Specificity of avian leukosis virus-induced hyperlipidemia.

Rous-associated virus 7 (RAV-7) is a subgroup C avian leukosis virus which does not transform cells in vitro or carry an oncogene. When injected into 1-day-old hatched chicks, RAV-7 causes a low incidence of lymphoid leukosis after a latent period of several months. In contrast, infection of 10-day-old chicken embryos with RAV-7 leads to a disease syndrome characterized by stunting, obesity, atrophy of the bursa and the thymus, high triglyceride and cholesterol levels, reduced thyroxine levels, and increased insulin levels (Carter et al., Infect. Immun. 39:410-422, 1983; J.K. Carter and R.E. S

5' avian leukosis virus sequences and osteopetrotic potential.

Recombinants of Rous-associated virus-0 and Br21 have been used to localize 5' viral sequences that affect the osteopetrotic potential of avian leukosis viruses. Rous-associated virus-0 is a benign subgroup E virus of endogenous origin that does not cause osteopetrosis. Br21 is a constructed subgroup E virus with high osteopetrotic potential. 5' sequences that affected osteopetrotic potential resided in an 834-bp region near the 5' LTR. Sequence analysis of this region revealed differences between Br21 and RAV-0 in the mRNA leader and codons for MA.

Rapid induction of lymphoid leukosis and ascites by avian leukosis virus from a lymphoid leukosis cell line.

To examine whether a lymphoid leukosis (LL) cell line releases an LL-specific avian leukosis virus (ALV) or not, two viral materials, culture fluid and a concentrated viral material from an LL-cell line, were inoculated into a total of 74 day-old chicks of line 15I in 5 experiments. Spectrum of diseases induced, their incidence and incubation periods to onset were examined. Fifteen chicks were inoculated with the culture fluid and 9 (60%) developed ascites [59-119 days post inoculation (dpi); geometric mean (GM) of dpi, GM: 89.6)], but LL was not induced in any chicks inoculated. Fifty-nine ch

Hypergammaglobulinemia in chickens congenitally infected with an avian leukosis virus.

Significantly elevated (2- to 5-fold higher than controls) serum levels of IgG were found in chickens congenitally infected with F42 strain of avian leukosis (ALV-F42) a subgroup A avian leukosis virus (ALV). A further increase in IgG levels in congenitally infected birds was found to be induced by injection of influenza virus in complete Freund's adjuvant(CFA). Serum immunoglobulin M (IgM) levels were not significantly elevated in ALV congenitally infected chickens except in those animals that had been injected with influenza virus in CFA. Hypergammaglobulinemia in ALV infected birds resulted

[Research progress in avian leukosis virus subgroup J in China].

Avian leukosis is a major neoplastic disease caused by avian leukosis viruses (ALVs), which are classified into 11 subgroups (ALV-A to ALV-K). Among them, ALV subgroup J (ALV-J) has undergone significant epidemiological changes since its introduction into China in 1999. It initially transmitted among broilers and then rapidly spread to commercial layer chickens and local chicken breeds. ALV-J infection typically induces myeloid leukosis in chickens and, in some layers, can also lead to hemangiomas. As a retrovirus, ALV-J exhibits high genetic variability. Compared with the prototype strain HPR

Myotropic avian leukosis virus subgroup J infection in a chicken.

The study describes a highly productive myotropic avian leukosis virus infection (ALV) in a 3-month-old female chicken. At necropsy, ascites, hepatic fibrosis and cardiomegaly were seen. Histologically, the most striking lesion was the presence of cytoplasmic basophilic inclusions in myocardial fibers. Immunostaining for ALV group specific antigen p27 revealed a diffuse presence of virus antigen in cardiac myofibers, in smooth muscle fibers of most of the organs, and in rare, pancreatic and ovarian theca cells. Ultrastructurally, myocardial inclusions consisted of clusters of 50-60 nm round pa

Selective integration of avian leukosis virus in different hematopoietic tissues.

Hematopoietic tissues obtained from avian leukosis virus (ALV)-infected Hyline SC chickens were analyzed for the presence of integrated viral DNA sequences. Cells were prepared from bone marrow, bursa, spleen, thymus, and peripheral blood. Following the removal of erythrocytes, cellular DNAs from each of these tissues were examined by Southern analysis. During the first few weeks of infection, DNA from the bone marrow contained as many as 0.5 copies of viral DNA per haploid genome. Cells from the bursa and peripheral blood contained between 0.05 and 0.15 copies per haploid genome. In contrast,

Establishment of interference in osteoblasts by an osteopetrosis-inducing avian leukosis virus.

Avian leukosis virus (ALV)-induced osteopetrosis is characterized by the presence of unintegrated viral DNA is diseased bone. To learn more about this phenomenon, ALVs with high (Br21) and low (RAV-1) osteopetrotic potential have been compared for the ability to establish superinfection resistance in cultures of calvarial-derived osteoblasts. Both viruses could establish interference in osteoblast cultures that were maintained as growing cells. This interference was maintained when cultures were induced to differentiate. These results suggest that the unintegrated viral DNA in osteopetrotic bo

Current knowledge on the epidemiology and prevention of Avian leukosis virus in China.

