Anoikis: To Die or Not to Die?

Epithelial, endothelial, and many connective tissue cells are normally attached to the extracellular matrix (ECM). These cells rely on the ECM for structural support, signaling, and regulation of their behavior. When these cells lose this attachment or are in an inappropriate location, these cells soon die by a mechanism called anoikis (homelessness). Anoikis is a programmed cell death of an apoptotic nature; however, it can, in certain cases, be overcome, and detached cells can survive in the absence of the correct signals from the ECM. This is the case of malignant cells, where anoikis resis

Assessment of resistance to anoikis in ovarian cancer.

Anoikis, a form of programmed cell death that occurs due to cell detachment from the extracellular matrix, is a critical mechanism in preventing ectopic cell growth. Acquisition of resistance to anoikis is a prerequisite for epithelial ovarian cancer cells to survive in ascitic fluids before forming metastatic foci. Here we describe a colorimetric method for monitoring the resistance of anoikis of ovarian cancer cells in vitro.

[The regulation of anoikis in tumor invasion and metastasis].

As a barrier to metastasis of cancer, cells that lost contact with the neighbouring cells or extracellular matrix(Extracellular matrix, ECM) will be subjected to apoptosis. This cell death process has been termed "anoikis". When normal epithelial cells or solid tumor cells without metastatic potential detach from the primary site, and then enter into the circulatory system, the anoikis mechanism will be activated. The significance of anoikis is to prevent the shedding cells from growing and implanting into other inappropriate sites. Tumor cells, especially several malignant cells that is prone

TGFBR3 supports anoikis through suppressing ATF4 signaling.

Epithelial morphogenesis and oncogenic transformation can cause loss of cell adhesion, and detached cells are eliminated by anoikis. Here, we reveal that transforming growth factor β receptor 3 (TGFBR3) acts as an anoikis mediator through the coordination of activating transcription factor 4 (ATF4). In breast cancer tissues, TGFBR3 is progressively lost, but elevated TGFBR3 is associated with a histologic subtype characterized by cellular adhesion defects. Dissecting the impact of extracellular matrix (ECM) deprivation, we demonstrate that ECM loss promotes TGFBR3 expression, which in turn cau

Anoikis resistance in gastric cancer: a comprehensive review.

Gastric cancer (GC) is a predominant malignant neoplasia responsible for cancer death worldwide. Because of the difficulty in early diagnosis as well as its high metastasis rate, GC shows an increasing incidence and poor prognosis. Conventional treatments for GC, such as chemotherapy, radiotherapy, and surgical resection, still fail to achieve curative effects because of drug resistance, a mechanism that leads to a reduction of 5-year survival for GC patients. Anoikis, a particular type of programmed cell death, is activated upon cancer cell detachment from the extracellular matrix, playing a

Shedding of NG2 by MMP-13 attenuates anoikis.

Disruption of cell-matrix interactions can lead to anoikis-apoptosis due to loss of matrix contacts. We previously showed that Nerve/glial antigen 2 (NG2) is a novel anoikis receptor. Specifically, overexpression of NG2 leads to anoikis propagation, whereas its suppression leads to anoikis attenuation. Interestingly, NG2 expression decreases in late anoikis, suggesting that NG2 reduction is also critical to this process. Thus, we hypothesized that NG2 undergoes cleavage to curtail anoikis propagation. Further, since matrix metalloproteinases (MMPs) cleave cell surface receptors, play a major r

PIK3CA silencing restores anoikis sensitivity in endothelial cells.

BACKGROUND: The development, differentiation, and homeostasis of adherent cells are regulated by interactions among cells, the extracellular matrix (ECM), and soluble factors. Loss of these contacts induces anoikis, a specific form of apoptosis. Tumor cells acquire resistance to anoikis, enabling them to survive without anchorage, invade the vascular system, and colonize distant organs in a process known as metastasis. Activation of the PI3K/Akt signaling pathway is the most common mechanism underlying the acquisition of anoikis resistance in cancer cells. The role of phosphoinositide 3-kinase

ABHD4-dependent developmental anoikis safeguards the embryonic brain.

