The majority of the prokaryotic microorganisms, especially bacteria cannot be cultivated under laboratory conditions. Irrespective of environmental samples, direct count of prokaryotes always exceeds viable count, a condition popularly coined as "Great Plate count anomaly". Our inability to culture them in isolation under laboratory conditions is due to many reasons. One possible reason could be the existence of a special group of extremophilic bacteria, such as, oligophilic bacteria, a group, that loves to grow in nutrient-deficient conditions. They are slow growing, showing unique physiologi

Nitric oxide is not only an obligatory intermediate in denitrification, but also a signalling and defence molecule of major importance. However, the basis of resistance to NO and RNS (reactive nitrogen species) is poorly understood in many microbes. The cellular targets of NO and RNS [e.g. metalloproteins, thiols in proteins, glutathione and Hcy (homocysteine)] may themselves serve as signal transducers, sensing NO and RNS, and resulting in altered gene expression and synthesis of protective enzymes. The properties of a number of such protective mechanisms are outlined here, including globins,

Biofilms are the predominant lifestyle for bacteria. Biofilm abundance makes understanding their behavior important for different research domains. These domains can have different information and reporting standards. Information reporting standards have been proposed for some biofilm research methods. Standard methods and consensus standards have also been promulgated. We propose dimensional analysis to encourage abundant information reporting and facilitate comparative analysis. Dimensional analysis utilizes physical system parameters to generate generalizable model data. We review dimension

Bacterial spores of the genera Bacillus and Clostridium are extremely resistant against desiccation, heat and radiation and involved in the spread and pathogenicity of health relevant species such as Bacillus anthracis (anthrax) or Clostridium botulinum. While the resistance of spores is very well documented, underlying mechanisms are not fully understood. In this study we show, by cryo-electron microscopy of vitreous sections and particular resin thin section electron microscopy, that dormant Bacillus spores possess highly ordered crystalline core structures, which contain the DNA, but only i

The continuous flow of billions of birds between Africa and Europe creates an "ecological bridge" between physically remote areas. Migratory birds fly south from their breeding grounds during late summer/fall and fly back in spring. These movements regulate the spread of internal and external parasites, as well as pathogens of potential public health concern. The aim of the present study was to investigate the possible introduction of exotic tick species and tick-borne pathogens into Europe via migratory birds. At the bird observatory of Ventotene island (Italy), 443 feeding ticks were collect

Cockroaches have the potential to disseminate bacteria in their environments and therefore a systematic review and meta-analysis was conducted to assess the state-of-the-art of our knowledge regarding bacterial contaminants of cockroaches. At least 78 bacterial species and 42 genera from 24 families and 11 orders of bacteria were reported to have contaminated cockroaches. At least 61, 42, 12, 13, 7, and 16 bacterial species have contaminated Blattella germanica (L.) (Blattaria: Ectobiidae), Periplaneta americana (L.) (Blattaria: Blattidae), Blatta orientalis (L.) Periodontal disease progression requires the simultaneous presence of high numbers of pathogens, low numbers of compatible or beneficial species, a conducive local environment, and a susceptible host. Effective therapy acts by altering one or more of these factors. Data from an ongoing study were used to examine the biological basis of treatment success or failure. Seventeen subjects showing disease progression were treated by Widman flap surgery at deep sites, scaling at shallow sites, and 1 of 4 randomlyassigned, systemically-administered adjunctive agents including amoxicillin/clavulanate p

Collective bacterial condensation arises from positive feedback between the ability of bacteria to generate chemical gradients in their environment and their chemotactic ability to follow those gradients. This feedback drives the spontaneous formation of local cell accumulations, characterized by sharp cell-density gradients, even in the absence of physical boundaries. By following the dynamics of bacterial condensation in uniform acidic environments, we show that condensation is critically constrained by the spontaneous emergence of correlated bacterial swimming and the associated active turb

Biofilms are a major contributor to antibiotic resistance and persistence in hospital environments. These bacterial communities form protective structures that shield microbes from various stressors, such as oxidative stress, pH fluctuations, osmotic pressure, and nutrient deprivation. As our understanding of biofilms has grown, it has become evident that metals play a crucial role in several aspects of biofilm biology. Metals are involved in regulating biofilm formation, facilitating communication and competition among bacteria, and supporting the structural integrity and adherence of bacteri

This paper summarizes the rationale for the application of microbiology in the management of destructive periodontal diseases. The subgingival microbiota in patients with severe periodontitis is complex and contains high numbers of obligate anaerobic bacteria as well as facultative micro-organisms. It has become clear that major differences exist in the composition of the subgingival microflora. These differences are not only quantitative but also qualitative. Difference in plaque composition is the basis for the application of clinical microbiology in the management of periodontal disease. Se

Mirror bacteria are hypothetical synthetic organisms built from molecules of opposite chirality to those used throughout nature. This primer discusses their potential dangers: because of their chiral mismatch to the natural environment, mirror bacteria could plausibly evade predation, immunity and other ecological control mechanisms to a degree never seen before. This property could simultaneously make them unprecedented invasive species and dangerous pathogens.

Bacteria commonly coexist within communities where they compete for limited space and resources. These communities play an essential role in supporting a diversity of critical ecosystems, from regulating metabolism within the gut to cycling nutrients within Earth's atmosphere. Mechanical forces have long been recognized as critical regulators of cell function within eukaryotic systems. Microbiology, however, has largely focused on the influence of the chemical environment, with most studies of bacterial populations performed in well-mixed cultures. An important aspect of bacteria living in con

Being the principal causative agent of bacterial pneumonia, otitis media, meningitis and septicemia, the bacterium Streptococcus pneumoniae is a major global health problem. To highlight the molecular basis of this problem, we have portrayed essential biological processes of the pneumococcal life cycle in eight watercolor paintings. The paintings are done to a consistent nanometer scale based on currently available data from structural biology and proteomics. In this review article, the paintings are used to provide a visual review of protein synthesis, carbohydrate metabolism, cell wall synth

Epigenetic phase variation (ePV) represents a sophisticated regulatory mechanism by which bacteria rapidly adapt to environmental stressors without permanent genetic alterations. This review examines the molecular mechanisms underlying bacterial ePV mediated by DNA methylation, with particular emphasis on restriction-modification systems, orphan methyltransferases, and their roles in stress adaptation. ePV systems enable bacteria to generate phenotypically diverse populations through reversible ON/OFF switching of gene expression, facilitated by phase-variable DNA methyltransferases. These sys

Gliding motility is one of the more elegant tricks in the microbial playbook, allowing cells to move smoothly along an external surface. Unlike swimming in bulk fluid, gliding requires contact with a biotic or abiotic surface, imposing strong physical constraints on how forces are generated, transmitted, and dissipated. It is an active, energy-dependent process that operates without flagella, instead relying on specialized molecular machines that couple dynamic surface adhesins to motion. In this mini-review, we summarize recent advances that illuminate how distinct molecular motors converge o

Post-transfusion bacterial sepsis is infrequent. It is, however, associated with a high mortality due to septic shock. This reflects the release of endotoxin from gram negative bacteria. Lesser transfusion reactions are usually under-reported. These are frequently caused by gram positive bacteria. Gram positive species such as staphylococci and other skin surface organisms may be cultured from platelets stored at room temperature. Typically, gram negative "psychrophyllic" species which survive storage at 4 degrees C are cultured from stored refrigerated blood implicated in transfusion reaction
