Food ingredients as anti-obesity agents: a review.

Overweight and obesity have a major impact on global health; their prevalence has rapidly increased in all industrialized countries in the past few decades and diabetes and hypertension are their direct consequences. Pharmacotherapy provides reinforcement for obesity treatment, but should be an adjunctive support to diet, exercise, and lifestyle modification. At present, only orlistat and sibutramine have been approved by the US Food and Drug Administration for long-term use, but sibutramine was withdrawn for sale by the European Medicines Agency. The development of functional foods for the pr

Sibutramine: a review of the pharmacology of a novel anti-obesity agent.

Sibutramine is a noradrenaline and 5-hydroxytryptamine reuptake inhibitor which causes weight loss in laboratory rodents via effects on both food intake and metabolic rate. Sibutramine's effects are predominantly mediated by two pharmacologically-active metabolites (its primary and secondary amines). Sibutramine and its active metabolites do not cause the release of monoamine neurotransmitters and do not have affinity for their receptors. Sibutramine dose-dependently inhibits 24 h food intake in rats by enhancing the natural physiological process of satiety. Sibutramine also stimulates thermog

The utility of animal models to evaluate novel anti-obesity agents.

The global incidence of obesity continues to rise and is a major driver of morbidity and mortality through cardiovascular and cerebrovascular diseases. Animal models used in the discovery of novel treatments for obesity range from straightforward measures of food intake in lean rodents to long-term studies in animals exhibiting obesity due to the continuous access to diets high in fat. The utility of these animal models can be extended to determine, for example, that weight loss is due to fat loss and/or assess whether beneficial changes in key plasma parameters (e.g. insulin) are evident. The potential of artemisinins as anti-obesity agents via modulating the immune system.

Artemisinin and its derivatives are the most effective antimalarial drugs. Besides anti-malarial activity, artemisinin and its derivatives have displayed wide-spectrum bioactivities such as anti-parasite, anti-tumor, and anti-obesity effects. Obesity is an epidemic worldwide which is a big threat to human health, but there are only a few approved anti-obesity drugs in the world. Also, these drugs are efficient to limited patients partly because their safety and efficacy are questioned. Anti-inflammatory therapies may be valuable in obesity treatment since growing evidence shows chronic metabol

Synthesis and biological evaluation of tert-butyl-5-methylpyrimidin-piperazine derivatives as anti-obesity agents.

A series of tert-butyl-5-methylpyrimidin-piperazine derivatives were synthesized and their anti-obesity activities were evaluated. Compounds 4g and 5j were found to have significant effects in down-regulating the triglyceride level of 3T3-L1 adipocytes. 5j exhibited remarkable therapeutic effects on the diet-induced obesity (DIO) mouse model at 20 mg kg(-1)  day(-1) for 4 weeks by decreasing the weights of body, liver, and fat. 5j also regulated serum biomarkers to appropriate ranges, exerted therapeutic activity of steatosis in liver tissue and ameliorated the obese-related symptoms. Oxygenated aromatic cadinane sesquiterpenoid dimers from Stahlianthus involucratus as a new class of anti-obesity agents.

Stahlianthus involucratus, a medicinal herb indigenous to Southwest China traditionally used for inflammation, remains unexplored for anti-obesity potential. Bioactivity-guided fractionation afforded an active fraction that markedly ameliorates high-fat diet (HFD)-induced obesity and related metabolic disorders. Phytochemical investigation of this active fraction afforded thirteen oxygenated aromatic cadinane-type sesquiterpenoid dimers (CSDs, 1-13), including eight new compounds. Their structures were established by extensive spectroscopic analyses, including NMR, HRESIMS, ECD and single-crys

Design, synthesis and structure-activity relationship of novel 2-pyrimidinylindole derivatives as orally available anti-obesity agents.

Due to the emerging global epidemic of obesity, developing safe and effective agents for anti-obesity is urgently needed. Our previous study found that 2-pyrimidinylindole derivative Wd3d exhibited potential anti-obesity activity. Herein, to further optimize the potential moiety, structural modifications were proceeded for two rounds in this study. Firstly, we designed, synthesized, and evaluated 36 new derivatives of 2-pyrimidinylindole scaffold with different substituents on the indole ring and pyrimidine ring to investigate their structure-activity relationship (SAR). Then, analogs with pot

Anti-consumption agents: Tirzepatide and semaglutide for treating obesity-related diseases and addictions, and improving life expectancy.

American culture encourages overconsumption, fueled by ubiquitous availability and pervasive marketing of ultra-processed foods and other addictive substances. This chronic overindulgence has contributed to rising rates of obesity, type 2 diabetes (T2D), substance abuse, mental health disorders and premature mortality. Glucose-like peptide-1 agonists (GLP-1RAs) affect the brain's reward pathway that mediates addiction to foods and various other substances. Evolving data suggest that tirzepatide and semaglutide may be the first effective "anti-consumption" agents with potential applications in

Flaxseed protein hydrolysate as a potential anti-obesity agent: Combined animal study and in silico investigation.

This study investigated the metabolic benefits of flaxseed protein hydrolysate (FPIH) in comparison with its parent protein, flaxseed protein isolate (FPI), and explored the underlying mechanisms using an integrated in vivo animal model and in silico approach. FPIH was produced via alcalase hydrolysis of FPI (final DH ∼ 21%), and both treatments were administered at 300 mg/kg body weight (BW) to Wistar rats fed a high-fat diet (HFD) for 10 weeks. FPIH showed an antioxidant potential higher than that of FPI and an inhibitory effect against pancreatic lipase in vitro (IC50 = 1.2 mg/mL). These fi

Combining kinetics and in silico approaches to evaluate bromhexine as an anti-pancreatic lipase agent for obesity management.

Industrialization, fast food intake and reduced physical activity, mainly in developed countries, exacerbate obesity and make it a major lifestyle disorder. A promising strategy for developing effective anti-obesity agents is to inhibit pancreatic lipase, thereby reducing lipid absorption. Currently, the only clinically approved pharmacological agent for pancreatic lipase inhibition is Orlistat. However, its undesirable gastrointestinal side effects have prompted the search for more effective and potent drugs. This study investigates the inhibitory mechanism of Bromhexine, a mucolytic drug, on

Coumarin Analogues as Promising Anti-Obesity Agents: In Silico Design, Synthesis, and In Vitro Pancreatic Lipase Inhibitory Activity.

A set of coumarin-3-carboxamide analogues were designed, synthesized, and evaluated for their ability to impede pancreatic lipase (PL) activity. Out of all the analogues, 5dh and 5de demonstrated promising inhibitory activity against PL, as indicated by their respective IC50 values of 9.20 and 11.4 μM, as compared to Orlistat (IC50 = 0.97 μM). It was found that analogue 5dh inhibited PL in a competitive manner with an inhibition constant (Ki) of 4.504 μM. Additionally, the docking analysis validated the interactions between the analogue 5dh (MolDock score of -140.251 kcal/mol) and key amino ac

Cell signaling mechanisms of gustatory perception of lipids: can the taste cells be the target of anti-obesity agents?

It has been well propounded that there exists five basic taste modalities, e.g., sweet, sour, bitter, salty and umami. Recent compelling evidence from rodents and human beings raises the possibility for an additional sixth taste modality devoted to the perception of lipids. A number of studies have suggested that lingual CD36, a glycoprotein, mainly expressed by circumvallate papillae of the tongue, might be implicated in the perception of dietary fat taste. G protein-coupled receptors (GPCRs) are important signaling molecules for many aspects of cellular function. It has been recently shown t
