Amelogenesis imperfecta.

Amelogenesis imperfecta (AI) represents a group of developmental conditions, genomic in origin, which affect the structure and clinical appearance of enamel of all or nearly all the teeth in a more or less equal manner, and which may be associated with morphologic or biochemical changes elsewhere in the body. The prevalence varies from 1:700 to 1:14,000, according to the populations studied. The enamel may be hypoplastic, hypomineralised or both and teeth affected may be discoloured, sensitive or prone to disintegration. AI exists in isolation or associated with other abnormalities in syndrome

Radioautographic studies on amelogenesis.

Radioautography has been used to visualize various aspects of morphogenesis and differentiation in the continuously erupting rat incisor. Formation of the entire incisor involves continuous production of "tooth segments" at the growing end of the tooth, each of which undergoes a similar history of development as it is carried by eruption towards the oral cavity. The sequence of differentiation which characterizes the life cycle of the ameloblasts was timed using 3H-thymidine, a precursor of DNA. The cells pass through presecretory, secretory and maturative stages whose collective activity resu

Amelogenesis imperfecta with taurodontism.

Reports of families having a combination of amelogenesis imperfecta and taurodontism are limited. This study of members of three families shows that the combination is inherited as an autosomal dominant trait. In each of the patients examined, neither condition was seen without the other. The enamel was rough and dysplastic and varied in color from white to yellow. Radiographically, taurodontism was present in the deciduous and permanent dentitions. The pulp chambers of the incisor teeth were larger than is usually seen at all ages. All patients had normal-appearing hair, fingernails, and bone

Amelogenesis Imperfecta: Case Study.

Amelogenesis imperfecta (AI) refers to a group of rare genetic disorders that involve tooth development and that are passed down through families as a dominant trait. This condition is characterized by abnormal enamel formation caused by gene mutations that alter the quality and/or quantity of enamel. This dental problem can impact both primary and permanent dentition, varies among affected individuals, and results in esthetic and functional problems. This condition caused the patient in the current case report to have a lack of confidence when speaking. The treatment for amelogenesis imperfec

Ultrastructural study of amelogenesis imperfecta.

An ultrastructural study of teeth with amelogenesis imperfecta revealed various aspects of microcavities in the enamel surface, which ranged from isolated imprints of ameloblasts corresponding to the mildest lesions at the end of amelogenesis, to pits caused by the death of 20 to 30 ameloblasts at the beginning of amelogenesis. Abnormalities in the shape of the prisms can be observed. Further, crystals are distributed randomly within a prism or at the junction of 2 contiguous prisms while intercrystalline spaces are widened, indicating in various places the lack of a preferred orientation of t

Analysis of a kindred with amelogenesis imperfecta.

Amelogenesis imperfecta (AI) is a group of hereditary disorders whose manifestations are generally considered as being confined to the teeth. Pedigree analysis of a kindred revealed 2 siblings affected with AI which was consistent with an autosomal recessive mode of inheritance. Cephalometric evaluation showed both children to have an obtuse gonial angle and steep mandibular plane. These skeletal changes were manifested as a severe anterior open bite. Histological evaluation of the teeth demonstrated numerous enamel changes including altered prism morphology, prism coalescence and disruption,

Novel ITGB6 Mutations Causing Amelogenesis Imperfecta.

BACKGROUND/OBJECTIVES: Amelogenesis imperfecta (AI) is a heterogeneous group of rare hereditary conditions mainly affecting the quantity and/or quality of tooth enamel. Its phenotypic expression is diverse, as is the mutational spectrum of the AI-causing genes and mutations. Integrins are cell-surface receptors that mediate adhesion between cells and between cells and the extracellular matrix. Among these, mutations in integrin αvβ6 have been shown to cause AI; however, phenotypic variation exists between the knockout mouse model and human cases, as well as among different human AI families.

ACP4 Variants in Hypoplastic Amelogenesis Imperfecta.

Amelogenesis imperfecta (AI) is a group of rare inherited conditions causing tooth enamel defects. Human acid phosphatase 4 (ACP4) is a transmembrane protein involved in maintaining appositional enamel growth. Variants in ACP4 cause recessive hypoplastic AI. Here we identify further families and review published ACP4 variants causing AI. In three Pakistani families, we identified a new ACP4 variant, c.254T > C, p.(Pro85Leu), which long-read sequencing revealed to be a founder variant. Two further families were homozygous for previously reported pathogenic ACP4 variants. Further details are als

Severely hypoplastic amelogenesis imperfecta with taurodontism.

BACKGROUND: The prominent dental feature of a boy was severely hypoplastic enamel in both primary and permanent teeth. CASE REPORT: Many permanent teeth were already infected while emerging in the oral cavity. Panoramic radiograph showed enlarged and elongated pulp chambers (taurodontism) in the permanent first molars. The clinical and radiological diagnosis was either hypomaturation-hypoplastic amelogenesis imperfecta with taurodontism (AIHHT) or tricho-dento-osseous syndrome (TDO). Histological examination of the upper right permanent first molar revealed thin lamellar or somewhat thicker a

[Amelogenesis imperfecta and hypothalamo-hypophyseal insufficiency].

