From predicted bacteriocins to ecological function.

Genome and metagenome mining have uncovered large repertoires of predicted bacteriocin loci, but ecological interpretation has lagged behind discovery. This review uses bacteriocins as a focused model for a broader problem in antimicrobial gene prediction: sequence identifies encoded potential, but it does not establish expression, product deployment, target engagement, or community-level consequence. We distinguish four forms of bacteriocin "silence": transcriptional silence, where loci are not detectably expressed under tested conditions; conditional silence, where expression or activity eme

[Bacteriophages and bacteriocins of the genus Listeria].

Since the discovery of the first Listeria bacteriophage by Schultz in 1945, more than 219 phages were isolated for L. monocytogenes, L. invanovii, L. innocua, L. seeligeri and L. welshimeri. To date, no phage for L. murrayi and L. grayi is described in the literature. Unless two phages found by Jasińska in a water sample, all the other phages, when the origin is mentioned, were isolated from lysogenic strains, with or without induction. Electron microscopic examination of 55 Listeria phages revealed that two L. innocua phages belonged to the Myoviridae family (tail with contractile sheath) whe

A bacteriocin of Actinobacillus actinomycetemcomitans.

An inhibitory factor from Actinobacillus actinomycetemcomitans Y4 was isolated, and its properties indicated that it was a bacteriocin (actinobacillicin). The bacteriocin was active against Streptococcus sanguis strains, Streptococcus uberis (FDC1), and Actinomyces viscosus T14 as well as other strains of A. actinomycetemcomitans, but not against other crevicular bacteria, including other streptococci and actinomycetes. The activity of this bacteriocin was inhibited by pronase, trypsin, and heat (45 min at 56 degrees C) but not by DNase, RNase, phospholipase, exposure to UV light, or low pH (1

Are bacteriocins underexploited? Novel applications for old antimicrobials.

Bacteriocins are ribosomally synthesized (poly)peptides produced by almost all prokaryotic lineages. Bacteriocins from lactic acid bacteria (LAB) and bacteriocin-producer probiotic organisms have been thoroughly studied due to their wide spectra of action, the long-term use in food fermentations and the consideration of these microorganisms as beneficial for human beings. Most of the studies on the biotechnological application of diverse bacteriocins have been focused on their use as food preservatives, nisin being the prototype successfully used in alimentation. However, bacteriocins from LAB

Purification of bacteriocins produced by lactic acid bacteria.

Bacteriocins are antibacterial substances of a proteinaceous nature that are produced by different bacterial species. Lactic acid bacteria (LAB) produce biologically active peptides or protein complexes that display a bactericidal mode of action almost exclusively toward Gram-positive bacteria and particularly toward closely related species. Generally they are active against food spoilage and foodborne pathogenic microorganisms including Bacillus cereus, Clostridium perfringens, Staphylococcus aureus, and Listeria monocytogenes. There is an increased tendency to use natural occurring metabolit

Bacteriocins for Control of Listeria spp. in Food.

Foodborne outbreaks of listeriosis caused by Listeria monocytogenes have contributed to public consciousness about bacterial pathogens involved with foodborne disease. Major concerns with L. monocytogenes are its high mortality rate, wide distribution on raw products, growth at low temperatures, and its ability to establish itself in various food-processing environments. These concerns have prompted the examination of novel approaches, including the use of antimicrobial peptides, or bacteriocins, to combat its survival in foods. Bacteriocins from lactic acid bacteria have received much attenti

Bacteriocin Typing of Vibrio cholerae.

Bacteriocins of Vibrio cholerae have been demonstrated against enterobacterial and vibrio indicator organisms by conventional techniques. Abundant bacteriocin production took place on casein hydrolysate-yeast extract, tryptic soy, digest broth, proteose-peptone, and neopeptone agars. Essential factors were a citrate-phosphate buffer concentration of 0.5 to 0.7%, at pH 7.5 to 7.6, and cold shock. Thermal treatment of indicator organisms at 45 C for 12 min increased the percentage of typable strains. The bacteriocins of V. cholerae appeared to be powerful diffusible bactericidal agents. By using

LlpB represents a second subclass of lectin-like bacteriocins.

