Salicylic acid (SA) is a plant hormone that is critical for resistance to pathogens1-3. The NPR proteins have previously been identified as SA receptors4-10, although how they perceive SA and coordinate hormonal signalling remain unknown. Here we report the mapping of the SA-binding core of Arabidopsis thaliana NPR4 and its ligand-bound crystal structure. The SA-binding core domain of NPR4 refolded with SA adopts an α-helical fold that completely buries SA in its hydrophobic core. The lack of a ligand-entry pathway suggests that SA binding involves a major conformational remodelling of the SA-

Regulation of protein function through oxidative modification has emerged as an important molecular mechanism modulating various biological processes. Here, we report a proteomic study of redox-sensitive proteins in Arabidopsis cells subjected to H(2)O(2) treatment. Four gel-based approaches were employed, leading to the identification of four partially overlapping sets of proteins whose thiols underwent oxidative modification in the H(2)O(2)-treated cells. Using a method based on differential labeling of thiols followed by immunoprecipitation and Western blotting, five of the six selected put

The plasma membrane (PM) is the primary interface between plant cells and their environment, and its resident proteins mediate key processes such as extracellular signal perception and downstream cellular reprogramming. Yet, PM proteins are typically underrepresented in total protein extracts, and existing enrichment strategies are often laborious and require extensive optimization. Here, a simple and robust workflow is described for enriching PM proteins from Arabidopsis thaliana seedlings using total microsomal membranes obtained by differential centrifugation as starting material. Sequentia

The lipodystrophy protein SEIPIN is important for lipid droplet (LD) biogenesis in human and yeast cells. In contrast with the single SEIPIN genes in humans and yeast, there are three SEIPIN homologs in Arabidopsis thaliana, designated SEIPIN1, SEIPIN2, and SEIPIN3. Essentially nothing is known about the functions of SEIPIN homologs in plants. Here, a yeast (Saccharomyces cerevisiae) SEIPIN deletion mutant strain and a plant (Nicotiana benthamiana) transient expression system were used to test the ability of Arabidopsis SEIPINs to influence LD morphology. In both species, expression of SEIPIN1

The common transition from out-crossing to self-fertilization in plants decreases effective population size. This is expected to result in a reduced efficacy of natural selection and in increased rates of protein evolution in selfing plants compared with their outcrossing congeners. Prior analyses, based on a very limited number of genes, detected no differences between the rates of protein evolution in the selfing Arabidopsis thaliana compared with the out-crosser Arabidopsis lyrata. Here, we reevaluate this trend using the complete genomes of A. thaliana, A. lyrata, Arabidopsis halleri, and

F-BOX STRESS INDUCED (FBS) proteins are substrate adaptors in SCF-type E3 ubiquitin ligases that regulate plant development and stress responses. Arabidopsis thaliana FBS1 (AT1G61340) interacts with multiple 14-3-3 proteins, but it is unknown whether other FBS family members share these interactions. Bimolecular fluorescence complementation (BiFC) assays in Nicotiana benthamiana showed that all four FBS proteins interact with multiple 14-3-3 proteins in vivo. However, FBS1 interactions were restricted to the nucleus, whereas FBS2 - FBS4 (AT4G21510, AT4G05010, AT4G35930) interactions were detec

Here we report proteins identified after conducting Tandem Affinity Purification (TAP) of the TOPLESS (TPL) corepressor from Arabidopsis. We generated transgenic plants harboring TPL fused to the GS-TAG, "Boosting tandem affinity purification of plant protein complexes" (Van Leene et al., 2008) [1]. Four independent biological replicates of a selected TPL-GS-TAG line were grown simultaneously, crosslinked with formaldehyde, and proteins were isolated from whole plant tissue via TAP. Purified proteins were treated with trypsin, and the peptides were analyzed via mass spectrometry. Datasets are

WRKY transcription factors are encoded by a large gene superfamily with a broad range of roles in plants. Recently, several groups have reported that proteins containing a short VQ (FxxxVQxLTG) motif interact with WRKY proteins. We have recently discovered that two VQ proteins from Arabidopsis (Arabidopsis thaliana), SIGMA FACTOR-INTERACTING PROTEIN1 and SIGMA FACTOR-INTERACTING PROTEIN2, act as coactivators of WRKY33 in plant defense by specifically recognizing the C-terminal WRKY domain and stimulating the DNA-binding activity of WRKY33. In this study, we have analyzed the entire family of 3

Accumulation of reactive oxygen species (ROS) is one of the early defense responses against pathogen infection in plants. The mechanism about the initial and direct regulation of the defense signaling pathway by ROS remains elusive. Perturbation of cellular redox homeostasis by ROS is believed to alter functions of redox-sensitive proteins through their oxidative modifications. Here we report an OxiTRAQ-based proteomic study in identifying proteins whose cysteines underwent oxidative modifications in Arabidopsis cells during the early response to salicylate or flg22, two defense pathway elicit

Optimizing the conditions for the overexpression and purification of membrane proteins for functional and structural studies is a complex and tedious process, which demands considerable time and effort. Membranes are made up of phospholipid bilayer embedded with proteins, cholesterol, and carbohydrates. While cholesterol regulates the fluidity, proteins and carbohydrates provide functionality to the membranes. Membrane proteins are responsible for transport, signaling, adhesion, and catalysis and have membrane spanning domains that tether them to the lipid bilayer. Due to the hydrophobic natur

