An In Vitro Tribological Astringency Assessment Method Based on Astringency Perception Mechanism.

Astringency is a complex oral sensation characterized by dryness and constriction in the mouth. It is typically induced by polyphenol-rich foods and beverages such as wine and tea. The quantitative assessment of astringency intensity has become a prominent research focus in the food science field. In this study, an in vitro oral tribological method was developed based on a soft-hard friction interface. An oral-like sensory structure was constructed using biomimetic materials to replicate tongue-palate contact. The frictional behavior carried out in this structure is similar to that of the pala

The role of TRPA1 and TRPV1 in the perception of astringency.

Astringency, commonly described as a drying, roughening, and/or puckering sensation associated with polyphenol-rich foods affects their palatability. While the compounds eliciting astringency are known, its mechanism of action is debated. This study investigated the role of transient receptor potential (TRP) channels A1 and V1 in astringency perception. If TRP A1 or V1 have a functional role in astringency perception, then desensitizing these receptors should decrease perceived astringency. Thirty-seven panelists underwent unilateral lingual desensitization of TRP A1 and V1 channels using must

Sensory Nutrition and Bitterness and Astringency of Polyphenols.

Recent studies have demonstrated that the interaction of dietary constituents with taste and olfactory receptors and nociceptors expressed in the oral cavity, nasal cavity and gastrointestinal tract regulate homeostasis through activation of the neuroendocrine system. Polyphenols, of which 8000 have been identified to date, represent the greatest diversity of secondary metabolites in plants, most of which are bitter and some of them astringent. Epidemiological studies have shown that polyphenol intake contributes to maintaining and improving cardiovascular, cognitive and sensory health. Howeve

TRPV2 and TRPC5 are potential targets for astringent phytochemicals.

Astringency is a multimodal sensory experience resulting from complex interactions between chemical compounds and the oral environment, involving tactile, chemosensory and thermosensory pathways. Recent human studies have examined the role of the polymodal transient receptor potential (TRP) channels TRPV1 and TRPA1 in astringency perception; however, other thermo- and mechanosensitive TRP channels expressed in oral epithelial cells and in trigeminal neurons innervating the mouth and tongue may also contribute to this complex sensation. This study explored the effects of structurally distinct r

Exploring the Relationship Between Astringency and Lingual Tactile Sensitivity.

When lingual tactile sensitivity has received less attention than taste, it plays a critical role in food textural preferences and eating behavior. The anterior tongue, rich in specialized mechanoreceptors, is particularly sensitive to tactile stimuli. This study investigated inter-individual variability in lingual tactile sensitivity and its potential relationship with astringency sensitivity. To this end, three distinct aspects of lingual tactile sensitivity as well as tongue strength were assessed in 39 subjects (26 F, mean age = 35 ± 5 years). The three tactile dimensions included: (i) lig

Evaluation of Astringent Compounds Using Electronic Tongue Technology.

The electronic tongue (E-tongue) is an emerging technology that enables the rapid and objective evaluation of various astringent compounds. This study investigated E-tongue measurements and human sensory evaluation to analyze the concentration-dependent astringency responses of seven compounds. Difference analysis, principal component analysis (PCA), and cluster analysis were applied to differentiate the compounds based on their taste profiles. The results revealed distinct relationships between E-tongue response values and concentration. Specifically, epigallocatechin gallate (EGCG) and epiga

New Insights into the Mechanisms of Bitterness and Astringency in Chitosan Oligosaccharide Complexes.

Chitosan oligosaccharides (COS) possess valuable bioactive properties but exhibit undesirable bitter and astringent tastes that severely restrict their applications in functional foods. This study comprehensively elucidates the formation mechanisms of these taste defects by systematically investigating COS fractions with varying molecular weights. It was found that COS astringency intensifies with increasing molecular weight, while bitterness increases with decreasing molecular weight. The astringency mechanism primarily involves COS binding to salivary proteins via electrostatic interactions,

Database, prediction, and antibacterial research of astringency based on large language models.

Astringency, a sensory experience causing mouth dryness, significantly impacts the taste of foods such as wine and tea, and astringent molecules may exhibit antibacterial properties. Traditional methods for predicting astringency are costly, and the connection between astringency and antibacterial activity remains largely unexplored. In this study, we present a pioneering computational approach that includes: (1) the creation of the first comprehensive astringency database comprising 238 molecules; (2) the development of a Ligand-Based Prediction (LBP) framework that combines large language mo

Astringent drugs for bleedings and diarrhoea: The history of Cynomorium coccineum (Maltese Mushroom).

ETHNOPHARMACOLOGICAL RELEVANCE: The botanical identity of the ancient vernacular cynomorium does not correspond to the modern scientific genus while it is not clear how many species of hipocistis (Cytinus sp.) were differentiated by the ancient physicians and whether Cynomorium coccineum was subsumed. The early history of therapeutic uses related to the herbal drugs derived from these parasitic taxa is therefore not easily accessible. Cynomorium coccineum became an important pharmaceutical commodity after the Siege of Malta but its importance decreased in the 18th century and now is considered

Characterization of a gene regulatory network underlying astringency loss in persimmon fruit.