Bacteriocins are secreted bacterial proteins that selectively kill related strains. Lectin-like bacteriocins are atypical bacteriocins not requiring a cognate immunity factor and have been primarily studied in Pseudomonas. These so-called LlpAs are composed of a tandem of B-lectin domains. One domain interacts with d-rhamnose residues in the common polysaccharide antigen of Pseudomonas lipopolysaccharide (LPS). The other lectin domain is crucial for interference with the outer membrane protein assembly machinery by interacting with surface-exposed loops of its central component BamA. Via genom

Genomic analyses of pneumococci reveal a wide diversity of bacteriocins - including pneumocyclicin, a novel circular bacteriocin.

BACKGROUND: One of the most important global pathogens infecting all age groups is Streptococcus pneumoniae (the 'pneumococcus'). Pneumococci reside in the paediatric nasopharynx, where they compete for space and resources, and one competition strategy is to produce a bacteriocin (antimicrobial peptide or protein) to attack other bacteria and an immunity protein to protect against self-destruction. We analysed a collection of 336 diverse pneumococcal genomes dating from 1916 onwards, identified bacteriocin cassettes, detailed their genetic composition and sequence diversity, and evaluated the

Therapeutic enhancement of newly derived bacteriocins against Giardia lamblia.

Trials for identifying efficient anti-giardial agents are still ongoing. Nowadays, bacteriocins have attracted the attention as potential antimicrobial compounds. For the first time, the current study evaluated the therapeutic efficacy of bacteriocins derived from newly isolated Egyptian strains of probiotics Lactobacilli; L. acidophilus (P106) and L. plantarum (P164) against Giardia lamblia. Bacteriocins' efficacy was evaluated both in vitro; by growth inhibition and adherence assays, and in vivo; through estimation of parasite density, intestinal histopathological examination and ultrastruct

Antibacterial activities of bacteriocins: application in foods and pharmaceuticals.

Bacteriocins are a kind of ribosomal synthesized antimicrobial peptides produced by bacteria, which can kill or inhibit bacterial strains closely-related or non-related to produced bacteria, but will not harm the bacteria themselves by specific immunity proteins. Bacteriocins become one of the weapons against microorganisms due to the specific characteristics of large diversity of structure and function, natural resource, and being stable to heat. Many recent studies have purified and identified bacteriocins for application in food technology, which aims to extend food preservation time, treat

Angicin, a novel bacteriocin of Streptococcus anginosus.

As a conserved defense mechanism, many bacteria produce antimicrobial peptides, called bacteriocins, which provide a colonization advantage in a multispecies environment. Here the first bacteriocin of Streptococcus anginosus, designated Angicin, is described. S. anginosus is commonly described as a commensal, however it also possesses a high pathogenic potential. Therefore, understanding factors contributing to its host colonization and persistence are important. A radial diffusion assay was used to identify S. anginosus BSU 1211 as a potent bacteriocin producer. By genetic mutagenesis the bac

Partial characterization of Yersinia intermedia bacteriocin.

Yersinia intermedia produces a temperature-dependent (25 degrees C) bactericidal substance that is active against other Yersinia species. Crude preparations of the inhibitory substance were inactivated by chymotrypsin, trypsin, pronase, and heating but were not affected by lipolytic enzymes, chloroform, or other organic solvents. These data suggest that the active molecule is a bacteriocin of a proteinaceous nature. The molecular weight of the bacteriocin was estimated to be greater than 14,000. Exposure of agar fragments containing the active component to a pH range of 3 to 11 did not affect

Combined Action of Antibiotics and Bacteriocins against Vancomycin-Resistant Enterococci.

