D2 autoreceptors gate vulnerability to cocaine use disorder.

A defining feature of substance use disorder is that repeated drug use does not always lead to addiction, motivating the search for biomarkers of vulnerability1. Reduced striatal dopamine D2/3 receptor availability is a robust PET correlate of problematic stimulant use2-5, but the signal may reflect high endogenous dopamine level, and it conflates presynaptic D2 autoreceptors on dopamine axons with postsynaptic D2/3 heteroreceptors on striatal projection neurons. We dissociated these contributions using cell type-specific Drd2 haploinsufficiency in dopamine neurons (autoD2KD), D2-expressing me

Autoreceptor control of serotonin dynamics.

BACKGROUND: Serotonin is a neurotransmitter that has been linked to a wide variety of behaviors including feeding and body-weight regulation, social hierarchies, aggression and suicidality, obsessive compulsive disorder, alcoholism, anxiety, and affective disorders. Full understanding involves genomics, neurochemistry, electrophysiology, and behavior. The scientific issues are daunting but important for human health because of the use of selective serotonin reuptake inhibitors and other pharmacological agents to treat disorders. This paper presents a new deterministic model of serotonin metabo

Presynaptic H3 autoreceptors modulate histamine synthesis through cAMP pathway.

Histamine H3 receptors modulate histamine synthesis, although little is known about the transduction mechanisms involved. To investigate this issue, we have used a preparation of rat brain cortical miniprisms in which histamine synthesis can be modulated by depolarization and by H3 receptor ligands. When the miniprisms were incubated in presence of forskolin, dibutyryl-cAMP, or 3-isobutyl-1-methylxanthine (IBMX), histamine synthesis was stimulated in 34, 29, and 47%, respectively. These stimulations could be prevented by the selective cAMP protein kinase blocker Rp-adenosine 3',5'-cyclic monop

Dopamine autoreceptor stimulation: clinical significance.

Recent studies of the effects of low doses of dopamine agonists, designed to stimulate dopamine autoreceptors and hence diminish the synthesis and release of dopamine, were based on a series of basic research studies which demonstrated the existence of autoreceptors on dopamine neurones of the nigrostriatal, mesolimbic and mesocortical dopaminergic neurones. Evidence for autoreceptors on the tuberoinfundibular dopamine neurones which participate in the regulation of prolactin and growth hormone secretion is lacking. Some recent reports have questioned the existence of dopamine autoreceptors on

Prejunctional alpha2A-autoreceptors in the human gastric and ileocolic arteries.

This study was undertaken to determine the subtype of prejunctional alpha2-autoreceptors in human blood vessels. Segments of gastric and ileocolic arteries were incubated with [3H]noradrenaline and subsequently perifused with modified Krebs-Henseleit solution containing cocaine (12 microM). Five periods of electrical stimulation (S1-S5) were applied (1 Hz, 1 ms, 50 V for 1 min). Concentration-response curves for the facilitatory effect of eight alpha-adrenoceptor antagonists [rauwolscine, 2-[2-(2-methoxy-1,4-benzodioxanyl)] imidazoline (RX821002), yohimbine, phentolamine, idazoxan, 2-(2',6'-di

Apomorphine anorexia: the role of dopamine cell body autoreceptors.

Anorectic effects of apomorphine were studied in a microstructural analysis paradigm. Systemic apomorphine reduced food intake by reducing both the rate of eating and the time spent eating. Peripheral administration of sulpiride reversed the apomorphine effect on both eating rate and eating time but central administration of this neuroleptic into the ventral tegmental area (VTA) selectively reversed the apomorphine effect on eating time, sparing eating rate. Administration of apomorphine directly into the VTA reduced eating time but not eating rate; the effect on eating time was blocked by per

Stimulation of dopamine autoreceptors elicits "premature ejaculation" in rats.

Following treatment with dopamine (DA) receptor agonists, such as apomorphine, N-n-propyl-norapomorphine, lisuride and 3-(3-hydroxyphenyl)-N-n-propyl-piperidine (3-PPP) (50, 2.5, 400 and 5000 micrograms/kg, respectively), male rats attain ejaculation with receptive females sooner and after fewer penile intromissions than controls. Since doses of DA agonists needed to produce "premature ejaculation" are within the low dose range needed to stimulate DA autoreceptors, it is suggested that "premature ejaculation" in rats results from inhibition of DA neurotransmission. This hypothesis is supported

Dopaminergic D-3 binding sites are not presynaptic autoreceptors.

Postsynaptic dopamine (DA) receptors have been classified biochemically and pharmacologically into two types: D-1 receptors mediate adenylate cyclase stimulation, demonstrating micromolar affinity for DA and butyrophenone antagonists; D-2 receptors mediate adenylate cyclase inhibition, demonstrating nanomolar affinity for DA and butyrophenone antagonists. D-1 receptors are labelled by 3H-thioxanthene antagonists, while D-2 receptors are labelled by both 3H-agonists and all 3H-antagonists. A third class of dopaminergic binding site, termed D-3, represents high-affinity 3H-agonist binding sites

The influence of dopamine autoreceptors on temperament and addiction risk.

As a major regulator of dopamine (DA), DA autoreceptors (DAARs) exert substantial influence over DA-mediated behaviors. This paper reviews the physiological and behavioral impact of DAARs. Individual differences in DAAR functioning influences temperamental traits such as novelty responsivity and impulsivity, both of which are associated with vulnerability to addictive behavior in animal models and a broad array of externalizing behaviors in humans. DAARs additionally impact the response to psychostimulants and other drugs of abuse. Human PET studies of D2-like receptors in the midbrain provide

Roxindole: psychopharmacological profile of a dopamine D2 autoreceptor agonist.

