Improvement of a recombinant avian avulavirus serotype 10 vectored vaccine by the addition of untranslated regions.

BACKGROUND: We have previously developed a recombinant avian avulavirus serotype 10 (rAAvV-10/HA) expressing the hemagglutinin (HA) gene of a highly pathogenic avian influenza virus (HPAIV) as an emergency vaccine for poultry. rAAvV-10/HA can overcome the activity of the anti-AAvV-1 (Newcastle disease virus) antibody acquired by commercial chickens upon routine vaccination. Most chickens do not have the anti-AAvV-10 antibody, which could interfere with the vaccine efficacy. However, the vaccine efficacy of rAAvV-10/HA is not satisfactory in chickens even though it affords protection against an

Effect of phytobiotics on humoral immune response and survival rate against avian Avulavirus challenge in broiler chickens.

BACKGROUND: Phytobiotics are natural plant-derived products that have gained recognition as superior alternatives to antibiotics that provide benefits without the risks of antibiotic resistance and residues. AIM: This study investigated the efficacy of phytobiotics supplemented in drinking water on the immune response and survival rate of broiler chickens against challenge from the Avian Avulavirus 1. METHOD: 200 one-day-old broiler chickens were selected. At first day, 20 chickens were sacrificed, and their blood sample was collected to estimate the maternal antibody titer against Avian Avu

Immunoprotective role of LaSota vaccine under immunosuppressive conditions in chicken challenged with velogenic avian avulavirus-1.

The first objective of the present study was to evaluate if the antibodies induced by the live LaSota and killed Newcastle disease (sub-genotype VIIi) vaccines protect the chickens against exposure with pathogenic avian avulavirus-1 (AAvV-1) of chicken and/or pigeon origins. The second objective was to study the effect of vaccines on stressed birds (dexamethasone, aflatoxin, and heat stressed) with respect to antibody production and protection against pathogenic AAvV-1 challenge. Sixty-one-day-old Hubbard chickens were divided into six groups (gA-gF) with ten animals each. All the groups recei

Cytokine expression in chicken embryo fibroblasts in response to infection with virulent or lentogenic avian avulavirus 1 (AAvV-1).

To investigate cytokine expression in chicken embryo fibroblast (CEF) cells, a virulent avian avulavirus 1 (AAvV-1) strain called SG10 that rapidly causes 100% mortality in its host, and a vaccine strain (La Sota) were characterized. Real-time quantitative PCR was performed on RNA samples from CEF cells, which were collected at 0, 24, 48 and 72 h post-infection. The dynamic expression patterns of ten cytokines (TNF-α, IFN-α, IFN-β, IL-1β, IL-2, IL-6, IL-10, IL-13, IL-15 and IL-18) were investigated. The results showed that infection with lentogenic La Sota induced significantly higher levels o

Biological characterization of wild-bird-origin avian avulavirus 1 and efficacy of currently applied vaccines against potential infection in commercial poultry.

Newcastle disease virus (NDV), the type member of the species Avian avulavirus 1 (formerly known as avian paramyxovirus serotype 1), causes a highly contagious and economically important disease in a myriad of avian species around the globe. While extensive vaccination programs have been implemented in ND-endemic countries, the disease is continuously spreading in commercial, backyard, and wild captive poultry. In order to investigate the evolution of the virus and assess the efficiency of the vaccine regimens that are currently being applied in commercial poultry, four wild-bird-origin NDV st

A Recombinant Turkey Herpesvirus Expressing F and HN Genes of Avian Avulavirus-1 (AAvV-1) Genotype VI Confers Cross-Protection against Challenge with Virulent AAvV-1 Genotypes IV and VII in Chickens.

Newcastle disease (ND) is responsible for significant economic losses in the poultry industry. The disease is caused by virulent strains of Avian avulavirus 1 (AAvV-1), a species within the family Paramyxoviridae. We developed a recombinant construct based on the herpesvirus of turkeys (HVT) as a vector expressing two genes: F and HN (HVT-NDV-F-HN) derived from the AAvV-1 genotype VI ("pigeon variant" of AAvV-1). This recombinant viral vaccine candidate was used to subcutaneously immunize one group of specific pathogen-free (SPF) chickens and two groups of broiler chickens (20 one-day-old bird

Avian paramyxovirus serotype 3 infection in Neopsephotus, Cyanoramphus, and Neophema species.

