Axons, dendrites and synapses.

Classical concepts of neuronal organization have been based on the motoneuron as a model. Recent work on the mitral cell of the olfactory bulb has revealed synaptic connections of dendrites and cell bodies that are not present in the motoneuron. Similar findings in many other parts of the nervous system suggest the need to revise our concept of synaptic relations and functions properties of axons and dendrites.--SHEPHERD, G. M. Axons, dendrites and synapses.

Lead uptake in motor axons.

In an attempt to determine whether lead (Pb) in striated muscle can be taken up by motor axon, mice were injected intramuscularly with a 5% Pb nitrate solution, and the passage of Pb through the tissues was traced with electron microscopy. Thirty minutes after injection in the tibialis anterior muscle, Pb was seen at the sarcolemma and axolemma of the neuromuscular junction (NMJ) and in the adjacent sarcoplasmic reticulum (SR). Pb was also present in the axoplasm and mitochondria of terminal and preterminal motor axons. The presence of Pb was confirmed with x-ray elemental microanalysis. The r

ALS Along the Axons - Expression of Coding and Noncoding RNA Differs in Axons of ALS models.

Amyotrophic lateral sclerosis (ALS) is a multifactorial lethal motor neuron disease with no known treatment. Although the basic mechanism of its degenerative pathogenesis remains poorly understood, a subcellular spatial alteration in RNA metabolism is thought to play a key role. The nature of these RNAs remains elusive, and a comprehensive characterization of the axonal RNAs involved in maintaining neuronal health has yet to be described. Here, using cultured spinal cord (SC) neurons grown using a compartmented platform followed by next-generation sequencing (NGS) technology, we find that RNA

Molecular signaling how do axons die?

Axons depend critically on axonal transport both for supplying materials and for communicating with cell bodies. This chapter looks at each activity, asking what aspects are essential for axon survival. Axonal transport declines in neurodegenerative disorders, such as Alzheimer's disease, amyotrophic lateral sclerosis, and multiple sclerosis, and in normal ageing, but whether all cargoes are equally affected and what limits axon survival remains unclear. Cargoes can be differentially blocked in some disorders, either individually or in groups. Each missing protein cargo results in localized lo

Glycine fluxes in squid giant axons.

1. The influx of a number of amino acids into squid giant axons has been studied. Particular emphasis has been placed on glycine and to a lesser extent glutamate. 2. To facilitate the study of the uptake of 14C-labelled amino acids a technique was devised in which the 14C taken up was measured directly in the intact axon with a glass scintillator fibre. This technique gave results similar to the usual technique in which the axoplasm was extruded for the assay of radioactivity. 3. The changes in glycine influx with extracellular glycine concentration suggests that two saturating components are

Pathway sampling by regenerating peripheral axons.

A century ago, Ramon y Cajal described the generalized response of regenerating peripheral axons to their environment. By using mice that express fluorescent proteins in their axons, we are now able to quantify the response of individual axons to nerve transection and repair. Sciatic nerves from nonexpressing mice were grafted into those expressing a yellow variant of green fluorescent protein, then examined at 5, 7, or 10 days after repair. Regeneration was found to be a staggered process, with only 25% of axons crossing the repair in the first week. In the setting of Wallerian degeneration,

Ionized calcium concentrations in squid axons.

Values for ionized [Ca] in squid axons were obtained by measuring the light emission from a 0.1-mul drop of aequorin confined to a plastic dialysis tube of 140-mum diameter located axially. Ionized Ca had a mean value of 20 x 10(-9) M as judged by the subsequent introduction of CaEGTA/EGTA buffer (ratio ca. 0.1) into the axoplasm, and light measurement on a second aequorin drop. Ionized Ca in axoplasma was also measured by introducing arsenazo dye into an axon by injection and measuring the Ca complex of such a dye by multichannel spectrophotometry. Values so obtained were ca. 50 x 10(-9) M as

Astrocyte-induced firing in primary afferent axons.

The large-caliber primary afferents innervating the spindles of the jaw-closing muscles have their cell bodies located centrally in the mesencephalic trigeminal nucleus (NVmes). We have shown, in an acid-induced jaw muscle chronic myalgia model, that these afferents exhibit increased excitability and ectopic discharges that emerge from subthreshold membrane oscillations (SMOs) supported by a persistent sodium current (I NaP) exquisitely sensitive to extracellular Ca2+-decreases. Here, we explore if the Ca2+-binding astrocytic protein, S100β, contributes to this hyperexcitability emergence and

Dopaminergic Axons: Key Recitalists in Parkinson's Disease.

Parkinson's disease (PD) is associated with dopamine depletion in the striatum owing to the selective and progressive loss of the nigrostriatal dopaminergic neurons, which results in motor dysfunction and secondary clinical manifestations. The dopamine level in the striatum is preserved because of the innervation of the substantia nigra (SN) dopaminergic neurons into it. Therefore, protection of the SN neurons is crucial for maintaining the dopamine level in the striatum and for ensuring the desired motor coordination. Several strategies have been devised to protect the degenerating dopaminerg

Mechanisms of paresthesias arising from healthy axons.

