Networking in microbes: conjugative elements and plasmids in the genus Alteromonas.

BACKGROUND: To develop evolutionary models for the free living bacterium Alteromonas the genome sequences of isolates of the genus have been extensively analyzed. However, the main genetic exchange drivers in these microbes, conjugative elements (CEs), have not been considered in detail thus far. In this work, CEs have been searched in several complete Alteromonas genomes and their sequence studied to understand their role in the evolution of this genus. Six genomes are reported here for the first time. RESULTS: We have found nine different plasmids of sizes ranging from 85 to 600 Kb, most of

Coevolution of marine phytoplankton and Alteromonas bacteria in response to pCO2 and coculture.

As a result of human activity, Earth's atmosphere and climate are changing at an unprecedented pace. Models based on short-term experiments predict major changes will occur in marine phytoplankton communities in the future ocean, but rarely consider how evolution or interactions with other microbes may influence these changes. Here, we experimentally evolved several phytoplankton in coculture with a heterotrophic bacterium, Alteromonas sp. EZ55, under either present-day or predicted future pCO2 conditions. Growth rates of phytoplankton generally increased over time under both conditions, but o

Multifunctionally diverse alkaline phosphatases of Alteromonas drive the phosphorus cycle in the ocean.

Phosphorus is a critically limiting nutrient in marine ecosystems, with alkaline phosphatases (APases) playing a vital role in liberating phosphate from organic compounds. However, the dominant taxa and APase families driving the marine phosphorus cycle, particularly in the deep ocean, remain poorly understood. Equally enigmatic remains the (multi)functional diversity and mechanisms of action of different APases. To address these gaps, this study combines global multi-omic analyses, biochemical studies of purified recombinant proteins, and laboratory experiments with proteomics and enzymatic r

Collaborative metabolic curation of an emerging model marine bacterium, Alteromonas macleodii ATCC 27126.

Inferring the metabolic capabilities of an organism from its genome is a challenging process, relying on computationally-derived or manually curated metabolic networks. Manual curation can correct mistakes in the draft network and add missing reactions based on the literature, but requires significant expertise and is often the bottleneck for high-quality metabolic reconstructions. Here, we present a synopsis of a community curation workshop for the model marine bacterium Alteromonas macleodii ATCC 27126 and its genome database in BioCyc, focusing on pathways for utilizing organic carbon and n

Genomic and transcriptomic characterization of methylmercury detoxification in a deep ocean Alteromonas mediterranea ISS312.

Methylmercury (MeHg) is one of the most worrisome pollutants in marine systems. MeHg detoxification is mediated by merB and merA genes, responsible for the demethylation of MeHg and the reduction of inorganic mercury, respectively. Little is known about the biological capacity to detoxify this compound in marine environments, and even less the bacterial transcriptional changes during MeHg detoxification. This study provides the genomic and transcriptomic characterization of the deep ocean bacteria Alteromonas mediterranea ISS312 with capacity for MeHg degradation. Its genome sequence revealed

Vesicle-associated exudates from Alteromonas enhance growth and survival of Prochlorococcus in batch culture.

The cyanobacterium Prochlorococcus MIT9312 requires helping functions from co-existing organisms for survival under stressful conditions. Here, we show that the helper strain Alteromonas macleodii EZ55 also facilitates greater exploitation of medium resources by Prochlorococcus. The presence of Alteromonas allowed Prochlorococcus to grow to greater cell densities, delay growth cessation, and avoid cell death relative to axenic cultures, phenotypes potentially associated with Alteromonas-driven degradation of autotoxic substances made by Prochlorococcus. We found that heat-labile high-molecular

Characterization of a novel alginate lyase Alg0392 with organic solvent-tolerance from Alteromonas sp. A1-6.

Enzymatic depolymerization of seaweed polysaccharides aroused great interest in the production of functional oligosaccharides and fermentable sugars. Alginate lyase Alg0392, a potential novel member of the polysaccharide lyase PL17 family, was cloned from Alteromonas sp. A1-6. The enzymatic properties, kinetic parameters, and hydrolytic products of Alg0392 were systematically characterized. Especially, the recombinant enzyme Alg0392 showed excellent tolerance to organic reagents. When treated with 5 mmol/L of TritonX-100 or 20%(v/v) of methanol, its relative enzyme activity could be maintained

Degradation of organic mercury in high salt environments by a marine aerobic bacterium Alteromonas macleodii KD01.

