Neuroscience and appetitive behavior research: 25 years.

Neuroscience techniques have made major contributions to the understanding of appetitive behavior. Highlights in six areas are summarized to illustrate progress during the 25 years of the Columbia Appetitive Behavior Seminar: (1) discovery of angiotensin and aldosterone in the control of thirst and salt appetite; (2) electrophysiological decoding of chemoreceptive information in the brain; (3) a new foundation in the hypothalamus built on peptides, such as neuropeptide Y and galanin, interacting with monoamines and steroids in the control of appetite for macronutrients; (4) discovery of numero

Clitoral stimulation modulates appetitive sexual behavior and facilitates reproduction in rats.

In rats, sexual reward, appetitive sexual behaviors and reproduction are modulated by the amount and rate of vaginocervical stimulation. Here the effect of clitoral stimulation (CLS) on proceptivity was assessed. In Exp 1, ovariectomized, hormone-primed Wistar females formed three groups: G1 (1 CLS every second), G2 (1 CLS every 5s) and G3 (no CLS). Precopulatory CLS consisted of 5cycles of 1min of stimulation with the tip of a cotton swab connected to a vibrator device, followed by 1-2min of rest. CLS increased proceptive behavior in G1 compared to G2, but not compared to G3. In Exp 2, gonada

The early presence of food-oriented appetitive behavior in developing rats.

The appetitive behavior of 3- to 6-day-old rat pups was studied by testing their ability to direct their ingestive behavior to a restricted food source. We found that, from 3 days of age, pups were able to feed efficiently from such a source. More specifically, pups that were deprived of nutrition but not of maternal care as well as pups that were dehydrated ingested significantly more than nondeprived animals, and did so whether liquid diet was spread over the entire floor surface beneath them or restricted to a fraction of the floor surface. However, pups that had been nutritionally and mate

Altered Appetitive Conditioning and Neural Connectivity in Subjects With Compulsive Sexual Behavior.

INTRODUCTION: There has been growing interest in a better understanding of the etiology of compulsive sexual behavior (CSB). It is assumed that facilitated appetitive conditioning might be an important mechanism for the development and maintenance of CSB, but no study thus far has investigated these processes. AIM: To explore group differences in neural activity associated with appetitive conditioning and connectivity in subjects with CSB and a healthy control group. METHODS: Two groups (20 subjects with CSB and 20 controls) were exposed to an appetitive conditioning paradigm during a functi

Furosemide, sodium appetite, and ingestive behavior.

Sodium appetite is often produced experimentally by using the diuretic furosemide (Furo) to induce a rapid loss of urinary sodium. The present experiments were designed to investigate the dose-dependent relationship between renal and behavioral responses to Furo. We compared the effects of five different Furo doses (0.5, 1, 2, 6, and 10 mg) on 3% NaCl intake, water intake, Na(+)-free chow intake, urine quantity, electrolyte balance, and weight gain in rats. The Na(+) loss produced by Furo injection was dose dependent from 0.5 to 10 mg and did not change across repeated depletions. There was on

Neurokinin type-3 receptor stimulation impairs ethanol-associated appetitive behavior in Wistar rats.

OBJECTIVES: Stimulating central neurokinin type-3 (NK-3) receptors decreases ethanol intake in rats. Although paraventricular nucleus of the hypothalamus (PVN) has a high density of NK-3 receptors, their influence on ethanol reinforcement has not been examined. This study's purpose was to assess the effects of intra-PVN infusion of senktide, a NK-3 receptor agonist, on ethanol self-administration. In a follow-up study, senktide's effects on ethanol self-administration after intracerebroventricular (ICV) infusion were examined. METHODS: Male Wistar rats were trained to self-administer 10% etha

Chronic social stress in puberty alters appetitive male sexual behavior and neural metabolic activity.

Repeated social subjugation in early puberty lowers testosterone levels. We used hamsters to investigate the effects of social subjugation on male sexual behavior and metabolic activity within neural systems controlling social and motivational behaviors. Subjugated animals were exposed daily to aggressive adult males in early puberty for postnatal days 28 to 42, while control animals were placed in empty clean cages. On postnatal day 45, they were tested for male sexual behavior in the presence of receptive female. Alternatively, they were tested for mate choice after placement at the base of

Effects of dopamine agonists on appetitive and consummatory male sexual behavior in Japanese quail.

The effects of pharmacological manipulations of dopaminergic transmission on appetitive and consummatory aspects of male sexual behavior were investigated in castrated male Japanese quail treated with exogenous testosterone. Appetitive male sexual behavior was assessed by measuring a learned social proximity response and consummatory behavior was assessed by measuring copulatory behavior per se. The nonselective dopamine receptor agonist, apomorphine, inhibited in a dose-dependent manner both components of male sexual behavior. Two indirect dopamine agonists were also tested. Nomifensine, a do

Prenatal imprinting of postnatal specific appetites and feeding behavior.

Epigenetic influences on the fetus's genotype have been shown to occur during intrauterine life. Experimentally imposed extracellular dehydration in pregnant rats (a model for human hyponatremia caused by gravidic vomiting) brings about a dramatic enhancement of salt appetite not only in the dam, but also in offspring when they reach adulthood. This phenomenon has been verified in human newborn infants and adults whose mothers experienced nausea and/or vomiting during pregnancy. Alcohol consumption during pregnancy enhances its palatability for the offspring. Ingestion of olfactory test substa

The role of the hypothalamic melanocortin system in behavioral appetitive processes.

