Access to 3-Azetidines via Halogenation of Titanacyclobutanes.

Azetidines are valuable nitrogenous heterocycles. Herein, we disclose a strategy for the modular assembly of 3-azetidines and related spirocyclic congeners featuring all-carbon quaternary centers. This approach leverages titanacyclobutanes generated from ketones or alkenes. Halogenation of these organotitanium species gives rise to functionalized alkyl dihalides that can be subsequently captured by amines to afford azetidine building blocks. This strategy facilitated the synthesis of a small molecule anti-tuberculosis drug. It also enabled access to carbon-13 isotopologs of diazaspiro[3.3]hept

Site-Selective Carbonylation of Azetidines via Copper-Catalyzed Difluorocarbene Insertion.

γ-Lactams are privileged five-membered pharmaco-phores in numerous bioactive compounds, but access to these motifs typically relies on cycloaddition/substitution chemistry involving activated substrates or CO carbonylations under harsh conditions. Here, we report a new route to functionalized γ-lactams through formal carbonylation of azetidines under nonprecious metal catalysis. The method leverages a copper-stabilized difluorocarbene to promote site-selective insertion followed by in situ hydrolysis to unmask the lactam group. In contrast to most difluorocarbene reactions that cause ring clea

Nitrate-Functionalized Azetidines for Synergistic Combustion in Ammonium Perchlorate-Based Propellant Formulations.

The development of high-performance energetic fuel components with rapid ignition characteristics is crucial for next-generation ammonium perchlorate (AP)-based composite propellants. Herein, we report a series of pyrimidine- and pyrazine-substituted azetidine nitrate esters (3, 6, and 9) and systematically investigate their structural, thermal, energetic, and combustion properties. Notably, compound 3 crystallized in two polymorphic forms (3 and 3'), revealing how crystal packing influences intermolecular interactions, density, and energetic performance despite identical molecular composition

Structural basis of malaria parasite phenylalanine tRNA-synthetase inhibition by bicyclic azetidines.

The inhibition of Plasmodium cytosolic phenylalanine tRNA-synthetase (cFRS) by a novel series of bicyclic azetidines has shown the potential to prevent malaria transmission, provide prophylaxis, and offer single-dose cure in animal models of malaria. To date, however, the molecular basis of Plasmodium cFRS inhibition by bicyclic azetidines has remained unknown. Here, we present structural and biochemical evidence that bicyclic azetidines are competitive inhibitors of L-Phe, one of three substrates required for the cFRS-catalyzed aminoacylation reaction that underpins protein synthesis in the p

Bicyclic azetidines kill the diarrheal pathogen Cryptosporidium in mice by inhibiting parasite phenylalanyl-tRNA synthetase.

Cryptosporidium is a protozoan parasite and a leading cause of diarrheal disease and mortality in young children. Currently, there are no fully effective treatments available to cure infection with this diarrheal pathogen. In this study, we report a broad drug repositioning effort that led to the identification of bicyclic azetidines as a new anticryptosporidial series. Members of this series blocked growth in in vitro culture of three Cryptosporidium parvum isolates with EC50 's in 1% serum of <0.4 to 96 nM, had comparable potencies against Cryptosporidium hominis and C. parvum, and was effec

Azetidine amino acid biosynthesis by non-haem iron-dependent enzymes.

Azetidine, a four-membered aza-cycle, is a crucial structure in many bioactive compounds and drugs. However, their biosynthesis is frequently enigmatic. Here we report the mechanism of azetidine amino acid (polyoximic acid) biosynthesis in the polyoxin antifungal pathway. Genetic, enzymological and structural experiments revealed that PolF is a member of haem-oxygenase-like dimetal oxidase and/or oxygenase (HDO) superfamily, and this enzyme alone is sufficient for the transformation of L-isoleucine (L-Ile) and L-valine to their azetidine derivatives via a 3,4-desaturated intermediate. Mechanis

Exploring the Mechanism of DABCO-Catalyzed Diastereoselective Cyclization for the Synthesis of Azetidine Nitrones.

The mechanistic details of the DABCO-catalyzed diastereoselective cyclization between β-alkyl nitroolefins and alkylidene malononitriles were elucidated by using dispersion-corrected density functional theory (DFT-D3) calculations at the M06-2X/6-311G(d,p) level with SMD solvation (dichloromethane). The reaction proceeds through four consecutive steps: (i) deprotonation and Michael addition; (ii) 1,3-proton transfer; (iii) Pinner-type cyclization; and (iv) ring contraction. Our computational results demonstrate DABCO's dual catalytic role in both proton abstraction and proton shuttle mechanism

Synthesis of 1,4-Benzodiazepines via Intramolecular C-N Bond Coupling and Ring Opening of Azetidines.

A facile and efficient synthesis of functionalized 1,4-benzodiazepine derivatives under mild conditions was developed. The CuI/N,N-dimethylglycine-catalyzed intramolecular cross-coupling reaction of 1-(2-bromobenzyl)azetidine-2-carboxamides proceeded smoothly under mild conditions to provide 1,4,9,10a-tetrahydroazeto[1,2-a]benzo[e][1,4]diazepin-10(2H)-ones. The resulting azetidine-fused 1,4-benzodiazepine compounds underwent consecutive N-methylation with methyl triflate and the opening of the four-membered heterocyclic ring by NaN3, KCN and PhSNa to produce diverse 1,4-benzodiazepine derivati

Photochemical Strategies Enable the Synthesis of Tunable Azetidine-Based Energetic Materials.

