Chemical crosslinking of bacteriophage phi 6 nucleocapsid proteins.

phi 6 is a lipid-containing dsRNA bacteriophage of Pseudomonas syringae. Its nucleocapsid (NC) has common features with Reoviridae core particles. We report here the crosslinking of phi 6 NC proteins with cleavable 12-A span chemical crosslinker, dithiobis(succinimidyl propionate). The crosslinked complexes were analyzed in two-dimensional polyacrylamide gels or by using monoclonal antibodies to uncleaved protein complexes in one-dimensional protein gels. The NC surface protein (P8) forms a series of multimeric homopolymers. The phi 6 lytic enzyme, protein P5, is associated with P8 on the NC s

Antiviral Characterization of Advanced Materials: Use of Bacteriophage Phi 6 as Surrogate of Enveloped Viruses Such as SARS-CoV-2.

The bacteriophage phi 6 is a virus that belongs to a different Baltimore group than SARS-CoV-2 (group III instead of IV). However, it has a round-like shape and a lipid envelope like SARS-CoV-2, which render it very useful to be used as a surrogate of this infectious pathogen for biosafety reasons. Thus, recent antiviral studies have demonstrated that antiviral materials such as calcium alginate hydrogels, polyester-based fabrics coated with benzalkonium chloride (BAK), polyethylene terephthalate (PET) coated with BAK and polyester-based fabrics coated with cranberry extracts or solidified han

Persistence of Bacteriophage Phi 6 on Porous and Nonporous Surfaces and the Potential for Its Use as an Ebola Virus or Coronavirus Surrogate.

The infection of health care workers during the 2013 to 2016 Ebola outbreak raised concerns about fomite transmission. In the wake of the coronavirus disease 2019 (COVID-19) pandemic, investigations are ongoing to determine the role of fomites in coronavirus transmission as well. The bacteriophage phi 6 has a phospholipid envelope and is commonly used in environmental studies as a surrogate for human enveloped viruses. The persistence of phi 6 was evaluated as a surrogate for Ebola virus (EBOV) and coronaviruses on porous and nonporous hospital surfaces. Phi 6 was suspended in a body fluid sim

Evaluating the Effect of Coated Filters and Pre-Filters on the Infectivity and Concentration of Aerosolized Phi6 Virus.

Development of efficient indoor air filtration technologies has gained prominence following the coronavirus disease 2019 (COVID-19) pandemic. The goal of this study is to evaluate the anti-viral efficacy of various copper and silver-based surface coatings for air filters using a standardized bioaerosol chamber and the enveloped bacteriophage Phi6 as a surrogate for respiratory viruses. Surface testing confirmed that all coatings exhibited anti-viral activity, with the specific copper-based coating formulation (B4) achieving complete inactivation of Phi6 virus within 10 min, A6 in 25 min, and V

Sunlight Inactivation of Influenza A Virus and Bacteriophages on an Acrylic Surface.

Seasonal epidemics and pandemics of influenza cause a large public health burden, with transmission occurring via contact with contaminated fomites. Sunlight has been shown to inactivate viruses in water, but its effect on viruses on surfaces remains poorly understood. This study investigated the inactivation of bacteriophage MS2, bacteriophage Phi6, and influenza A virus H1N1 (IAV) on an acrylic surface exposed to both natural and simulated sunlight. Findings demonstrate that sunlight can rapidly inactivate viruses on an acrylic surface, with the time required for 99% inactivation ranging fro

Migration enhances adaptation in bacteriophage populations evolving in ecological sinks.

Migration between populations can be a major evolutionary force. However, some disagreement exists as to precisely how migration affects population adaptation. Some theories emphasize the inhibitory effects of gene flow between locally adapted populations, whereas others propose that migration can enhance adaptation. Migration has also been theorized to rescue sink populations from extinction. In our experiments, we serially passaged bacteriophage Φ6 host range mutants under sink conditions on a novel host while manipulating the source and number of migrants into these experimental populations

Optimization of An Enveloped Virus Surrogate, Bacteriophage Phi6, Recovery from Hands.

