Evaluating the Compositional Quality of Probiotics Containing Bacillus clausii in India.

INTRODUCTION: The quality of probiotics is crucial for their effectiveness and safety. Many studies have found discrepancies between the claimed and the actual content of microorganisms in probiotics. However, only few studies have assessed the quality of these products in India. This study aimed to evaluate the quality of various probiotics containing Bacillus clausii in India. METHODS: Eleven commercially available probiotics containing Bacillus clausii were selected: Enterogermina® (reference product), Bifilac clausii™, Entromax®, Progermina™, Entroflora™, Novogermina™, Tufpro™, Gutgermina

In vitro and in vivo evaluation of Bacillus clausii against Schistosoma mansoni.

Experimental studies and clinical trials have been showing that probiotics are promising in the prevention and control of parasite infections. B. clausii, obtained from Enterogermina®, was cultured to obtain cell-free culture supernatant (CFS) and spores to evaluate its schistosomicidal effect in vitro and in vivo against Schistosoma mansoni, respectively. For in vitro and in vivo analysis mice were infected with 120 and 50 cercariae, respectively. Couples of adult worms, recovered on day 45 of infection, were exposed to CFS. The in vivo assay was performed for 100 days, where all animals were

Bacillus clausii Attenuates 5-Fluorouracil-Induced Intestinal Mucositis in Mice.

5-Fluorouracil (5-FU) is an antimetabolite widely prescribed in cancer treatments, but its use in highly proliferative tissues can cause significant problems such as mucositis. Bacillus clausii is a probiotic commonly used for protection against acute diarrhea, gastrointestinal dysbiosis and inflammatory bowel diseases. We investigated the effect of B. clausii on 5-FU intestinal mucositis in mice. B. clausii was administered concomitantly with 5-FU on the first day and alone for the other two days. After the third day of 5-FU (450 mg/kg, ip), the animals were euthanized. Ileum samples were rem

Probiotic Bacillus Clausii Protects against Amoxicillin-evoked Reproductive Dysfunction through Oxidative Stress Modulation.

Antibiotics are frequently used in livestock, aquaculture, and poultry, resulting in their accumulation in these animals and the surrounding environment. This contamination can present health risks to humans. Research on the harmful effects of antibiotics, especially their potential to disrupt hormones in the male reproductive system, is still in its early phases. This study investigated the impact of exposure to the antibiotic amoxicillin (AMO) on male reproductive health. The aim is to shed light on the potential dangers antibiotics pose to male reproductive health and to emphasize how Bacil

Protective action of Bacillus clausii probiotic strains in an in vitro model of Rotavirus infection.

Rotavirus is the most common cause of acute gastroenteritis (AGE) in young children. Bacillus clausii (B. clausii) is a spore-forming probiotic that is able to colonize the gut. A mixture of four B. clausii strains (O/C, T, SIN and N/R) is commonly used for the treatment of AGE, and it has been demonstrated that it can reduce the duration and severity of diarrhea in children with AGE. Few studies have sought to characterize the mechanisms responsible for such beneficial effects. Intestinal effects of probiotics are likely to be strain-specific. We conducted a series of in vitro experiments inv

Accidental Intravenous Administration of Oral Bacillus clausii Probiotic (Enterogermina) in a Toddler: A Case Report.

Acute diarrheal illness is a common cause of hospitalization in toddlers, and probiotics such as oral Bacillus clausii suspension (Enterogermina) are frequently prescribed to restore gut flora. This report describes the first documented case in the United Arab Emirates of accidental intravenous administration of oral Bacillus clausii probiotic in a 17-month-old toddler. The previously healthy child presented with rotavirus-positive gastroenteritis and moderate dehydration. On the day of admission, 3 mL of Enterogermina containing approximately 2 billion spores was inadvertently given intraveno

Bacillus clausii effect on gene expression pattern in small bowel mucosa using DNA microarray analysis.

BACKGROUND: Probiotics are widely used for the cure or prevention of several clinical conditions. However, clinical decisions need to be substantiated by an analysis of the complex bacteria-host interplay in the intestinal lumen. AIMS: To identify the gene expression pattern induced by Bacillus clausii in the intestinal mucosa of healthy individuals. METHODS: Six male patients (mean age 38+/-5 years) affected by endoscopically confirmed mild oesophagitis were treated for one month with esomeprazole, and were randomly selected to receive or not B. clausii (groups I and II, respectively). Surfactin-like lipopeptides from Bacillus clausii efficiently bind to spike glycoprotein of SARS-CoV-2.

