Immunization with bacterial antigens: pasteurellosis.

Pasteurella piscicida is the aetiological agent of pasteurellosis or pseudotuberculosis, one of the most threatening diseases of wild and cultured marine fish. This bacterium has been reported from many geographical areas including USA, Japan, and the Mediterranean countries. In this review, the biochemical, serological, and molecular characteristics of the pathogen are described. In addition, its main virulence mechanisms, such as the presence of capsule, the iron uptake system, and the phospholipase activity, as well as their putative role in the pathogenicity of P. piscicida are also discus

Interactions of lymphocytes with bacterial antigens.

Several observations indicate that smooth (S) and rough (R) Salmonella strains display the capacity to spontaneously adhere to lymphoid cell membrane. Such a phenomenon is confined to T lymphocytes and affects both CD4+ and CD8+ cells. As far as receptor structures on lymphocytes surface are concerned, the lipopolysaccharides (LPS) of the bacterial cell wall play a key role in human and murine cytoadherence. In addition, evidence has been provided that LPS of gut flora induce bacterial binding as assessed by the evaluation of cyto-adherence at different anatomical sites. Interestingly, cells m

Mucosal delivery of antigens using adsorption to bacterial spores.

The development of new-generation vaccines has followed a number of strategic avenues including the use of live recombinant bacteria. Of these, the use of genetically engineered bacterial spores has been shown to offer promise as both a mucosal as well as a heat-stable vaccine delivery system. Spores of the genus Bacillus are currently in widespread use as probiotics enabling a case to be made for their safety. In this work we have discovered that the negatively charged and hydrophobic surface layer of spores provides a suitable platform for adsorption of protein antigens. Binding can be promo

Mite-related bacterial antigens stimulate inflammatory cells in rosacea.

BACKGROUND: Patients with papulopustular rosacea have a higher density of Demodex folliculorum mites on their faces than normal subjects but the role, if any, of their mites in initiating inflammation is disputed. Selective antibiotics are effective in reducing the inflammatory changes of papulopustular rosacea, but their mode of action is unknown. OBJECTIVES: To investigate whether a D. folliculorum-related bacterium was capable of expressing antigens that could stimulate an inflammatory immune response in patients with rosacea. METHODS: A bacterium (Bacillus oleronius) was isolated from a

Whipple's disease: morphologic and immunofluorescence characterization of bacterial antigens.

The bacterial etiology of Whipple's disease is generally accepted. However, the exact identity of the "Whipple bug" has remained elusive. Indeed, the isolation of several types of bacteria from different patients with Whipple's disease has caused some to speculate that Whipple's disease may have polymicrobial etiology. Our light and electron microscopic studies document the presence of bacilliform organisms lying free in the lamina propria of the duodenal mucosa. Intact and partially degraded bacterial organisms were seen in the phagosomes of macrophages. Indirect immunofluorescence studies de

Identification of circulating bacterial antigens by in vivo microbial antigen discovery.

UNLABELLED: Detection of microbial antigens in clinical samples can lead to rapid diagnosis of an infection and administration of appropriate therapeutics. A major barrier in diagnostics development is determining which of the potentially hundreds or thousands of antigens produced by a microbe are actually present in patient samples in detectable amounts against a background of innumerable host proteins. In this report, we describe a strategy, termed in vivo microbial antigen discovery (InMAD), that we used to identify circulating bacterial antigens. This technique starts with "InMAD serum," w

Reactive arthritis: the role of bacterial antigens in inflammatory arthritis.

