Ameloblastic fibrosarcoma transformation from ameloblastic fibroma.

Ameloblastic fibrosarcoma (AFS) is an extremely rare malignant odontogenic tumor. It is composed of benign odontogenic epithelium, resembling that of ameloblastoma, and a mesenchymal part exhibiting features of fibrosarcoma. The development of this lesion in the jaws is either de novo or from preexisting ameloblastic fibroma which has been well documented. The most commonly affected site within the jaw is the posterior mandible. These tumors show local aggressiveness and a high tendency to recur. We present a case of a 33-year-old female patient with swelling of the right posterior mandible fo

WDR72 regulates vesicle trafficking in ameloblasts.

As the hardest tissue in the human body, tooth enamel formation is a highly regulated process involving several stages of differentiation and key regulatory genes. One such gene, tryptophan-aspartate repeat domain 72 (WDR72), has been found to cause a tooth enamel defect when deleted or mutated, resulting in a condition called amelogenesis imperfecta. Unlike the canonical genes regulating tooth development, WDR72 remains intracellularly and is not secreted to the enamel matrix space to regulate mineralization, and is found in other major organs of the body, namely the kidney, brain, liver, and

Ameloblastic Fibro-Odontoma.

Introduction: Ameloblastic fibro-odontoma (AFO) is a benign odontogentic tumor without an aggressive behavior, unlike the similar ameloblastic fibroma. Case Presentation: A 9-year-old boy, with tooth eruption failure, underwent enucleation and curettage of a well-defined variable radiolucent and radio-opaque right mandible lesion. There was odontogenic epithelium with peripheral palisading in a loose myxoid stroma as well as a disorganized component of dentin, enamel, and cementum, features of an AFO. Conclusion: AFO is an odontogenic mixed tumor of epithelium and mesenchyme.

Ameloblastic fibro-odontoma.

Ameloblastic fibro-odontoma is a slow growing, benign, expansile epithelial odontogenic tumour with odontogenic mesenchyme, accounting for 0.3-1.7% of jaw tumours, signifying its rarity. The WHO defines it as "a neoplasm composed of proliferating odontogenic epithelium in a cellular ectomesenchymal tissue with varying degrees of inductive changes and dental hard tissue formation". We report a case of an 11-year-old girl who presented to the Department of Maxillo-Facial Medicine and Radiology for the evaluation of a swelling in the left posterior mandible. Her clinical chart and investigations

Giant ameloblastic fibroma.

A 10 year-old male was referred to our department for a delay in the eruption of his lower canine, premolars and molars on the right side. The panoramic radiograph showed a multilocular radiolucent lesion approximately 3.5 x 7.5 cm in diameter including the right canine, first and second premolar and second mandibular molar. The lesion was clinically diagnosed as dentigerous cyst. It was enucleated via curettage of the bone bed and diagnosed as ameloblastic fibroma at the histopathological examination. After twenty-one months, radiographs showed that the surgical defect had filled with new bon

Ameloblasts express type I collagen during amelogenesis.

Enamel and enameloid, the highly mineralized tooth-covering tissues in living vertebrates, are different in their matrix composition. Enamel, a unique product of ameloblasts, principally contains enamel matrix proteins (EMPs), while enameloid possesses collagen fibrils and probably receives contributions from both odontoblasts and ameloblasts. Here we focused on type I collagen (COL1A1) and amelogenin (AMEL) gene expression during enameloid and enamel formation throughout ontogeny in the caudate amphibian, Pleurodeles waltl. In this model, pre-metamorphic teeth possess enameloid and enamel, wh

Tooth morphogenesis and the differentiation of ameloblasts.

All vertebrate organs are formed from several cell types, and it is currently believed that interactions between the different components constitute the most important mechanism in the regulation of organ morphogenesis. In developing teeth morphogenetic interactions occur between the epithelium covering the facial processes and the underlying neural crest-derived mesenchyme. Morphogenesis is accompanied by differentiation of the various dental cell types, including the ameloblasts. Although ameloblasts differentiate terminally and start the deposition of enamel matrix only after the completion

Ameloblastic Fibro-Odontoma.

The ameloblastic fibro-odontoma (AFO) is a rare, mixed odontogenic tumor exhibiting the histological characters of the ameloblastic fibroma and complex odontoma. It is comprised of proliferating ectodermal and mesenchymal components of odontogenic tissue as well as enamel and dentin. AFO normally presents as an asymptomatic swelling of the posterior maxilla or mandible and is usually associated with developing teeth, occurring predominantly in children and adolescents. Such lesions are generally found upon radiographic examination of patients whose tooth eruption is delayed. This lesion often

Ameloblastic fibro-odontoma.

INTRODUCTION: Ameloblastic fibro-odontoma (AFO) is a quite rare, mixed odontogenic tumour generally seen in the early stages of life. Frequent signs of this tumour are asymptomatic swelling, delayed tooth eruption and mixed radiological appearance within well-defined borders. Management of the lesion includes enucleation of the tumour and long-term follow-up. PRESENTATION OF CASE: A 10-year-old girl was referred to our oral and maxillofacial surgery clinic with an incidental radiological finding of radiopaque mass in the posterior region of maxilla. OPG showed unerupted tooth bud of upper rig

Ameloblastic carcinoma of the mandible.

BACKGROUND: Ameloblastic carcinoma is a rare, aggressive odontogenic neoplasm of the jaws in which the epithelial cells exhibit cytologic features of recognizable ameloblastoma and malignancy. Cases with metastasis have been infrequently reported. METHODS: A case of a 64-year-old white woman with mandibular ameloblastic carcinoma with documented distant metastasis is presented. The patient's presenting symptoms included facial asymmetry of the right jaw over 2 months and the development of moderate trismus. Clinical manifestations, pathology, treatment, and biologic behavior are discussed. Th

Ameloblastic fibrodentinoma, ameloblastic fibro-odontoma and odontoameloblastoma: Distinct entities or a morphological spectrum?

BACKGROUND: The classification of mixed odontogenic tumors remains one of the most debated topics in oral and maxillofacial pathology. Although ameloblastic fibrodentinoma (AFD), ameloblastic fibro-odontoma (AFO), and odontoameloblastoma were historically recognized as distinct entities, their biological nature and taxonomic status have been repeatedly questioned in successive editions of the World Health Organization (WHO) Classification of Head and Neck Tumours. OBJECTIVE: To critically evaluate the current evidence regarding the classification, pathogenesis, and biological behavior of AFD,

Malignant transformation of ameloblastic fibroma to ameloblastic fibrosarcoma: case report and review of the literature.

INTRODUCTION: Ameloblastic fibrosarcoma is a rare malignant odontogenic tumour and is regarded as the malignant counterpart of the ameloblastic fibroma. The epithelial component remains benign, but the mesenchymal component becomes malignant. The diagnosis is made by histopathology. PATIENT: The case of a 26-year-old man who underwent curettage of an ameloblastic fibroma and died of an ameloblastic fibrosarcoma is presented, and the course of malignant transformation is analysed retrospectively. CONCLUSION: One-third of ameloblastic fibrosarcoma cases seem to have developed from recurrent am