The coronavirus disease 2019 (COVID-19) rapidly spread across the globe, infecting millions and causing hundreds of deaths. It has been now around three years but still, it remained a serious threat worldwide, even after the availability of some vaccines. Bio-surfactants are known to have antiviral activities and might be a potential alternative for the treatment of SARS-CoV-2 infection. In the present study, we have isolated and purified, a surfactin-like lipopeptide produced by a probiotic bacterial strain Bacillus clausii TS. Upon purification and characterization with MALDI analysis, the m

Water miscible mono alcohols' effect on the proteolytic performance of Bacillus clausii serine alkaline protease.

In this study, our investigations showed that the increasing concentrations of all examined mono alcohols caused a decrease in the Vm, kcat and kcat/Km values of Bacillus clausii GMBE 42 serine alkaline protease for casein hydrolysis. However, the Km value of the enzyme remained almost the same, which was an indicator of non-competitive inhibition. Whereas inhibition by methanol was partial non-competitive, inhibition by the rest of the alcohols tested was simple non-competitive. The inhibition constants (KI) were in the range of 1.32-3.10 M, and the order of the inhibitory effect was 1-propan

Molecular and biochemical characterization of the chromosome-encoded class A beta-lactamase BCL-1 from Bacillus clausii.

A chromosomal beta-lactamase gene from Bacillus clausii NR, which is used as a probiotic, was cloned and expressed in Escherichia coli. It encodes a clavulanic acid-susceptible Ambler class A beta-lactamase, BCL-1, with a pI of 5.5 and a molecular mass of ca. 32 kDa. It shares 91% and 62% amino acid identity with the chromosomally encoded PenP penicillinases from B. clausii KSM-K16 and Bacillus licheniformis, respectively. The hydrolytic profile of this beta-lactamase includes penicillins, narrow-spectrum cephalosporins, and cefpirome. This chromosome-encoded enzyme was inducible in B. clausii

Antiadhesive and antibiofilm potential of Bacillus clausii derived biosurfactant against Streptococcus mutans, Enterococcus faecalis, and Candida albicans biofilms.

BACKGROUND: Oral biofilms are principal drivers of oral and dental diseases such as caries, periodontal disease, endodontic infections, and candidiasis. Conventional biocides can reduce microbial load but may irritate tissues and disturb oral homeostasis, limiting prolonged use. Biosurfactants represent a sustainable alternative strategy by modulating adhesion and biofilm structure rather than relying solely on direct bactericidal activity. Lipopeptide biosurfactants are particularly attractive due to their amphiphilic structure, surface activity, and reported antibiofilm properties. F7 is a c

Protective effects of Bacillus clausii in cyclophosphamide-induced immunosuppression and intestinal mucositis in rats: histopathological and inflammatory evaluation.

BACKGROUND: Bacillus clausii (B. clausii) is a Gram-positive, spore-forming probiotic widely used for the prevention and treatment of gastrointestinal disorders and may have protective effects against chemotherapy-induced intestinal mucositis. Cyclophosphamide (CP) can induce immunosuppression and intestinal mucosal injury, leading to inflammation and alterations in intestinal homeostasis. This study evaluated the protective effects of B. clausii on intestinal histopathology, inflammation, pro-inflammatory cytokines, and miR-146a expression in a rat model of CP-induced immunosuppression and in

Therapeutic efficacy of Bacillus clausii via NFκB/Nrf-2/HO-1 pathway modulation in a CFA-induced arthritis model.

BACKGROUND: Rheumatoid arthritis (RA) is a chronic autoimmune disease characterised by dysregulated cytokine signalling, oxidative stress, and persistent synovial inflammation. Nearly 0.24-1 % of people worldwide suffer from RA, caused by complex genetic and environmental factors leading to joint destruction and inflammation cascades. METHODS: Using a complete Freund's adjuvant (CFA)-induced murine model of arthritis, this study investigated the immunomodulatory and antioxidant properties of Bacillus clausii (1 × 108 CFU/animal/day, orally) administered as a pre-treatment, concurrent, and adj