Eleven patients with hypopituitarism, medio-facial hypoplasia and amelogenesis imperfecta were examined. The hypothalamo-pituitary dysfunction affected somatotrophic, gonadotrophic, corticotrophic and thyrotrophic function. The facial anomalies included medio-facial hypoplasia with hypertelorism and amelogenesis imperfecta. The latter--hypoplastic in nature--was noted in all anterior teeth and affected both dentitions. The enamel defect was confirmed on microscopic examination of an impacted tooth. The spectrum of malformation may also present as amelogenesis imperfecta in association with som

Is amelogenesis imperfecta an indication for renal examination?

BACKGROUND: The term 'amelogenesis imperfecta' (AI) describes a diverse group of hereditary conditions primarily affecting the quality and/or quantity of dental enamel. CASE REPORT: This paper describes a case in which hypoplastic AI with delayed/failure of eruption of the permanent teeth was shown to be associated with renal calcification. CONCLUSION: Given the importance of the renal involvement, the authors suggest that paediatric dentists consider referring all children with this dental phenotype for renal ultrasound examination.

Association of Amelogenesis Imperfecta and Bartter's Syndrome.

Bartter's syndrome is an autosomal recessive renal tubular disorder characterized by hypokalemia, hypochloremia, metabolic alkalosis, and hyperreninemia with normal blood pressure. Bartter's syndrome is associated with hypercalciuria and nephrocalcinosis. Amelogenesis imperfecta (AI) is a group of hereditary disorders that affect dental enamel. AI could be part of several syndromes. The enamel renal syndrome is the association of AI and nephrocalcinosis. We report two patients of AI with Bartter's syndrome.

Prosthodontic management of patients with amelogenesis imperfecta.

INTRODUCTION: Amelogenesis Imperfecta (AI) is an heterogenous genetic disorder that disturbs the developing enamel structure. This rare ectodermal defect leads to a variety of clinical manifestations due to agenesis, hypoplasia, and/or hypomineralisation of the enamel. AIMS AND OBJECTIVES: To describe the prosthodontic management of dental anomalies commonly associated with AI. METHODS: By using the classification of Witkop and Rao (1971), the variation in clinical presentation of the different Types of AI are illustrated and discussed, in particular Type I AI and Type 4 AI. RESULTS AND CON

[Structure and composition of the enamel in amelogenesis imperfecta].

Amelogenesis imperfecta (AI) represents a broad spectrum of genetic diseases affecting the enamel formation. Suspected changes in the composition and structure of the enamel can hamper the establishment of an adequate adhesive bonding to the substrate, i.e. by influencing the conditioning. It was the purpose of this article to dwell on this problematic issue and to present a case with a hypomaturation type of AI. Using dual etch biopsies, the composition and structure of the affected enamel was studied and findings compared to healthy control teeth, which were extracted for orthodontic reasons

Molecular-based phenotype variations in amelogenesis imperfecta.

Amelogenesis imperfecta (AI) is one of the typical dental genetic diseases in human. It can occur isolatedly or as part of a syndrome. Previous reports have mainly clarified the types and mechanisms of nonsyndromic AI. This review aimed to compare the phenotypic differences among the hereditary enamel defects with or without syndromes and their underlying pathogenic genes. We searched the articles in PubMed with different strategies or keywords including but not limited to amelogenesis imperfecta, enamel defects, hypoplastic/hypomaturation/hypocalcified, syndrome, or specific syndrome name. Ameloblasts express type I collagen during amelogenesis.

Enamel and enameloid, the highly mineralized tooth-covering tissues in living vertebrates, are different in their matrix composition. Enamel, a unique product of ameloblasts, principally contains enamel matrix proteins (EMPs), while enameloid possesses collagen fibrils and probably receives contributions from both odontoblasts and ameloblasts. Here we focused on type I collagen (COL1A1) and amelogenin (AMEL) gene expression during enameloid and enamel formation throughout ontogeny in the caudate amphibian, Pleurodeles waltl. In this model, pre-metamorphic teeth possess enameloid and enamel, wh

LAMB3 mutations causing autosomal-dominant amelogenesis imperfecta.

Amelogenesis imperfecta (AI) can be either isolated or part of a larger syndrome. Junctional epidermolysis bullosa (JEB) is a collection of autosomal-recessive disorders featuring AI associated with skin fragility and other symptoms. JEB is a recessive syndrome usually caused by mutations in both alleles of COL17A1, LAMA3, LAMB3, or LAMC2. In rare cases, heterozygous carriers in JEB kindreds display enamel malformations in the absence of skin fragility (isolated AI). We recruited two kindreds with autosomal-dominant amelogenesis imperfecta (ADAI) characterized by generalized severe enamel hypo
