Disrupted cells of Bdellovibrio bacteriovorus exhibited adenosine triphosphatase activity, 60 to 80% of which was in the soluble fraction. Dicyclohexylcarbodiimide did not inhibit the adenosine triphosphatase activity in membrane particles. The particles did not show energy-linked transhydrogenase activity. The activity of non-energy-linked transhydrogenase as well as the rate of oxygen consumption were higher in membrane particles of the host-independent strain than in the host-dependent strains. The uptake of amino acid uptake was inhibited by cyanide and by carbonyl cyanide p-trifluorometho

Predatory bacteria such as Bdellovibrio bacteriovorus exert strong top-down control on Gram-negative bacterial populations, yet the prey surface determinants governing susceptibility to predation remain incompletely resolved. Lipopolysaccharide (LPS) is a dominant structural component of the Gram-negative outer membrane and has long been implicated in predator attachment and invasion, but how specific features of LPS architecture influence predation efficiency remains unclear. Here, the susceptibility of Escherichia coli K-12 mutants bearing defined truncations in LPS core biosynthesis was sys

Bdellovibrio bacteriovorus is a gram-negative Proteobacterium that has evolved to use a novel, predatory, intrabacterial lifestyle. Once encountered under a microscope, in a lecture, or a textbook, it is seldom forgotten. B. bacteriovorus speedily collides with, enters, modifies, and kills other gram-negative bacteria. Residing inside the periplasm of the dead prey for several hours, it uses prey molecules for growth and replication, only bursting out from that temporary home when its progeny are fully formed predators themselves. Rapid motility whilst hunting contrasts with sessility during p

We evaluated the bactericidal activity of Bdellovibrio bacteriovorus, strain HD100, within blood sera against bacterial strains commonly associated with bacteremic infections, including E. coli, Klebsiella pneumoniae and Salmonella enterica. Tests show that B. bacteriovorus HD100 is not susceptible to serum complement or its bactericidal activity. After a two hour exposure to human sera, the prey populations decreased 15- to 7,300-fold due to the serum complement activity while, in contrast, the B. bacteriovorus HD100 population showed a loss of only 33%. Dot blot analyses showed that this is

Cows, horses, pigs and ducks have been found to contain Bdellovibrio bacteriovorus in their intestine and to constantly excrete them with feces into the environment. These microorganisms have not been detected in the feces of man, white mice, frogs and fish. Bdellovibrio, if introduced together with Shigella or after them, prevent the development of keratoconjunctivitis in some of the rabbits. No manifestations of the lytic activity of Bdellovibrio in relation to Salmonella and Vibrio cholerae have been observed in the intestine of white mice and young rabbits.

Predation of Chromobacterium piscinae by Bdellovibrio bacteriovorus HD100 was inhibited in dilute nutrient broth (DNB) but not in HEPES. Experiments showed that the effector responsible was present in the medium, as cell-free supernatants retained the ability to inhibit predation, and that the effector was not toxic to B. bacteriovorus Violacein, a bisindole secondary metabolite produced by C. piscinae, was not responsible. Further characterization of C. piscinae found that this species produces sufficient concentrations of cyanide (202 µM) when grown in DNB to inhibit the predatory activity o

BACKGROUND: Neonatal sepsis is associated with high rates of morbidity and mortality, long hospital stays and high cost of care, thereby inflicting a burden on health care systems. Oral care with breast milk has been shown to modify the intestinal tract microbiota and immune system. Herein, we attempted to identify probiotics that may be beneficial to prevent or treat neonatal sepsis.
METHODS: This was a secondary analysis comparing the microbiota during oropharyngeal care in very-low-birth-weight infants with and without clinical sepsis. Oral samples were collected before oral feeding was in

Bdellovibrio and like organisms (BALOs) are a unique bacterial group that live by predating on other bacteria, consuming them from within to grow and replicate before the progeny come out to complete the life cycle. The mechanisms by which these predators recognize their prey and differentiate them from nonprey bacteria, however, are still not clear. Through genetic knockout and complementation studies in different Escherichia coli strains, we found that Bdellovibrio bacteriovorus strain 109J recognizes outer membrane porin F (OmpF) on the E. coli surface and that the activity of the E. coli E

Multiple drug-resistant bacterial infections have led to an increasing need for an effective disease outbreak response system, such as the One Health platform. While interactions among humans, animals, and ecosystems are crucial and inevitable for survival, the intercommunity transfer of drug resistance genes and the resulting diseases are matters of concern. The concept of bacterial predation and its application in different fields have attracted interest as a natural alternative to traditional antibiotics. Bdellovibrio bacteriovorus, an obligate bacterial predator, preys on a wide range of o

