CRESS domain-based molecular characterization of Begomovirus gossypimultanense.

BACKGROUND: Cotton leaf curl disease (CLCuD) is a serious threat to cotton production across the Indian subcontinent, especially in Rajasthan, Haryana, and Punjab. The disease is caused by monopartite single-stranded DNA begomoviruses along with their associated satellite DNAs. Among these viruses, Begomovirus gossypimultanense (Cotton leaf curl Multan virus; CLCuMuV) is one of the primary viruses responsible for widespread infections and significant yield losses. METHODS AND RESULTS: Twenty-six symptomatic cotton leaf samples collected from diverse agro-ecological regions of northwestern Ind

Genome characterization of a begomovirus isolated from Brazilian Carica papaya plants.

Papaya (Carica papaya L.) plants are susceptible to several viral pathogens. However, information regarding papaya-associated DNA viruses in Latin America is limited. In this study, we collected symptomatic papaya plants in Baraúna, Rio Grande do Norte, Brazil, and screened for begomovirus infection. Three of ten samples tested positive through PCR using universal primers, and were further analyzed using rolling circle amplification and Oxford Nanopore sequencing. This approach allowed us to recover complete circular viral genomes with the typical DNA-A segment organization of begomoviruses. W

The Begomovirus Disease Tetrahedron: Weeds as the Missing Dimension in Virus Epidemiology.

Begomoviruses are among the most destructive plant viruses, causing substantial yield losses across diverse cropping systems. Their epidemiological success is driven by high genetic plasticity, broad host range, and efficient transmission by the whitefly vector Bemisia tabaci. Traditional epidemiological models based on the classical disease triangle (virus-host-vector) fail to fully capture the ecological and evolutionary complexity of begomovirus pathosystems. Increasing evidence highlights the critical role of non-cultivated plants, particularly weeds, as persistent reservoirs that maintain

Molecular characterization of a distinct Begomovirus infecting Euphorbia pulcherrima in China.

Virus isolate G35 was obtained from Euphorbia pulcherrima showing leaf curl and vein thickening symptoms in Tianyang, Guangxi Province, China. The virus was transmitted by whiteflies to Nicotiana tabacum, Lycopersicon esculentum, Datura stramonium and E. pulcherrima. DNA-A contains 2746 nucleotides, with two open reading frames (ORFs) in the virion-sense DNA and four ORFs in the complementary-sense DNA. When compared with the DNA-A sequence of other begomo-viruses, the total DNA-A of isolate G35 was most closely related to that of Ageratum enation virus (79.9% sequence identity). However, the

Into the wild: a long-term study of begomovirus community dynamics in a natural landscape.

Begomoviruses have been the subject of many evolutionary studies that explored their temporal dynamics; however, most of the studies relied on sequences retrieved from public repositories rather than hypothesis-driven sampling. These datasets are often temporally or spatially unbalanced, which can bias molecular clock calibration, especially when the population exhibits strong genetic structure. Furthermore, most studies have focused on agricultural environments where anthropogenic activity may influence viral diversification. Longitudinal studies with systematic sampling in the natural enviro

Molecular Characterization of a Begomovirus Associated with Tomato Leaf Curl Disease in Uganda.

During the summer of 2003, leaf curl symptoms were observed in tomato (Lycopersicon esculentum) plantings in the Iganga District of Uganda. Begomoviral infection was suspected. Twelve symptomatic samples were collected. Begomoviral DNA was extracted and amplified using polymerase chain reaction (PCR) with the begomovirus-specific degenerate primer pair PAL1v1978/PAR1c715 (4). The expected 1.4-kb PCR products were obtained from 11 of 12 samples. The 1.4-kb PCR product of one of the samples was cloned and sequenced. Based on the sequence of the 1.4-kb DNA product, specific primers were designed

Spot Inoculation of Begomovirus by Single Whitefly (Bemisia tabaci) Transmission in Mungbean Using Microcage.

The Begomovirus genus is a prominent group of viruses that are transmitted by the whitefly vector, Bemisia tabaci, and have been known to cause significant yield reductions in several economically important crops worldwide. Among these viruses, the mungbean yellow mosaic India virus is particularly problematic, causing substantial yield losses in mungbean and proving difficult to control. Therefore, developing new techniques to study the vector-virus systems involving begomoviruses is a research priority for mungbean and other crops affected by these viruses. In this chapter, we present a tech

Begomovirus Transmission to Tomato Plants Is Not Hampered by Plant Defenses Induced by Dicyphus hesperus Knight.

Plants can respond to insect infestation and virus infection by inducing plant defenses, generally mediated by phytohormones. Moreover, plant defenses alter host quality for insect vectors with consequences for the spread of viruses. In agricultural settings, other organisms commonly interact with plants, thereby inducing plant defenses that could affect plant-virus-vector interactions. For example, plant defenses induced by omnivorous insects can modulate insect behavior. This study focused on tomato yellow leaf curl virus (TYLCV), a plant virus of the family Geminiviridae and genus Begomovir

Mutation of AV2 Start Codon Impairs the Infectivity of a Bipartite Begomovirus Under High Temperature.

