Three acetylcholine receptors in Aplysia neurones.

1. In the pleural ganglion of Aplysia californica, groups of cells were identified that respond differently to an iontophoretic injection of ACh. The anterior neurones are excited by ACh, whereas the medial cells are inhibited. The inhibitory response is biphasic, consisting of a short-latency, rapid component and a longer-latency, slow component. Homologous responses (e.p.s.p.s in the anterior cells and a two-component inhibitory response in the medial cells) are evoked in these same cell groups by stimulation of an identifiable presynaptic neurone.2. The e.p.s.p. and the corresponding ACh po

Electroporation of neurons and growth cones in Aplysia californica.

Specific labeling of individual neurons and neuronal processes is virtually an everyday task for neuroscientists. Many traditional ways for delivery of intracellular dyes have limitations in terms of speed, efficiency and reproducibility. Electroporation is a fast, reliable and efficient method to deliver microscopic amounts of polar and charged molecules into neurons and their compartments such as individual neurites and growth cones. Here, we present a simple and highly effective procedure for intracellular labeling of individual Aplysia neurons both in intact ganglia and in cell culture. Pl

Lipochondria and the light response of Aplysia giant neurons,

The ultrastructure, absorbance, and elemental content of lipochondria present in the cytoplasm of Aplysia giant neurons have been investigated before and after 30-1,200 sec doses of white light at intensities which produce saturated light responses. The effects of exposure to the calcium ionophore A-23187 and to EGTA were also examined. The lipochondria of nonilluminated neurons are membrane-bound, and contain lipids, protein, Na, K, Mg Ca, Si, Cl, Br, P, and a pigment which is probably beta-carotene. The cytoplasm appeared to have little pigment. When neurons were illuminated for 20 min, 60-7

Regulation of statoconia mineralization in Aplysia californica in vitro.

Statoconia are calcium carbonate inclusions in the lumen of the gravity-sensing organ, the statocyst, of Aplysia californica. The aim of the present study was to examine the role of carbonic anhydrase and urease in statoconia mineralization in vitro. The experiments were performed using a previously described culture system (Pedrozo et al., J. Comp. Physiol. (A) 177:415-425). Inhibition of carbonic anhydrase by acetazolamide decreased statoconia production and volume, while inhibition of urease by acetohydroxamic acid reduced total statoconia number, but had no affect on statoconia volume. Orientation of Aplysia californica to distant food sources.

The behavior of the marine mollusc Aplysia was examined under different experimental conditions designed to determine the food searching strategy of the animals. In a small, open field tank with still water, the animals took an average of 42 min to find a piece of seaweed, even though the stimulus was never located more than 30 cm away from the animal. Observations of the animals indicated that their search was not directed, without a clear tendency towards the food, and during the course of a search, they often crawled through most of the area of the tank. The search time, the distance travel

A Cl- conductance activated by hyperpolarization in Aplysia neurones.

Although many voltage-gated cation channels have been described and extensively studied in biological membranes, there are very few examples of voltage-gated anion channels. Chloride conductances activated by depolarization have been observed in skate electroplaque and in frog and chick skeletal muscle. A Cl- conductance activated by hyperpolarization has been suggested both for frog muscle treated with acid (pH 5) solutions, and for crayfish muscle where it could account for the fact that the pronounced inward-going rectification of the I-V curve disappears if the fibres have been soaked in a

An immunohistochemical study of the neuropeptidergic bag cells of Aplysia.

The bag cell neurosecretory system of Aplysia californica synthesizes and releases a neuropeptide, the egg-laying hormone (ELH), which can induce egg-laying behavior. While ELH acts as a neurohormone on distant neuronal and non-neuronal targets, it also can moderate the electrical activity of neurons within the abdominal ganglion. Using antibodies generated against pure ELH, our immunohistochemical study provides direct morphological support for this dual function of ELH by defining the distribution of this neuropeptide within the abdominal ganglion of A. californica. We also find ELH cross-re

Aplysia gigantea toxicosis in 72 dogs in Western Australia.

OBJECTIVES: This study aimed to: (1) confirm a temporal association between exposure to the sea hare Aplysia gigantea and the development of a neurotoxicosis in dogs and (2) further characterise the clinical signs, treatment and outcomes in dogs with this suspected toxicosis. METHODS: The medical records from four veterinary practices within the Geraldton region of Western Australia were searched for dogs that had been exposed to A. gigantea and subsequently presented to a veterinarian during the period of January 2001 to March 2011. Signalment, exposure history, clinical signs, treatment and

Halogenated Metabolites from the Diet of Aplysia dactylomela Rang.

Invertebrates are an important source of structurally-diverse and biologically-active halogenated metabolites. The sea hare Aplysia dactylomela Rang has long been known to possess halogenated metabolites of dietary origin that are used as a self-defense mechanism. The compounds from Aplysia dactylomela Rang are comprised mainly of terpenoids and small percentages of C-15 acetogenins, indoles, macrolides, sterols and alkaloids with potent cytotoxic, anti-microbial and anti-inflammatory properties. For decades the metabolites discovered have been investigated for their medical and pharmaceutical

Barbiturate effects on acetylcholine-activated channels in Aplysia neurons.