Transcriptome analysis of a persimmon population segregating for an astringency trait in fruit suggested central roles for a limited number of transcriptional regulators in the loss of proanthocyanidin accumulation. Persimmon (Diospyros kaki; 2n = 6x = 90) accumulates a large amount of proanthocyanidins (PAs) in its fruit, resulting in an astringent taste. Persimmon cultivars are classified into four types based on the nature of astringency loss and the amount of PAs at maturity. Pollination constant and non-astringent (PCNA)-type cultivars stop accumulating PAs in the early stages of fruit de

Interfacial and colloidal drivers of wine drying astringency: the role of wine-saliva aggregates.

Astringency in red wine is associated with lubrication changes driven by tannin-saliva interactions, yet the physicochemical basis of astringency subqualities remains unclear. Focusing on drying, a key quality-related subquality, we examined whether wine-saliva aggregates can contribute to lubrication stability rather than only increasing friction. Red wines spanning non-drying to highly drying profiles were mixed 1:1 with unstimulated human saliva. Film thickness and friction were measured under controlled, oral-inspired sliding conditions, and mixtures were centrifuged to obtain soluble (sup

Inhibitory effect of chlorogenic acid on tannase-mediated astringency removal and its mechanism.

BACKGROUND: Phenolic acids, such as chlorogenic acid (CGA) and rosmarinic acid (RA), are added to plant-based beverages as nutritional supplements to enhance their health benefits. However, these compounds can also interfere with the astringency-reducing effect of tannase. This study employed electronic tongue analysis, enzyme inhibition kinetics, spectroscopy, molecular docking and molecular dynamics simulations to investigate the inhibitory mechanisms of CGA and RA on tannase-mediated deastringency. RESULTS: Our research results indicate that CGA can inhibit tannase-mediated deastringency.

Astringent flavanol fires the locus-noradrenergic system, regulating neurobehavior and autonomic nerves.

Astringency is a characteristic exhibited by only a limited number of polyphenolic compounds which show high electrochemical activity and are susceptible to oxidation at neutral pH conditions like the oral cavity and small intestine. Large-scale intervention studies have demonstrated that the astringent flavanol (FLs) can restore hippocampal-dependent memory. However, due to the low bioavailability of FLs, the mechanism of action remains unclear. In this study, we aimed to elucidate the mechanism by which FLs acts on the nervous system through the gastrointestinal tract. Following a single gav

Correlation Analysis and Spiking Validation of Flavor Compounds Modulating Bitterness and Astringency in Jasmine Tea.

To elucidate the chemical basis underlying bitterness and astringency modulation in jasmine tea, four green tea bases and their corresponding jasmine teas were analyzed using weighted gene co-expression network analysis (WGCNA) combined with spiking validation experiments. WGCNA revealed that catechin, gallocatechin gallate, aspartic acid (Asp), glutamic acid (Glu), arginine (Arg), and alanine (Ala) were positively associated with sweetness and umami but negatively associated with bitterness and astringency. Spiking experiments showed that taste modulation was both concentration- and matrix-de

The Different Contributors to Antioxidant Activity in Thermally Dried Flesh and Peel of Astringent Persimmon Fruit.

In the thermal-drying processing of astringent persimmon fruit, the tissue-specific changes in the key antioxidants have hardly been investigated, while they have been well investigated in the flesh. We report here the different patterns of the antioxidant activities in the thermally processed flesh and peel of astringent persimmon, with analyses of the carotenoids, the condensed and hydrolysable tannins, and the total phenolics and flavonoids. The persimmon powders presented different colors on the basis of the drying temperatures: brown in 30 °C; light yellow in 60 °C; and dark brown in 90 °

A sclerosant with astringent properties developed in China for oesophageal varices: comparison with ethanolamine oleate and polidocanol.

BACKGROUND: Xiao zhi ling (XZL), which consists of Chinese nutgalls and aluminium potassium sulphate, is used as a local injection for the treatment of internal haemorrhoids in China. It is also used for endoscopic sclerotherapy of oesophageal varices. To date, however, it has not been compared with other sclerosants with regard to its safety and efficacy. METHODS: In the present study, the effect on the endothelium and the haemolytic and thrombosing effects of XZL were compared with those of 5% ethanolamine oleate and 1% polidocanol, using an endothelial cell line and red blood cells taken f

Integrated Transcriptomics and Metabolomics Analysis of the Formation Mechanism of Low Bitterness and Astringency in "Huangjinya" Tea.

The flavor variation among different tea plant cultivars significantly influences their economic value. 'Huangjinya' (HJY), a typical light-sensitive etiolated tea cultivar, is prized for its fresh and mellow taste with low bitterness and astringency, and commands a high market value, serving as excellent germplasm for breeding superior-flavor tea varieties. Previous studies have largely focused on the mechanisms underlying fresh and umami tastes, whereas the regulatory mechanisms underlying bitterness and astringency remain poorly understood. In this study, two tea plant (Camellia sinensis (L

Targeted Removal of Galloylated Flavanols to Adjust Wine Astringency by Using Molecular Imprinting Technology.

Excessive galloylated flavanols not only cause instability in the wine but also lead to unbalanced astringency. Although clarification agents are always used to precipitate unstable tannins in wine, the non-specific adsorption of tannins results in the failure to precisely regulate the tannin composition of the wine. In this work, molecularly imprinted polymers (MIPs) with template molecules of galloylated flavanols were designed to specifically adsorb gallotannins to reduce wine astringency. The results showed that the "pores" on the surface of the MIPs are the structural basis for the specif