The putative, selective dopamine (DA) dopamine-2 autoreceptor agonist roxindole, which also exhibits serotonin-1A-agonistic and 5-hydroxytryptamine reuptake-inhibiting properties, was examined for its behavioral effects in rats and mice. Roxindole inhibited apomorphine-induced climbing in mice and stereotyped behavior in rats with ED50 values of 1.4 mg/kg s.c. and 0.65 mg/kg s.c., respectively, and inhibited conditioned avoidance response in rats (ED50 = 1.5 mg/kg s.c.). Presynaptic Release-Regulating Alpha2 Autoreceptors: Potential Molecular Target for Ellagic Acid Nutraceutical Properties.

Polyphenol ellagic acid (EA) possesses antioxidant, anti-inflammatory, anti-carcinogenic, anti-diabetic and cardio protection activities, making it an interesting multi-targeting profile. EA also controls the central nervous system (CNS), since it was proven to reduce the immobility time of mice in both the forced swimming and the tail-suspension tests, with an efficiency comparable to that of classic antidepressants. Interestingly, the anti-depressant-like effect was almost nulled by the concomitant administration of selective antagonists of the noradrenergic receptors, suggesting the involve

Effects of the D3 and autoreceptor-preferring dopamine antagonist (+)-UH232 in schizophrenia.

The aminotetralin derivative (+)-UH232 is a dopamine-receptor antagonist with complex pharmacological properties, including a 4:1 selectivity for the D3 vs. D2 receptor and a preference for the autoreceptors. Its behavioral profile differs markedly from that of other dopamine antagonists in exhibiting both stimulant and inhibitory features. In an effort to elucidate the role of different dopamine receptor subtypes in psychosis, we administered (+)-UH232 to drug-free schizophrenic patients. Six patients received single doses of (+)-UH232 over a dose range of 80 to 180 mg in a rising-dose, doubl

Calcium permeable AMPA receptors and autoreceptors in external tufted cells of rat olfactory bulb.

Glomeruli are functional units of the olfactory bulb responsible for early processing of odor information encoded by single olfactory receptor genes. Glomerular neural circuitry includes numerous external tufted (ET) cells whose rhythmic burst firing may mediate synchronization of bulbar activity with the inhalation cycle. Bursting is entrained by glutamatergic input from olfactory nerve terminals, so specific properties of ionotropic glutamate receptors on ET cells are likely to be important determinants of olfactory processing. Particularly intriguing is recent evidence that AMPA receptors o

EMD 23,448: effects of a putative dopamine autoreceptor agonist in chorea.

A novel dopamine agonist, which stimulates presynaptic dopamine receptors in the experimental animal, was administered acutely to four patients with chorea. Abnormal involuntary movements worsened, plasma prolactin levels fell, and plasma growth hormone concentrations increased in all patients tested. These effects resemble those associated with a postsynaptic rather than presynaptic dopamine agonist, and thus raise some question about the ability of conventional dopaminomimetic screens to predict clinical activity.

All three alpha2-adrenoceptor types serve as autoreceptors in postganglionic sympathetic neurons.

Postganglionic sympathetic neurons and brain noradrenergic neurons use alpha(2A)- and alpha(2C)-adrenoceptors as presynaptic autoreceptors. The present experiments were carried out in order to see whether they possess presynaptic alpha(2B)-autoreceptors as well. Pieces of atria, vasa deferentia, the occipito-parietal cortex and the hippocampus were prepared from either wildtype (WT) mice or mice in which both the alpha(2A)- and the alpha(2C)-adrenoceptor gene had been disrupted (alpha(2AC)KO). The pieces were incubated with (3)H-noradrenaline and then superfused and stimulated electrically. In

Synaptojanin-1 supports VPS35-dependent trafficking of dopamine D2 autoreceptors at presynaptic terminals.

Synaptic dysfunction is a hallmark of early Parkinson's disease (PD), but molecular mechanisms underlying dopaminergic synaptic impairment and vulnerability remain poorly understood. Here, we identify a functional interaction between two PD genes, Synaptojanin1 and VPS35, in regulating endosomal sorting of the dopamine D2 short (D2S) autoreceptor, thereby modulating dopamine signaling. We show that Synaptojanin1 deficiency results in intracellular retention of the D2S, impaired gating of dopamine release and reduced behavioral responsiveness to a D2-like agonist. VPS35 is recruited to D2S-cont

Are the pharmacology and physiology of α₂ adrenoceptors determined by α₂-heteroreceptors and autoreceptors respectively?

α(2)-Adrenoceptors are important mediators of physiological responses to the endogenous catecholamines noradrenaline and adrenaline. In addition, α(2)-adrenoceptors are pharmacological targets for the treatment of hypertension, sympathetic overactivity and glaucoma. α(2)-Adrenoceptors are also targeted to induce sedation and analgesia in anaesthesia and intensive care. α(2)-Adrenoceptors were first described as presynaptic receptors inhibiting the release of various transmitters from neurons in the central and peripheral nervous systems. In addition to these presynaptic neuronal receptors, α(2

In vivo evidence for the existence of autoreceptors on dopaminergic, serotonergic, and cholinergic neurons in the brain.

Intrastriatal infusions as well as systemic administration of the selective D-2 antagonist (-)-sulpiride caused similar increases in the dialysate levels of dopamine (DA) to about 180% of controls. A similar conclusion was drawn when the selective D-2 agonist (-)-N-0437 was infused intrastriatally or administered systemically: both routes of administration caused a decrease in the release of DA to about 40-50% of controls. In order to evaluate the properties of synthesis-modulating autoreceptors on dopaminergic and serotonergic neurons we have estimated the synthesis rate of serotonin (5-HT) o
