BACKGROUND: Sulphidoleukotrienes (slt) are important mediators in allergic diseases that are synthesized after allergen-specific stimulation.
OBJECTIVES: The aim of this study is to determine in vitro slt production after allergen-specific (Dermatophagoides pteronyssinus) stimulus of peripheral blood leucocytes and to observe whether histamine release in whole blood with the same allergen correlates with slt production. We also wanted to evaluate whether a correlation exists between the release of slt and histamine and other diagnostic procedures as well as various clinical situations.

Mite dialysate prepared from an extract of house dust mite, Dermatophagoides farinae, suppressed 48-h homologous passive cutaneous anaphylaxis (PCA) in rats caused by a non-dialyzable fraction of the mite extract (mite antigen), and the suppression was dose-dependent with a high specificity for the antigen. INTRODUCTION: 'Gold standard' in the diagnosis of atopic disease are skin prick tests and specific IgE evaluation. Well-established in vitro tests, such as the histamine release test, the leukotriens release test and the flow cytometric basophil activation test can be very helpful in diagnostics, especially when the skin prick test is contraindicated. This study demonstrated that antigens of the parasitic mite Sarcoptes scabiei (SS) cross-react with antigens of the house dust mite Dermatophagoides pteronyssinus (DP). Crossed immunoelectrophoresis (CIE) reaction of SS extract with rabbit anti-DP serum resulted in multiple immunoprecipitates. Reciprocal CIE reactions gave similar results. BACKGROUND: In recent years considerable interest in the pathogenetic role of aeroallergens exacerbating atopic dermatitis (AD) has emerged. The 'atopy patch test' with aeroallergens was introduced by Platts-Mills et al. as an experimental model and as a diagnostic tool. Dust mites are a medically important group of animals commonly found in carpets and mattresses in houses. Antigens in their feces cause allergic reactions such as asthma and contact dermatitis. Dermatophagoides had been established as an antigen in house dust by many investigators. An untapped source of cross antigenicity is presented in this paper, namely, Otodectes cyanotis, the mite found in the ears of millions of cats. House dust samples for mite species in other parts of the world may not be similar to samples in the United States since in most other countries the cat exists outdoors, whereas in this country it is a household pet.
