Effects of bariatric surgery on apolipoprotein A1, apolipoprotein B, and apolipoprotein B/ apolipoprotein A1 ratio: Findings from a prospective cohort.

BACKGROUND: Bariatric surgery is associated with significant improvements in cardiometabolic health. The apolipoprotein B (ApoB)/apolipoprotein A1 (ApoA1) ratio has been demonstrated to be a strong predictor of cardiovascular disease risk. However, the long-term impact of sleeve gastrectomy (SG) on the ApoB/ApoA1 ratio remains unexplored. METHODS: Observational study including data from a prospective cohort of patients undergoing SG with ApoA1, ApoB, and ApoB/ApoA1 ratio data at baseline, 1 year, and in the long term (3- to 10 years). RESULTS: A total of 376 participants with baseline and 1-

Associations between maternal mid-pregnancy apolipoprotein A-1, apolipoprotein B, apolipoprotein B/apolipoprotein A-1 ratio and preterm birth.

BACKGROUND AND AIMS: Elevated lipid levels during pregnancy have been shown to be related to the risk of preterm birth. Despite the importance of apolipoprotein (Apo) in lipid metabolism and transportation, evidence regarding apolipoprotein levels during pregnancy and preterm birth is still limited. Therefore, we aim to investigate the associations between maternal ApoA-1, ApoB, ApoB/ApoA-1 ratio and preterm birth. MATERIALS AND METH: Data were extracted from the information system of Guangdong Women and Children Hospital. Lipoprotein levels were tested using Beckman Coulter AU5800 in mid-pre

Apolipoproteins A-I and B-markers in coronary risk evaluation.

OBJECTIVES: To investigate if apoB, apoA-I and apoB/apoA-I ratios are independent risk factors for coronary heart disease and to determine their value in relationship with serum lipid fractions in evaluating the risk of coronary events. MATERIALS AND METHODS: We carried out a comparative observational study on 289 subjects divided into two groups: 144 subjects with old myocardial infarction, and 145 subjects without coronary heart disease, but with cardiovascular risk factors. None of the subjects received lipid-lowering drugs in the previous 3 months. RESULTS: The mean values of lipid fract

Apolipoprotein B, the villain in the drama?

Low-density lipoprotein (LDL) is the major atherogenic lipoprotein and the primary target of lipid-lowering therapy for treating ischemic cardiovascular disease. Apolipoprotein B (apoB), an important structural component of LDL, plays a key role in cholesterol transport and removal in vascular wall. On the other hand, under pathological process, apoB interacts with the arterial wall to initiate the cascade of events that leads to atherosclerosis. However, interactions between apoB and vascular wall remain to be determined. Here, we address a pathological role of apoB per se and whole LDL parti

Apolipoprotein E and apolipoprotein B genotypes and risk for spina bifida.

BACKGROUND: Altered cholesterol metabolism and defects in cholesterol biosynthesis may influence abnormal central nervous system (CNS) development. During early stages of embryonic development, high levels of cholesterol are needed by rapidly proliferating cells that utilize cholesterol as a key cell membrane component. Alterations in cholesterol levels are influenced by variations in the apolipoprotein E (apoE) and apolipoprotein B (apoB) genes. The purpose of our study was to explore the possible association between infant genetic variations in the apoE and apoB genes and spina bifida (SB) r

Genetics of apolipoprotein B and apolipoprotein AI and premature coronary artery disease.

Increased low-density lipoprotein (LDL) and decreased high-density lipoprotein cholesterol (HDL-C) predict premature coronary artery disease, as do elevated levels of apolipoprotein B or reduced levels of apolipoprotein AI. Probands were studied of families with common genetic forms of dyslipidaemia to determine if apo B or apo AI define genetic groups and if apo B or apo AI levels relate to premature coronary artery disease risk. Elevated apo B was characteristic of familial hypercholesterolaemia, familial combined hyperlipidaemia (FCHL), and was seen in individuals with elevated Lp(a). Norma

Slower removal of intestinal apolipoprotein B-48 than of apolipoprotein B-100 in severely hypertriglyceridemic subjects.

Apolipoprotein B, the major structural protein of triacylglycerol-rich lipoprotein, occurs in two immunologically distinct forms, termed apolipoproteins B-100 and B-48. In man, the former is associated with triacylglycerol-rich lipoproteins of hepatic origin and the latter with intestinal triacylglycerol-rich lipoproteins. We have studied the rates of removal of the two proteins when 125I-labelled triacylglycerol-rich lipoproteins were reinjected into six severely hypertriglyceridemic subjects showing hyperchylomicronemia, and into two normal subjects. The specific radioactivities of apolipopr

Association between Apolipoproteins AI and B and Ultrasound Indicators of Carotid Atherosclerosis.

BACKGROUND: Apolipoproteins A-I and B (apoA-I and apoB) may be better indicators of the risk of cardiovascular and cerebrovascular diseases than conventional risk factors (RFs). The onset of ischemic stroke (IS) may be preceded by the development of atherosclerotic changes in carotid arteries, which can be detected by ultrasound. Only a certain % of patients with IS have an (underlying) carotid etiology. OBJECTIVE: The aim of our study was to determine the association between ultrasound indicators of carotid atherosclerosis and the presence of apolipoproteins and other biomarkers in patients

Determination of apolipoproteins B-48 and B-100 in triglyceride-rich lipoproteins by analytical SDS-PAGE.

