Vannella russkaya sp. nov. (Amoebozoa, Discosea) - a new marine amoeba from a terrestrial habitat.

A new amoeba species, Vannella russkaya sp. nov., is described based on morphological (light and transmission electron microscopy) and molecular (SSU rRNA and Cox1) analyses. The strain was isolated from soil near a brackish stream, Solonik (Novgorod Region, Russia), and was cultivated under marine conditions. Phylogenetic analysis placed the new species within a clade comprising predominantly marine Vannella, with the closest affinity to V. salarenaria, which was also found in soil and cultivated under marine conditions. The studied strain exhibits a broad tolerance to salinity from 5‰ to 50‰

Genotyping of Candidatus Syngnamydia salmonis (chlamydiales; Simkaniaceae) co-cultured in Paramoeba perurans (amoebozoa; Paramoebidae).

Candidatus Syngnamydia salmonis (Chlamydiales, Simkaniaceae) was described as an epitheliocystis-causing bacterium from the gills of Atlantic salmon (Salmo salar) in Norway. A bacterium showing 99.2% 16S rRNA identity to Cand. S. salmonis is able to multiply in Paramoeba perurans and based on the classification criteria this bacterium could represent the same species as Cand. S. salmonis. Sequencing the genome of the cultured bacterium has made it possible to fulfill the minimal standards for genetic characterization of species within the order Chlamydiales. The complete rRNA genes, the amino

High genetic diversity of Sappinia-like strains (Amoebozoa, Thecamoebidae) revealed by SSU rRNA investigations.

Sappinia diploidea is known as a free-living amoeba of worldwide distribution and has also been reported as causative agent of a brain infection in an immunocompetent young man. In the current study, we were able to isolate eight strains of S. diploidea-like amoebae identified by light microscopy from different habitats. Cultures of all strains were established successfully for molecular characterization. The small subunit ribosomal RNA genes of all strains were sequenced and compared to one another, to the neotype of S. diploidea, and to strains of Sappinia pedata, the only other Sappinia spe

Vannella mustalahtiana sp. nov. (Amoebozoa, Vannellida) and rainbow trout nodular gill disease (NGD) in Russia.

An outbreak of nodular gill disease (NGD) in farmed rainbow trout Oncorhynchus mykiss (Walbaum, 1792) was recorded in Ladoga Lake (Karelia, north-western Russia) in Spring 2020. The disease was characterised by typical clinical signs including asphyxiation, distended opercula, loss of activity and swimming upside down under the water surface. Maximum monthly mortality was 15.2%. The histological examination of the gills showed deformation and clubbing of lamellae, epithelial hypertrophy and hyperplasia, lamellar fusion and fusion of filaments. Granulomas were located within the epithelial laye

All Different All Equal: High Polymorphism in Nebela (Amoebozoa: Arcellinida) Revealed by Additional Sampling and Barcoding.

Hyalospheniids are a conspicuous group of testate amoebae whose rich morphology has allowed their classification based on the characterization of their shells. However, recent studies showed the occurrence of convergences and phenotypic plasticity, which complicate their taxonomy. Molecular barcoding is the method of choice to resolve this diversity in an objective manner and accurately delineate species. In the genus Nebela, species diagnosis is based on small differences in shell size and shape, but sampling is scarce and restricted to a few locations, which could underestimate the real phen

Tuberomeba capitovia gen. et sp. nov. (Amoebozoa, Tubulinea) accommodates an incertae sedis lineage of monotactic, fish-associated amoebae.

A new species from a yet undescribed group of small monotactic amoebae was isolated from a cage with farmed rainbow trout (Oncorhynchus mykiss). Artificial salinization of the original water sample to 18‰ was required to recognize and isolate this amoeba, whereas in cultures with a salinity level corresponding to freshwater, the amoebae were in an encysted state. The studied amoeba is closely related to the clade of strains isolated from the internal tissues of various fish: the liver of tench (Tinca tinca), the liver of carp (Cyprinus carpio), and the kidney of freshwater stinging catfish (He

Leptomyxa echinata n. sp. - a remarkable species of the genus Leptomyxa (Amoebozoa: Tubulinea) from the sediment of an artificial freshwater pond.