Much evidence suggests that the hypothalamic melanocortin (MC) system plays an important role in the control of food intake. However, investigations of the potential behavioral mechanisms have been limited to measures of aversion. The purpose of the present experiment was to assess whether other behavioral consequences of administration of MC peptides were similar to those produced by 0- or 24-h food deprivation, respectively. Rats were first trained while food deprived that a tone predicted the delivery of peanut oil. They then received exposure to oil under food deprivation, satiation, intra

Dissociating the incubation of appetitive and consummatory behavior in a model of oral cocaine self-administration.

Cocaine use disorder remains a persistent public health dilemma that currently lacks effective treatment strategies. One key impediment to successful treatment outcomes is increased drug craving that occurs over the course of abstinence and subsequent relapse to drug use. This phenomenon, known as the incubation of drug craving, has been modeled extensively in rodent models of intravenous drug self-administration. As commonly implemented, the design of intravenous self-administration preclinical studies precludes disentangling appetitive and consummatory behaviors as drug seeking (appetitive)

Appetitive and Aversive Motivation in Dysregulated Behaviors: A Meta-Analysis.

Appetitive and aversive motivation are prominent in theories of dysregulated behaviors. The authors conducted a meta-analysis of the association between individual differences in appetitive and aversive motivation and several dysregulated behaviors (i.e., alcohol use, marijuana use, tobacco use, binge eating, aggression, gambling, and nonsuicidal self-injury). Alcohol use (r = .17, k = 141), marijuana use (r = .13, k = 23), aggression (r = .22, k = 52), and gambling (r = .08, k = 55) were all significantly positively related to appetitive motivation. Binge eating (r = .28, k = 34) and self-inj

Appetitive maternal behavior in mice is resistant to perinatal chronic variable stress and perinatal chronic corticosterone treatment.

Perinatal stress is a risk factor for emotional and cognitive disturbances in both caregivers and their infants. Here, we compared the impact of two stressors, perinatal chronic variable stress (CVS) and perinatal chronic corticosterone treatment (C-CORT), on maternal exploratory behavior, maternal caregiving, and hypothalamic neural activity. We stressed mouse dams with each paradigm for 13 days starting at gestational day 11 (GD11). We examined open field exploration behavior on day 12 of stress (or post-partum day 3, PP3) and pup-directed behavior on day 13 of stress (PP4). We collected bra

Electrophysiological responses to appetitive and consummatory behavior in the rostral nucleus tractus solitarius in awake, unrestrained rats.

As the intermediate nucleus in the brainstem receiving information from the tongue and transmitting information upstream, the rostral portion of the nucleus tractus solitarius (rNTS) is most often described as a "taste relay". Although recent evidence implicates the NTS in a broad neural circuit involved in regulating ingestion, there is little information about how cells in this structure respond when an animal is eating solid food. Here, single cells in the rNTS were recorded in awake, unrestrained rats as they explored and ate solid foods (Eating paradigm) chosen to correspond to the basic

Infusions of ascorbic acid into the medial preoptic area facilitate appetitive sexual behavior in the female rat.

Ascorbic acid (AA), also known as Vitamin C, enhances dopamine (DA) transmission in mesolimbic and nigrostriatal terminals and augments DA-mediated behaviors. It is not yet known whether AA has a similar influence in other DA terminals, in particular terminals of the incertohypothalamic system that modulate the function of the medial preoptic area (mPOA). In female rats, DA in the mPOA plays a critical role in the generation of appetitive sexual responses, notably solicitations, hops, and darts, and we have shown previously that the role of DA in this region on female sexual behavior changes d

Lateral hypothalamic LEPR neurons drive appetitive but not consummatory behaviors.

Assigning behavioral roles to genetically defined neurons within the lateral hypothalamus (LH) is an ongoing challenge. We demonstrate that a subpopulation of LH GABAergic neurons expressing leptin receptors (LHLEPR) specifically drives appetitive behaviors in mice. Ablation of LH GABAergic neurons (LHVGAT) decreases weight gain and food intake, whereas LHLEPR ablation does not. Appetitive learning in a Pavlovian conditioning paradigm is delayed in LHVGAT-ablated mice but prevented entirely in LHLEPR-ablated mice. Both LHVGAT and LHLEPR neurons bidirectionally modulate reward-related behaviors

An empirical examination of appetite hormones and cognitive and behavioral bulimic symptomatology.

PURPOSE: Existing literature has demonstrated that appetite hormones are frequently dysregulated in individuals with bulimic-spectrum eating disorders (BN-EDs). Although dysregulations in appetite hormones may maintain BN-EDs, very limited research has examined the association between dysregulated appetite hormones and cognitive and behavioral bulimic symptoms. We hypothesized that greater frequency of behavioral symptoms and severity of cognitive symptoms of BN-EDs would correlate with greater dysregulation in appetite hormones. METHODS: The association between ghrelin, cortisol, leptin, GLP
