Nitrothiadiazolo[3,2-a]pyrimidines as promising antiglycating agents.

Managing the advanced glycation end-products (AGEs) concentration is a reliable approach to achieve control over the pathogenesis of diabetic vascular complications. Inhibition of dipeptidyl peptidase-4 (DPP-4) is also an attractive way to tackle type 2 diabetes mellitus (T2DM). We showed previously that azoloazine heterocycles have the potential to prevent the formation of AGEs and in this work, we conducted docking studies with DPP-4 of 5-alkylamino-6-nitro-1,3,4-thiadiazolo[3,2-a]pyrimidines. Consequently, we have developed a synthetic approach to these structures by chlorodeoxygenation and

Ameliorating Effect of Akebia quinata Fruit Extracts on Skin Aging Induced by Advanced Glycation End Products.

The accumulation of free radicals and advanced glycation end products (AGEs) in the skin plays a very important role in skin aging. Both are known to interact with each other. Therefore, natural compounds or extracts that possess both antioxidant and antiglycation activities might have great antiageing potential. Akebia quinata fruit extract (AQFE) has been used to treat urinary tract inflammatory disease in traditional Korean and Chinese medicines. In the present study, AQFE was demonstrated to possess antioxidant and antiglycation activity. AQFE protects human dermal fibroblasts (HDFs) from

[Antiglycation Activity of Isoindole Derivatives and Its Prediction Using Frontier Molecular Orbital Energies].

The extracellular matrix (ECM) provides structural support and regulates cell activity. ECM dysfunction due to metabolic pathologies or aging can lead to disease. Developing ECM protectors is crucial for the etiological prevention and treatment of pathologies associated with ECM alterations. Key mechanisms of pathological changes in the ECM include nonenzymatic reactions, such as glycation and glycoxidation. The potential of agents as ECM protectors can be assessed by their capability of inhibiting these processes. Compounds based on heterocyclic scaffolds with partially hydrogenated isoindole

Preliminary evaluation of the antiglycoxidant activity of verapamil using various in vitro and in silico biochemical/biophysical methods.

Introduction: Glycoxidative stress is essential for linking glucose disturbances and cardiovascular diseases. Unfortunately, contemporary antidiabetic drugs do not have an antiglycative effect but only lower blood glucose levels. Therefore, there is an intense search for substances that could inhibit protein glycation and prevent diabetic complications. A potential antioxidant activity has been demonstrated with verapamil, a phenylalkylamine derivative belonging to selective calcium channel blockers. Verapamil has a well-established position in cardiology due to its wide range of indications a

Olive leaf extract concentrated in hydroxytyrosol attenuates protein carbonylation and the formation of advanced glycation end products in a hepatic cell line (HepG2).

Glycation takes place both at the cellular level and at the extracellular matrix level and generates, consequently, advanced glycation end-products (AGEs) associated with chronic diseases and the aging process. Two olive leaf extracts concentrated in (i) oleuropein (OLE-A; 93.9 mg oleuropein g-1) and (ii) hydroxytyrosol (OLE-B; 54.5 mg hydroxytyrosol g-1) were evaluated according to their antiglycative and antioxidant capacity in vitro. OLE-B exerted the highest anti-AGE effect in different glycation models (IC50: 0.25-0.29 mg mL-1). OLE-B showed the highest antioxidant capacity and methylglyo

Multi-spectroscopic and in silico approaches reveal the α-amylase and α-glucosidase inhibitory mechanism of anacardic acids and their effects on advanced glycation end products.

Anacardic acids (AAs) are a class of alkyl phenols widely distributed in the Anacardiaceae family and known for their diverse biological properties. However, their potential against type 2 diabetes mellitus (T2DM) has never been thoroughly investigated. In this study, a fraction isolated from a Pistacia vera shell extract, enriched in AAs, exhibited potent inhibitory activity against α-amylase (α-Amy) and α-glucosidase (α-Glu). Pure (15:0)-AA and (17:1)-AA also acted as strong inhibitors of both enzymes, displaying IC₅₀ values significantly lower than those of the marketed anti-diabetic drug a

Drug repurposing: In-vitro anti-glycation properties of 18 common drugs.

Drug repositioning or repurposing, i.e. identifying new indications for existing drugs, has gained increasing attention in the recent years. This approach enables the scientists to discover "new targets" for known drugs in a cost and time efficient manner. Glycation, the non-enzymatic reaction of sugars with proteins or nucleic acids to form early glycation (Amadori or fructosamine) products, is a key molecular basis of diabetic complications. Inhibiting the process of non-enzymatic protein glycation is one of the key strategies to prevent glycation-mediated diabetic complications. The present