An outbreak of avian paramyxovirus serotype 3 (APMV-3) infection occurred in a private bird collection consisting mainly of Neophema species. Two Bourke's parrots (Neopsephotus bourkii) and one red-crowned parakeet (Cyanoramphus novaezelandiae) were examined after developing torticollis and circling. The 3 birds were euthanatized and submitted for necropsy. Brain and liver samples from the 3 birds were homogenized and inoculated into the allantoic cavity of 10-day-old embryonated specific-pathogen-free chicken eggs. An APMV-3 isolate replicated but did not induce embryonic mortality after up t

SEROLOGICAL SURVEY FOR SELECT INFECTIOUS AGENTS IN WILD MAGELLANIC PENGUINS (SPHENISCUS MAGELLANICUS) IN ARGENTINA, 1994-2008.

Despite being the most numerous penguin species in South America, exposure of the Magellanic Penguin (Spheniscus magellanicus) to pathogens has not yet been thoroughly assessed. We collected serum from 1,058 Magellanic Penguins at 10 breeding colonies along the entire latitudinal range of this species in Argentina. The work spanned 10 breeding seasons over 15 yr (1994-2008). Sera were tested for antibodies to select infectious agents. Antibodies reacting against 16 pathogens were detected (seroprevalence): Aspergillus sp. (15.1%), Chlamydia psittaci (6.5%), Salmonella Pullorum (3.1%), Salmonel

Avian Orthoavulavirus 1 Vaccination Failure in Minnesota Turkeys.

Minnesota is the leading state in number of turkeys produced in the United States. Turkey flocks in the field are usually vaccinated several times with live avian orthoavulavirus 1 (AOAV-1) vaccines starting as early as 2 wk of age (WOA). During the years 2018-2019, many turkey flocks were diagnosed with low-virulence AOAV-1 infection around 9 WOA that led to respiratory disease, although they were previously vaccinated. This study was designed to investigate the immunity against AOAV-1 in Minnesota turkey flocks in the field and experimentally after vaccination. We reviewed antibody titers ag

Prevalence of antibodies to different avian paramyxoviruses in commercial poultry in the United States.

Avian paramyxoviruses (APMVs) other than Newcastle disease virus have been isolated from feral avian species and have the potential to cause clinical disease. The objective of this study was to investigate the APMV antibody prevalence in commercial poultry settings. Sera from 100 commercial, layer-type and broiler-type (broiler-breeder and broiler) chicken flocks were analyzed on a random basis. Pooled serum samples from each flock were tested for the presence of antibodies to APMV-1, -2, -3, -4, -6, -7, -8, and -9 by hemagluttination inhibition (HI) test. Reactions with HI titers > or = 1:64

Recovery of Recombinant Avian Paramyxovirus Type-3 Strain Wisconsin by Reverse Genetics and Its Evaluation as a Vaccine Vector for Chickens.

A reverse genetic system for avian paramyxovirus type-3 (APMV-3) strain Wisconsin was created and the infectious virus was recovered from a plasmid-based viral antigenomic cDNA. Green fluorescent protein (GFP) gene was cloned into the recombinant APMV-3 genome as a foreign gene. Stable expression of GFP by the recovered virus was confirmed for at least 10 consecutive passages. APMV-3 strain Wisconsin was evaluated against APMV-3 strain Netherlands and APMV-1 strain LaSota as a vaccine vector. The three viral vectors expressing GFP as a foreign protein were compared for level of GFP expression

Long term circulation of avian paramyxovirus 4 in Australia revealed by historical and contemporary genomes from wild and domestic birds.

Avian paramyxovirus 4 (APMV4) is detected sporadically in wild birds and poultry, globally. While predominantly detected in waterfowl, the natural reservoir and ecology of APMV4 remains unclear. Herein we report historical and contemporary detections of APMV4 in Australia, and through sequencing demonstrate the likely long-term presence in a diversity of Australian wild bird species. This is evidenced by contemporary Australian genome sequences being more similar to historical Australian genomes rather than contemporary Asian genomes and that these genomes form a single lineage. This suggests

Co-Circulation and Excretion Dynamics of Diverse Rubula- and Related Viruses in Egyptian Rousette Bats from South Africa.

