Atropa belladonna, commonly known as belladonna or deadly nightshade, ranks among one of the most poisonous plants in Europe and other parts of the world. The plant contains tropane alkaloids including atropine, scopolamine, and hyoscyamine, which are used as anticholinergics in Food and Drug Administration (FDA) approved drugs and homeopathic remedies. These alkaloids can be very toxic at high dose. The FDA has recently reported that Hyland's baby teething tablets contain inconsistent amounts of Atropa belladonna that may have adverse effects on the nervous system and cause death in children,

Acute angle closure glaucoma is a sight-threatening condition that may lead to blindness. This is a case report of a woman who presented to the emergency department (ED) with acute angle closure glaucoma following use of an over-the-counter (OTC) homeopathic eye drop containing atropa belladonna (deadly nightshade). A 55-year-old woman presented to the ED with a 5-day history of left eye redness, swelling, tearing, and foreign-body sensation that had acutely worsened in the last two days. Her exam revealed mild left conjunctival injection with watery tearing and a hazy appearance of her left c

Atropa belladonna (A. belladonna), commonly known as deadly nightshade, is a poisonous plant belonging to the Solanaceae family. The toxic effects of A. belladonna are attributable to its alkaloid content, which possesses potent anticholinergic properties. These alkaloids are responsible for the plant's toxicity and can cause a range of adverse effects in humans and animals upon ingestion or contact. In this report, we describe two atypical cases of A. belladonna poisoning resulting from accidental ingestion of the plant's raw leaves, which were referred to the emergency room of a poisoning ce

Ornithine decarboxylase (ODC) plays an important role in various biological processes; however, its role in plant secondary metabolism, especially in the biosynthesis of tropane alkaloids (TAs) such as pharmaceutical hyoscyamine, anisodamine, and scopolamine, remains largely unknown. In this study, we characterized the physiological and metabolic functions of the ODC gene of Atropa belladonna (AbODC) and determined its role in TA production using metabolic engineering approaches. Feeding assays with enzyme inhibitors indicated that ODC, rather than arginine decarboxylase (ADC), plays a major r

Elicitation of transgenic Atropa baetica overexpressing the h6h gene with salicylic acid (SA), acetylsalicylic acid (ASA),or methyl jasmonate (MeJ) was conducted to boost tropane alkaloid yields. Scopolamine (1) amounts increased after treatment with ASA and MeJ, but not with SA. The highest enhancement of 1 was achieved with MeJ followed by ASA dissolved in EtOH. Transcriptomic analyses showed a direct relationship between content of 1 and gene expressions;the engineered h6h gene and other biosynthetic genes were stimulated. ASA dissolved in EtOH showed a high h6h gene expression, increasing

Homeopathic products are still a controversial issue in modern medicine, understood as complementary or alternative medicine (CAM). In this particular case, homeopathic products prepared from Atropa belladonna extracts may present specific problems due to the effects derived from its components. This article applies a simple, rapid, reliable method to the analysis of different homeopathic products obtained from Atropa belladonna; drugs containing high concentration of plant extracts; and Atropa belladonna seeds. The method was based on a simple solid-phase preconcentration method followed by u

The Solanaceae family comprises numerous species of economic and pharmacological importance, including the toxic deadly nightshade (Atropa belladonna), bittersweet nightshade (Solanum dulcamara), and black nightshade (Solanum nigrum). These plants are native to Europe, North Africa, and Western Asia, and have been used since ancient times in both ritual practices and medicine. A. belladonna contains potent tropane alkaloids (atropine, hyoscyamine, and scopolamine) and is widely utilized in modern pharmacology, although accidental poisonings still occur, often due to confusion with edible plant

Atropa belladonna L. is the officially medicinal plant species and the main commercial source of scopolamine and hyoscyamine in China. In this study, we reported the simultaneous overexpression of two functional genes involved in biosynthesis of scopolamine, which respectively encoded the upstream key enzyme putrescine N-methyltransferase (PMT; EC 2.1.1.53) and the downstream key enzyme hyoscyamine 6beta-hydroxylase (H6H; EC 1.14.11.11) in transgenic hair root cultures of Atropa belladonna L. HPLC results suggested that four transgenic hair root lines produced higher content of scopolamine at

Hyoscyamine, an active tropane alkaloid primarily extracted from the medicinal plant Atropa belladonna, exhibits analgesic and sedative effects and is widely used as a precursor in pharmaceutical formulations. However, the low yield of hyoscyamine and the complex composition of tropane alkaloids in wild-type A. belladonna plants pose significant challenges for industrial production, making it imperative to modify the metabolic pathway to enhance hyoscyamine yield and purity. In this study, a pull-block metabolic engineering strategy, which enhanced hyoscyamine biosynthesis by co-overexpressing

Two series of Atropa belladonna hairy root lines were obtained, the first one transformed via Agrobacterium tumefaciens harboring rol C and npt II genes, and the other transformed with rol ABC and npt II genes. Thirteen hairy root lines were obtained and selected on hormone-free medium. The transformation was confirmed by PCR analysis, and these root lines were first examinated for their growth rate. Then hyoscyamine and scopolamine production was measured after 3 and 4 weeks of culture to evaluate the possible role of rol C gene in tropane alkaloid formation. The rol C gene alone played a sig

Atropa belladonna L. (Solanaceae) is a significant source of the pharmacologically important tropane alkaloids (TAs), particularly hyoscyamine and scopolamine. Although rolB and rolC from Rhizobium rhizogenes are known to enhance plant secondary metabolism, their functional specificities and combined actions in TAs biosynthesis remain unclear. Here, we generated transgenic A. belladonna overexpressing 35S::rolB, 35S::rolC, and 35S::rolB + rolC to systematically investigate their effects on TAs accumulation, plant morphology, and drought tolerance. Co-expression of rolB and rolC exhibited metab

A description of the case of a 57 year old woman is given who showed the symptoms of an atropine poisoning after drinking stinging nettle tea. The analyses of the tea specimen proved bad defilements among other things with elements of the "belladonna" (Atropa belladonna). Several similar reports became known recently and it seems to be necessary to discuss checking the purity of the tea species.

We describe the Italian contribution to the description and treatment of parkinsonism following encephalitis lethargica (EL): postencephalitic parkinsonism (PEP). Special attention is devoted to the description of postencephalitic symptoms by Giuseppe Panegrossi (1871-1953) and to the treatment based on Atropa belladonna introduced in Italy and extensively supported by Arturo Nannizzi (1887-1961), who was charged by the queen of Italy with conducting research into this plant and advocating its cultivation for healing purposes. This article gives us the unique opportunity to revisit the figure

Some activities of seven vegetable extracts and an association of them given by oral route were tested on the C.N.S. of the mouse. Among these, Crataegus oxyacantha and, less clearly, Valeriana officinalis show some sedative activity, whereas the extract from Passiflora incarnata gives some anxiolytic effect. Matricaria chamomilla and Piscidia erythrina stand in an intermediate position between the previous ones. Hyoscyamus niger proved to be active in only one of the tests performed, whereas Atropa belladonna did not show any activity on the C.N.S. The association of the seven extracts seemed
