Embryotoxic effects of nonylphenol and octylphenol in sea urchin Arbacia lixula.

Nonylphenol (NP) and octylphenol (OP), both of which are biodegradation products of alkylphenols, are widely used in industrial applications and in some domestic products. These chemicals are found widely in surface water and aquatic sediments. We have carried out a comparative embryotoxicity analysis of the effects of increasing concentrations of NP (seven concentrations ranging from 0.937 to 18.74 microg/l) and OP (six concentrations ranging from 5 to 160 microg/l) on embryos of the sea urchin Arbacia lixula. The indicators evaluated were larval malformations, developmental arrest and embryo

Discovery of bioactive peptides from Arbacia lixula as dipeptidyl peptidase-IV inhibitors: An in silico approach.

Dipeptidyl peptidase-IV (DPP-IV) inhibition is a validated therapeutic mechanism for type 2 diabetes mellitus. Marine-derived bioactive peptides offer a diverse array of potential inhibitors but often exhibit pharmacokinetic limitations that impede clinical translation. This study aimed to identify and evaluate candidate peptides from Arbacia lixula as potential DPP-IV inhibitors using an integrated in silico approach. A total of twenty peptides were evaluated in silico for absorption, distribution, metabolism, excretion, and toxicity (ADMET) profiles using SwissADME, pkCSM, and ProTox. Molecu

Embryotoxicity of polystyrene microplastics, alone and conjugated with bisphenol A, in the black sea urchin Arbacia lixula: A multi-biomarker approach.

The widespread occurrence of microplastics (MPs) and endocrine-disrupting compounds as bisphenol A (BPA) poses risks to aquatic life. This study evaluates the effects of polystyrene (PS) MPs and BPA, individually and conjugated, on embryonic development of black sea urchin Arbacia lixula, an ecologically relevant bioindicator. Embryos were exposed to 1 and 5 µm PS MPs (10 µg/mL), BPA (5 and 25 µM), and BPA-sorbed PS MPs. A multi-endpoint approach was employed, integrating embryotoxicity assays, morphological evaluation, metabolomics, and enzymatic biomarkers of oxidative stress and neurotoxici

Ca2+ spikes in the flagellum control chemotactic behavior of sperm.

The events that occur during chemotaxis of sperm are only partly known. As an essential step toward determining the underlying mechanism, we have recorded Ca2+ dynamics in swimming sperm of marine invertebrates. Stimulation of the sea urchin Arbacia punctulata by the chemoattractant or by intracellular cGMP evokes Ca2+ spikes in the flagellum. A Ca2+ spike elicits a turn in the trajectory followed by a period of straight swimming ('turn-and-run'). The train of Ca2+ spikes gives rise to repetitive loop-like movements. When sperm swim in a concentration gradient of the attractant, the Ca2+ spike

Heatwave conditions increase the toxicity of phthalates in marine organisms.

Climate change- driven marine heatwaves are major risk for marine organisms already facing other anthropogenic hazards, such as chemical contamination in coastal areas. In this study we analyzed the impacts of marine heatwaves and phthalic acid esters (PAEs) pollution as single and combined stressors on development of the sea urchin Arbacia lixula. We tested whether the temperature suggested as optimal for development (24 °C) of this thermophilus species would enhance tolerance to PAEs pollution compared to that showed under ambient temperature (18 °C). Embryo-larval bioassays were conducted i

Optimizing preservation protocols to extract high-quality RNA from different tissues of echinoderms for next-generation sequencing.

Transcriptomic information provides fundamental insights into biological processes. Extraction of quality RNA is a challenging step, and preservation and extraction protocols need to be adjusted in many cases. Our objectives were to optimize preservation protocols for isolation of high-quality RNA from diverse echinoderm tissues and to compare the utility of parameters as absorbance ratios and RIN values to assess RNA quality. Three different tissues (gonad, oesophagus and coelomocytes) were selected from the sea urchin Arbacia lixula. Solid tissues were flash-frozen and stored at -80 °C until

Toxicity of oil spill response agents and crude oils to five aquatic test species.

The majority of aquatic toxicity data for petroleum products has been limited to a few intensively studied crude oils and Corexit chemical dispersants, and acute toxicity testing in two standard estuarine test species: mysids (Americamysis bahia) and inland silversides (Menidia beryllina). This study compared the toxicity of two chemical dispersants commonly stock piled for spill response (Corexit EC9500A®, Finasol®OSR 52), three less studied agents (Accell Clean®DWD dispersant; CytoSol® surface washing agent; Gelco200® solidifier), and three crude oils differing in hydrocarbon composition (Do

Evaluation of effectiveness of EDTA and sodium thiosulfate in removing metal toxicity toward sea urchin embryo-larval applying the TIE.

