Antigen presentation for central tolerance induction.

The extent of central T cell tolerance is determined by the diversity of self-antigens that developing thymocytes 'see' on thymic antigen-presenting cells (APCs). Here, focusing on insights from the past decade, we review the functional adaptations of medullary thymic epithelial cells, thymic dendritic cells and thymic B cells for the purpose of tolerance induction. Their distinct cellular characteristics range from unconventional phenomena, such as promiscuous gene expression or mimicry of peripheral cell types, to strategic positioning in distinct microenvironments and divergent propensities

Endo-lysosomal proteases in antigen presentation.

Endo-lysosomal proteases have long been attractive, yet elusive, targets for medicinal chemistry. They have found to play key roles in health and disease; with protease under- and over-activity having been implicated in cancer, osteoporosis and Alzheimer's disease. Here we will discuss their role in the adaptive immune response. The crucial roles of these enzymes multiple processes in antigen presentation will be discussed: from activating MHC-II receptors, to the production of epitopes from antigens and the activation of Toll-like receptors. The early efforts at pharmacological interventions

In Situ Polymerization-Mediated Antigen Presentation.

Activating antigen-presenting cells is essential to generate adaptive immunity, while the efficacy of conventional activation strategies remains unsatisfactory due to suboptimal antigen-specific priming. Here, in situ polymerization-mediated antigen presentation (IPAP) is described, in which antigen-loaded nanovaccines are spontaneously formed and efficiently anchored onto the surface of dendritic cells in vivo through co-deposition with dopamine. The resulting chemically bound nanovaccines can promote antigen presentation by elevating macropinocytosis-based cell uptake and reducing lysosome-r

Antigen Presentation of Vacuolated Apicomplexans--Two Gateways to a Vaccine Antigen.

For parasites that sequester themselves within a vacuole, new rules governing antigen presentation are coming into focus. Components of the host's autophagy machinery and the parasite's membranous nanotubular network within the parasitophorous vacuole play a major role in determining antigenicity of Toxoplasma proteins. As such, both parasite and vaccinologist may exploit these pathways to regulate the ever important CD8 T cell response to apicomplexan parasites.

Evidence for antigen presentation by human neutrophils.

Neutrophils are the first migrating responders to sterile and infectious inflammation and act in a powerful but nonspecific fashion to kill a wide variety of pathogens. It is now apparent that they can also act in a highly discriminating fashion; this is particularly evident in their interactions with other cells of the immune system. It is clear that neutrophils are present during the adaptive immune response, interacting with T cells in complex ways that differ between tissue types and disease state. One of the ways in which this interaction is mediated is by neutrophil expression of HLA mol

Medullary Thymic Epithelial Cell Antigen-presentation Assays.

Medullary thymic epithelial cells (mTEC) are bona fide antigen-presenting cells that play a crucial role in the induction of T-cell tolerance. By their unique ability to express a broad range of tissue-restricted self-antigens, mTEC control the clonal deletion (also known as negative selection) of potentially hazardous autoreactive T cells and the generation of Foxp3+ regulatory T cells. Here, we describe a protocol to assess major histocompatibility complex (MHC) class II antigen-presentation capacity of mTEC to CD4+ T cells. We detail the different steps of thymus enzymatic digestion, immuno

Ethanol inhibits antigen presentation by dendritic cells.

Previous studies suggest that altered virus-specific T-cell responses observed during chronic ethanol exposure may be due to abnormal functioning of dendritic cells (DCs). Here we explored the effects of ethanol on exogenous antigen presentation by DCs. BALB/c, C57BL/6, and CBA/caj mice were fed ethanol or an isocaloric control diet for 8 weeks. The splenic DC population was expanded using an Flt3L expression plasmid via tail vein injection. DCs were purified and assessed for antigen presentation and processing and for peptide-major histocompatibility complex class I and II (MHCI and MHCII) fo

[CD1: A new paradigm for antigen presentation].

Molecules of the major histocompatibility complex (MHC) bind protein-derived peptide antigens and present them to T cells. This has been a central dogma in modern immunology, and our appreciation of a variety of cell-mediated immune responses has been based only on this paradigm. However, we now know that T cell recognition also involves non-peptide antigens. Studies over the past several years have established a new paradigm that non-MHC-encoded molecules of the CD1 family mediate presentation of lipid antigens to T cells, and unraveled their significant role in microbial immunity, tumor immu

Impaired antigen presentation in toxic epidermal necrolysis.

BACKGROUND AND DESIGN--The pathophysiology of toxic epidermal necrolysis (TEN) remains largely unknown. Toxic epidermal necrolysis is considered a hypersensitivity reaction to drugs, but direct evidence of an immunologic mechanism is still lacking. Lymphopenia and a decrease in the numbers of circulating CD3- and CD4-positive lymphocytes have been reported in acute phase, but, to our knowledge, no study of cellular immune functions of patients with TEN has been reported so far. Herein, we investigated several T-cell functions in a series of 11 patients with TEN. Peripheral blood mononuclear ce

Antigen presentation in human autoimmune thyroid disease.

Monocyte/macrophage function in patients with autoimmune thyroiditis was investigated by their presentation of two distinct antigens; sheep red blood cells (SRBC) and human thyroglobulin (hTg) using in vitro systems designed for antibody induction. Purified peripheral blood monocyte/macrophages were primed by prefeeding with antigen for 60 min at 37 degrees C, washed, and co-cultured with autologous lymphocytes under a variety of incubation conditions. The most successful system employed 5% monocyte/macrophages with autologous T-B cells in the presence of the mitogen Staphylococcus aureus and

Presentation of a membrane cysticercus antigen and its homology with excretory--secretory antigen.

