New Ru(II) complexes encompassing α-diimine ligands were synthesized by reacting ruthenium precursors with α-diimine hydrazones. The new ligands and Ru(II) complexes were analyzed by analytical and various spectroscopic methods. The molecular structures of L1 and complexes 1, 3, and 4 were determined by single-crystal XRD studies. The results reveal a distorted octahedral geometry around the Ru(II) ion for all complexes. Moreover, the new ruthenium complexes show efficient catalytic activity toward the C-N and C-C coupling reaction involving alcohols. Particularly, complex 3 demonstrates effec

A new class of potent electrophilic "R(+)" alkylating agents has been developed using weakly nucleophilic carborane anions as leaving groups. These reagents, R(CHB(11)Me(5)X(6)) (R = Me, Et, and i-Pr; X = Cl, Br), are prepared via metathesis reactions with conventional alkylating agents such as alkyl triflates, using the high oxophilicity of silylium ion-like species, Et(3)Si(carborane), as the driving force to obtain increased alkyl electrophilicity. The crystal structure of the isopropyl reagent, i-Pr(CHB(11)Me(5)Br(6)), has been determined, revealing covalence in the alkyl-carborane bonding

The palladium-catalyzed allylic alkylation of non-stabilized ketone enolates was thought for a long time to be not as efficient as the analogous reactions of stabilized enolates, e. g. of malonates and β-ketoesters. The field has experienced a rapid development during the last two decades, with a range of new, highly efficient protocols evolved. In this review, the early developments as well as current methods and applications of palladium-catalyzed ketone enolate allylations will be discussed.

The diversity of mechanisms for enantiodiscrimination and of bond types that can be formed make Pd-catalyzed asymmetric allylic alkylation a powerful key step for simplification of synthetic strategy to complex molecular targets. Using a wide range of different classes of compounds including alkaloids, polyhydrofurans, nucleosides and carbanucleosides, cyclohexitols and cyclopentitols, chromanes, cyclopentanoids, amino acids, barbiturates, etc., novel synthetic strategies emerge that provide short efficient asymmetric syntheses.

The first metal-free method for alkylation of quinoxalinones using cheap and stable aryl alkyl ketones as nucleophilic alkylation reagents is reported. This strategy greatly broadens the application channels of aryl alkyl ketones through carbon-carbon bond activation. In addition, the protocol has the advantages of simple operation, broad substrate scope, and good functional group tolerance.

Herein, we have developed a new and efficient catalytic system for the C-H/N-H cross-coupling N-alkylation of azoles under metal-free, base-free, and room temperature conditions. In this transformation, DDQ serves as a catalyst to promote the coupling process via single electron transfer and hydrogen atom transfer, and BPO acts as an oxidant to regenerate DDQ. This method offers an economical and practical protocol with a broad substrate scope.

Hydrogen borrowing describes a one-pot multistep sequence in which an alcohol is used as an alkylating agent. In comparison to a traditional alkylation reaction using alkyl halides, this is an attractive strategy: alcohol substrates are commercially abundant and stable, the process uses catalytic amounts of metal and base, and water is generated as the sole byproduct. Since seminal reports in the early 2000s, the field has been investigated extensively, but most hydrogen borrowing reactions operate under a high-temperature regime (76-200 °C), particularly those involving carbon-carbon bond for

Functional molecules derived from stereogenic phosphorus centers have important applications in the discovery of drugs and agrochemicals. They are also widely utilized as chiral ligands or organocatalysts for diverse asymmetric transformations. However, access to P-stereogenic motifs has always been regarded as a highly challenging yet desirable goal in organic synthesis. The development of general and practical methods for the stereoselective construction of synthetically versatile P(III)-stereogenic phosphines is particularly appealing but remains elusive. Herein, we describe a nickel-cataly

Ketones are among the most useful functional groups in organic synthesis, and they are commonly encountered in a broad range of compounds with various applications. Herein, we describe the mesoionic carbene-catalyzed coupling reaction of aldehydes with non-activated secondary and even primary alkyl halides. This metal-free method utilizes deprotonated Breslow intermediates derived from mesoionic carbenes (MICs), which act as super electron donors and induce the single-electron reduction of alkyl halides. This mild coupling reaction has a broad substrate scope and tolerates many functional grou