Mercury (Hg), particularly organic mercury, poses a global concern due to its pronounced toxicity and bioaccumulation. Bioremediation of organic mercury in high-salt wastewater faces challenges due to the growth limitations imposed by elevated Cl- and Na+ concentrations on microorganisms. In this study, an isolated marine bacterium Alteromonas macleodii KD01 was demonstrated to degrade methylmercury (MeHg) efficiently in seawater and then was applied to degrade organic mercury (MeHg, ethylmercury, and thimerosal) in simulated high-salt wastewater. Results showed that A. macleodii KD01 can rapi

Characterization and Genome Sequence of Marine Alteromonas gracilis Phage PB15 Isolated from the Yellow Sea, China.

A novel marine Alteromonas gracilis siphovirus, phage PB15, was isolated from the surface water of the Yellow Sea in August 2015. It has a head diameter of 58 ± 5 nm head and a contractile tail approximately 105 ± 10 nm in length, and overall, the morphology suggests that PB15 belongs to the family Siphoviridae. PB15 phage is stable at over the temperature range 0-60 °C. The best MOI of these phage was 0.1, and infectivity decreased above 60 °C. The results suggest that phage is stable at pH value ranging between 3.0 and 11.0. Chloroform test shows that PB15 is not a lipid-containing phage. A

A Novel Metallo-β-Lactamase AMM-1 From Alteromonas mangrovi Reveals a Cryptic Environmental Reservoir of Carbapenem Resistance.

Carbapenem resistance driven by metallo-β-lactamases (MBLs) poses a formidable global challenge as these enzymes can degrade a wide range of β-lactam antibiotics, including last-line carbapenems. Despite extensive documentation of MBL-producing pathogens, their evolutionary origins and ecological reservoirs are still poorly understood. Here, we report the discovery and in-depth characterisation of AMM-1, a previously unrecognised B1.2 MBL identified within a metagenome-assembled genome of Alteromonas mangrovi obtained from the Yangtze River Estuary. Role of the N-terminal polycystic kidney disease domain in chitin degradation by chitinase A from a marine bacterium, Alteromonas sp. strain O-7.

AIMS: The aim of study was to clarify whether the polycystic kidney disease (PKD) domain of chitinase A (ChiA) participates in the hydrolysis of powdered chitin. METHODS AND RESULTS: Site-directed mutagenesis of the conserved aromatic residues of PKD domain was performed by PCR. The aromatic residues, W30, Y48, W64 and W67, were replaced by alanine, and single- and double-mutant chitinases were produced in Escherichia coli XL10 and purified with HisTrap column. Single mutations were not quite effective on the hydrolysing activities against chitinous substrates when compared with wild-type Chi

Prevalence of Heterotrophic Methylmercury Detoxifying Bacteria across Oceanic Regions.

Microbial reduction of inorganic divalent mercury (Hg2+) and methylmercury (MeHg) demethylation is performed by the mer operon, specifically by merA and merB genes, respectively, but little is known about the mercury tolerance capacity of marine microorganisms and its prevalence in the ocean. Here, combining culture-dependent analyses with metagenomic and metatranscriptomic data, we show that marine bacteria that encode mer genes are widespread and active in the global ocean. We explored the distribution of these genes in 290 marine heterotrophic bacteria (Alteromonas and Marinobacter spp.) is

[Gram-negative eubacteria isolated from the water, molluscs and algae of the Black Sea].

Microorganisms strains (384) have been isolated from water, algae and molluscs collected in the Karadag reserve and the village of Katsiveli (Black Sea shore of the Crimea); 81 of the above strains are gram-negative oxidase-positive rods which require sodium ions for their growth. A group of nonfermenting mobile bacteria deprived of arginine dihydrolase have been distinguished among the latter; as to their physiological and biochemical properties and sensitivity to some antibiotics these bacteria were referred to Alteromonas, Marinomonas, Pseudoalteromonas genera.

