Synergistic antibacterial activity between L-norvalyl-L-1-aminoethylphosphonic acid and nocardicin A.

The phosphonopeptide L- norvalyl -L-1- aminoethylphosphonic acid [ Nva -Ala(P)] has been studied in combination with 12 beta-lactam antibiotics for activity against Pseudomonas aeruginosa. Nocardicin A was found to give the most potent synergistic combination with Nva -Ala(P). This interaction was widely observed in clinical isolates of P. aeruginosa in vitro and in a mouse septicemia model. Synergy was also observed in vitro and in vivo in several other species, including Proteus mirabilis, indole-positive Proteus spp., and Serratia marcescens. The interaction between Nva -Ala(P) and nocardic

High-performance liquid chromatographic determination of 2-aminoethylphosphonic acid and 2-amino-3-phosphonopropionic acid in seawater matrix using precolumn fluorescence derivatization with o-phthalaldehyde-ethanethiol.

2-Aminoethylphosphonic acid (2-AEP) and 2-amino-3-phosphonopropionic acid (2-AP3) are two types of abundant and ubiquitous naturally occurring phosphonates used as sources of phosphorus by many prokaryotic lineages. The potential utilization mechanism of 2-AEP and 2-AP3 in eukaryotic phytoplankton is currently under investigation. However, the lack of suitable analytical methods in saline samples are the limitation of such researches. Herein, a high-performance liquid chromatography (HPLC) method for monitoring 2-AEP and 2-AP3 using precolumn fluorescence derivatization with o-phthalaldehyde-e

Detection of 2-aminoethylphosphonic acid in suspended particles in an ultraoligotrophic lake: a two-dimensional nuclear magnetic resonance (2D-NMR) study.

Particulate organic phosphorus (P) compounds were examined in ultraoligotrophic Lake Saiko, Japan. A cartridge filter was used to collect sufficient amount of suspended particles for analysis by a two-dimensional NMR (1H-31P heteronuclear multiple bond correlation). 2-Aminoethylphosphonic acid (2-AEP), a phosphonate, was detected in suspended particles in Lake Saiko. The identity of the phosphonate was confirmed by comparison with a commercially available compound. Because 2-AEP is bioavailable, microorganisms can store and use this compound under extremely P-limited conditions. This is the fi

Structures of an alanine racemase from Bacillus anthracis (BA0252) in the presence and absence of (R)-1-aminoethylphosphonic acid (L-Ala-P).

Bacillus anthracis, the causative agent of anthrax, has been targeted by the Oxford Protein Production Facility to validate high-throughput protocols within the Structural Proteomics in Europe project. As part of this work, the structures of an alanine racemase (BA0252) in the presence and absence of the inhibitor (R)-1-aminoethylphosphonic acid (L-Ala-P) have determined by X-ray crystallography to resolutions of 2.1 and 1.47 A, respectively. Difficulties in crystallizing this protein were overcome by the use of reductive methylation. Alanine racemase has attracted much interest as a possible

An Oxidative Pathway for Microbial Utilization of Methylphosphonic Acid as a Phosphate Source.

Methylphosphonic acid is synthesized by marine bacteria and is a prominent component of dissolved organic phosphorus. Consequently, methylphosphonic acid also serves as a source of inorganic phosphate (Pi) for marine bacteria that are starved of this nutrient. Conversion of methylphosphonic acid into Pi is currently only known to occur through the carbon-phosphorus lyase pathway, yielding methane as a byproduct. In this work, we describe an oxidative pathway for the catabolism of methylphosphonic acid in Gimesia maris DSM8797. G. maris can use methylphosphonic acid as Pi sources despite lackin

2-Aminoethylphosphonate utilization by the cold-adapted Geomyces pannorum P11 strain.

Cold-adapted strain of Geomyces pannorum P11 was found to mineralize of phosphorus-carbon bond-containing compound--2-aminoethylphosphonic acid (2-AEP, ciliatine). The biodegradation process proceeded in the phosphate-independent manner. Ciliatine-metabolizing enzymes' activity was detectable in cell-free extracts prepared from psychrophilic G. pannorum pregrown on 4 mM 2-AEP. Phosphonoacetaldehyde hydrolase (phosphonatase) activity in a partially purified extract was demonstrated at 10 °C.

Organophosphonates revealed: new insights into the microbial metabolism of ancient molecules.

