Characteristics of inapparent Aleutian disease virus infection in mink.

Inapparent of nonprogressive Aleutian disease virus (ADV) infection is a subclinical but persistent virus infection of mink. Mink with the inapparent type of ADV infection when subjected to stress did not develop the progessive form of the disease. However, when challenged with a large dose of the virus, these mink did develop progressive Aleutian disease indicating that they were not highly resistant to the virus. Sera of mink with either the progressive of the inapparent type of ADV infection did not neutralise the virus. The anti-ADV antibody activity in mink with inapparent type of ADV inf

Isolation of Aleutian mink disease virus by affinity chromatography.

Affinity chromatography was used to isolate the Aleutian disease virus of mink. Dissociation of the immunoadsorbent-virus complex with 0.75 molar sodium chloride and then with a glycine-hydrochloride gradient released infective particles resembling picornaviruses. The elution profile suggests that two different types of virus-antibody complexes are formed, one dissociated by sodium chloride and another that requires glycine-hydrochloride in addition to sodium chloride for release of virus.

Intrapulmonary lymphoid tissue in mink infected with Aleutian disease virus.

Pulmonary lymphoid tissue was studied histologically in mink infected experimentally and naturally with Aleutian disease virus, an infection characterised by systemic lymphoid tissue hyperplasia. Lymphoid tissue hyperplasia was observed in the lamina propria of the bronchial tree, in perivascular tissue, in interlobular septae and subpleurally by day 42 after experimental aerosol infection. Four forms of pulmonary lymphoid tissue were recognised: lymphoepithelial nodules or bronchus-associated lymphoid tissue; lymphoid clusters; lymph nodes and diffuse lymphoid tissue. The cel components of th

Drivers Shaping Spillover of Aleutian Mink Disease Virus Introduced With American Mink Among Native Mustelids.

Invasive alien species pose a major threat to ecosystems by outcompeting native species for resources, altering habitats, enabling potential genetic hybridisation and introducing pathogens into the environment. An understanding of the factors that determine virus transfer between invasive and native species is crucial to the mitigation of the negative impact of the pathogens introduced. This study presents a comprehensive analysis of factors influencing Aleutian mink disease virus (AMDV) infection in native mustelids in Poland, following its introduction by feral American mink. AMDV seropreval…

Transcriptomic profiling of mink spleen infected by Aleutian mink disease virus: Insight into immune response.

Aleutian disease (AD) is one of the most challenging mink diseases that causes high mortality and affects several economically important traits. Aleutian mink disease virus (AMDV) targets multiple organs, the spleen being one of the major targeted organs. Transcriptomics has been widely used to reveal genes and biological pathways and to identify biomarkers for early detection or prevention of diseases. This study aimed to identify genes and pathways related to the host response to AD infection in the spleen using transcriptomics. For this purpose, twelve AMDV-free black male mink (3 sets of 4…

Long-term antibody production and viremia in American mink (Neovison vison) challenged with Aleutian mink disease virus.

BACKGROUND: Selecting American mink (Neovison vison) for tolerance to Aleutian mink disease virus (AMDV) has gained popularity in recent years, but data on the outcomes of this activity are scant. The objectives of this study were to determine the long-term changes in viremia, seroconversion and survival in infected mink. Mink were inoculated intranasally with a local isolate of Aleutian mink disease virus (AMDV) over 4 years (n = 1742). The animals had been selected for tolerance to AMDV for more than 20 years (TG100) or were from herds free of AMDV (TG0). The progenies of TG100 and TG0, and …

Restricted heterogeneity of the early antibody response to Aleutian disease virus in mink kits.

Mink kits born of Aleutian disease (AD) negative dams were infected neonatally with different isolates of Aleutian disease virus (ADV). They were then sacrificed at different days after infection (10-45 days) and their sera were analysed by different techniques for concentrations of immunoglobulins and quality of the produced antibodies to ADV. The infected mink kits produced significantly higher concentrations of IgM and IgG immunoglobulins than non-infected-age matched control mink (p less than 0.001). Eight percent of the sera from the infected mink kits exhibited a restricted heterogeneous

Anti-deoxyribonucleic acid antibody associated with persistent infection of mink with Aleutian disease virus.

Anti-deoxyribonucleic acid (DNA) antibody was quantitated in sera from mink infected with Aleutian disease virus (ADV). During the course of the disease after experimental infection, the amount of anti-DNA antibody increased 60% initially, but then decreased to an intermediate level when measured 2.5 months later. The percentage of serum immunoglobulin, however, steadily increased over 3.5-fold during this period, resulting in the characteristic gammopathy. Correlation between the level of anti-DNA antibody and hypergammaglobulinemia was demonstrated with sera from chronically infected mink. C

Aleutian mink disease virus (AMDV) enhances viral replication by downregulating JNK protein to promote apoptosis.

Aleutian mink disease virus (AMDV) is a pathogen that causes severe illness in minks, leading to substantial economic losses in the mink farming industry. This study investigated the influence of c-Jun N-terminal kinase (JNK), a host cell protein, on the pathological effects of AMDV in infected cells. AMDV infection caused extrinsic apoptotic pathway with Bax translocation, Cytochrome C release, and caspase-9/3 activation. Proteomic and Western blot analyses confirmed that JNK protein was downregulated after infection. JNK mRNA expression was unaffected at 12-48 h post-infection, whereas cyclo…

Genetic polymorphism of IgG in the mink. 7. Expression of the C gamma-allotypes in domestic mink infected with the Aleutian disease virus.

