoting the asymmetric conjugate addition of hydrogen cyanide (generated in situ from trimethylsilyl cyanide) to aromatic enals via an iminium activation pathway. Biocatalytic asymmetric aldol addition into unactivated ketones.

Enzymes are renowned for their catalytic efficiency and selectivity, but many classical transformations in organic synthesis have no biocatalytic counterpart. Aldolases are prodigious C-C bond-forming enzymes, but their reactivity has only been extended past activated carbonyl electrophiles in special cases. To probe the mechanistic origins of this limitation, we use a pair of aldolases whose activity is dependent on pyridoxal phosphate. Our results reveal how aldolases are limited by kinetically favourable proton transfer with solvent, which undermines aldol addition into ketones. We show how