Shewanella infection in humans: Epidemiology, clinical features and pathogenicity.

The genus Shewanella consists of Gram-negative proteobacteria that are ubiquitously distributed in environment. As the members of this genus have rapidly increased within the past decade, several species have become emerging pathogens worldwide, attracting the attention of the medical community. These species are also associated with severe community- and hospital-acquired infections. Patients infected with Shewanella spp. had experiences of occupational or recreational exposure; meanwhile, the process of infection is complex and the pathogenicity is influenced by a variety of factors. Here, a

Hydrocarbon-degrading microbial populations in permanently cold deep-sea sediments in the NW Atlantic.

Permanently cold deep-sea sediments (2500-3500 m water depth) with and without indications of thermogenic hydrocarbon seepage were exposed to naphtha to examine the presence and potential of cold-adapted aerobic hydrocarbon-degrading microbial populations. Monitoring these microcosms for volatile hydrocarbons by GC-MS revealed sediments without in situ hydrocarbons responded more rapidly to naphtha amendment than hydrocarbon seep sediments overall, but seep sediments removed aromatic hydrocarbons benzene, toluene, ethylbenzene and xylene (BTEX) more readily. Naphtha-driven aerobic respiration

Hepatic cholangiocarcinoma and testicular mesothelioma in a wild-caught blue shark, Prionace glauca (L.).

An adult male blue shark, Prionace glauca (L.), caught in July 2000 by a recreational fisherman off Long Island, New York, USA, had a retained fishing hook from a previous capture. The hook penetrated the gastric wall and lacerated the right liver lobe. Macroscopic lesions consisted of transmural gastritis and peritonitis. Alteromonas sp. and Vibrio alginolyticus were isolated from the peritoneal fluid. In addition, a well delineated, sessile mass was found on the otherwise normal serosa of the right testis. Histopathological findings included mesothelial hyperplasia and hypertrophy involving

Metabolomic Comparison and Assessment of Co-cultivation and a Heat-Killed Inducer Strategy in Activation of Cryptic Biosynthetic Pathways.

Co-cultivation has been used as a promising tool to turn on or up-regulate cryptic biosynthetic pathways for microbial natural product discovery. Recently, a modified culturing strategy similar to co-cultivation was investigated, where heat-killed inducer cultures were supplemented to the culture medium of producer fermentations to induce cryptic pathways. In the present study, the repeatability and effectiveness of both methods in turning on cryptic biosynthetic pathways were unbiasedly assessed using UHPLC-HRESIMS-based metabolomics analysis. Both induction methods had good repeatability, an

Fatty acid composition of bacteria associated with the toxic dinoflagellate Ostreopsis lenticularis and with Caribbean Palythoa species.

The fatty acid composition of a Pseudomonas sp. (Alteromonas) and its host, the dinoflagellate Ostreopsis lenticularis, vectors in ciguatera fish poisoning, has been studied. The major fatty acids in O. lenticularis were 16:0, 20:5n-3, and 22:6n-3, but 18:2n-6, 18:3n-3, and 18:n-3 were also identified. In contrast to other dinoflagellates, 1 8:5n-3 was not detected in O. lenticularis. Even-chain fatty acids such as 9-16:1, 11-18:1, and 13-20:1 predominated in the Pseudomonas sp. from O. lenticularis, but 1 6-20% of (E)-11-methyl-12-octadecenoic acid was also identified. We performed hybridizations between labeled rRNAs from seven representative members of the family Pasteurellaceae and from three other taxa on the one hand and DNAs from 53 strains known or presumed to belong to the Pasteurellaceae on the other hand. The members of the Pasteurellaceae are most closely related to members of the Enterobacteriaceae, the Vibrionaceae, the Aeromonadaceae, and the genus Alteromonas. The family Pasteurellaceae is very heterogeneous. There are at least seven rRNA branches. Several organisms with the same genus name are dispersed over the entire dendrogram. The "Histop