Avian leukosis virus (ALV) is an enveloped retrovirus with a single-stranded RNA genome, belonging to the genus Alpharetrovirus within the family Retroviridae. The disease (Avian leukosis, AL) caused by ALV is mainly characterized by tumor development and immunosuppression in chickens, which increases susceptibility to other pathogens and leads to significant economic losses in the Chinese poultry industry. The government and poultry industry have made lots of efforts to eradicate ALV, but the threat of which remains not vanished. This review provides a summary of the updated understanding of

Detection and characterization of avian leukosis virus in Marek's disease vaccines.

Avian leukosis virus (ALV) infection in chickens is known to induce increased mortality, tumors, delayed growth, and suboptimal egg production. Countries importing specified pathogen-free eggs, vaccines, and poultry breeding stock require freedom of infection or contamination with ALV in such products among other avian pathogens. Recently, ALV was found as a contaminant in a limited number of commercial poultry vaccines, even after routine quality assurance procedures cleared the vaccines for commercialization. The contaminated vaccines were promptly withdrawn from the market, and no direct de

Lability of leukosis virus enhancer-binding proteins in avian hematopoeitic cells.

Bursal lymphomas induced by avian leukosis virus (ALV) are characterized by integration of long terminal repeat (LTR) enhancer sequences next to the myc proto-oncogene and by subsequent myc hyperexpression. Nuclear runoff transcription analyses have shown that protein synthesis inhibition specifically decreases transcription of LTR-enhanced genes in bursal lymphoma cell lines (M. Linial, N. Gunderson, and M. Groudine, Science 230:1126-1132, 1985). Here, we show that LTR-enhanced transcription is also labile in nontransformed bursa, bone marrow, and spleen but not in other ALV-infected tissues

Experimental infection with avian leukosis virus isolated from Marek's disease vaccines.

Recently, avian leukosis virus (ALV) was isolated from four lots of Marek's disease vaccine produced by two laboratories. The ALVs isolated were characterized by examination of their interactions with cells of two phenotypes (C/E and C/A,E), subgroup-specific polymerase chain reaction (PCR), virus neutralization, envelope gene sequencing, and phylogenetic analysis. All four ALVs are exogenous, belong to subgroup A, and appear to be virtually identical to each other based on PCR and envelope gene nucleotide sequences. We describe herein the characterization of the contaminant viruses in vivo by

Yellow fever vaccination, avian leukosis virus, and cancer risk in man.

Comparison was made between 2659 veterans who died of cancer, during 1950 to 1954 or 1959 to 1963, and matched controls, based on the frequency of yellow fever immunization during World War II. The vaccine was produced from chick embryos that almost certainly contained avian leukosis-sarcoma viruses. Among the veterans, no relation was found between vaccination and leukemia, lymphoma, or other cancer.

Diagnosis of myeloid leukosis induced by a recombinant avian leukosis virus in commercial white leghorn egg laying flocks.

Commercial white leghorn egg layer flocks being used to produce fertile eggs for human vaccine production exhibited dramatically low peaks in egg production, two to four times higher than normal weekly mortality, and high numbers of cull, nonlaying birds after the onset of sexual maturity. These lower production characteristics could not be associated with management-related problems. Gross lesions of cull and fresh dead birds necropsied showed approximately 60% lacked ovarian activity and had lesions of a bacterial bursitis or synovitis, whereas the other 40% had tumors of the viscera but not

Avian leukosis virus contamination in live vaccines: A retrospective investigation in China.

Avian leukosis (AL) is one of the most pandemic immunosuppressive diseases and has been widely spread between 2006 and 2009 in China. The contamination of avian leukosis virus (ALV) in attenuated vaccine is considered as one of the possible transmission routes of this disease. Based on a retrospective survey of 918 batches of attenuated vaccine produced before 2010, three of them were identified as ALV-positive and corresponding ALV strains were successfully isolated from a live Fowlpox virus vaccine, a live Newcastle disease virus vaccine and a live Infectious Bursal Disease virus vaccine, re

Association of the major histocompatibility complex with avian leukosis virus infection in chickens.

1. Association of the B blood group, the major histocompatibility complex (MHC) in chickens, with avian leukosis virus (ALV) infection shown by shedding of group-specific (gs) antigen was studied in an Australorp line selected for short oviposition interval to improve egg production. Three haplotypes (B8a, B9a and B21) were segregating in this line at frequencies of 66.7, 15.6 and 17.8%, respectively, averaged over three generations. 2. The relative risk (odds ratio) of a hen becoming a gs-antigen shedder was calculated for progenies of the dams shedding gs-antigen and those of non-shedding da
