Allantoin uptake in both growing and resting cultures of Saccharomyces cerevisiae occurs by a low-Km (ca. 15 micrometer) transport system that uses energy that is likely generated in the cytoplasm. This conclusion was based on the observation that transport did not occur in the absence of glucose or the presence of dinitrophenol, carbonyl cyanide-m-chloro-phenyl hydrazine, fluoride, or arsenate ions. Normal uptake was observed, however, in the presence of cyanide. The rate of accumulation was maximal at pH 5.2. In contrast to the urea transport system, allantoin uptake appeared to be unidirect

Rabbit renal cortical slices were incubated at 25 degrees C, pH 7.4, for 2 hr under 100% O2 in a phosphate-buffered medium. [14C]allantoin was accumulated to a tissue water/medium ratio of 1.38 +/- 0.03 (mean +/- S.E., n = 22). Under 100% N2, the tissue water/medium ratio fell significantly to 0.90 +/- 0.02 (n = 14, P less than .001). Probenecid (an anion) inhibited the uptake of allantoin at doses which had no effect on the water content of the slices, i.e., did not appear to have a toxic effect on the tissue. Its effect was specific. In contrast, salicylate (another anion) and quinine (a cat

We previously described the only satisfactory procedure yet achieved for separating uric acid and allantoin from rat liver. The procedure was based on trichloroacetic acid (TCA) extraction, acid hydrolysis, treatment with Hg-acetate, and cation- and anion-exchange chromatography. After separation, allantoin was quantified by a colorimetric method, and uric acid enzymatically using uricase. Since this procedure is too time-consuming, we propose an improved version which avoids the need for anion-exchange chromatography and the complex assay of catabolic compounds. The new method consists of a v

Allantoin increases in different stress conditions and environment such as physical activity, amniotic fluids and oxidative stress. So, we inspired to explore the role of allantoin as a metabolic by-product in health improvement and protection using irradiation as simulator for oxidative stress. Allantoin was injected i.p. (100 mg/kg) in senile male rats in irradiated and non-irradiated groups in comparison to sham operated group. The studied parameters were superoxide dismutase, Glutathione reductase, Glutathione, total antioxidant capacity, collagenase, urea, creatine kinase, alanine transam

Alzheimer's disease (AD) is characterised by hallmark pathology of amyloid beta (Aβ) plaques and hyperphosphorylated tau neurofibrillary tangles in the brain. Ergothioneine (ET) is a potent antioxidant, and anti-inflammatory compound that has shown potential as a therapeutic for neurodegenerative disease*. Cross-sectional analyses of cognitively normal individuals aged 65-90yrs from the Kerr Anglican Retirement Village Initiative in Ageing Health (KARVIAH) cohort were stratified by amyloid status (Aβ+, SUVR >1.35). Plasma ET, its metabolites and urinary allantoin, were quantified by liquid chr

The oxidative catabolism of uric acid produces 5-hydroxyisourate (HIU), which is further degraded to (S)-allantoin by two enzymes, HIU hydrolase and 2-oxo-4-hydroxy-4-carboxy-5-ureidoimidazoline decarboxylase. The intermediates of the latter two reactions, HIU and 2-oxo-4-hydroxy-4-carboxy-5-ureidoimidazoline, are unstable in solution and decay nonstereospecifically to allantoin. In addition, nonenzymatic racemization of allantoin has been shown to occur at physiological pH. Since the further breakdown of allantoin is catalyzed by allantoinase, an enzyme that is specific for (S)-allantoin, an

This study aims to investigate the acute effect of local allantoin treatment on experimental distraction osteogenesis. A total of 14 adult New Zealand rabbits underwent mandibular distraction under general anesthesia. The control group received saline washes, while the experimental group received local allantoin (1 mg/kg). Computed tomography was used to measure bone mineral density (Hounsfield Units). Stereological analysis of decalcified tissues was performed using the Cavalieri Method, which assessed the volumes of new bone, connective tissue, and capillaries (mm3). Immunohistochemistry emp