Organophosphonates are ancient molecules that contain the chemically stable C-P bond, which is considered a relic of the reducing atmosphere on primitive earth. Synthetic phosphonates now have a wide range of applications in the agricultural, chemical and pharmaceutical industries. However, the existence of C-P compounds as contemporary biogenic molecules was not discovered until 1959, with the identification of 2-aminoethylphosphonic acid in rumen protozoa. Here, we review advances in our understanding of the biochemistry and genetics of microbial phosphonate metabolism, and discuss the role

Discovery of a New, Recurrent Enzyme in Bacterial Phosphonate Degradation: (R)-1-Hydroxy-2-aminoethylphosphonate Ammonia-lyase.

Phosphonates represent an important source of bioavailable phosphorus in certain environments. Accordingly, many microorganisms (particularly marine bacteria) possess catabolic pathways to degrade these molecules. One example is the widespread hydrolytic route for the breakdown of 2-aminoethylphosphonate (AEP, the most common biogenic phosphonate). In this pathway, the aminotransferase PhnW initially converts AEP into phosphonoacetaldehyde (PAA), which is then cleaved by the hydrolase PhnX to yield acetaldehyde and phosphate. This work focuses on a pyridoxal 5'-phosphate-dependent enzyme that

Genomic context analysis enables the discovery of an unusual NAD-dependent racemase in phosphonate catabolism.

Phosphonates are organic molecules containing a direct carbon-phosphorus (C-P) bond. They are chemically sturdy compounds that can, however, be degraded by environmental microorganisms. In the frame of bacterial phosphonate catabolism, we recently reported the discovery of (R)-1-hydroxy-2-aminoethylphosphonate ammonia-lyase (PbfA), a lyase acting on the natural compound (R)-2-amino-1-hydroxyethylphosphonate (R-HAEP). PbfA converts R-HAEP into phosphonoacetaldehyde (PAA), which can be subsequently processed and cleaved by further enzymes. However, PbfA is not active toward S-HAEP (the enantiome

Biogenic Phosphonate Utilization by Globally Distributed Diatom Thalassiosira pseudonana.

Phosphonates are a class of organic phosphorus (P) compounds that contribute ~25% of dissolved organic P. Recent studies reveal the important role of phosphonates mediated by prokaryotes in the marine P redox cycle. However, its bioavailability by eukaryotic phytoplankton is under debate. 2-Aminoethylphosphonic acid (2-AEP) and 2-amino-3-phosphonopropionic acid (2-AP3) are two biogenic phosphonates in the marine environment. Here, Thalassiosira pseudonana, a common diatom species in the ocean, is able to recover growth from P starvation when provided with 2-AEP and 2-AP3. Moreover, 2-AEP cultu

Inhibition of rabbit brain 4-aminobutyrate transaminase by some taurine analogues: a kinetic analysis.

The use of the antiepileptic drug, 4-aminobutyrate transaminase (GABA-T) inhibitor vigabatrin (VIGA), has been recently cautioned because it is associated to irreversible field defects from damage of the retina. Since novel GABA-T inhibitors might prove useful in epilepsy or other CNS pathologies as VIGA substitutes, the aim of the present investigation was to characterize the biochemical properties of some taurine analogues (TA) previously shown to act as GABA-T inhibitors. These include (+/-)piperidine-3-sulfonic acid (PSA), 2-aminoethylphosphonic acid (AEP), (+/-)2-acetylaminocyclohexane su

Probing a coral genome for components of the photoprotective scytonemin biosynthetic pathway and the 2-aminoethylphosphonate pathway.

Genome sequences of the reef-building coral, Acropora digitifera, have been decoded. Acropora inhabits an environment with intense ultraviolet exposure and hosts the photosynthetic endosymbiont, Symbiodinium. Acropora homologs of all four genes necessary for biosynthesis of the photoprotective cyanobacterial compound, shinorine, are present. Among metazoans, these genes are found only in anthozoans. To gain further evolutionary insights into biosynthesis of photoprotective compounds and associated coral proteins, we surveyed the Acropora genome for 18 clustered genes involved in cyanobacterial

A New Microbial Pathway for Organophosphonate Degradation Catalyzed by Two Previously Misannotated Non-Heme-Iron Oxygenases.