A specialized neurogenic niche along the ventricles accumulates millions of progenitor cells in the developing brain. After mitosis, fate-committed daughter cells delaminate from this germinative zone. Considering the high number of cell divisions and delaminations taking place during embryonic development, brain malformations caused by ectopic proliferation of misplaced progenitor cells are relatively rare. Here, we report that a process we term developmental anoikis distinguishes the pathological detachment of progenitor cells from the normal delamination of daughter neuroblasts in the devel

Role and Mechanism of BRIP1 in Anoikis Resistance of Gastric Cancer.

To assess the therapeutic relevance of BRIP1 in gastric cancer (GC), we examine its functional role in conferring resistance to anoikis within GC cells and elucidate the oncogenic signaling pathways modulated by BRIP1. By integrating the Cancer Genome Atlas (TCGA) and Gene Set Enrichment Analysis (GSEA) databases with Least Absolute Shrinkage and Selection Operator (LASSO) regression, a novel risk score to stratify GC patients based on prognosis was generated, and a significantly differentially expressed gene, BRIP1, was validated through reverse transcription quantitative polymerase chain rea

Pathogenesis of Thyroid Cancer with Particular Emphasis on the Role of Anoikis.

Thyroid cancer (TC) is the most prevalent malignancy of the endocrine system, and its incidence has been increasing worldwide in recent years. Although it generally has a favorable prognosis, aggressive forms, such as anaplastic TC, are associated with high mortality rates. Cell-microenvironment interactions largely influence the progression and metastatic behavior of TC however, the precise mechanisms underlying these processes remain inadequately understood. One critical factor influencing metastasis is anoikis, a form of programmed cell death triggered by the detachment of cells from the ex

Identification of STAT3 and BIRC5 as anoikis-related biomarkers in psoriasis.

Anoikis is a programmed cell death that occurs when cells detach from the extracellular matrix (ECM). Its role in psoriasis remains unclear. This study aims to explore the relationship between psoriasis and apoptosis, focusing on anoikis-related mechanisms. Differentially expressed genes (DEGs) in psoriasis were identified using the GEO database and overlapped with anoikis-related genes from GeneCards to find differentially expressed anoikis-related genes (DE-ARGs). A PPI network was constructed, revealing eight hub DE-ARGs. These were then analyzed using GO, KEGG and ssGSEA. Immune infiltrati

YAP activation in melanoma contributes to anoikis resistance and metastasis.

Melanoma is inherently heterogeneous, providing resistance to apoptosis. Anoikis resistance is a hallmark feature of metastatic melanoma to escape apoptosis when cells lose contact with adjacent cells or extracellular matrix. The yes-associated protein transcription co-activator is the effector of Hippo pathway. Herein, we investigated the function of yes-associated protein in anoikis resistance of melanoma cells. When melanoma cells were grown under anchorage-independent condition, anoikis-resistant cells displayed higher levels of yes-associated protein activation than the cells that were at

Roles of anoikis in hepatocellular carcinoma: mechanisms and therapeutic potential.

Hepatocellular carcinoma (HCC), the most common primary liver cancer, is a highly aggressive malignancy with limited viable therapeutic options. For early HCC, resection surgery is currently the most effective treatment. However, in advanced stages, resection alone does not sufficiently address the disease, so finding a method with a better prognosis is necessary. Anoikis, known as matrix detachment-induced apoptosis or detachment-induced cell death, is crucial for tissue development and homeostasis. Cancer cells develop means to evade anoikis, e.g. anoikis resistance, thereby allowing for cel

A prognostic model for anoikis-related genes in pancreatic cancer.

Anoikis, a distinct form of programmed cell death, is crucial for both organismal development and maintaining tissue equilibrium. Its role extends to the proliferation and progression of cancer cells. This study aimed to establish an anoikis-related prognostic model to predict the prognosis of pancreatic cancer (PC) patients. Gene expression data and patient clinical profiles were sourced from The Cancer Genome Atlas (TCGA-PAAD: Pancreatic Adenocarcinoma) and the International Cancer Genome Consortium (ICGC-PACA: Pancreatic Ductal Adenocarcinoma). Non-cancerous pancreatic tissue gene expressio

Resistance to anoikis in transcoelomic shedding: the role of glycolytic enzymes.