The effects of pentobarbital, phenobarbital, amobarbital, and diphenylbarbituric acid were studied by examining the average lifetime and conductance of acetylcholine-activated sodium channels in Aplysia neurons. Although none of the barbiturates tested modified the conductance of single-ion channels, pentobarbital and amobarbital had profound effects on channel lifetime. In the absence of barbiturate, relaxations in response to voltage jumps during steady-state current responses to acetylcholine have a single-exponential time course. In the presence of pentobarbital (75-500 microM), current re

Functional organization and adaptability of a decision-making network in aplysia.

Whereas major insights into the neuronal basis of adaptive behavior have been gained from the study of automatic behaviors, including reflexive and rhythmic motor acts, the neural substrates for goal-directed behaviors in which decision-making about action selection and initiation are crucial, remain poorly understood. However, the mollusk Aplysia is proving to be increasingly relevant to redressing this issue. The functional properties of the central circuits that govern this animal's goal-directed feeding behavior and particularly the neural processes underlying the selection and initiation

Designing the Aplysia punctata Arginine-Depleting Enzyme for Tumor Targeting.

l-Amino acid oxidases (LAAO) deaminate amino acids to α-keto acids and generate hydrogen peroxide, a reactive oxygen species (ROS) with potential value for cancer therapy. We recombinantly expressed the LAAO from Aplysia punctata, called APIT (Cuvier 1803). The resulting wild-type APIT (APITwt) was conjugated to polyethylene glycol (APIT-PEG). Furthermore, an APIT mutant with an affibody targeting the human epidermal growth factor receptor 2 (HER2; zHER2-APIT) was genetically engineered resulting in a binding affinity KD of ∼ 2.2 nM to the HER2 receptor ectodomain. Further, we evaluated if the

Magnetic magic: How stimulation alters feeding patterns in Aplysia californica.

Repetitive transcranial magnetic stimulation (rTMS) is emerging as a groundbreaking treatment for eating disorders, such as anorexia and bulimia nervosa, though its underlying neurological mechanisms remain elusive. To shed light on the effects of rTMS on neural control of feeding behavior, we investigated the feeding responses in the marine mollusk Aplysia californica. Using in vitro preparations, we elicited feeding responses and performed dual nerve recordings to identify motor programs associated with rejection and ingestion. We applied rTMS to the cerebral ganglion, the neural center resp

Multiple mating, sperm storage, and mating preference in Aplysia californica.

Aplysia californica, a simultaneous hermaphrodite, has the ability to store and digest allosperm and frequently mates with multiple partners. Using controlled matings and behavioral observations, we characterized the mating behavior of multiply-mated A. californica. Regardless of their initial mating role, all animals mated a second time. Individuals acting as the initial sperm donor showed no preference for second mating role when paired with a "virgin" partner. However, individuals acting as the initial sperm recipient showed a highly significant preference for acting as the sperm donor in s

Polypeptide secretion from the isolated parietovisceral ganglion of Aplysia californica.

In vitro studies of the secretory behavior of the parietovisceral ganglion in Aplysia californica were performed. The aim of these studies was to investigate the release of polypeptides in response to depolarizing stimuli, and, in particular, to determine if a specific polypeptide known to induce egg laying in the intact animal is secreted into the bathing medium. During continuous perfusion of a ganglion preincubated in leucine-(3)H the application of either high-potassium medium or a burst of electrical stimuli (via the pleurovisceral connective nerve) evoked a marked increase in the amount

Neuron-specific membrane glycoproteins promoting neurite fasciculation in Aplysia californica.

We have generated a library of mouse monoclonal antibodies against membrane proteins of the nervous system of the marine snail Aplysia californica. Two of these antibodies, 4E8 and 3D9, recognize a group of membrane glycoproteins with molecular masses of 100-150 kD. We have called these proteins ap100, from the molecular mass of the most abundant species. Based on Western blots, these proteins appear to be specific for the nervous system. They are enriched in the neuropil of central nervous system ganglia, and are present on the surface of neurites and growth cones of neurons in culture. They

Multiple sensory neuronal correlates of site-specific sensitization in Aplysia.

Noxious stimulation of a restricted site on the skin of Aplysia (training) causes site-specific sensitization of the tail-withdrawal reflex that is associated with several sensory correlates that are evident both 10 min and 2 hr after training. First, extracellularly recorded afferent activity evoked by test stimulation of the trained site increases, indicating peripheral sensory changes. Second, central sensory alterations are manifested by tail sensory neurons within the pleural VC cluster that innervate the trained site and are activated during training. These mechanosensory/nociceptive cel

Trifluoperazine and calcium antagonists accelerate cholinergic desensitization in Aplysia neurons.

The onset of desensitization of excitatory acetylcholine responses in isolated, voltage clamped Aplysia RB neurons to microperfused agonist was accelerated by trifluoperazine and the calcium antagonists SKF-525A and D-600. In the absence of these drugs the time course of desensitization is described as the sum of two exponentials plus a constant. All 3 compounds accelerated the fast exponential component of desensitization, but had opposite effects on the slow component: trifluoperazine and SKF-525A accelerated the slow component, whereas this phase was greatly slowed by D-600. These results p
