The odontoclasts of Ambystoma mexicanum.

The resorption of teeth in Ambystoma mexicanum during postembryonal ontogenesis and induced metamorphosis occurs by means of light-microscopic detectable giant-cells. These have morphological and functional characters similar to those of odontoclasts of other vertebrates. The multinucleated odontoclasts resorb not only the pedicel (base), but the stalk of the tooth, too. When active, the cells form a ruffled border and a sealing zone. In this way they are able to demineralize the hard tissues of teeth (dentin and mineral of the pedicel) and to dissolve the extracellular matrix. Resorption of e

Eimeria in the Xochimilco axolotl Ambystoma mexicanum.

Coccidia are protozoal parasites that can cause coccidiosis, a disease linked to high mortality in young, stressed, and overcrowded animals. In the order Urodela, only 21 coccidia species have been registered, and none in the Critically Endangered Ambystoma mexicanum. The present study presents the detection and follow-up of Eimeria in A. mexicanum. The monitoring was conducted at CIBAC (Centro de Investigaciones Biológicas y Acuícolas de Cuemanco), analyzing fecal samples from 90 juveniles using sedimentation, Faust, and modified Zajíček McMaster techniques. An overall prevalence of 65.6% was

Management of Hydrocoelom in an Axolotl (Ambystoma mexicanum).

A 3-year-old axolotl (Ambystoma mexicanum) presented with anorexia, weakness and abdominal swelling due to hydrocoelom. According to the anamnesis, the symptoms had occurred 3 months prior. The radiographic and ultrasonographic examinations revealed that these symptoms resulted from the fluid accumulating in the coelomic cavity. Then, the habitat of the axolotl and the fluid in the coelomic cavity were inspected. Then, the patient's hydrocoelomic fluid was drained and medical treatment was performed. In addition, habitat arrangement and diet were changed. At the follow-up visit 1 week later, t

Resegmentation in the Mexican axolotl, Ambystoma mexicanum.

The segmental series of somites in the vertebrate embryo gives rise to the axial skeleton. In amniote models, single vertebrae are derived from the sclerotome of two adjacent somites. This process, known as resegmentation, is well-studied using the quail-chick chimeric system, but the presumed generality of resegmentation across vertebrates remains poorly evaluated. Resegmentation has been questioned in anamniotes, given that the sclerotome is much smaller and lacks obvious differentiation between cranial and caudal portions. Here, we provide the first experimental evidence that resegmentation

An update on the characterization of immunoglobulin loci in Ambystoma mexicanum.

BACKGROUND: We previously reported the genomic characterization of immunoglobulin loci in Ambystoma mexicanum of the laboratory d/d white strain, where the IGH locus gene orientation was incompatible with VDJ recombination, suggesting scaffold orientation errors. A novel 29.1 Gbp A. mexicanum genome derived from an F1 cross between A. mexicanum and A. tigrinum (UKY_AmexF1_1) has recently been released, containing only 220 unmapped scaffolds. Here, we present an updated description of the immunoglobulin loci based on this improved genome assembly. METHODS: Using our previously annotated AmbMex

Histological reinterpretation of paraphysis cerebri in Ambystoma mexicanum.

Intraventricular and extraventricular choroid plexuses are neuroepithelial folds which arise from the roof of the diencephalon. We describe the circumventricular structure of the diencephalon roof (paraphysis cerebri) during the various development stages of Ambystoma mexicanum. The parasagittal sections of the larvae epithalamus exhibit the presence, in addition to the epiphysis, of two dorsal primordia in nearby areas, which appear to be extraventricular saccular evaginations of different origin that give rise to two structures we define as the anterior extraventricular choroid plexus (AEP)

Natural triploidy in salamanders related to Ambystoma jeffersonianum.

Female salamanders with large erythrocytes and erythrocyte nuclei produced triploid larvae; females with small erythrocytes and erythrocyte nuclei produced diploid larvae. Larval chromosome counts, dimorphism in cell size, and sex inheritance indicate distinct, persisting populations of triploid females in parts of the range of the Ambystoma jeffersonianum complex.

FGF2 rescues denervation-induced muscle atrophy in Ambystoma mexicanum.

BACKGROUND: Denervation occurs as a consequence of disease or injury and is typically accompanied by skeletal muscle atrophy. Although the relationship between nerves and muscle atrophy has been studied, the direct molecular contributions of nerves remain unclear. METHODS: We used the axolotl (Ambystoma mexicanum), a vertebrate with robust regenerative capacity and optically accessible musculature, to investigate the role of nerve-derived signals in muscle maintenance. RESULTS: Forelimb denervation produced significant muscle atrophy in axolotls. Quantitative imaging showed that reduced muscle

Identification of enterobacteriaceae causing septicemia in the axolotl Ambystoma mexicanum.

