Arthropod venoms and cancer.

Many active principles produced by animals, plants and microorganisms have been employed in the development of new drugs to treat diseases such as cancer. Among the animals that produce pharmacologically active molecules capable of interfering in human cellular physiology, the highlights are venomous arthropods, such as scorpions, bees, wasps, spiders, ants and caterpillars. The substances found in the venom of these animals present great potential as anti-tumor agents. In this review, we present the main results of years of research involving the active compounds of arthropods venoms that hav

Arthropod venoms: a vast arsenal of insecticidal neuropeptides.

Arthropods are the most diverse animal group on the planet, and occupy almost all ecological niches. Venomous arthropods are a rich source of bioactive compounds evolved for prey capture and defense against predators and/or microorganisms. These highly potent chemical arsenals represent an available source for new insecticidal compounds as they act selectively on their molecular targets. These toxins affect the invertebrate nervous system and, until the moment, several insecticidal compounds belonging to the class of peptides or polyamine-like compounds have been purified and characterized fro

Cytotoxic effects of arthropod venoms on various cultured cells.

The action of arthropod venoms is important to predators in search of prey and to humans as incidental victims or as a source for pharmacologically active compounds. Venoms from 30 arthropods (including 26 spider species) were assessed for cytotoxicity using cultured cells from one insect (Sf9) and three mammalian (murine neuroblastoma and macrophages and human osteosarcoma) sources. The most cytotoxic venoms to the four cell lines were from predatory jumping spiders (Salticidae, Phidippus sp.) and a centipede (Scolopenra sp.), with concentrations for 50% response of 1-8 microg venom per ml. Antimicrobial peptides from arthropod venoms exhibit activity against Sporothrix species.

BACKGROUND: Sporotrichosis is a fungal infection caused by species of the Sporothrix schenckii complex. Antifungal treatment with itraconazole and amphotericin B is limited by increasing resistance, adverse effects, and prolonged treatment courses, highlighting the need for novel antifungal strategies. METHODS: The antifungal activity of seven antimicrobial peptides derived from arthropod toxins against Sporothrix species was evaluated: six synthetic peptides derived from the spider Lycosa erythrognatha toxin and one peptide isolated from apitoxin (melittin). Minimum inhibitory concentrations

Structural diversity of arthropod venom toxins.

Arthropods are a diverse and ancient group of invertebrate animals, which constitute approximately 75-85% of all known species on earth. Many arthropod species, such as spiders, scorpions and even some crustaceans, contain venoms that can be very complex, representing natural combinatorial libraries of bioactive compounds. Characterization of the compounds in these libraries has helped the development of tools for pharmacology, and the discovery of lead molecules for therapeutic treatments. A critical aspect in the characterization of venom compounds is determination of three-dimensional struc

Structure and experimental uses of arthropod venom proteins.

In summary, the initial studies conducted thus far into the components of venoms of parasitic wasps and other arthropods have already yielded a number of interesting properties of the proteins therein. These properties have already offered the possibilities of additional principles operating in the evolution of venoms. That so many unexpected rewards have already surfaced with the relatively little experimental digging conducted thus far generates great anticipation that indeed there remains a pharmacological gold mine awaiting to be discovered in components of the other insect venoms as yet u

Arthropod venom citrate inhibits phospholipase A2.

Citrate has been identified as a major component of honey bee (Apis mellifera) venom by gas liquid chromatography-mass spectrometry. A citrate concentration of 9% was found for dried bee venom by a coupled enzyme assay, aconitase-isocitric dehydrogenase. A liquid honey bee venom would contain 140 mM citrate concentration (if the solids content were 30%). Bee venom phospholipase was inhibited at a 43% level with a citrate concentration of 20 mM and calcium ion at 3 mM with the enzyme assay. Citrate was also found in the venoms of bumble bee, Bombus fervidus, 7%; yellow jacket, Vespula maculifro

Snake and arthropod venoms: Search for inflammatory activity in human cells involved in joint diseases.

