Molecular characterization of planktic cyanobacteria of Anabaena, Aphanizomenon, Microcystis and Planktothrix genera.

Toxic and non-toxic cyanobacterial strains from Anabaena, Aphanizomenon, Calothrix, Cylindrospermum, Nostoc, Microcystis, Planktothrix (Oscillatoria agardhii), Oscillatoria and Synechococcus genera were examined by RFLP of PCR-amplified 16S rRNA genes and 16S rRNA gene sequencing. With both methods, high 16S rRNA gene similarity was found among planktic, anatoxin-a-producing Anabaena and non-toxic Aphanizomenon, microcystin-producing and non-toxic Microcystis, and microcystin-producing and non-toxic Planktothrix strains of different geographical origins. The respective sequence similarities we

Rare cyanobacteria drive nitrogen-fixation and cyanotoxin production in an Aphanizomenon-dominated bloom.

Late summer, recurring cyanobacterial blooms in Lake of the Woods (LOW) are polycyanobacterial and typically dominated by Aphanizomenon flos-aquae. LOW waters are typically nitrogen limited, relative to phosphorus. As such, the dominance of Aphanizomenon flos-aquae, a putative nitrogen-fixing cyanobacterium, suggests that its ability to fix nitrogen may be advantageous and aid in its ability to bloom. This study sought to quantify nitrogen fixation rates and identify cyanotoxin-producing species during the blooms. Throughout the 2021 season, we quantified nutrients, N-fixation rates, microbial

Genomic insights into the taxonomy, safety and biotechnological potential of Aphanizomenon flos-aquae and related Nostocales.

The Nostocales are filamentous cyanobacteria that include bloom-forming, nitrogen-fixing species with major ecological and biotechnological relevance. Owing to their metabolic diversity, nitrogen-fixing capabilities and nutritional profile, they hold potential for various biotechnological applications including the production of nutraceuticals and biofertilizers. However, the capacity of certain Nostocales species to produce cyanotoxins also raises health concerns. This study provides a comparative bioinformatic analysis of 161 Nostocales genomes from the National Center for Biotechnology Info

Cyanophage lysis reshapes nitrogen cycling and microbiome composition in diazotrophic cyanobacterium Aphanizomenon flos-aquae.

Although it is presumed that viruses play a significant role in nutrient cycling and bacterial community dynamics, this has only rarely been addressed in studies of diazotrophic cyanobacteria. In this study, we therefore examined how cyanophage infection affects the expression of nitrogen (N) cycling genes, N₂ fixation rates, and population structure in the diazotrophic cyanobacterium Aphanizomenon flos-aquae, a prevalent bloom-forming species in temperate brackish and freshwater ecosystems. We also assessed the influence of A. flos-aquae lysis on co-occurring bacterial assemblages throughout

Aeruginosin 525 (AER525) from Cyanobacterium Aphanizomenon Sp. (KUCC C2): A New Serine Proteases Inhibitor.

Aeruginosins (AERs) are one of the most common classes of cyanobacterial peptides synthesised through a hybrid non-ribosomal peptide synthase/polyketide synthase pathway. They have been found in Microcystis, Nodularia spumigena, Oscillatoria/Plantothrix, and Nostoc. The presence of AER in Aphanizomenon isolated from the Curonian Lagoon was reported for the first time in our previous work. Here, the structure of aeruginosin 525 (AER525), isolated from Aphanizomenon sp. KUCC C2, was characterised based on high-resolution mass spectrometry. This new AER variant shows potent activity against throm

NOS-derived NO modulates photosynthetic integrity, carbon metabolism and cellular homeostasis in the cyanobacterium Aphanizomenon flos-aquae under methyl viologen stress.

Photosynthetic organisms often face environmental challenges that disrupt cellular homeostasis. Of the various protective mechanisms, nitric oxide (NO) is a crucial signaling molecule with diverse regulatory functions. In mammalian cells, NO is enzymatically synthesized by nitric oxide synthase (NOS) and regulates several physiological processes such as neuronal differentiation, vascular relaxation, synaptic signaling, and immune regulation. Although recent studies have reported the presence of canonical NOS homologs in photosynthetic microorganisms, their function is yet to be elucidated. Hen

The response and detoxification strategies of three freshwater phytoplankton species, Aphanizomenon flos-aquae, Pediastrum simplex, and Synedra acus, to cadmium.

The response and detoxification mechanisms of three freshwater phytoplankton species (the cyanobacterium Aphanizomenon flos-aquae, the green alga Pediastrum simplex, and the diatom Synedra acus) to cadmium (Cd) were investigated. The cell growth of each species was measured over 10 days, and chlorophyll a fluorescence, Cd bioaccumulation (including surface-adsorbed and intracellular Cd), and phytochelatin (PC) synthesis were determined after 96-h exposures. The growth of the three phytoplankton species was significantly inhibited when Cd concentrations were ≥5 mg L(-1). Compared with P. simple

Deciphering the impact of NOS-derived NO on nitrogen metabolism and carbon flux in the heterocytous cyanobacterium Aphanizomenon flos-aquae 2012/KM1/D3.

Nitric oxide synthases (NOSs) are heme-based monooxygenases that catalyze the NADPH-dependent oxidation of L-arginine to produce NO and L-citrulline. Over the past five years, the identification and characterization of NOS homologs in cyanobacteria have significantly advanced our understanding of these enzymes. However, the precise mechanisms through which NOS-derived NO influences nitrogen metabolism remain incompletely elucidated. Therefore, the present study aims to investigates the impact of NOS-derived NO on nitrogen metabolism, heterocyte development, and carbon utilization dynamics in A

Allergenic (sensitization, skin and eye irritation) effects of freshwater cyanobacteria--experimental evidence.