Since the development of the TIE (Toxicity Identification and Evaluation) in 1988 it has been assumed that the capacity of EDTA and sodium thiosulfate to complex some metals, and thus remove their toxicity, can be applied to both freshwater and seawater ecotoxicological tests and the results subsequently interpreted. However, it is now known that there is a wide variability in the extent of this complexation. In this context, the removal of toxicity caused by the presence of Hg(2+), Cd(2+), Cu(2+), Cr(6+), Zn(2+), Ni(2+), Pb(2+), Ag(1+) and Se(2+), through metal complexation by EDTA and sodium

Unravelling Hidden Trophic Interactions Among Sea Urchin Juveniles and Macroinvertebrates by DNA Amplification.

Rocky reefs may shift between two distinct stable states: productive algal forests, characterised by high abundance and biodiversity of macrofauna, and impoverished barrens, dominated by overgrazing sea urchins. Barren states may persist despite the recovery of adult sea urchin predators, suggesting additional stabilising mechanisms. Sea urchin settlers equally colonise forests and barrens in large numbers, but in forests only a few of them reach adult size, suggesting that post-settlement predation might play a crucial role in determining sea urchin population density. Visual assessment of pr

Functional mimicry of sea urchin biomineralization proteins with CaCO3-binding peptides selected by phage display.

The intricate process of biomineralization, e.g. in sea urchins, involves the precise interplay of highly regulated mineralization proteins and the spatiotemporal coordination achieved through compartmentalization. However, the investigation of biomineralization effector molecules, e.g. proteins, is challenging, due to their very low abundance. Therefore, we investigate the functional mimicry in the bioinspired precipitation of calcium carbonate (CaCO3) with artificial peptides selected from a peptide library by phage display based on peptide-binding to calcite and aragonite, respectively. The

A simplified high-pressure liquid chromatography method for determining lipophilicity for structure-activity relationships.

A highly deactivated octadecyl-bonded silica column and a mobile phase consisting of an water-methanol mixture in the range of blood pH and ionic strength are used to correlate log kappa' with biological activity for a series of sulfonamides and barbiturates. The results were compared to literature methods by using retention volume (VR) and retention indexes (I). For the nine sulfonamides tested, log VR and log kappa' were used with and without correction for ionization. For each biological end point (protein binding and minimum inhibitor concentration against Eschericia coli from two sources)

Ecotoxicity of oilfield production chemicals and their mitigation by activated carbon adsorption and UV photolysis.

Produced water (PW) represents the largest wastewater stream in offshore oil and gas production, often containing chemical additives that cause significant ecological risks upon discharge. This study evaluated the acute and chronic toxicity of seven PW additives at operational dosages using Artemia sp. (acute) and embryos of the sea urchins Echinometra lucunter and Arbacia lixula (chronic). Furthermore, the effectiveness of activated carbon (AC) adsorption and UV photolysis in mitigating toxicity was investigated. The biocide (2,2-dibromo-3-nitrilopropionamide) was identified as the most hazar

Reproductive toxicity assessment of cellulose nanofibers, citric acid, and branched polyethylenimine in sea urchins: Eco-design of nanostructured cellulose sponge framework (Part B).

In the framework of a safe-by-design approach, we previously assessed the eco-safety of nanostructured cellulose sponge (CNS) leachate on sea urchin reproduction. It impaired gamete quality, gamete fertilization competence, and embryo development possibly due to the leaching of chemical additives used during the CNS synthesis process. To extend this observation and identify the component(s) that contribute to CNS ecotoxicity, in the present study, we individually screened the cytotoxic effects on sea urchin Arbacia lixula and Paracentrotus lividus gametes and embryos of the three main constitu

A novel cross-species inhibitor to study the function of CatSper Ca2+ channels in sperm.

BACKGROUND AND PURPOSE: Sperm from many species share the sperm-specific Ca2+ channel CatSper that controls the intracellular Ca2+ concentration and, thereby, the swimming behaviour. A growing body of evidence suggests that the mechanisms controlling the activity of CatSper and its role during fertilization differ among species. A lack of suitable pharmacological tools has hampered the elucidation of the function of CatSper. Known inhibitors of CatSper exhibit considerable side effects and also inhibit Slo3, the principal K+ channel of mammalian sperm. The compound RU1968 was reported to suppr

Identification and partial characterization of the receptor for speract.

Here, we report the first identification of a sperm receptor for an egg-associated peptide (speract). Gly-Gly-Gly-Gly-Tyr-Asp-Leu-Asn-Gly-Gly-Gly-Val-Gly (GGG[Y2]-speract), was chemically synthesized and shown to possess equivalent respiration-stimulating activity and the same receptor binding characteristics as speract (Gly-Phe-Asp-Leu-Asn-Gly-Gly-Gly-Val-Gly). The incubation of Strongylocentrotus purpuratus intact sperm cells with radioiodinated GGG[Y2]-speract and the chemical cross-linking reagent, disuccinimidyl suberate, resulted in the appearance of a single radiolabeled band with an es