The genus Leptomyxa unifies two morphologically distinct groups of species: reticulate and non-reticulate. Previously, it was believed that only large reticulate species were capable of adopting a fan-shaped form. In recent years, fan-shaped cells have been discovered in four small non-reticulate species. In this study, we describe Leptomyxa echinata n. sp., isolated from a bottom sediment sample from Sobachiy Pond, Izmailovsky Park, Moscow, Russia. Amoebae of this species are non-reticulate, flattened, and branched, and can adopt a monopodial form. Interestingly, the morphology of monopodial

Understanding the Evolution and Clade Diversity of Mayorella (Amoebozoa): Description of a Novel Species From Freshwater Near Cape Coast, Ghana.

Mayorella species are globally distributed amoeboid protists, yet their diversity remains poorly resolved because of pronounced morphological plasticity and limited molecular data. Freshwater ecosystems of West Africa remain particularly underexplored, with virtually no integrative taxonomic or molecular studies addressing Mayorella and broader microbial eukaryotic diversity. Here, we describe Mayorella budamensis sp. nov. isolated from Budamu Stream, Ghana, using an integrative approach combining live-cell morphology and SSU rDNA phylogenetic analyses. The new species exhibits extensive morph

The sexual side of parasitic protists.

Much of the vast evolutionary landscape occupied by Eukaryotes is dominated by protists. Though parasitism has arisen in many lineages, there are three main groups of parasitic protists of relevance to human and livestock health: the Apicomplexa, including the malaria parasite Plasmodium and coccidian pathogens of livestock such as Eimeria; the excavate flagellates, encompassing a diverse range of protist pathogens including trypanosomes, Leishmania, Giardia and Trichomonas; and the Amoebozoa, including pathogenic amoebae such as Entamoeba. These three groups represent separate, deep branches

Molecular identification of bacterial endosymbionts of Sappinia strains.

The genus Sappinia comprises free-living amoebae occurring worldwide in a variety of habitats such as soils, plant matter and freshwater ponds, but also animal faeces, and includes at present three species, S. pedata, S. diploidea and S. platani. The genus is potentially pathogenic, as indicated by the identification of S. pedata in a case of human amoebic encephalitis. Electron microscopy studies on some strains already revealed intracellular bacteria in Sappinia. In the current study, we performed 16S ribosomal RNA gene (rDNA) analysis of these bacterial endosymbionts. We first inferred rela

Evolution of the TSC1/TSC2-TOR signaling pathway.

The TSC1/TSC2-TOR signaling pathway [the signaling pathway that includes the heterodimeric TSC1 (tuberous sclerosis 1 protein)-TSC2 (tuberous sclerosis 2 protein) complex and TOR (target of rapamycin)] regulates various cellular processes, including protein synthesis, in response to growth factors and nutrient availability. Homologs of some pathway components have been reported from animals, fungi, plants, and protozoa. These observations led to the perception that the whole pathway is evolutionarily conserved throughout eukaryotes. Using complete genome sequences, we show that, contrary to th

Fatal gastric amebiasis in a Linnaeus's two-toed sloth associated with Naegleria australiensis infection.

Here we describe a case of fatal amebic gastritis associated with Naegleria australiensis infection in an 11-mo-old Linnaeus's two-toed sloth (Choloepus didactylus). The sloth had a history of weight loss and intermittent diarrhea for 18 d, and subsequently died despite empirical treatment. Postmortem findings included emaciation, gastric dilation with fluid content, and fibrinonecrotic gastritis with intralesional amebic trophozoites and cysts in the glandular region of the fundus. Transmission electron microscopy ruled out Amoebozoa of the family Entamoebidae based on the presence of mitocho

Conserved actin machinery drives microtubule-independent motility and phagocytosis in Naegleria.

