Amanita phalloides is the most dangerous, poisonous mushroom species in our climatic conditions. It is the cause of 90-95% of all deaths due to mushroom poisoning, a-Amanitin, a polymerase RNA II inhibitor, is mainly responsible for the Amanita phalloides toxic property. Inhibition of polymerase RNA II functioning in a transcription process is connected with inhibition biosynthesis of structural and enzymatic proteins in cells. A lethal dose of a-amanitin is 0.1 mg/kg b.w. for humans. One of the medical problems in Amanita phalloides poisonings is a relatively prolonged latency period (8-24 h)

A pregnant woman at the 28th gestational week and three other members of the family had mushrooms for dinner. One of these mushrooms was an Amanita mushroom. The morning after, three of them suffered from nausea, vomiting and diarrhoea. The pregnant woman was admitted to hospital, and therapy with Penicillin and Silibinin was started immediately. Hepatic and renal blood parameters were observed repeatedly for early detecting of organ failure. Fetal lung maturation was forced by administration of betamethasone. After five days at the intensive care unit mother and unborn child were dismissed at

This study investigates potential inhibitors of nitric oxide (NO) production, α-glucosidase, and protein tyrosine phosphatase 1B (PTP1B) from Amanita abrupta and Amanita pantherina. Sixteen compounds (1-16) were isolated by chromatographic techniques and high-performance liquid chromatography (HPLC), and their structures were elucidated by nuclear magnetic resonance (NMR) and literature comparison. Biological activities were evaluated via in vitro assays: the Griess assay for NO inhibition, the p-nitrophenol assay for α-glucosidase, and enzymatic assays for PTP1B. Molecular docking was employe

BACKGROUND: Renal failure as a consequence of eating mushrooms has been reported repeatedly after ingestion of webcaps of the Cortinarius orellanus group. But mushrooms of the genus Amanita can also cause renal failure: Amanita smithiana (North America) and Amanita proxima (Mediterranean area). Here, we discuss poisonings caused by other white amanitas. A German and--independently--two Portuguese patients reported the ingestion of completely white mushrooms with ring. Similar to intoxications with A. smithiana or A. proxima, the clinical picture was characterized by nausea and vomiting 10-12 h

From the experiments detailed in this paper it may be concluded that Amanita phalloides contains besides phallin, or the haemolytic principle of Kobert, another body of toxic nature. Phallin is thermolabile, and is destroyed by the action of pepsin and pancreatin. The other toxic body is thermostable and is resistant to pepsin and pancreatin. The two substances, moreover, possess different toxophoric and haptophoric groups, since an antiserum produced by the immunization of animals to the thermostable body has no neutralizing effects upon phallin. The thermolable body, phallin, produces the su

Amanita phalloides is responsible for about 90 per cent of all fatal cases of mushroom intoxication. The amatoxins, the main toxic component of these fungi, are responsible for gastro-intestinal symptoms as well as hepatic and renal failure. Three brothers with Amanita phalloides poisoning were admitted with gastro-intestinal symptoms beginning 12 h after ingestion. Jaundice, hepatomegaly and neurological symptoms were not present, but liver enzymes were moderately increased. Alfa-amanitin was detected in sera of all patients. All patients underwent charcoal hemoperfusion and two of them had a

A case of a 72-year-old female who ingested panther cap (Amanita pantherina) was presented. During the ingestion of mushrooms she developed short-time diarrhea and severe transient neurological disorders; short-term hallucination followed by deep coma, skeletal muscle flaccidity with hyporeflexia. Also significant facial asymmetry was noted. A head CT showed a small ischaemic focus in capsula interna. After a neurological consult it was concluded that this small malformation was not the cause of severe patient's condition. The sporological test revealed numerous spores of panther cap in patien

