Perceived barriers to the adoption of alternatives to laboratory animal use for rabies diagnosis.

The use of laboratory animals is still common practice, but some uses can be replaced by alternative methods, such as Virus Isolation in Cell Culture (VICC) instead of the Mouse Inoculation Test (MIT) for rabies diagnosis. The objective of this work was to describe current rabies diagnosis methods in Brazil and other countries, and the constraints associated with replacing this use of mice with alternative methods. Nine out of 12 Brazilian and 14 out of 43 non-Brazilian respondents reported that they currently used the MIT. Respondents in countries other than Brazil, male respondents, and thos

An alternative to the use of animals to teach diabetes mellitus.

We developed an alternative approach to teach diabetes mellitus in our practical classes, replacing laboratory animals. We used custom rats made of cloth, which have a ventral zipper that allows stuffing with glass marbles to reach different weights. Three mock rats per group were placed into metabolic cages with real food and water and with test tubes containing artificial urine, simulating a sample collection of 24 h. For each cage, we also provided other test tubes with artificial blood and urine, simulating different levels of hyperglycemia. The artificial "diabetic" urine contained differ

Human Leukemia T-Cell Lines as Alternatives to Animal Use for Detecting Biologically Active Staphylococcal Enterotoxin Type B.

Staphylococcal enterotoxin type B (SEB) is associated with food poisoning. Current methods for the detection of biologically active SEB rely upon its ability to cause emesis when administered to live kittens or monkeys. This technique suffers from poor reproducibility and low sensitivity and is ethically disfavored over concerns for the welfare of laboratory animals. The data presented here show the first successful implementation of an alternative method to live animal testing that utilizes SEB super-antigenic activity to induce cytokine production for specific novel cell-based assays for qua

[Global trends in drug development using alternatives to animal testing].

In recent years, declining success rates and rising development costs have posed major challenges in drug development, highlighting the need for more efficient and rational approaches to evaluating drug efficacy and safety. Although non-clinical studies have traditionally relied primarily on animal experiments, adverse events that are difficult to predict continue to emerge during clinical trials and post-marketing, underscoring limitations in human extrapolation. In this context, new approach methodologies (NAMs), including human cell-based models and in silico analyses, have gained increasin

Use of blood matrices and alternative biological fluids for antibody detection in animal tuberculosis.

Bovine tuberculosis (bTB) control programs can be improved by implementation of advanced ante-mortem testing algorithms. Serodiagnostic methods using traditional blood or blood-derived specimens may benefit from the use of less invasive alternative biological fluids, provided those mirror systemic antibody responses. In the present study, we used Dual Path Platform (DPP) and Multiantigen Print Immunoassay (MAPIA) to compare antibody levels in ten sample types including whole blood (fresh and hemolyzed), plasma (fresh and leftover from Bovigam testing), serum, saliva, broncho-alveolar lavage, u

Alternative methods to reduce the animal use in quality controls of inactivated BTV8 Bluetongue vaccines.

The acceptance of serology data instead of challenge for market release of new batches of commercial vaccine is under evaluation by regulatory agencies in order to reduce the use of animals and costs for manufacturers. In this study two vaccines for Bluetongue virus serotype 8 were submitted to quality controls required by the European Pharmacopoeia and tested on sheep in comparison with a commercial inactivated vaccine. Body temperature, antibody titres and viraemia of vaccinated and controls sheep were recorded. In addition IL4 and IFNγ in sera and supernatant derived from in vitro stimulati

Alternative in vitro and in silico models to reduce animal use: a crucial technological advance in dermatological sciences.

INTRODUCTION: Alternative models are tools to replace and reduce the number of animals used in biomedical sciences, for either research or tests of industrial products. Several new alternative models have been developed in the most diverse fields. Their implementation has led to significant advances in the dermatological cosmetic industry, enabling chemical and molecular screening without animal use. However, limitations remain, particularly regarding tissue microenvironment complexity and systemic metabolic responses. OBJECTIVES: The objective of this viewpoint is to present the existing alt

The Overt and Hidden Use of Animal-Derived Products in Alternative Methods for Skin Sensitisation: A Systematic Review.