Viruses, a group of non-cellular parasites, pose serious threats to human health and agriculture. While viral genetic variations modulating virus adaptation to biotic stresses have been reported, whether and how they affect virus interactions with abiotic stresses such as high temperature remain unknown. Here, we report the impact of AV2 mutations on the life cycle of squash leaf curl China virus (SLCCNV), a bipartite begomoviruses. We first conducted sequence analysis and identified a naturally-occurring mutation at position 35 in AV2 that changes the codon TGG to TAG. Followed analysis revea

Pervasive mixed infections and recurrent begomovirus co-detection in symptomatic pumpkin plants from Guangxi, China.

INTRODUCTION: Pumpkin production in Guangxi, China, is increasingly affected by complex viral diseases, but the regional viral spectrum and mixed-infection structure in symptomatic pumpkin plants remain insufficiently characterized. METHODS: We analyzed 289 symptomatic pumpkin samples collected across Guangxi during 2022-2025 using small-RNA sequencing for virus discovery and targeted PCR/RT-PCR assays for individual-sample confirmation. Virus prevalence, mixed-infection profiles, regional and host-associated differences, and virus-virus and symptom-virus associations were evaluated. RESULTS

Genetic variability of blainvillea yellow spot virus (Begomovirus blainvilleae) reveals recombinant variants and a distinct nonanucleotide motif.

Viruses from wild plants may emerge as new economically important agricultural pathogens through spillover events. Blainvillea yellow spot virus (Begomovirus blainvilleae, BlYSV) is a bipartite geminivirus which is ubiquitous in the non-cultivated plant Blainvillea rhomboidea. To better understand the ecological and evolutionary aspects of this virus, we sampled B. rhomboidea in three Brazilian states between 2020 and 2023. Following rolling-circle amplification, cloning and Sanger sequencing, a dataset was assembled with 23 complete DNA-A sequences plus an additional set of 30 BlYSV sequences

Molecular characterization and evolutionary dynamics of a mixed begomovirus infection associated with chilli leaf curl disease in Odisha, India.

Begomoviruses are one of the most genetically diverse plant-infecting DNA viruses. They significantly limit chilli (Capsicum annuum L.) production in tropical and sub-tropical areas. Their evolution is largely influenced by recombination and mixed infections. However, the relationship between genetic diversity and selection pressure in natural populations of chilli-infecting begomoviruses remains poorly understood. We collected chilli plants showing leaf curl disease symptoms from Bargarh district, Odisha, India, and tested them for begomovirus presence. Molecular analysis showed the presence

Identification and characterization of a novel begomovirus, Withania leaf curl virus associated with leaf curl disease of Withania somnifera.

Regeneration in the peripheral nervous system (PNS) is widely studied both for its relevance to human disease and to understand the robust regenerative response mounted by PNS neurons thereby possibly illuminating the failures of CNS regeneration(1). Sciatic nerve crush (axonotmesis) is one of the most common models of peripheral nerve injury in rodents(2). Crushing interrupts all axons but Schwann cell basal laminae are preserved so that regeneration is optimal(3,4). This allows the investigator to study precisely the ability of a growing axon to interact with both the Schwann cell and basal

Mutation and trans-activation analyses suggest novel functional motifs for AC2 of the begomovirus Sri Lankan cassava mosaic virus.

This study was aimed to have a better understanding of the functional domains of the product of AC2, a protein having a trans-activation function on the late promoters of begomoviruses. The bipartite begomovirus Sri Lankan cassava mosaic virus (SLCMV) was chosen for this study. Infectious SLCMV clones carrying mutations in AC2 were analysed for infectivity, accumulation of viral DNA and trans-activation of the CP promoter in the experimental host Nicotiana benthamiana. The mutations were designed to express truncated AC2 proteins (containing N-terminal 36, N-terminal 101 or C-terminal 22 out o

A glycine rich protein (NbRBGA) confers broad-spectrum tolerance to begomovirus-betasatellite complex through activation of salicylic acid signalling pathway.

Begomoviruses, the largest genus of the family Geminiviridiae, are ssDNA viruses transmitted by whiteflies. Betasatellite molecules are occasionally found to be associated with begomoviruses and greatly influence viral pathogenesis. During viral invasion, plants activate a well-coordinated antiviral immune response to inhibit the propagation of the pathogen. In this study, we have reported an RNA-binding glycine-rich protein (RBGA) getting differentially expressed during infection due to begomovirus-betasatellite complex in Nicotiana benthamiana. Over-expression of the protein successfully res

Tomato curly stunt virus, a New Begomovirus of Tomato Within the Tomato yellow leaf curl virus-IS Cluster in South Africa.

Tomato yellow leaf curl virus (TYLCV) causes a serious disease of tomato in many countries throughout the world. Preliminary reports suggested that TYLC disease was present in 1997 in South Africa. In 1998 140 ha of tomato fields in the Onderberg area were assessed for possible presence of TYLCV. Symptoms like those caused by TYLCV isolates in Israel were observed in most fields, and disease incidence ranged from <1 to 50%. Yield losses in individual plants ranged from negligible to 100% and appeared related to the age of the plants at time of infection. Two isolates of the suspect virus were