Detachment of cells from the extracellular matrix into the peritoneal cavity initiates a cascade of metabolic alterations, leading usually to cell death by apoptosis, so-called anoikis. Glycolytic enzymes enable the switch from oxidative phosphorylation to aerobic glycolysis and allow resistance to anoikis of shed tumour cells. These enzymes also have moonlighting activities as protein kinases and transcription factors. Phosphoglycerate kinase (PGK) and pyruvate kinase are the only glycolytic enzymes generating ATP in the hexokinase pathway. Hypoxia, EGFR activation, expression of K-Ras G12V a

Induction of Anoikis in Human Colorectal Cancer Cells by Fucoxanthinol.

Fucoxanthin (Fx), one of the major xanthophylls in brown algae, is known to be effective for colorectal cancer (CRC) chemoprevention through inhibiting cell growth, cell cycle and caspase activation. Recently, we observed fucoxanthinol (FuOH), an anti-cancer active metabolite of Fx, treatment of human CRC cells resulted in plenty of living floating cells several hours after exposure, and induced apoptosis. In the present study, we investigated whether FuOH induced anchorage-dependent apoptosis, that is "anoikis", along with integrin signal suppression in human CRC cells. We found that cells ex

Role of CD24 in anoikis resistance of ovarian cancer cells.

This study examined the effect of CD24 on anoikis of ovarian cancer cells. The expression of CD24 was detected by RT-PCR and Western blotting in ovarian cancer cells with high metastatic potential (HO-8910PM cells) and low metastatic potential (A2780 cells). Cell viability and cell proliferation were detected by MTT assay in suspension culture and adhesion culture. Soft agar culture was used to observe the colony formation. Anoikis was flow cytometrically detected. The results showed that the expression levels of CD24 mRNA and protein were significantly higher in HO-8910PM cells than in A2780

Relationship between keratinocyte adhesion and death: anoikis in acantholytic diseases.

Loss of attachment to the substratum may trigger apoptosis in epithelial cells (anoikis). It is less clear whether apoptosis may be triggered by disruption of cell-cell contacts, as happens in acantholytic diseases. Biopsy specimens were obtained from the border of skin lesions from four patients with pemphigus vulgaris (PV), four patients with pemphigus foliaceus (PF), three patients with Darier's disease (DD), two patients with Darier's-type Grover's disease (GD), and two patients with benign familial pemphigus Hailey-Hailey disease (HH). CAMK1 Suppresses Anoikis Resistance and Liver Metastasis in Colorectal Cancer.

Metastasis represents a major challenge in the treatment of clinical colorectal cancer (CRC), and anoikis resistance enables cancer cell dissemination. This study identifies Calcium/calmodulin-dependent protein kinase I (CAMK1) as a crucial regulator of anoikis in CRC progression. We analyzed CAMK1 expression in CRC tissues and its correlation with patient prognosis. In vitro, CAMK1 was overexpressed in CRC cell lines to assess its effects on proliferation, migration, invasion, and anoikis resistance. In vivo, mouse models were established to evaluate the impact of CAMK1 overexpression on tumo

FOXM1 regulates platelet-induced anoikis resistance in pancreatic cancer cells.

BACKGROUND: Resistance to anoikis, a form of programmed cell death that occurs after detachment from the surrounding extracellular matrix, is a prerequisite for the survival of circulating tumor cells (CTCs) in the bloodstream. Platelets can interact with these CTCs and protect them from cytokine- and immune cell-mediated cell death. Whether platelets can regulate anoikis resistance by controlling intrinsic gene expression changes in tumor cells that contribute to metastasis has not been studied in detail in pancreatic cancer cells. METHODS: Pancreatic cancer cells were cultured under attache