Despite their favorable properties, azetidines are often overlooked as lead compounds across multiple industries. This is often attributed to the challenging synthesis of densely functionalized azetidines in an efficient manner. In this work, we report the scalable synthesis and characterization of seven azetidines with varying regio- and stereochemistry and their application as novel azetidine-based energetic materials, enabled by the visible-light-mediated aza Paternò-Büchi reaction. The performance and stark differences in the physical properties of these new compounds make them excellent p

TLR9-activating cholesterol azetidine derivative-assisted therapeutic vaccines for cancer immunotherapy.

Therapeutic cancer vaccines show great promise for de novo induction of antigen-specific T cell responses against tumors. However, weak coordination between innate immune activation and antigen delivery remains a major obstacle to vaccine efficacy. Here, we present a cholesterol azetidine derivative-assisted polymeric carrier, Aze-Chol NP, which stimulates innate immunity and primes tumor-specific CD8+ T cell responses. Mechanistically, Aze-Chol NP induces maturation and activation of dendritic cells (DCs) through the Toll-like receptor 9 (TLR9) pathway while simultaneously delivering protein

Modular synthesis of functionalized azetidines through visible-light copper-catalyzed carboamination of 1,3-dienes with azabicyclo[1.1.0]butanes.

The strain-release reactivity of azabicyclo[1.1.0]butanes (ABBs) offers a powerful entry to substituted azetidines, yet modular access to highly functionalized, allylic amine-containing frameworks remains underdeveloped. Here, we report a visible-light copper-catalyzed four-component coupling that effects 1,4-carboamination of aryl-substituted 1,3-dienes, assembling azetidines bearing a quaternary carbon center from dienes, aryl amines, and electrophiles using ABBs. Synergistic copper/photoredox catalysis unites strain-release generation of an azetidinyl radical with allylic C-N bond formation

Electrocatalytic Access to Azetidines via Intramolecular Allylic Hydroamination: Scrutinizing Key Oxidation Steps through Electrochemical Kinetic Analysis.

Azetidines are prominent structural scaffolds in bioactive molecules, medicinal chemistry, and ligand design for transition metals. However, state-of-the-art methods cannot be applied to intramolecular hydroamination of allylic amine derivatives despite their underlying potential as one of the most prevalent synthetic precursors to azetidines. Herein, we report an electrocatalytic method for intramolecular hydroamination of allylic sulfonamides to access azetidines for the first time. The merger of cobalt catalysis and electricity enables the regioselective generation of key carbocationic inte

Azetidine-2-carboxylic acid-induced oligodendrogliopathy in vitro and the pathogenesis of multiple sclerosis.

Azetidine 2-carboxylic acid (Aze) is consumed by humans and can be misincorporated in place of proline (Pro) in myelin basic protein (MBP). In systemically treated mice Aze induced distinct oligodendroglial (OL) alterations mimicking those in multiple sclerosis (MS) patient normal-appearing white matter. Here, Aze induced an unfolded protein response (UPR), cytoplasmic MBP aggregation, apoptosis and tumor necrosis factor secretion in the human OL lineage MO13.3 cell line. These alterations were counteracted by equimolar Pro suggesting that they are due to Aze substitution for Pro in OL protein

Bicyclic azetidines target acute and chronic stages of Toxoplasma gondii by inhibiting parasite phenylalanyl t-RNA synthetase.

Toxoplasma gondii commonly infects humans and while most infections are controlled by the immune response, currently approved drugs are not capable of clearing chronic infection in humans. Hence, approximately one third of the world's human population is at risk of reactivation, potentially leading to severe sequelae. To identify new candidates for treating chronic infection, we investigated a series of compounds derived from diversity-oriented synthesis. Bicyclic azetidines are potent low nanomolar inhibitors of phenylalanine tRNA synthetase (PheRS) in T. gondii, with excellent selectivity. B

Simple and efficient access to N-tosyl beta-amino ketones and their conversion into 2,4-disubstituted azetidines.

Treatment of N-tosylaldimines with acetophenone at room temperature in the presence of BF(3).OEt(2) as a catalyst furnished the corresponding N-tosyl beta-amino ketones in high yields (77-86%) within 6-9 h. Subsequent reduction and cyclization of these compounds afforded 2,4-disubstituted N-tosylazetidines, comprising a three-step, high-yielding synthesis starting from aldimines.

Directed biosynthesis of the designer peptidyl nucleoside antibiotics featuring a simplified azetidine-containing moiety.

Fungal and fungal-like pathogen infections pose escalating threats to the global one-health, thus underscoring the urgent need to reverse this trend by accelerating the development of novel antifungal drugs. Polyoxin (POL), a group of structurally related nucleoside antibiotics, is an eco-friendly fungicide targeting fungal cell wall biosynthesis. However, the chemical diversity and therapeutic potential of this antibiotic has remained underexplored. Here, we report the rational design of a series of POL analogs that feature a simplified azetidine-containing moiety, and we have further realize

Conventional strain energies of azetidine and phosphetane: can density functional theory yield reliable results?

The conventional strain energies for azetidine and phosphetane are determined within the isodesmic, homodesmotic, and hyperhomodesmotic models. Optimum equilibrium geometries, harmonic vibrational frequencies, and corresponding electronic energies and zero-point vibrational energies are computed for all pertinent molecular systems using self-consistent field theory, second-order perturbation theory, and density functional theory and using the correlation consistent basis sets cc-pVDZ, cc-pVTZ, and cc-pVQZ. Single point fourth-order perturbation theory, CCSD, and CCSD(T) calculations using the