Bdellovibrio bacteriovorus is a bacterium which preys upon and kills Gram-negative bacteria, including the zoonotic pathogens Escherichia coli and Salmonella. Bdellovibrio has potential as a biocontrol agent, but no reports of it being tested in living animals have been published, and no data on whether Bdellovibrio might spread between animals are available. In this study, we tried to fill this knowledge gap, using B. bacteriovorus HD100 doses in poultry with a normal gut microbiota or predosed with a colonizing Salmonella strain. In both cases, Bdellovibrio was dosed orally along with antaci

Fourteen strains of Bdellovibrio-like organisms were isolated from cultured fish ponds using Aeromonas hydrophila J-1 as host, one of them formed large plaques after 48 h of incubation at 28 degrees C on a double layer plate, designated as Bdellovibrio C-1. The Bdellovibrio was confirmed by electron microscopy and PCR amplification of Bdellovibrio-specific 16S rDNA. The optimum temperature for the growth of BdC-1 was between 15-37 degrees C and with optimal activity at temperatures of 25-30 degrees C. The ability of BdC-1 to lyse A. hydrophila was similar in the pH range of 6.5 to 8.5. It can

During intraperiplasmic growth of Bdellovibrio bacteriovorus 109J, the substrate cell surface becomes more hydrophobic. This was shown (i) by comparing the sensitivity to hydrophobic antibiotics of wild-type and lipopolysaccharide mutant strains of Salmonella typhimurium to that of the bdellovibrio growing on these strains and (ii) by measuring the binding efficiency of these strains, Escherichia coli, and their derived bdelloplasts to octyl Sepharose. The kinetics of increase in surface hydrophobicity was similar to the kinetics of the conversion of the substrate cell peptidoglycan to a lysoz

The paper describes a predatory Gram-negative bacterium from the genus Bdellovibrio, which was isolated from water of the Lyubozhikha River. As revealed by electron microscopy, the bacterium is an intracellular predator of Gram-negative microorganisms. Its prey range includes Pseudomonas tolaasii, the phytopathogen responsible for brown spot disease in the cultivated button mushroom (Agaricus bisporus). Based on the results of a 16S rRNA gene sequence analysis, the bacterium was identified as Bdellovibrio bacteriovorus strain LR3. We characterized the predator-prey dynamics between B. bacterio

The contribution of predation between bacteria to microbial community dynamics in agricultural soil has hardly been investigated. Here, dynamics of general prokaryotes (GEP) and the predatory Bdellovibrionales (Bd) and Bacteriovoracales (Bac) (Bdellovibrio-and-Like Organisms, BALOs) were studied in two agricultural fields differing in organic and mineral inputs for 1 year. Season, but not fertilization, affected absolute sizes of GEP and of BALO communities. 16S rRNA gene analysis identified numerous previously undetected Bd and Bac lineages, with none of the BALOs related to characterized iso

Pseudomonas aeruginosa is a significant opportunistic pathogen affecting both humans and aquaculture animals. However, there has been no documented evidence linking P. aeruginosa to hepatopancreatic necrosis disease in Chinese mitten crabs Eriocheir sinensis. In the present study, P. aeruginosa isolate HX-1 was identified as a causative agent of hepatopancreatic necrosis disease in E. sinensis, and its pathogenic characteristics in terms of pathogenicity-related genes, antimicrobial resistance profiles, and virulence was comprehensively analyzed. Additionally, the in vitro and in vivo efficaci

The objective of this study was to determine the potential of Bdellovibrio bacteriovorus 109J as an alternative non-chemotherapeutic treatment of infectious bovine keratoconjunctivitis (IBK). To accomplish this, various parameters of B. bacteriovorus predation of Moraxella bovis were determined in vitro. Initial passage of B. bacteriovorus using M. bovis as prey required 10 d for active cultures to develop compared with 2 d for culture on normal Escherichia coli prey; however by the 5th passage, time to active predatory morphology was reduced to 2 d. This high passage B. bacteriovorus culture

Bdellovibrio bacteriovorus is a small Gram-negative bacterium and an obligate predator of other Gram-negative bacteria. Prey resistance to B. bacteriovorus attack is rare and transient. This consideration together with its safety and low immunogenicity makes B. bacteriovorus a valid alternative to antibiotics, especially in the treatment of multidrug resistant pathogens. In this study we developed a novel technique to estimate B. bacteriovorus sensitivity against antibiotics in order to make feasible the development and testing of co-therapies with antibiotics that would increase its antimicro

UNLABELLED: Bdellovibrio are obligate predators and have been described as living antibiotics since their predatory lifestyle enables them to kill pathogenic bacteria, making Bdellovibrio a promising biological control agent. Bdellovibrio strains were isolated from sampling sites in an urban watershed and tested for their killing activity on Salmonella enterica strains associated with human infections. An enrichment technique was used to isolate Bdellovibrio from surface water samples from local tributaries to the Oconee River, employing Salmonella Infantis as the prey bait. The isolated Bdell