The assignment of biochemical functions to hypothetical proteins is challenged by functional diversification within many protein structural superfamilies. This diversification, which is particularly common for metalloenzymes, renders functional annotations that are founded solely on sequence and domain similarities unreliable and often erroneous. Definitive biochemical characterization to delineate functional subgroups within these superfamilies will aid in improving bioinformatic approaches for functional annotation. We describe here the structural and functional characterization of two non-h

Sub-3 nm Lanthanide-Doped Double Perovskite Quantum Dots: A Multifunctional Platform for Thermal-Enhanced Upconversion Nanothermometry and Magnetic Resonance Imaging.

Conventional perovskite microcrystals commonly exhibit luminescence thermal quenching and functional singularity, severely limiting their practical utility. The inherent toxicity and instability of lead-based variants further restrict their biomedical applicability. To address these challenges, we synthesized ultrasmall (∼2.5 nm) lead-free Cs2NaGdCl6:Yb3+,Er3+ double perovskite quantum dots (QDs) through an optimized variable-temperature hot-injection approach. These QDs display an anomalous thermal enhancement in upconversion luminescence, attributed to temperature-dependent desorption of sur

Phosphonopeptides as antibacterial agents: alaphosphin and related phosphonopeptides.

Alaphosphin, l-alanyl-l-1-aminoethylphosphonic acid, was selected from a range of phosphonopeptides for evaluation in humans on the basis of its antibacterial activity, pharmacokinetics, and stability to intestinal and kidney peptidases. In vitro, the antibacterial action was antagonized by small peptides, resulting in low activity on peptone media. On an antagonist-free medium alaphosphin was bactericidal and rapidly lysed most susceptible gram-negative bacteria, but it was largely bacteriostatic and essentially nonlytic against gram-positive organisms. Its spectrum included most strains norm

Methane production by phosphate-starved SAR11 chemoheterotrophic marine bacteria.

The oxygenated surface waters of the world's oceans are supersaturated with methane relative to the atmosphere, a phenomenon termed the 'marine methane paradox'. The production of methylphosphonic acid (MPn) by marine archaea related to Nitrosopumilus maritimus and subsequent decomposition of MPn by phosphate-starved bacterioplankton may partially explain the excess methane in surface waters. Here we show that Pelagibacterales sp. strain HTCC7211, an isolate of the SAR11 clade of marine α-proteobacteria, produces methane from MPn, stoichiometric to phosphorus consumption, when starved for phos

New ways to break an old bond: the bacterial carbon-phosphorus hydrolases and their role in biogeochemical phosphorus cycling.

Phosphonates are organophosphorus molecules that contain the highly stable C-P bond, rather than the more common, and more labile, C-O-P phosphate ester bond. They have ancient origins but their biosynthesis is widespread among more primitive organisms and their importance in the contemporary biosphere is increasingly recognized; for example phosphonate-P is believed to play a particularly significant role in the productivity of the oceans. The microbial degradation of phosphonates was originally thought to occur only under conditions of phosphate limitation, mediated exclusively by the poorly

Multiple Phosphonate Degradation Pathways in Amphidinium carterae-Associated Bacteria Potentially Contribute to Host Adaptation to Phosphorus Limitation.

Phosphonates in the ocean can serve as an alternative phosphorus (P) source for microorganisms when phosphate is scarce. Dinoflagellates cannot utilize phosphonates, but some associated bacteria can degrade these compounds and release phosphate. However, the community composition of these bacteria and their phosphonate degradation pathways remain poorly understood. In this study, the dinoflagellate Amphidinium carterae was cultured with 2-aminoethylphosphonic acid (2-AEP) as the exclusive P source. Metagenomic and genomic analyses were conducted to identify bacterial taxa and genes related to

Mechanisms of ZnO Nanoparticles Enhancing Phototransformation of Biologically Derived Organic Phosphorus in Aquatic Environments.

Zinc oxide nanoparticles (ZnO NPs), as the highly efficient photocatalysts, could enhance the transformation of biogenic organic phosphorus (OP) to orthophosphate (PO43-) by photodegradation, accelerating eutrophication. Conversely, orthophosphate can also transform ZnO NPs and thus potentially alter their catalytic and chemical properties. Here, we investigated the transformation mechanisms of three biogenic OP compounds and ZnO NPs under ultraviolet light (UV) illumination: inositol phosphates (IHPs), nucleic acids (DNA), and aminoethylphosphonic acid (AEP). The physicochemical characteristi