Paresthesias are common manifestations of central and peripheral pathological processes and are due to ectopic impulse activity in cutaneous afferents or their central projections. Cutaneous afferents are more excitable than motor axons, due to differences in their biophysical properties. These differences probably include more persistent Na(+) conductance and inward rectification on cutaneous afferents, properties which probably confer greater protection from impulse-dependent conduction failure but create a greater tendency to ectopic activity. Ectopic discharges can be induced in normal aff

Effects of intravenous anaesthetics on nerve axons.

The effects of ketamine, etomidate, alphaxalone and alphadolone acetate on nodal sodium and potassium currents of frog myelinated nerve fibre have been examined. The four general anaesthetics reversibly depressed ionic currents, although they tended to be more effective in blocking potassium than sodium current. The potassium current was reduced by etomidate and alphaxalone in a time-dependent manner. This was not found for ketamine and alphadolone acetate. In addition to blocking effects on sodium current, etomidate, alphaxalone and alphadolone acetate, but not ketamine, induced a negative sh

Glycogen accumulation in axons after stretch injury.

Thin-section cytochemistry has been used to demonstrate the formation of glycogen deposits within axons after stretch injury to the optic nerve of adult guinea pigs in a model of focal axonal injury. Glycogen deposits occurred within 17% of structurally normal but, we suggest, damaged fibres within the stretched optic nerve. Adjacent fibres did not stain for glycogen. Small numbers of beta glycogen particles were present 15 min after injury within damaged axons and increasing numbers of particles occurred until 72 h. Degeneration bulbs formed by 72 h, but beta glycogen particles were sparse wi

Early development of olfactory receptor cell axons.

An electron microscope study was done on development of olfactory receptor cell axons in rat fetuses 13-17 days after conception (E13-E17). Initiation of axon outgrowth was first seen on E13. On E14, small bundles of olfactory axons, accompanied by epithelial cells, grow out of the epithelium and, by so doing, breach the basal lamina. Close examination of these epithelial cells from E14 fetuses has revealed that they can be grouped into two types, one containing a ribosome-rich, dense cytoplasm, the other containing fewer free ribosomes and a more lucent cytoplasm. The first of these types is

Peripheral Nerve unmyelinated axons following lumbar sympathectomy.

One year after bilateral lumbar sympathectomy for Raynaud's syndrome, a young woman underwent fascicular biopsy of the sural nerve to help elucidate the cause of lower limb pain. The morphological features, density, and diameter spectrum of unmyelinated fibers were normal. The sympathetic nervous system appears to contribute few, if any, axons to the total population of unmyelinated axons in this cutaneous nerve.

Quantitation of sprouting of dorsal root axons.

The axonal sprouting that occurs after denervation resulting from a spinal hemisection can be quantified. Rats were subjected to hemisection of the spinal cord at birth, and the myelinated and unmyelinated axons in dorsal roots three segments cranial and three segments caudal to the lesion were counted 1 month after surgery. The number of unmyelinated axons in the dorsal root on the side of the hemisection increased 22 percent for the roots one segment from the lesion and 13 percent for the roots two and three segments from the lesion.

Axonal cytoskeletal changes after nondisruptive axonal injury. II. Intermediate sized axons.

Earlier studies of axonal cytoskeletal responses to stretch injury in the guinea pig optic nerve, a model of nondisruptive axonal injury such as occurs in human diffuse axonal injury, have demonstrated different cytoskeletal responses between the smallest and largest axons. But these form only approximately 3% of the total number of axons in the optic nerve. It was then posited that the pathology described in the latter axons may not be representative of the pathology in the majority of axons after stretch injury. In order to test this hypothesis, we carried out a quantitative, morphological a

Axons morphometry in the human spinal cord.

Due to the technical challenges of large-scale microscopy and analysis, to date only limited knowledge has been made available about axon morphometry (diameter, shape, myelin thickness, volume fraction), thereby limiting our understanding of neuronal microstructure and slowing down research on neurodegenerative pathologies. This study addresses this knowledge gap by establishing a state-of-the-art acquisition and analysis framework for mapping axon morphometry, and providing the first comprehensive mapping of axon morphometry in the human spinal cord. We dissected, fixed and stained a human sp

Abnormal axons in the albino optic tract.

PURPOSE: There have been suggestions that the misrouting of axons at the optic chiasm leading to the abnormal ratio of crossed to uncrossed axons in the optic tract of albinos is a consequence of abnormal timing of ganglion cell axon outgrowth. The sequence of genesis of ganglion cell classes and their axon outgrowth is correlated with the deep-to-superficial distribution of their axons by size in the mammalian optic tract. Optic tract axon order in albino and normally pigmented ferrets was, therefore, examined to determine whether an abnormal pattern of retinal ganglion cell genesis and axon

Regeneration of axons in the visual system.

This review will describe the unique advantages that are offered by the visual system of mammals and other vertebrates for studying the regenerative responses of the central nervous system (CNS) to injury, and recent insights provided by such studies. In the mouse and rat visual system a variety of experimental paradigms promote survival of retinal ganglion cells (RGC) and optic nerve regeneration, probably through stimulation by neurotrophic factors (NTF) either directly, or indirectly through retinal astrocyte/Müller cell intermediary activation. NTF induce disinhibition of axon growth throu