Antibiotics have been one of the most important discoveries in the area of applied medical microbiology; however, as a result of various factors, we are currently facing a dramatic and relatively dangerous increase in the number of cases of antibiotic resistance, and the need for new types of antimicrobials continues to grow. New approaches are needed to combat antibiotic-resistant pathogens. Bacteriocins, as part of the group of antimicrobial peptides, can be considered as alternatives and/or complements to known antibiotics. Their narrow spectra of activity can be explored for the control of

Broadening and Enhancing Bacteriocins Activities by Association with Bioactive Substances.

Bacteriocins are antimicrobial peptides some of which are endowed with antiviral, anticancer and antibiofilm properties. These properties could be improved through synergistic interactions of these bacteriocins with other bioactive molecules such as antibiotics, phages, nanoparticles and essential oils. A number of studies are steadily reporting the effects of these combinations as new and potential therapeutic strategies in the future, as they may offer many incentives over existing therapies. In particular, bacteriocins can benefit from combination with nanoparticles which can improve their

Bacteriocins in Veterinary Medicine: From Antibiotic Limitations to Targeted Solutions.

Antimicrobial resistance (AMR) has emerged as one of the foremost global threats to public health, with the veterinary sector, responsible for nearly three-quarters of global antimicrobial consumption, representing an underappreciated epicenter of this crisis. Despite the extensive literature on bacteriocins as antibiotic alternatives, most reviews focus on human medicine or food preservation, leaving a conspicuous gap in evidence specific to veterinary medicine. The present review addresses this gap by examining the molecular basis of bacteriocin activity (lipid II, bacterial RNA polymerase,

The clinical praxis of bacteriocins as natural anti-microbial therapeutics.

In recent decades, the excessive use of antibiotics has resulted in a rise in antimicrobial drug resistance (ADR). Annually, a significant number of human lives are lost due to resistant infectious diseases, leading to around 700,000 deaths, and it is estimated that by 2050, there could be up to 10 million casualties. Apart from their possible application as preservatives in the food sector, bacteriocins are gaining acknowledgment as potential clinical treatments. Not only this, these antimicrobial peptides have revealed in modulating the host immune system producing anti-inflammatory and anti

Promising Antimicrobial Activity and Synergy of Bacteriocins Against Mycobacterium tuberculosis.

In this study, we assessed the potential of bacteriocins and their in vitro synergistic effects in combination with anti-tuberculosis drugs against Mycobacterium tuberculosis. We evaluated the in vitro activity of chemically synthesized bacteriocins in combination with rifampicin (RIF), ofloxacin, and moxifloxacin against the reference M. tuberculosis H37Rv and a clinical-resistant strain. We first screened the bacteriocin PARAGEN collection and found active bacteriocins. We then determined their minimal inhibitory concentration (MIC), minimal bactericidal concentration, and their fractional i

Novel Selective Inhibition of Lactobacillus iners by Lactobacillus-Derived Bacteriocins.

Lactobacillus iners is often associated with vaginal dysbiosis and bacterial vaginosis (BV), which are risk factors for adverse gynecological and obstetric outcomes. To discover natural inhibitors of L. iners, cell-free culture supernatants (CFSs) from 77 vaginal human Lactobacillus strains and 1 human intestinal strain were screened for inhibitory activity. Three active strains were identified, and Lactobacillus paragasseri K7 (K7), a human intestinal strain, produced the most potent L. iners-inhibitory activity. The active material was purified from the K7 CFS and yielded three active peptid

Antimicrobial activity of Enterococcus-derived bacteriocins against multidrug-resistant Pseudomonas aeruginosa.

Pseudomonas aeruginosa is a key concern in clinical settings due to its high level of resistance to antibiotics, making infections given rise to this bacterium very problematic to treat. The rise of multidrug-resistant bacteria poses a danger to treatments and stresses the necessity to find new antimicrobial drugs. In a neoteric study, P. aeruginosa was found in a suction machine tube, affirming the importance of identifying and managing potential sources of infection in healthcare facilities-many strains of Enterococcus faecium output bacteriocins, which are antimicrobial compounds.