Surfaces contaminated with enveloped viruses, such as severe acute respiratory syndrome coronavirus 2 and influenza virus, can potentially spread illness via hand contact. Often, the efficacy of hand hygiene interventions relies on virus recovery from hands. However, the recovery of bacteriophage phi6 (Φ6), a recommended surrogate for enveloped viruses, from the entire hands using the ASTM E2011-21 standard has not been optimized. For Φ6 recovery from the hands, three eluents [lysogeny broth (LC), tryptic soy broth (TSB), and 1.5% beef extract (BE)] and three recovery methods [glove juice meth

Rapid virucidal activity of an air sanitizer against aerosolized MS2 and Phi6 phage surrogates for non-enveloped and enveloped vertebrate viruses, including SARS-CoV-2.

An air sanitizer was evaluated using an aerobiology protocol, compliant with the U.S. Environmental Protection Agency's Air Sanitizer Guidelines, for virucidal activity against bacteriophages Phi6 and MS2 (used as surrogates for enveloped and non-enveloped human pathogenic viruses). The phages were suspended in a medium containing a tripartite soil load simulating body fluids and aerosolized using a six-jet Collison nebulizer in an enclosed 25 m3 aerobiology chamber at 22 ± 2°C and 50 ± 10% relative humidity. The air sanitizer was sprayed into the chamber for 30 s. Viable phages in the air wer

Experimental evidence that source genetic variation drives pathogen emergence.

A pathogen can readily mutate to infect new host types, but this does not guarantee successful establishment in the new habitat. What factors, then, dictate emergence success? One possibility is that the pathogen population cannot sustain itself on the new host type (i.e. host is a sink), but migration from a source population allows adaptive sustainability and eventual emergence by delivering beneficial mutations sampled from the source's standing genetic variation. This idea is relevant regardless of whether the sink host is truly novel (host shift) or whether the sink is an existing or rela

Narrowed host ranges do not constrain future host range expansion in RNA phage Φ6.

RNA viruses frequently shift between different host species and emerge on novel hosts. Part of this evolutionary process can involve specialization, when viruses adapt to increase their fitness on a particular host, often at the expense of their ability to infect other hosts. This trajectory has the potential to lead to extreme host narrowing, which excludes all other previously accessible hosts. The consequences of the evolutionary history of host specialization on a virus's future evolutionary potential are understudied. Using previously evolved specialized strains of model dsRNA bacteriopha

Temperature-modulated persistent dsRNA phage infection selectively excludes superinfection and promotes generation of new viral variants.

UNLABELLED: Persistent viral infections are common in nature and can play a significant role in the ecology and evolution of the host organism. However, our understanding of these prolonged virus-host interactions remains limited. This study characterizes the persistent infection strategy of Pseudomonas phage phi6, an enveloped virus with a tri-segmented, double-stranded (ds)RNA genome that infects Pseudomonas syringae. Phi6 is predominantly lytic but can alternatively establish a non-productive chronic infection (carrier cell state) in which viral particles assemble and replicate viral RNA wi

A bioassay-based protocol for chemical neutralization of human faecal wastes treated by physico-chemical disinfection processes: A case study on benzalkonium chloride.

In situ physico-chemical disinfection of high risk faecal waste is both effective and widely used as a sanitation management strategy for infection prevention and control. Systematic tests where the performance of alternative physico-chemical disinfection methods is systematically compared and optimized must be based on reliable protocols. These protocol are currently not adequately addressing the neutralization related issues: the neutralization of the tested disinfectant after specified conditions of concentration and contact time (CT) is necessary to prevent continued disinfection after the

Inhibition of coxsackievirus B3 and related enteroviruses by antiviral short interfering RNA pools produced using phi6 RNA-dependent RNA polymerase.

Coxsackievirus B3 (CBV3) is a member of the human enterovirus B species and a common human pathogen. Even though much is known about the enteroviral life cycle, no specific drugs are available to treat enterovirus infections. RNA interference (RNAi) has evolved to be an important tool for antiviral experimental therapies and gene function studies. We describe here a novel approach for RNAi against CBVs by using a short interfering (siRNA) pool covering 3.5 kb of CBV3 genomic sequence. The RNA-dependent RNA polymerase (RdRP) of bacteriophage phi6 was used to synthesize long double-stranded RNA

Virus leakage through natural membrane condoms.