The present work describes a procedure for determining apolipoproteins (apo) B-100 and B-48 in subfractions of triglyceride-rich lipoproteins by analytical sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) with Coomassie staining. The chromogenicity of the two apoB species was found to be almost equal, and independent of lipoprotein particle size. Both proteins were sensitive to overloading of the gel, which resulted in low dye uptake. This was particularly evident for apoB-48. The precision of this analytical SDS-PAGE-based procedure to determine the plasma concentrations o

Apolipoprotein B - An ideal biomarker for atherosclerosis?

This review article describes the pathophysiological mechanisms linking Apolipoprotein B (Apo-B) and atherosclerosis, summarizes the existing evidence on Apo B as a predictor of atherosclerotic cardiovascular disease and recommendations of (inter)national treatment guidelines regarding Apo B in dyslipidemia management. A single Apo B molecule is present in every particle of very low-density lipoprotein, intermediate density lipoprotein, low density lipoprotein, and lipoprotein(a). This unique single Apo B per particle ratio makes plasma Apo B concentration a direct measure of the number of cir

Structural heterogeneity of apolipoprotein B-100.

Cardiovascular disease is a major cause of human morbidity and mortality. Drug strategies for the prevention of the disease are largely centered on the interaction of low-density lipoprotein receptor (LDLR) with the apolipoprotein B-100 (apoB-100) protein on low-density lipoprotein (LDL). Recently, the structure of apoB-100 on LDL was determined in the absence and presence of LDLR, using cryo-electron microscopy. A remarkable structural feature of apoB-100 is the lack of any significant tertiary structure within the C-terminal two-thirds of the protein (>3000 residues). Instead, apoB-100 forms

Familial defective apolipoprotein B-100.

Abnormal interaction between low density lipoprotein receptors (LDLR) and their ligands, apolipoprotein E and B, causes decreased catabolism of lipoproteins which carry these apolipoproteins (VLDL, IDL and/or LDL) and thereby increased plasma concentrations of these. In familial hypercholesterolemia (FH), abnormal interaction is due to mutations in the LDLR gene, and in type III hyperlipidemia due to mutations in the apo E gene. A few mutations in the apolipoprotein B (apo B) gene have been described, of which the apo B-3,500Arg-Gln seems by far the most frequent, that causes defective binding

Knockdown of apolipoprotein B, an atherogenic apolipoprotein, in HepG2 cells by lentivirus-mediated siRNA.

ApoB is an important determinant of atherosclerosis susceptibility and a potential pharmaceutical target for lowering atherogenic lipoproteins. In the present study, we used a lentiviral vector to express short hairpin RNAs for inhibition of apoB production in HepG2 cells. We first demonstrated that lentivirus could efficiently deliver transgene into HepG2 cells by using GFP lentivirus. We then made three lentiviral siApoB constructs, two of which were highly efficient for silencing apoB expression in HepG2 cells. We showed that siApoB lentivirus specifically knocked down apoB but had no effec

Increased level of apolipoprotein B/apolipoprotein A1 ratio as a potential risk for osteonecrosis.

OBJECTIVE: This study was performed to investigate whether a high ratio of apolipoprotein B to apolipoprotein A1 (apo B/apo A1 ratio) is significantly associated with the risk of developing non-traumatic osteonecrosis of the femoral head (ON). METHODS: Fifty consecutive non-traumatic ON cases were compared with 50 age and sex matched controls, using both univariate and stepwise discriminant analyses, regarding the factors of corticosteroid, alcohol, cigarettes, cholesterol, triglyceride, and apo B/apo A1 ratio. To eliminate the possibility that ON or osteoarthritic change itself can increase

A truncated species of apolipoprotein B, B-83, associated with hypobetalipoproteinemia.

Familial hypobetalipoproteinemia, a syndrome associated with low plasma cholesterol levels, can be caused by apoB gene mutations. We identified a healthy 42-year-old man whose total plasma cholesterol level was 80 mg/dl. His plasma very low density lipoprotein (VLDL) contained a unique truncated apoB species, apoB-83, in addition to the normal B apolipoproteins, apoB-100 and apoB-48. Virtually no apoB-83 was detectable in his low density lipoprotein (LDL). From the subject's kindred, we identified nine other hypocholesterolemic subjects whose VLDL contained apoB-83. Apolipoproteins A-I and B as predictors of complications in gallbladder lithiasis surgical patients.

Specific serum proteins have been traditionally used in the assessment of protein-energy malnutrition. Some short half-life serum proteins have been related to the appearance of postoperative complications in surgical patients with low-risk pathologies that, far from undernutrition, showed a trend toward overweight and obesity. Apolipoproteins have been proposed as potential tools to assess protein status and nutritional recovery, so we investigated apolipoproteins A-I and B as new tools with prognostic value to detect postoperative complications. We analyzed the interrelation between apolipop

Familial defective apolipoprotein B-100 in hypercholesterolemic Chinese Canadians: identification of a unique haplotype of the apolipoprotein B-100 allele.

Familial defective apo B-100 (FDB) is an autosomal dominant condition resulting in hypercholesterolemia. It is generally observed in 1-6% of hypercholesterolemic subjects in Caucasian populations studied. There are, thus far, no reports characterizing the frequency and phenotype of FDB in a Chinese population. We report on the frequency of the FDB (Arg(3500)--> Gln) mutation and the associated haplotype among 160 hypercholesterolemic (TC > or = 6.2 mmol/l) Chinese Canadians including 36 subjects with a clinical diagnosis of familial hypercholesterolemia (FH). Screening for the FDB mutation was