Microtubule organization during cell division requires coordinated regulation by microtubule-associated proteins and protein kinase signaling pathways. In animals, TPX2 (Targeting Protein for Xklp2) regulates spindle assembly by binding to and activating Aurora A kinase. In Arabidopsis, individual TPX2-like (TPXL) proteins have been implicated in Aurora-associated functions, but whether the TPXL family members share common microtubule-associated properties and how TPXL-Aurora interactions differ across the family remain incompletely understood. Here, we investigated the structural and cellular

Plasmodesmata (PD) are plasma membrane-lined cytoplasmic nanochannels that mediate cell-to-cell communication across the cell wall. A range of proteins are embedded in the PD plasma membrane and endoplasmic reticulum (ER), and function in regulating PD-mediated symplasmic trafficking. However, knowledge of the nature and function of the ER-embedded proteins in the intercellular movement of non-cell-autonomous proteins is limited. Here, we report the functional characterization of two ER luminal proteins, AtBiP1/2, and two ER integral membrane proteins, AtERdj2A/B, which are located within the

The spider mite Tetranychus urticae is an economically important agricultural pest, which feeds on a broad spectrum of plant species. In an RNAseq experiment performed in our laboratory, 4 of the 15 members of the RIN4-like/NOI family of Arabidopsis thaliana were significantly overexpressed after T. urticae infestation. Two of them (NOI3 and NOI5) are shorter and harbour one NOI domain, which characterises this family, and the other two (NOI10 and NOI11) have two-NOI domains. The only member of this family characterized is RIN4, a two-NOI intrinsically disordered protein anchored to the plasma

Dynamin-related proteins (DRPs) are large GTPases involved in a wide range of cellular membrane remodeling processes. In Arabidopsis thaliana, two paralogous land plant-specific type DRPs, DRP2A and DRP2B, are thought to participate in the regulation of post-Golgi trafficking. Here, we examined their molecular properties and functional relationships. qRT-PCR and GUS assays showed that DRP2A and DRP2B were expressed ubiquitously, although their expressions were strongest around root apical meristems and vascular bundles. Yeast two-hybrid, bi-molecular fluorescent complementation, and co-immunop

The major latex proteins (MLP) are a protein family first identified in the latex of opium poppy. They are found only in plants and have 24 identified members in Arabidopsis alone as well as in other plants such as peach, strawberry, melon, cucumber, and soybean. While the function of the MLPs is unknown, they have been associated with fruit and flower development and in pathogen defense responses. Based on modest sequence similarity, they have been characterized as members of the Bet v 1 protein superfamily; however, no structures have yet been reported. As part of an ongoing structural genom

Cadmium (Cd) toxicity requires plants to coordinate growth restraint with cellular protection, but the upstream regulatory mechanism remains unclear. Here, we identify the gibberellin (GA)-DELLA module as a positive regulator of Cd tolerance in Arabidopsis thaliana. Cd exposure reduced endogenous GA levels, shifted GA metabolic gene expression toward a low-GA state, and increased DELLA protein abundance; exogenous GA₃ attenuated this increase. Genetic and pharmacological analyses showed that elevated DELLA activity improved growth under Cd stress, whereas loss of DELLA function increased sensi

Arabidopsis thaliana possesses hitherto unknown Occludin domain containing ELL ortholog identified as CARP9. The AtCARP9 protein similar to ELL orthologs across species, interacts with both the Arabidopsis EAF orthologs. Transcription is one of the early steps during which the expression of most protein-coding genes is controlled in cells. Accumulating evidence suggests that elongation of transcription is another dynamic step, which is regulated by 'transcription elongation factors'. ELL (eleven nineteen lysine rich leukemia) belongs to a family of conserved transcription elongation factors, w

The phloem is a specialized tissue that facilitates systemic transport of carbohydrates and signal molecules, making it a common target for viruses, phloem-feeding insects, and cyst nematodes. In Arabidopsis, the stationary phloem-associated SIEVE ELEMENT-LINING CHAPERONE1 (SLI1) and RESTRICTED TEV MOVEMENT (RTM) proteins restrict insect phloem-feeding and potyviruses systemic transport, respectively. However, their broader roles in plant-attacker interactions remain largely unexplored. We investigated the roles of SLI1, RTM1, RTM2, and RTM3 in tobacco etch virus (TEV) infection, as well as in

Protein S-acylation, predominately in the form of palmitoylation, is a reversible lipid post-translational modification on cysteines that plays important roles in protein localization, trafficking, activity, and complex assembly. The functions and regulatory mechanisms of S-acylation have been extensively studied in mammals owing to remarkable development of high-resolution proteomics and the discovery of the S-acylation-related enzymes. However, the advancement of S-acylation studies in plants lags behind that in mammals, mainly due to the lack of knowledge about proteins responsible for this

Protein carbonylation is an irreversible form of post-translational modification triggered by reactive oxygen species in animal and plant cells. It occurs either through the metal-catalyzed oxidation of Lys, Arg, Pro, and Thr side chains or the addition of α, β-unsaturated aldehydes and ketones to the side chains of Cys, Lys, and His. Recent genetic studies concerning plants pointed to an implication of protein carbonylation in gene regulation through phytohormones. However, for protein carbonylation to stand out as a signal transduction mechanism, such as phosphorylation and ubiquitination, i