Amanita muscaria has a bright red or orange cap covered with small white plaques. It contains the isoxazole derivatives ibotenic acid, muscimol and muscazone and other toxins such as muscarine. The duration of clinical manifestations after A. muscaria ingestion does not usually exceed 24 hours; we report on a 5-day paranoid psychosis after A. muscaria ingestion. A 48-year-old man, with no previous medical history, gathered and ate mushrooms he presumed to be A. caesarea. Half an hour later he started to vomit and fell asleep. He was found comatose having a seizure-like episode. On admission fo

Mushroom poisoning from the genus Amanita is being reported with increasing frequency in the United States. Endocrine hormone abnormalities were investigated in four patients who ingested mushrooms of the Amanita genus ("death cap", "destroying angel"). Marked abnormalities were found in the hormones controlling glucose, calcium, and thyroid homeostasis. Insulin and C-peptide concentrations were elevated at admission, indicating that the hypoglycemia associated with Amanita poisoning may not be solely secondary to hepatic failure. Serum calcitonin concentrations were elevated in conjunction wi

Mushroom poisonings in Poland are quite common, especially in summer and autumn, but fly agaric (Amanita muscaria) and panther cap (Amanita pantherina) are rather rare cause of these intoxications. Fly agaric is a cause of deliberate poisoning, whereas panther cap poisoning also happens accidentally. The main toxins of these two mushrooms are ibotenic acid (pantherine, agarine), muscimol, muscazone and muscaridine. The other bioactive substances are stizolobic and stizolobinic acids and aminodicarboxyethylthiopropanoic acids. All these compounds are responsible for diverse picture of intoxicat

This paper reported a case of poisoning caused by ingestion of Amanita neoovoidea. The patient experienced nausea, vomiting, oliguria, acute renal function injury, and was discharged after symptomatic support treatment and blood purification treatment. Given the different toxicity of different mushrooms, species identification of poisonous mushrooms can help clinicians in diagnosis and treatment.

HISTORY AND ADMISSION FINDINGS: A 41-year-old patient was found in his flat in a state of coma. After emergency treatment his vital signs were stable and he was transferred to an acute hospital with possible cannabis intoxication. The patient, a hobby gardener, was previously well and had an adversion to the use of any chemical substances. The main symptom showed a cholinergic syndrome with deep coma. We assumed plant ingestion because of the clinical picture and history.
INVESTIGATIONS: The laboratory results were within normal limits apart from a slight rise of the serum creatinine kinase l

A study was performed on the hepatic reaction of mice to acute intoxication with virotoxins (alaviroidin, viroisin, deoxoviroisin, viroidin, deoxoviroidin) and phalloidin, cyclic peptides isolated from Amanita virosa mushrooms. Purified fractions were administered intraperitoneally at various dosages to determine the LD50 which ranged from 1.0 to 5.3 mg/kg, with viroidin, phalloidin, and viroisin being the most potent. The virotoxins and phalloidin induced hemorrhagic necrosis of the liver. The development of hepatic lesions was followed by enhanced serum alanine aminotransferase (ALT) activit

Five fatal cases of poisoning from ingestion of Amanita phalloides, a very common mushroom in central Italy, are reported. The fact that four of the cases occurred simultaneously enabled uniform collection of clinical, pathology and toxicology data, which is presented with particular emphasis on the histological aspects. The fifth case involved a six-year-old girl, and is discussed with reference to differential diagnosis with respect to Reye's syndrome, which was the initial diagnosis, demonstrated incorrect by the histology, pathology and toxicology findings. The typical liver and kidney alt

Orthotopic liver transplantation in patients with acute toxic hepatitis has not, to our knowledge, been reported. Our recent experience with orthotopic liver transplantation in a 3-year-old girl with acute hepatic failure secondary to Amanita poisoning is described. Aspects of Amanita toxin-induced pathophysiology pertinent to orthotopic liver transplantation are discussed. The results in this case provide important implications for patients with fulminant hepatic failure secondary to other hepatotoxic agents in whom liver transplantation is considered.