Much of our understanding of actin-driven phenotypes in eukaryotes has come from the "yeast-to-human" opisthokont lineage and the related amoebozoa. Outside of these groups lies the genus Naegleria, which shared a common ancestor with humans >1 billion years ago and includes the "brain-eating amoeba." Unlike nearly all other known eukaryotic cells, Naegleria amoebae lack interphase microtubules; this suggests that actin alone drives phenotypes like cell crawling and phagocytosis. Naegleria therefore represents a powerful system to probe actin-driven functions in the absence of microtubules, ye

Environmental Free-Living Amoebae Isolated from Soil in Khon Kaen, Thailand, Antagonize Burkholderia pseudomallei.

Presence of Burkholderia pseudomallei in soil and water is correlated with endemicity of melioidosis in Southeast Asia and northern Australia. Several biological and physico-chemical factors have been shown to influence persistence of B. pseudomallei in the environment of endemic areas. This study was the first to evaluate the interaction of B. pseudomallei with soil amoebae isolated from B. pseudomallei-positive soil site in Khon Kaen, Thailand. Four species of amoebae, Paravahlkampfia ustiana, Acanthamoeba sp., Naegleria pagei, and isolate A-ST39-E1, were isolated, cultured and identified ba

Immersion challenge of naïve Atlantic salmon with cultured Nolandella sp. and Pseudoparamoeba sp. did not increase the severity of Neoparamoeba perurans-induced amoebic gill disease (AGD).

Amoebic gill disease (AGD) is one of the main health issues impacting farmed Atlantic salmon. Neoparamoeba perurans causes AGD; however, a diversity of other amoeba species colonizes the gills and there is little understanding of whether they are commensal or potentially involved in different stages of gill disease development. Here, we conduct in vivo challenges of naïve Atlantic salmon with cultured Nolandella sp. and Pseudoparamoeba sp. to investigate their pathogenicity to Atlantic salmon gills. Additionally, we assessed whether the presence of Nolandella sp. and Pseudoparamoeba sp. influe

DAPI staining and DNA content estimation of nuclei in uncultivable microbial eukaryotes (Arcellinida and Ciliates).

Though representing a major component of eukaryotic biodiversity, many microbial eukaryotes remain poorly studied, including the focus of the present work, testate amoebae of the order Arcellinida (Amoebozoa) and non-model lineages of ciliates (Alveolata). In particular, knowledge of genome structures and changes in genome content over the often-complex life cycles of these lineages remains enigmatic. However, the limited available knowledge suggests that microbial eukaryotes have the potential to challenge our textbook views on eukaryotic genomes and genome evolution. In this study, we develo

Characterizing the spatial distributions of soil biota at a legacy base metal mine using environmental DNA.

Characterizing the distribution of biota in response to contaminants is a critical element of site risk assessments. In this study we investigated the spatial distributions of biota and soil chemistry data in surface soil from Sunny Corner, a legacy base metal sulfide mine, Australia. Our results showed that copper (Cu), zinc (Zn), arsenic (As) and lead (Pb) in the surface soil exceeded Australian national soil quality guidelines and posed risks to the environment. Environmental (e)DNA metabarcoding of prokaryote and eukaryote composition confirmed the suggestion of environmental risk posed by

Molecular mechanisms of perilipin protein function in lipid droplet metabolism.

Perilipins are abundant lipid droplet (LD) proteins present in all metazoans and also in Amoebozoa and fungi. Humans express five perilipins, which share a similar domain organization: an amino-terminal PAT domain and an 11-mer repeat region, which can fold into amphipathic helices that interact with LDs, followed by a structured carboxy-terminal domain. Variations of this organization that arose during vertebrate evolution allow for functional specialization between perilipins in relation to the metabolic needs of different tissues. We discuss how different features of perilipins influence th