The Aleutian disease (AD), i.e., viral plasmocytosis in mink can be used as a model of the natural development of immune complex pathology in man. The immunogenetic aspect of AD was studied with the help of genetic markers of the constant region of the mink immunoglobulin gamma-heavy chain (the C gamma allotypes H2, H3, H4, H6, H7 and H8). The frequencies of 2 of the 6 allotypes, H3 and H4, were significantly higher in the AD-infected than in normal minks from the same population. This supports and extends the data in the literature indicating that the frequencies of certain human Gm allotypes

Polymorphism of the RNF165 Gene in American Mink (Neogale vison) as a Potential Factor Responsible for Resistance to Infection with the Aleutian Mink Disease Virus.

Background: Advances in American mink (Neogale vison) genomics have identified candidate genes linked to disease resistance, including RNF165, which is involved in immune response and virus-host interactions. Objectives: This study aimed to characterize genetic variation within the RNF165 (Ring Finger Protein 165) gene in farmed and wild mink to identify polymorphisms potentially associated with resistance to Aleutian mink disease virus (AMDV). Methods: Two groups of mink were analyzed: farmed animals from Latvia (n = 50) and wild animals (n = 20) from Poland. The presence of viral DNA was ver…

A Novel Amdoparvovirus of Badgers and Foxes and the Perpetuation of Aleutian Mink Disease Virus 3 in the Wildlife of Denmark.

Amdoparvoviruses, encompassing the well-characterized Aleutian mink disease viruses (AMDV) as well as less investigated viruses infecting both captive and wild animals, are important carnivoran viruses that are significant pathogens in the mink farming industry. We investigated the molecular epidemiology of amdoparvoviruses among Danish wildlife. Spleen samples from 118 animals of seven carnivoran species were screened with a pan-amdoparvovirus PCR, and the identified viruses were molecularly characterized. In one of five European badgers (Meles meles), we identified an AMDV-3 strain whose anc…

Studies on the sequential development of acute interstitial pneumonia caused by Aleutian disease virus in mink kits.

We studied different parameters during the development of acute interstitial pneumonia in mink kits caused by neonatal infection with Aleutian disease virus (ADV). When histological lesions, presence of intranuclear inclusion bodies, and intranuclearly localized ADV antigen were correlated with levels of single-stranded virion and duplex replicative forms of ADV DNA in the different tissues, it was concluded that the lung, probably alveolar type II cells, is the major primary target for viral replication and cytopathology. The presence of the duplex dimeric replicative-form DNA, a strong marke

Development of an antigen-capture enzyme-linked immunosorbent assay for diagnosis of Aleutian mink disease virus.

Aleutian mink disease (AMD), caused by Aleutian mink disease virus (AMDV), is a very important infectious disease of mink. Currently, elimination of antibody- or antigen-positive animals is the most successful strategy for eradicating AMD, but the claw-cutting method of blood sampling is difficult to perform and painful for the animal. In this study, we aimed to establish an antigen capture enzyme-linked immunosorbent assay (AC-ELISA) method for the efficient detection of AMDV antigens using fecal samples. A purified mouse monoclonal antibody (mAb) was used as the capture antibody, and a rabbi…

Analysis of the quantity of antiviral antibodies from mink infected with different Aleutian disease virus strains.

Mink persistently infected with Aleutian disease virus (ADV) develop hypergammaglobulinaemia and immune complex disease. Radiolabelled antibodies from mink infected with ADV-G, DK, Pullman , and Utah I strains of ADV were reacted against all four ADV strains in radioimmunoassay (RIA). The amount of anti-ADV antibody in two equally hypergammaglobulinaemic serum pools varied from 13% (anti- Pullman ) to 57% (anti-Utah I). Serum pools from two other sources (anti-DK and anti-ADV-G), although less hypergammaglobulinaemic , had 5% and 13%, respectively, indicating that 43-95% of the Ig in the sera

Molecular epidemiology of Aleutian mink disease virus causing outbreaks in mink farms from Southwestern Europe: a retrospective study from 2012 to 2019.

BACKGROUND: Aleutian mink disease virus (AMDV) causes major economic losses in fur-bearing animal production. The control of most AMDV outbreaks is complex due to the difficulties of establishing the source of infection based only on the available on-farm epidemiological data. In this sense, phylogenetic analysis of the strains present in a farm may help elucidate the origin of the infection and improve the control and biosecurity measures. OBJECTIVES: This study had the following aims: characterize the AMDV strains from most outbreaks produced at Spanish farms between 2012-2019 at the molecu…

Pathogenesis of Aleutian Mink Disease Virus Infection-Comparison of Natural Transmission with Experimental Aerosol and Intraperitoneal Inoculation.

Though intraperitoneal (IP) inoculation is not the natural pathway of Aleutian Mink Disease Virus (AMDV) infection in mink, it is frequently used experimentally. To investigate AMDV pathogenesis, we compared the effects of IP, aerosol (AE), and natural infection in mink. Forty-six sapphire mink were divided into groups: negative controls, IP and AE AMDV-inoculated mink, and sentinels exposed to IP-inoculated mink for two-week periods. Mink in the control, IP, and AE groups were euthanized 2, 5, or 10 weeks post-inoculation. The mink were tested for AMDV antibodies and by PCR on serum samples t…

Identification of aleutian mink disease parvovirus capsid sequences mediating antibody-dependent enhancement of infection, virus neutralization, and immune complex formation.

Aleutian mink disease parvovirus (ADV) causes a persistent infection associated with circulating immune complexes, immune complex disease, hypergammaglobulinemia, and high levels of antiviral antibody. Although antibody can neutralize ADV infectivity in Crandell feline kidney cells in vitro, virus is not cleared in vivo, and capsid-based vaccines have proven uniformly ineffective. Antiviral antibody also enables ADV to infect macrophages, the target cells for persistent infection, by Fc-receptor-mediated antibody-dependent enhancement (ADE). The antibodies involved in these unique aspects of A