In vitro methods that can replace animal testing in the identification of skin sensitisers are now a reality. However, as cell culture and related techniques usually rely on animal-derived products, these methods may be failing to address the complete replacement of animals in safety assessment. The objective of this study was to identify the animal-derived products that are used as part of in vitro methods for skin sensitisation testing. Thus, a systematic review of 156 articles featuring 83 different in vitro methods was carried out and, from this review, the use of several animal-derived pr

The impact of sex, species, environmental context, and alternative reinforcers in animal models of cocaine use disorders.

Cocaine use disorder (CUD) remains a major public health challenge with no effective pharmacological treatments to date. Although extensive preclinical research using animal models has advanced our understanding, these findings have yet to translate into clinical success. This review highlights key independent variables that should be incorporated to improve the validity and translational relevance of animal studies. The discussion is organized around three primary domains: the agent; the host; and the environment. Most studies in this review focus on cocaine as the agent and host factors such

The use of a decision tree based on the rabies diagnosis scenario, to assist the implementation of alternatives to laboratory animals.

Brazilian federal legislation makes the use of alternatives mandatory, when there are validated methods to replace the use of laboratory animals. The objective of this paper is to introduce a novel decision tree (DT)-based approach, which can be used to assist the replacement of laboratory animal procedures in Brazil. This project is based on a previous analysis of the rabies diagnosis scenario, in which we identified certain barriers that hinder replacement, such as: a) the perceived higher costs of alternative methods; b) the availability of staff qualified in these methods; c) resistance to

Control of Salmonella and Pathogenic E. coli Contamination of Animal Feed Using Alternatives to Formaldehyde-Based Treatments.

This study compared a novel non-formaldehyde combination product developed for pathogen control in animal feed Finio (A), with a panel of three commonly used organic acid feed additive products: Fysal (B), SalCURB K2 (C) and Salgard (D). Products were evaluated for their ability to reduce Salmonella Typhimurium DT104 and avian pathogenic Escherichia coli in poultry feed. A commercial layer-hen mash was treated with each product and then mixed with feed previously contaminated (via inoculated meat and bone meal) with either Salmonella or E. coli. After 24 hours at room temperature, 10 replicate

Development of a multi-analysis model using an epithelial-fibroblast co-culture system as an alternative to animal testing.

The evaluation of respiratory chemical substances has been mostly performed in animal tests (OECD TG 403, TG 412, TG 413, etc.). However, there have been ongoing discussions about the limited use of these inhalation toxicity tests due to differences in the anatomical structure of the respiratory tract, difficulty in exposure, laborious processes, and ethical reasons. Alternative animal testing methods that mimic in vivo testing are required. Therefore, in this study, we established a co-culture system composed of differentiated epithelial cells under an air-liquid interface (ALI) system in the

Establishment of a strategy using alternative methods to the use of animals to evaluate the ocular irritant potential of cosmetics for styling, braiding, or fixing hair.

Assessing the safety of cosmetic products requires rigorous analysis before marketing, with emphasis on the potential for ocular irritation, given the sensitivity of the eye and the complexity of the toxicological mechanisms involved. Ocular irritation is a complex process involving multiple toxicity mechanisms; alternative methods can be combined within a testing strategy for a complete prediction of ocular safety. This study proposed a sequential testing strategy to assess the irritating potential of hair pomades, products widely used for hair styling but associated with adverse ophthalmolog

A genomic biomarker signature can predict skin sensitizers using a cell-based in vitro alternative to animal tests.

BACKGROUND: Allergic contact dermatitis is an inflammatory skin disease that affects a significant proportion of the population. This disease is caused by an adverse immune response towards chemical haptens, and leads to a substantial economic burden for society. Current test of sensitizing chemicals rely on animal experimentation. New legislations on the registration and use of chemicals within pharmaceutical and cosmetic industries have stimulated significant research efforts to develop alternative, human cell-based assays for the prediction of sensitization. The aim is to replace animal exp

Testing the biocompatibility of a glutathione-containing intra-ocular irrigation solution by using an isolated perfused bovine retina organ culture model - an alternative to animal testing.

The effects of a glutathione-containing intra-ocular irrigation solution, BSS Plus©, on retinal function and on the survival of ganglion cells in whole-mount retinal explants were studied. Evidence is provided that the perfused ex vivo bovine retina can serve as an alternative to in vivo animal testing. Isolated bovine retinas were prepared and perfused with an oxygen-saturated standard irrigation solution, and an electroretinogram was recorded to assess retinal function. After stable b-waves were detected, the isolated retinas were perfused with BSS Plus for 45 minutes. To investigate the eff