Carbon-oxygen bonds are commonplace in organic molecules, including chiral bioactive compounds; therefore, the development of methods for their construction with simultaneous control of stereoselectivity is an important objective in synthesis. The Williamson ether synthesis, first reported in 18501, is the most widely used approach to the alkylation of an oxygen nucleophile, but it has significant limitations (scope and stereochemistry) owing to its reaction mechanism (SN2 pathway). Transition-metal catalysis of the coupling of an oxygen nucleophile with an alkyl electrophile has the potential

The development of efficient and selective C-N bond-forming reactions from abundant feedstock chemicals remains a central theme in organic chemistry owing to the key roles of amines in synthesis, drug discovery, and materials science. Herein, we present a dual catalytic system for the N-alkylation of diverse aromatic carbocyclic and heterocyclic amines directly with carboxylic acids, by-passing their preactivation as redox-active esters. The reaction, which is enabled by visible-light-driven, acridine-catalyzed decarboxylation, provides access to N-alkylated secondary and tertiary anilines and

After anaerobic reductive activation by either NADPH cytochrome P-450 reductase (EC 1.6.2.4) or xanthine oxidase (EC 1.2.3.2), mitomycin C readily alkylated DNA. When the mitomycin C-alkylated DNA is digested by DNase, snake venom phosphodiasterase, and alkaline phosphatase, only partial release of the monofunctionally linked mitomycin C nucleotide adduct occurs. Cross-linked adducts are not released into dinucleotides but resist nuclease digestion and remain in oligonucleotides and insoluble precipitates. Kinetic analyses show that the nuclease-resistant fraction which is indicative of DNA cr

The utilization of economical and environmentally benign transition metals in crucial catalytic processes is pivotal for sustainable advancement in synthetic organic chemistry. Iron, as the most abundant transition metal in the Earth's crust, has gained significant attention for this purpose. A combination of FeCl2 (5 mol%) in the presence of phenanthroline (10 mol%) and NaOtBu (1.5 equivalent) proved effective for the C(α)-alkylation of oxindole, employing challenging secondary alcohol as a non-hazardous alkylating agent. The C(α)-alkylation of oxindole was optimized in green solvent or under

Alkyl glucosides are surfactants synthesized through the condensation of long-chain fatty alcohols and glucose, extracted from vegetal, renewable sources. Although available for more than 4 decades, they have been rediscovered in recent years because of their eco-friendly character. They are used in various leave-on and rinse-off cosmetics and are considered of low irritancy and allergenicity. However, since the early 2000s, cases of allergic contact dermatitis to this family of molecules have been repeatedly reported. Decyl glucoside was found to be a "hidden" allergen in the sunscreen ingred

Cysteine is one of the most reactive amino acids. This is due to the electronegativity of sulphur atom in the side chain of thiolate group. It results in cysteine being present in several distinct redox forms inside the cell. Amongst these, reversible oxidations, S-nitrosylation and S-sulfenylation are crucial mediators of intracellular redox signalling, with known associations to health and disease. Study of their functionalities has intensified thanks to the development of various analytical strategies, with particular contribution from differential alkylation-based proteomics methods. Prese

A catalytic, enantioselective method for direct α-alkylation of ketones with unactivated alkyl halides is realized by employing an α-enolizable ketone in a nickel-catalyzed C(sp3)-C(sp3) cross-coupling reaction. The key to the success is attributed to a unique bimetallic ligand. A variety of acyclic ketones and unactivated alkyl iodides can serve as suitable substrates under mild conditions to generate chiral ketones with α-quaternary carbon stereocenters in high yields with good enantioselectivities. A range of transformations based on the ketone moiety are also demonstrated to show the poten

Defluorinative alkylation of alkenes is a useful reaction for converting trifluoromethyl compounds into diverse materials. Photoredox chemistry has been established as an efficient protocol to achieve difluoroalkylation reactions of alkenes using trifluoromethyl arenes as reagents. In comparison, electrochemistry has not achieved comparable progress. In this work, we reported a zinc-cation-promoted electrochemical monodefluorinative alkylation of alkenes using trifluoromethyl arenes as reagents. The reaction tolerated a series of functional groups, including pyridine, silyl, aniline, and cyclo

Control of the regioselectivity of α-alkylation of carbonyl compounds is a longstanding topic of research in organic chemistry. By using stoichiometric bulky strong bases and carefully adjusting the reaction conditions, selective alkylation of unsymmetrical ketones at less-hindered α-sites has been achieved. In contrast, selective alkylation of such ketones at more-hindered α-sites remains a persistent challenge. Here we report a nickel-catalysed alkylation of unsymmetrical ketones at the more-hindered α-sites with allylic alcohols. Our results indicate that the space-constrained nickel cataly
