A hallmark of plasma cells is the expression of syndecan-1, which has major functions in epithelial cells, in particular as the coreceptor of heparin-binding growth factors. We previously found that heparin-binding epidermal growth factor-like growth factor (HB-EGF) is a growth factor for malignant plasma cells. As amphiregulin (AREG) is another heparin-binding factor of the EGF family, we investigated its role in multiple myeloma (MM). Using Affymetrix DNA microarrays, we show here that the AREG gene was expressed by purified primary myeloma cells from 65 patients and that the expression was

Amphiregulin (AREG) is an epidermal growth factor (EGF)-like growth factor that binds exclusively to the EGF receptor (EGFR). Treatment with luteinizing hormone (LH) and/or human chorionic gonadotropin dramatically induces the expression of AREG in the granulosa cells of the preovulatory follicle. In addition, AREG is the most abundant EGFR ligand in human follicular fluid. Therefore, AREG is considered a predominant propagator that mediates LH surge-regulated ovarian functions in an autocrine and/or paracrine manner. In addition to the well-characterized stimulatory effect of LH on AREG expre

Although type 2 immune responses contribute to allergy and asthma, these responses are essential for clearing intestinal helminth infestations by mechanisms that include increased epithelial shedding. We show that T helper 2 cells (T(H)2), but not other T cell subsets, express amphiregulin, a member of the epidermal growth factor (EGF) family. EGF receptor ligands directly induce epithelial cell proliferation, and lack of amphiregulin delayed expulsion of the nematode Trichuris muris. This newly recognized link between T(H)2 cells and epithelial proliferation should help in planning therapeuti

Amniotic epithelial cells (AECs) exert potent paracrine immunoregulatory functions, yet whether inflammatory cues enhance the biological activity of their extracellular vesicles (EVs) remains unclear. To address this question, we characterized the secretome of ovine AECs under basal and lipopolysaccharide (LPS)-stimulated conditions using label-free proteomics and genetic approaches. Inflammatory stimulation profoundly remodeled the microvesicles (MVs) cargo, enriching 101 proteins associated with metabolic and regulatory pathways. Among the top 5% were two immune-related proteins, including a

Dimethylarsinic acid (DMA) is a major metabolite in the urine of humans and rats exposed to inorganic arsenicals, and is reported to induce rat bladder carcinogenesis. In the present study, we focused on early pathways of carcinogenesis triggered by DMA that were also active in tumors. RNA expression in the bladder urothelium of rats treated with 0 and 200 ppm DMA in the drinking water for 4 weeks and in bladder tumors of rats treated with 200 ppm DMA for 2 years was initially examined using microarray analysis and Ingenuity Pathway Analysis (IPA). Expression of 160 genes was altered in both t

Adhesion of the human pathogen Neisseria gonorrhoeae has established effects on the host cell and evokes a variety of cellular events including growth factor activation. In the present study we report that infection with N. gonorrhoeae causes altered amphiregulin processing and release in human epithelial cells. Amphiregulin is a well-studied growth factor with functions in various cell processes and is upregulated in different forms cancer and proliferative diseases. The protein is prototypically cleaved on the cell surface in response to external stimuli. We demonstrate that upon infection,

Amphiregulin (AREG) is an EGF-like ligand that binds EGFR to activate signaling pathways governing cellular proliferation, differentiation, tissue repair, and immune responses. This review integrates AREG's structural characteristics and signaling mechanisms with its biological functions derived from cell-based and animal models. We critically evaluate AREG's mechanistic involvement in cardiovascular, respiratory, intestinal, neoplastic, and infectious diseases, highlighting its dual roles in tissue protection and pathogenesis. The broader significance lies in discussing AREG's translational p

OBJECTIVE: Diabetic retinopathy is a common diabetic microvascular problem. Its diagnosis and classification are based on visible changes in clinical fundus examination. However, the discovery of possible vitreous biomarkers in patients with proliferative and nonproliferative diabetic retinopathy may guide both the differentiation and degree of retinopathy. Biomarkers that will be accepted can be also a treatment target. Amphiregulin (AREG) promotes proliferative and regenerative activity and repairs most cell types by binding and activating epidermal growth factor receptors. Progranulin (PGRN

BACKGROUND/PURPOSE: Hepcidin is an antimicrobial peptide that regulates iron metabolism. Recently, hepcidin was reported to promote wound healing. However, the mechanism underlying hepcidin signaling in the oral mucosa remains unclear. In the present study, we examined the mechanism by which hepcidin accelerates healing in a rat model of oral ulcerative mucositis.
MATERIALS AND METHODS: In male Wistar rats, 50 % acetic acid was applied to the labial fornix region of the inferior incisors to create a model of oral ulcerative mucositis. The ulcerative mucositis severity was evaluated using an o

Amphiregulin (AREG) serves as a ligand for the epidermal growth factor receptor (EGFR) and is involved in vital biological functions, including inflammatory responses, tissue regeneration, and immune system function. Upon interaction with the EGFR, AREG initiates a series of signaling cascades necessary for several physiological activities, such as metabolism, cell cycle regulation, and cellular proliferation. Recent findings have provided evidence for the substantial role of AREG in maintaining the equilibrium of homeostasis in damaged tissues and preserving epithelial cell structure in the c

BACKGROUND: Luteinizing hormone (LH) can stimulate mural granulosa cells to produce Amphiregulin (AREG), which can induce the resumption of meiosis in oocytes. Theca cells are present in the outer layer of follicles, providing communication with the pituitary axis through the established vascular system around the follicle. As LH target cells, it is unknown whether theca cells can produce AREG after LH stimulation.
METHODS: Primary cultured human theca cells were treated with LH (with or without the inhibitor of PKA, H89), or agonists of adenylate cyclase (forskolin or db-cAMP). The mRNA and

Thymic injury associated with disease or cancer treatment reduces T cell production and makes patients more vulnerable to infections and cancers. Here, we examined the role of regulatory T (Treg) cells on thymic regeneration. Treg cell frequencies increased in the thymus in various acute injury models. Depletion of Treg cells impaired thymic regeneration, impacting both the thymocyte compartment and the stromal cell compartment; adoptive transfer of Treg cells enhanced regeneration. Expansion of circulating Treg cells, as opposed to that of tissue resident or recent thymic emigrants, explained

BACKGROUND & AIMS: The loss of parietal cells from the gastric mucosa (oxyntic atrophy) is a critical step in the pathogenesis of chronic gastritis and gastric adenocarcinoma. Parietal cells are known to secrete epidermal growth factor receptor (EGFR) ligands, which are critical regulators of differentiation in the gastric mucosa. Although all of the actions of EGFR ligands are mediated through a common EGFR protein, individual ligands may produce different physiologic responses. Previous investigations have suggested that a deficit in EGFR signaling in waved-2 mice accelerates the emergence o

The primary pathological mechanism underlying ventricular remodeling and cardiac dysfunction following myocardial infarction (MI) is predominantly mediated by cardiomyocyte apoptosis. Pannexin1 (PANX1) channels, which open during apoptosis, facilitate the release of ATP from dying cells. However, the functional significance of PANX1 in mediating cardiomyocyte apoptosis and its contribution to myocardial infarction progression remain to be fully elucidated. To investigate the regulatory role of PANX1 in cardiomyocyte apoptosis following MI and elucidate its underlying molecular mechanisms. We c