The Ambystoma mexicanum axolotl is a highly threatened amphibian and a valuable research model, with very little information about bacterial diseases affecting it. The aim of this study was to perform an identification of bacteria responsible for septicemia in three individuals. For all of them, necropsies were made, bacteria classification was performed by traditional and DNA-based molecular methods and tissues were histologically examined. All animals showed edema and ascites, and other tissues such as the lungs, spleen, liver, and kidney were also affected, dermatitis also occurred, in one

Stabilizing a Femur Osteotomy with a Plate Fixation in Ambystoma mexicanum.

The axolotl (Ambystoma mexicanum) is a promising model organism for regenerative medicine due to its remarkable ability to regenerate lost or damaged organs, including limbs, brain, heart, tail, and others. Studies on axolotl shed light on cellular and molecular pathways ruling progenitor activation and tissue restoration after injury. This knowledge can be applied to facilitate the healing of regeneration-incompetent injuries, such as bone non-union. In the current protocol, the femur osteotomy stabilization using an internal plate fixation system is described. The procedure was adapted for u

Characterization of immunoglobulin loci in the gigantic genome of Ambystoma mexicanum.

BACKGROUND: The axolotl, Ambystoma mexicanum is a unique biological model for complete tissue regeneration. Is a neotenic endangered species and is highly susceptible to environmental stress, including infectious disease. In contrast to other amphibians, the axolotl is particularly vulnerable to certain viral infections. Like other salamanders, the axolotl genome is one of the largest (32 Gb) and the impact of genome size on Ig loci architecture is unknown. To better understand the immune response in axolotl, we aimed to characterize the immunoglobulin loci of A. mexicanum and compare it with

Pathological features of olfactory neuroblastoma in an axolotl (Ambystoma mexicanum).

A one-year-old, female Mexican axolotl (Ambystoma mexicanum) had a rough-surfaced, polypoid, pink tumor mass of approximately 10 mm in diameter in the oral cavity. Histologically, the tumor extended from the ethmoturbinate region and into the oral cavity and had replaced some of the maxillary bone tissue. The tumor mass was composed of a lobular architecture of small round-shaped tumor cells with occasional Flexner-Wintersteiner-like rosette formation. There were no metastatic lesions in the other organs. Immunohistochemically, the tumor cells were partly positive for several neural markers (c

Genomic characterization of the T-cell receptor loci in Ambystoma mexicanum.

BACKGROUND: Amphibians are valuable models for comparative immunology. In the caudate Ambystoma mexicanum, the architecture of immunoglobulin loci resembles that of the anuran Xenopus tropicalis, although some antibody gene features are absent. Evidence supports the presence of T lymphocytes in axolotl, the expression of T cell receptor alpha, beta, and delta chains, and a restricted diversity in the delta chain. Here, we describe the T cell receptor loci in the A. mexicanum genome and compare them with X. tropicalis and other tetrapods. METHODS: T cell receptor loci were mapped and annotated

Gonadal Development in Larvae of the Paedomorphic Ambystoma mexicanum (Amphibia: Urodela).

The axolotl (Ambystoma mexicanum) is an endemic amphibian of the order Urodela, which has the ability to reproduce while retaining its larval characteristics. The primary objective of this study was to outline the stages of gonadal development leading to sexual maturity. Gonads from both male and female larvae, aged 4-12 months, were histologically processed and characterized according to their developmental stage. Histological analysis of the ovaries revealed that larvae aged 4-8 months were immature. At 9 and 10 months, the oocytes showed early signs of maturity, characterized by an increase

Systematic Necropsy and Tissue Sampling in the Neotenic Axolotl (Ambystoma mexicanum).

Axolotls (Ambystoma mexicanum) are neotenic salamanders widely used as experimental models in regenerative biology, developmental research, toxicology, and pharmacology. Despite their extensive use in laboratory research and conservation programs, standardized necropsy procedures for this species remain limited and poorly documented. The absence of systematic protocols limits the consistency of postmortem examinations and complicates the interpretation of pathological findings in both experimental and diagnostic contexts. Here, we present a detailed and reproducible necropsy protocol for the n

Tyrp1 is the mendelian determinant of the Axolotl (Ambystoma mexicanum) copper mutant.

Several dozen Mendelian mutants have been discovered in axolotl (Ambystoma mexicanum) populations, including several that affect pigmentation. Four recessive mutants have been described in the scientific literature and genes for three of these have been identified. Here we describe and genetically dissect copper, a mutant with an albino-like phenotype known only from the pet trade. We performed a cross segregating copper and wildtype color phenotypes and used bulked segregant RNA-Seq to identify a region on chromosome 6 that was enriched for single-nucleotide polymorphisms (SNPs) between the c