The authors determined virus leakage from condoms made from processed sheep caecum using two viral probes simultaneously. They poured a mixture of two viruses, the bacteriophage, phi X174 (4 X 10(7) pfu/ml), and the human pathogen, herpes simplex virus (about 1 X 10(6) pfu/ml), in a buffered solution into condoms, which were suspended into beakers also containing buffered solution. The authors then assayed aliquots from the beakers to measure the extent of virus leakage from the condoms. With one brand of condom, 10 out of 24 samples leaked small amounts of phi X174; with the other brand of co

Disinfection of an ambulance using a compact atmospheric plasma device.

AIMS: The worldwide spread of the coronavirus SARS-CoV-2 has highlighted the need for fast and simple disinfection processes, amongst others for ambulance cars on site. To overcome current drawbacks regarding room disinfection, the use of cold atmospheric plasma in remote operation represents a promising alternative for the disinfection of larger volumes. In this study, a compact plasma system was evaluated regarding its disinfection efficiency inside an ambulance car. METHODS AND RESULTS: The developed plasma device is based on a dielectric barrier discharge (DBD) and operates with ambient a

UV Inactivation of SARS-CoV-2 across the UVC Spectrum: KrCl* Excimer, Mercury-Vapor, and Light-Emitting-Diode (LED) Sources.

Effective disinfection technology to combat severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) can help reduce viral transmission during the ongoing COVID-19 global pandemic and in the future. UV devices emitting UVC irradiation (200 to 280 nm) have proven to be effective for virus disinfection, but limited information is available for SARS-CoV-2 due to the safety requirements of testing, which is limited to biosafety level 3 (BSL3) laboratories. In this study, inactivation of SARS-CoV-2 in thin-film buffered aqueous solution (pH 7.4) was determined across UVC irradiation wavelengths

Handwashing and Ebola virus disease outbreaks: A randomized comparison of soap, hand sanitizer, and 0.05% chlorine solutions on the inactivation and removal of model organisms Phi6 and E. coli from hands and persistence in rinse water.

To prevent Ebola transmission, frequent handwashing is recommended in Ebola Treatment Units and communities. However, little is known about which handwashing protocol is most efficacious. We evaluated six handwashing protocols (soap and water, alcohol-based hand sanitizer (ABHS), and 0.05% sodium dichloroisocyanurate, high-test hypochlorite, and stabilized and non-stabilized sodium hypochlorite solutions) for 1) efficacy of handwashing on the removal and inactivation of non-pathogenic model organisms and, 2) persistence of organisms in rinse water. Oral Immunization of Rabbits with S. enterica Typhimurium Expressing Neisseria gonorrhoeae Filamentous Phage Φ6 Induces Bactericidal Antibodies Against N. gonorrhoeae.

All Neisseria gonorrhoeae strains whose DNA sequences have been determined possess filamentous phage DNA sequences. To ascertain if phage encoded proteins could form the basis of a gonococcal vaccine, rabbits were orally infected with S. enterica Typhimurium strain χ3987 harboring phagemid NgoΦ6 fm. The elicited sera contained large quantities of anti-phage IgG and IgA antibodies that bound to the surface of N. gonorrhoeae cells, as shown by indirect fluorescent analysis and flow cytometry. Persistence of Coronavirus Surrogates on Meat and Fish Products during Long-Term Storage.

Multiple pathways of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) transmission have been examined, and the role of contaminated foods as a source of SARS-CoV-2 exposure has been suggested. As many cases of SARS-CoV-2 have been linked to meat processing plants, it may be that conditions in live animal markets and slaughterhouses or meat processing plant procedures transfer viral particles to meat, poultry, and seafood during animal slaughter, processing, storage, or transport. Because of the potential for contamination of foods such as beef, chicken, pork, or fish, the goal of t

Molecular evolution of bacteriophages: evidence of selection against the recognition sites of host restriction enzymes.

Restriction enzymes produced by bacteria serve as a defense against invading bacteriophages, and so phages without other protection would be expected to undergo selection to eliminate recognition sites for these enzymes from their genomes. The observed frequencies of all restriction sites in the genomes of all completely sequenced DNA phages (T7, lambda, phi X174, G4, M13, f1, fd, and IKe) have been compared to expected frequencies derived from trinucleotide frequencies. Attention was focused on 6-base palindromes since they comprise the typical recognition sites for type II restriction enzyme