Many studies have been published on the human health effects of cyanobacterial toxicity. As a public health concern, we have to mention that the development of contact dermatitis, asthmalike symptoms, and symptoms resembling hay fever during bathing in cyanobacterial blooms have been also described. Microcystis aeruginosa, Anabaena flos-aquae, Aphanizomenon flos-aquae, and Cylindrospermopsis raciborskii are the most common species found in Hungarian freshwaters. A sensitization test on albino guinea pigs, and intradermal reactivity, and occular irritation test on albino rabbits were carried ou

Deciphering the key factors determining spatio-temporal heterogeneity of cyanobacterial bloom dynamics in the Nakdong River with consecutive large weirs.

Following the construction of eight large weirs in a 200-km section of the Nakdong River, which is a major water source for the region, harmful cyanobacterial blooms have been occurring annually, causing severe problems with water quality. The present study investigated the community structure of harmful cyanobacteria and identified temporal and spatial patterns in harmful cyanobacterial blooms and their dynamic relationships with physicochemical, hydrological, and meteorological variables in the eight weir sections for 6 years from 2013 to 2018. The dominant harmful cyanobacteria in the eight

Dominance and toxicity without lethality: Exploring biomass, cyanometabolites, and Daphnia responses across Cyanobacterial strains.

Cyanobacteria are cosmopolitan, important components of biomass with a remarkable ability to synthesize a diverse array of bioactive compounds, some of which may have toxic effects on other organisms and ecosystems. In this study, filamentous cyanobacterial strains isolated predominantly from temperate European freshwaters (Aphanizomenon, Chrysosporum, Cuspidothrix, Dolichospermum, Planktothrix, Raphidiopsis), with a subset from Arctic waters (Microcoleus, Phormidesmis), were qualitatively screened for commonly studied cyanometabolites, using immunoassays and chromatographic techniques. Anti-cancer effect of vitexin onto the CFPAC-1 and MIAPaCa-2 pancreatic cancer cells.

Microalgal food supplements are becoming increasingly popular due to their promising biological effects and high nutritional value, evidenced in in vitro, in vivo, and human studies. Some products of this kind have, however, raised controversies concerning their safety. At the same time, not much is known about the frequency of adverse events following the use of microalgal supplements, potential factors that may influence them, and general characteristics and behaviours of the consumer group. The present study aimed to fill this gap and surveyed a group of Polish consumers of microalgal produ

Synergistic interactions between biogenic organic matter and microbial dynamics during simulated senescent cyanobacterial blooms in freshwater mesocosms.

One of the major consequences of a phytoplanktonic bloom is the massive release of autochthonous organic matter (OM) into the water column, stimulating heterotrophic microbial activity and disrupting the trophic web. To better understand the consequences of such biotic stress, the senescent phase of a bloom was simulated under semi-controlled conditions through the addition of cyanobacterial-derived OM from Microcystis aeruginosa and Aphanizomenon gracile (30% enrichment as eq C) into lake water mesocosms. By following both the autochthonous OM and the microbial communities during 28 days, we

The effects of immunostimulatory herbal supplements on autoimmune skin diseases.

The use of herbal supplements that promise to improve immune health has gained popularity among dermatology patients. However, there is little to no evidence that herbal supplements improve dermatologic conditions. Several in vitro and in vivo studies have shown that Spirulina platensis, Aphanizomenon flos-aqua, Chlorella, Echinacea, and alfalfa activate immune cells via certain cytokines and chemokines. Case reports suggest the association of ingesting immunostimulatory herbs and the clinical onset or flares of diseases characterized by an exaggerated immune response such as lupus erythematos

Toxin mixture in cyanobacterial blooms--a critical comparison of reality with current procedures employed in human health risk assessment.

Cyanobacteria are the oldest life forms on earth known to produce a broad spectrum of secondary metabolites. The functions/advantages of most of these secondary metabolites (peptides and alkaloids) are unknown, however, some of them have adverse effects in humans and wildlife, especially when ingested, inhaled or upon dermal exposure. Surprisingly, some of these cyanobacteria are ingested voluntarily. Indeed, for centuries mankind has used cyanobacteria as a protein source, primarily Spirulina species. However, recently also Aphanizomenon flos-aquae are used for the production of so called blu

Environmental triggers of dermatomyositis: a narrative review.

Dermatomyositis (DM) is an autoimmune disease that affects the skin, lungs, and muscle. Although the pathogenesis of DM is not completely understood, several environmental triggers have been linked to DM onset or flare. This article specifically examines the effects of herbal supplements, drugs, infections, ultraviolet (UV) radiation, and environmental pollutants on the onset or exacerbation of DM. Herbal supplements such as Spirulina platensis, Aphanizomenon flos-aquae, Chlorella, Echinacea, and Alfalfa have been implicated and are frequently used in health foods. Medications such as hydroxyu

Activation of autoimmunity following use of immunostimulatory herbal supplements.

BACKGROUND: Evidence for the scientific basis of purported therapeutic effects and adverse effects of herbal supplements continues to grow. Many herbal supplements are touted for their immunostimulatory properties, and both in vitro and in vivo experiments have supported this claim. Although this explains their beneficial effects in preventing or curtailing disease, to our knowledge, no immunostimulatory herbal supplements have been reported to exacerbate disorders of immune system overactivity. OBSERVATIONS: We describe 3 patients whose autoimmune disease onset and/or flares correlated with