A cysticercus antigen was purified from the membrane. It is a glycoprotein containing hexoses and sialic acids with a molecular weight of 70 Kd and isoelectric focussing point 4.8. In western blot studies, the purified antigen bound with antibodies present in cerebrospinal fluid of all neurocysticercosis patients tested. Amino acid composition of the antigen showed a high content of Glutamate, Leucine and Aspartate while Proline and Hydroxyproline were absent. When cysts recovered from pigs were cultured for eight days, the excretory-secretory antigen obtained bound strongly with the antibody

Oligosaccharide adjuvant-deferred antigen endocytosis promotes cross-presentation.

Antigen cross-presentation by dendritic cells (DCs) is essential for inducing cytotoxic immune responses, which are crucial for eliminating infected or malignant cells. Although microbial signal processing reveals that the temporal relationship between DC activation and antigen uptake dictates cross-presentation efficiency, these two fundamental events are poorly coordinated in current vaccine design. Here, we report an oligosaccharide (OG) adjuvant that induces rapid DC activation but deferred antigen uptake, by sequentially activating two separate pathogen-recognition receptors on the same c

Phagosomes are competent organelles for antigen cross-presentation.

The ability to process microbial antigens and present them at the surface of cells is an important aspect of our innate ability to clear infections. It is generally accepted that antigens in the cytoplasm are loaded in the endoplasmic reticulum and presented at the cell surface on major histocompatibility complex (MHC) class I molecules, whereas peptides present in endo/phagocytic compartments are presented on MHC class II molecules. Despite the apparent segregation of the class I and class II pathways, antigens from intracellular pathogens including mycobacteria, Escherichia coli, Salmonella

Spotlight on TAP and its vital role in antigen presentation and cross-presentation.

In the late 1980s and early 1990s, the hunt for a transporter molecule ostensibly responsible for the translocation of peptides across the endoplasmic reticulum (ER) membrane yielded the successful discovery of transporter associated with antigen processing (TAP) protein. TAP is a heterodimer complex comprised of TAP1 and TAP2, which utilizes ATP to transport cytosolic peptides into the ER across its membrane. In the ER, together with other components it forms the peptide loading complex (PLC), which directs loading of high affinity peptides onto nascent major histocompatibility complex class

Virus-like particle vaccines: from antigen architecture to antigen presentation and adaptive immune responses.

INTRODUCTION: Virus-like particles (VLPs) mimic key structural features of native viruses while lacking a replication-competent viral genome, combining favorable safety with multivalent antigen presentation and strong immunogenicity. Yet, how VLP design influences the quality and persistence of adaptive immunity remains incompletely understood. AREAS COVERED: This review examines relevant literature identified through targeted searches on recent advances in VLP engineering, with emphasis on particle architecture, antigen-display strategies, and structural determinants of immune recognition. Methods for Probing NK Cell Surveillance of Antigen Presentation.

Natural killer (NK) cells are critical components of the innate immune system, capable of rapidly identifying and eliminating virus-infected and tumor cells without prior antigen sensitization. This rapid cytotoxic response is regulated by a balance between activating and inhibitory receptor signals, a mechanism known as "missing self." Inhibitory receptors recognize HLA class I molecules, preventing unwanted activation against healthy cells, while activating receptors detect stress-induced ligands on abnormal cells, triggering the release of perforin and granzyme B.Recent evidence shows that

Development of a novel assay for antigen presentation measurement.

Accurate measurement of antigen presentation is essential for understanding immune responses to infections and tumors. However, current methods are cumbersome, time-consuming, and rely on known peptide sequences and antibodies, leading to unstable antigen presentation, antigen loss during processing and editing, and inconsistent results. We developed a novel, cost-effective method for examining antigen presentation using Click chemistry, which utilizes a bioorthogonal reaction between azides and alkynes/cyclooctenes. Antigens were pre-labeled with azides or alkynes to facilitate their uptake b

Birinapant Reshapes the Tumor Immunopeptidome and Enhances Antigen Presentation.

Birinapant, an antagonist of the inhibitor of apoptosis proteins, upregulates MHCs in tumor cells and displays a better tumoricidal effect when used in combination with immune checkpoint inhibitors, indicating that Birinapant may affect the antigen presentation pathway; however, the mechanism remains elusive. Based on high-resolution mass spectrometry and in vitro and in vivo models, we adopted integrated genomics, proteomics, and immunopeptidomics strategies to study the mechanism underlying the regulation of tumor immunity by Birinapant from the perspective of antigen presentation. Firstly,

Engineering Cell Lines for Specific Human Leukocyte Antigen Presentation.

Epitope-specific immunotherapies have enabled the targeted treatment of a variety of diseases, ranging from cancer, infection, and autoimmune disorders. For CD8+ T cell-based therapies, the precise identification of immunogenic peptides presented by human leukocyte antigen (HLA) class I is essential which can be achieved by immunopeptidomics. Here, using lentivirus-mediated transduction and cell sorting approaches, we present a method to engineer a cell line that does not express its native HLA but instead expresses an HLA of interest (in this instance HLA-A*02:01). This technique can be used