PURPOSE: Gastritis is found to be one of the most common gastrointestinal diseases worldwide. However, current therapeutic agents cause side effects, interaction, and recurrence. Allantoin has anti-inflammatory and wound healing properties. In this study, the therapeutic effect of allantoin has been assessed on the histopathological indices and gastric mucosal barrier of male rats.
METHODS: Male rats were equally divided into control, ethanol-induced gastritis, and allantoin groups. The therapeutic groups consisted of gastritis plus 12.5 mg/kg allantoin, gastritis plus 25 mg/kg allantoin, and

Background Oxidative stress (OS) represents the primary mediator of chronic heart failure (CHF) development and progression. It is well established that homocysteine is able to generate reactive oxygen species. Small amounts of allantoin in human serum result from free radical action on urate and may provide a stable marker for in vivo free radical activity. To investigate whether some easily measurable indexes such as antioxidants (uric acid, glutathione) and related molecules (allantoin, homocysteine and cysteine) can serve as OS biomarkers. Methods We investigated 75 stable CHF patients. Am

Glycolic acid and allantoin are known to be responsible, at least in part, for the main activities of snail slime, such as moisturizing, skin regeneration, antioxidant, soothing and anti-inflammatory. Accordingly, a hydrophilic interaction liquid chromatography (HILIC) method for the analysis of allantoin and glycolic acid in snail slime samples was developed by a conventional UV-Vis detector and the diol-type Luna® HILIC column. An optimized eluent made up of acetonitrile/water/methanol/formic acid (90:5:5:0.1 v/v/v/v) allowed the separation of the two investigated compounds from each other a

Cordycepin is a high-value bioactive compound with significant therapeutic potential. In this study, we report a transposon-based genome engineering strategy for establishing an allantoin-inducible cordycepin production platform in Saccharomyces cerevisiae. We first optimized the allantoin-inducible system by refining the culture medium's components to improve cell growth and achieved an approximately 50-fold on/off ratio after reaching a plateau, thereby enhancing the robustness of engineered yeast strains. Next, a multiselection marker transposon system was implemented to achieve combinatori

Xanthine dehydrogenase/xanthine oxidase (XDH/XO) catalyses the hydroxylation of hypoxanthine to xanthine and finally to uric acid in purine degradation. These reactions generate H(2)O(2) yielding allantoin from uric acid when reactive oxygen species accumulates. The presence of XO in the human epidermis has not been shown so far. As patients with vitiligo accumulate H(2)O(2) up to mm levels in their epidermis, it was tempting to examine whether this enzyme and consequently allantoin contribute to the oxidative stress theory in this disease. To address this question, reverse transcription-polym

Gastric ulcer affects people worldwide, and its inefficacy and recurrence have fueled the search for new therapeutic strategies. Despite the well-known use of allantoin in medicines and cosmetic products, its effect has not yet been studied with regard to gastric ulcer. Hence, the aim of the present study was to explore the pharmaco-mechanistic efficacy of allantoin against commonly harmful agents that cause injuries to the stomach. Ethanol, indomethacin, and stress-induced gastric ulcer models were adopted, in addition to pylorus ligature, a quantification of vascular permeability, glutathion

Ureides are nitrogen-rich compounds, derived from purine catabolism. A dual role for ureides, and for allantoin in particular, in both nitrogen recycling and the abiotic stress response has been recently identified. Previous work on the effect of allantoin on cadmium (Cd)-exposed Arabidopsis revealed that high concentration of allantoin in allantoinase-negative mutant (aln-3) leaves alleviates Cd toxicity via inducing antioxidant mechanisms in these plants. In the present study, we evaluate whether allantoin has a similar protective role in roots. Both wild type and aln-3 roots contain higher

Plants apply various molecular, physiological and morphological strategies in response to undesirable environmental conditions. One of the possible responses which may contribute to surviving stressful conditions is the accumulation of ureides. Ureides are recognized as important nitrogen-rich compounds involved in recycling nitrogen in plants to support growth and reproduction. Amongst them, allantoin not only serves as a transportable nitrogen-rich compound, but has also been suggested to protect plants from abiotic stresses via minimizing oxidative damage. This work focuses on the effect of