The Egyptian rousette bat (Rousettus aegyptiacus) has previously been implicated as the natural host of a zoonotic rubulavirus; however, its association with rubulaviruses has been studied to a limited extent. Urine, spleen, and other organs collected from the R. aegyptiacus population within South Africa were tested with a hemi-nested RT-PCR assay targeting a partial polymerase gene region of viruses from the Avula- and Rubulavirus genera. Urine was collected over a 14-month period to study the temporal dynamics of viral excretion. Diverse rubulaviruses, including viruses related to human mum

Experimental pigeon paramyxovirus-1 infection in chicken: evaluation of infectivity, clinical and pathological manifestations and diagnostic methods.

Several pigeon paramyxovirus-1 (PPMV-1) outbreaks in feral pigeons were described recently in Switzerland. The potential of PPMV-1 to induce the notifiable Newcastle disease in chickens is discussed controversially. Therefore, in order to study epidemiologically relevant parameters such as the kinetics of PPMV-1 replication and shedding as well as seroconversion after infection, chickens were infected experimentally with a Swiss PPMV-1 isolate. This generated also defined sample material for the comparison of diagnostic tests. The infectivity of the Swiss PPMV-1 isolate for chickens was demons

Immunogenicity of Newcastle Disease Vaccine in Southern Ground-hornbill (Bucorvus leadbeateri).

The southern ground-hornbill (Bucorvus leadbeateri; hereafter SGH) is endangered in South Africa, Namibia, and Swaziland. Through a conservation program established in South Africa by the Mabula Ground Hornbill Project, wild populations are being re-established by the reintroduction of captive-reared birds. The SGH is susceptible to infection with avian avulavirus 1, which causes Newcastle disease (ND). Four different vaccines to protect against ND were administered through various vaccination schedules and evaluated by serologic monitoring to assess the efficiency and safety of various combin

Epidemiology, control, and prevention of Newcastle disease in endemic regions: Latin America.

Newcastle disease (ND) infects wild birds and poultry species worldwide, severely impacting the economics of the poultry industry. ND is especially problematic in Latin America (Mexico, Colombia, Venezuela, and Peru) where it is either endemic or re-emerging. The disease is caused by infections with one of the different strains of virulent avian Newcastle disease virus (NDV), recently renamed Avian avulavirus 1. Here, we describe the molecular epidemiology of Latin American NDVs, current control and prevention methods, including vaccines and vaccination protocols, as well as future strategies

First detection and biological characterization of an avian metaavulavirus 8 isolated from a migratory swan goose in Qinghai Lake, Northwest China.

Avian metaavulavirus 8 (AMAV-8), formerly known as avian paramyxovirus 8 (APMV-8), has been detected sporadically in wild birds worldwide since it was first identified in a Canadian goose in 1976. However, the presence of AMAV-8 in birds has never been reported in China. To understand the epidemiological situation of AMAV-8 and its ability to infect chickens, we conducted a surveillance study and in vivo analysis of the AMAV-8 isolate identified in total of 14,909 clinical samples collected from wild and domestic birds from 2014 to 2022 in China. However, in 2017, only one AMAV-8 virus (Y7) wa

Replication, neurotropism, and pathogenicity of avian paramyxovirus serotypes 1-9 in chickens and ducks.

Источник пометил публикацию как отозванную

Avian paramyxovirus (APMV) serotypes 1-9 have been isolated from many different avian species. APMV-1 (Newcastle disease virus) is the only well-characterized serotype, because of the high morbidity, mortality, and economic loss caused by highly virulent strains. Very little is known about the pathogenesis, replication, virulence, and tropism of the other APMV serotypes. Here, this was evaluated for prototypes strains of APMV serotypes 2-9 in cell culture and in chickens and ducks. In cell culture, only APMV-1, -3 and -5 induced syncytium formation. In chicken DF1 cells, APMV-3 replicated with

Baculovirus expression of the avian paramyxovirus 2 HN gene for diagnostic applications.

Avian paramyxovirus 2 (APMV-2) infections are associated with respiratory diseases in poultry worldwide. The hemagglutination inhibition (HI) test is a useful tool for surveillance and monitoring of this virus. In this study, full-length hemagglutinin (HN) gene of APMV-2 was chemically synthesized based on its published sequence, cloned and expressed in Spodoptera frugiperda insect cells using recombinant baculoviruses. The biological, antigenic and immunogenic properties of the expressed protein were evaluated to assess its ability to produce diagnostic reagents for HI testing. Recombinant AP