Most anti-inflammatory drugs currently adopted to treat chronic inflammatory joint diseases can alleviate symptoms but they do not lead to remission. Therefore, new and more efficient drugs are needed to block the course of joint inflammatory diseases. Animal venoms, rich in bioactive compounds, can contribute as valuable tools in this field of research. In this study, we first demonstrate the direct action of venoms on cells that constitute the articular joints. We established a platform consisting of cell-based assays to evaluate the release of cytokines (IL-6, IL-8, TNFα, IL-1β, and IL-10)

A microinjection technique using Drosophila melanogaster for bioassay-guided isolation of neurotoxins in arthropod venoms.

Modern analytical techniques permit isolation and structural determination of neurotoxins at the picomole level. However, bioassay-guided fractionation of the sample often relies on simple injection assays using insects, vertebrates or crustaceans of a fairly large size, thus consuming quite a large amount of the samples being investigated. In order to investigate samples of very small size, we have devised an insect microinjection method using glass micropipettes and Drosophila melanogaster adults as test insects. The validity of the method was tested with a series of six buthoid scorpion ven

Tropical dermatology: Venomous arthropods and human skin: Part I. Insecta.

Although many tropical insects carry infectious diseases, cutaneous injury can occur by other mechanisms, for example erucism (envenomation by caterpillars) or lepidopterism (dermatitis from moths). Pararama is a unique form of erucism seen in workers in contact with rubber trees in the Amazon, and it is caused by Premolis larvae, resulting in progressive periarticular fibrosis, ankylosis, and the loss of articulation. Ants and aquatic insects of the Belostomatidae family can cause painful bites and stings. Anaphylactic shock and death can result from the venom of bees and wasps. Beetles can c

Arthropod venom peptides: Pioneering nanotechnology in cancer treatment and drug delivery.

Cancer is the second leading cause of death globally, claiming >10 million lives in 2020 and a projected increase to 13 million by 2030. Traditional treatments such as chemotherapy, radiation, and surgery can be effective but often lead to systemic toxicity and drug conflict due to their lack of selectivity. Nanotechnology is a promising alternative that targets cancer cells and reduces harm to healthy tissues. We aimed to conduct a review to present an inclusive examination of nanotechnology, macromolecules, and related methodologies, focusing on therapeutic strategies utilizing arthropod ven

Weaponization of a Hormone: Convergent Recruitment of Hyperglycemic Hormone into the Venom of Arthropod Predators.

Arthropod venoms consist primarily of peptide toxins that are injected into their prey with devastating consequences. Venom proteins are thought to be recruited from endogenous body proteins and mutated to yield neofunctionalized toxins with remarkable affinity for specific subtypes of ion channels and receptors. However, the evolutionary history of venom peptides remains poorly understood. Here we show that a neuropeptide hormone has been convergently recruited into the venom of spiders and centipedes and evolved into a highly stable toxin through divergent modification of the ancestral gene.

Mass spectrometry strategies for venom mapping and peptide sequencing from crude venoms: case applications with single arthropod specimen.

Due to their complexity and diversity, animal venoms represent an extensive source of bioactive compounds such as peptides and proteins. Conventional approaches for their characterization often require large quantities of biological material. Current mass spectrometry (MS) techniques now give access to a wealth of information in a short working time frame with minute amounts of sample. Such MS approaches may now be used for the discovery of novel compounds, and once their structure has been determined they may be synthesized and tested for functional activity. Molecular mass fingerprints of ve

Natural Occurrence in Venomous Arthropods of Antimicrobial Peptides Active against Protozoan Parasites.

Arthropoda is a phylum of invertebrates that has undergone remarkable evolutionary radiation, with a wide range of venomous animals. Arthropod venom is a complex mixture of molecules and a source of new compounds, including antimicrobial peptides (AMPs). Most AMPs affect membrane integrity and produce lethal pores in microorganisms, including protozoan pathogens, whereas others act on internal targets or by modulation of the host immune system. Protozoan parasites cause some serious life-threatening diseases among millions of people worldwide, mostly affecting the poorest in developing tropica