For more than 100 years it has been suspected that bacteria or products derived from them are deposited in joints and cause arthritis without suppuration. Over this time a vast amount of evidence, much of which is still unchallenged, has accumulated to demonstrate that whole bacteria and subcellular bacterial elements do pass, under certain circumstances, from sites of mucosal colonization or infection into the circulation and thence into joints. Similarly, experimental studies have demonstrated that the deposition of both inert material and bacterial components within synovium is sometimes, b

Pronounced changes occur in the kidneys carp injected with extracellular product (ECP), lipopolysaccharide (LPS) of A. salmonicida subsp. salmonicida, Freund's complete adjuvant (FCA). ECP caused 50-70% decrease in macrophages and neutrophils, whereas blasts and lymphocyte-like cells increased FCA-elicited increased percentages of blasts and a left shift of neutrophils after LPS injection. After both ECP and FCA injections, the density of major leucocyte types and red blood cells (RBC) in the kidney decreased. LPS generally increased the density of leucocytes. Granulocytes were larger after LP

Focused specificity of intestinal TH17 cells towards commensal bacterial antigens.

T-helper-17 (TH17) cells have critical roles in mucosal defence and in autoimmune disease pathogenesis. They are most abundant in the small intestine lamina propria, where their presence requires colonization of mice with microbiota. Segmented filamentous bacteria (SFB) are sufficient to induce TH17 cells and to promote TH17-dependent autoimmune disease in animal models. However, the specificity of TH17 cells, the mechanism of their induction by distinct bacteria, and the means by which they foster tissue-specific inflammation remain unknown. Here we show that the T-cell antigen receptor (TCR)

Lymphocyte proliferative responses to bacterial antigens in B27-associated arthropathies.

Lymphocyte transformation tests to Yersinia enterocolitica 0:3 and 0:6, Klebsiella pneumoniae and Streptococcus faecalis antigens have been carried out in ankylosing spondylitis, reactive arthritis/Reiter's disease and controls. Ankylosing spondylitis cases gave significantly lower responses than reactive arthritis/Reiter's to Yersinia enterocolitica 0:6 (p = 0.003) and Klebsiella pneumoniae (p = 0.008), and less than controls to S. faecalis (p = 0.008). It appears, therefore, that within the B27 arthritis population there is heterogeneity of cell-mediated response to certain enteric bacteria,

[Determination of bacterial antigens by latex agglutination tests in cultured cerebrospinal fluid specimens from bacterial meningitis].

Between December 1988 and February 1991, the latex agglutination test was evaluated for its ability to detect the antigens of N. meningitidis, S. pneumoniae and H. influenzae in the samples of cerebrospinal fluid (CSF) of 57 patients with bacterial meningitis. Totally antigens were detected in 34 of 42 (80.9%) patients with bacterial meningitis proven by culture. Latex agglutination was positive in 60% of CSFs for N. meningitidis, 88.2% of CSFs for S. pneumoniae and 100% of CSFs for H. influenzae. Antigens were detected in 7 of 9 (77.8%) patients with no bacterial organisms grown on CSF cultur

Coated Bacterial Vaccine: A New Approach for Antigen Display on Bacterial Surface.

Bacterial vaccines using recombinant antigens displayed on the bacterial surface represent a major advancement in vaccine development. This approach leverages bacteria's natural ability to induce immune responses while improving vaccine targeting and efficacy. Existing platforms, such as live bacterial vaccines, inactivated bacterial vaccines, and bacterial-like particles (BLPs), face challenges like genetic modifications, antigen dilution, and degradation. To address these limitations, a novel system called coated bacterial vaccine (CBV) is proposed, combining the properties of inactivated ba

Immunoglobulin E antibody reactivity to bacterial antigens in atopic dermatitis patients.

BACKGROUND: Atopic dermatitis (AD) is a chronic inflammatory skin disease affecting up to 20% children and 9% adults world-wide. AD patients are often sensitized against a broad variety of allergens and more than 90% of them suffer from skin superinfections with Staphylococcus aureus. OBJECTIVE: In this study, we searched for the presence of specific IgE antibodies against S. aureus and Escherichia coli antigens in AD patients. METHODS: Sera from AD patients (n=79), patients suffering only from allergic rhinoconjunctivitis (n=41) or allergic asthma (n=37) were tested for IgE reactivity to ni
