Rabbit blastocysts accumulate [3H]quinuclidinyl benzilate in vitro.

Day-6 rabbit blastocysts were able to accumulate [3H]quinuclidinyl benzilate (QNB) from their environment. This accumulation was reduced approximately 50% in the presence of 1.5 x 10(-4) M atropine (an accepted antagonist for ligands which bind to muscarinic cholinergic receptors). The accumulation of QNB was sensitive to temperature and was apparently saturable. In the presence of 2 nM QNB, Day-6 blastocysts accumulated 30.3 +/- 2.0 fmoles per blastocyst. When the cellular elements alone were examined, lesser amounts of specific binding were detected. Owing to the complexity of this multicomp

Stereoselective benzilic acid rearrangements: new advances on an old story.

The benzilic acid rearrangement (BAR) is the oldest rearrangement on record. It is a powerful synthetic tool for accessing significant biologically active molecules. The reaction is both catalytic (generally Lewis acid) and stereoselective, recently the first catalytic asymmetric version was reported with astonshing results (ees of up to 97%) to give chiral tartronic esters. In this unique highlight, we look at the progress made over the last 10 years on the stereoseletive aspects of this synthetic transformation, showing interesting examples of this rearrangment in both acyclic and cyclic sys

A Multi-Stimuli Transformational Network of Benzil-Based Pd(II) Assemblies.

Achieving precise control over supramolecular topology and nuclearity in coordination-driven self-assemblies through integration of multiple stimuli remains a challenge. We here introduce ligand LA, based on a flexible and reactive 1,2-dicarbonyl benzil-based backbone, able to adopt several conformations. A range of homo- and heteroleptic Pd(II) assemblies is formed with different nuclearities and topologies, connected through distinct transformation pathways. In homoleptic systems, folded and open conformations of LA afford the mononuclear complex PdLA 2 and the lantern-shaped cage Pd2LA 4. I

Chiral Benzoins via Asymmetric Transfer Hemihydrogenation of Benzils: The Detail that Matters.

The synthesis of enantiomerically pure benzoins by hydrogenation of readily available benzils has been long thwarted by their base-sensitivity. We show here that an iron(II) hydride complex catalyzes the asymmetric transfer hydrogenation of benzils from 2-propanol. When strictly base-free conditions are granted, excellent enantioselectivity is achieved even with o-substituted substrates, which are particularly challenging to prepare with other methods. Hence, under optimized reaction conditions, chiral benzoins were prepared in good yields (up to 83%) and excellent enantioselectivity (up to 98

Structural diversity of benzil bis(benzoylhydrazone): Mononuclear, binuclear and trinuclear complexes.

The coordination behaviour of the Schiff-base, benzil bis(benzoylhydrazone), LH(2) towards divalent nickel, lead, cadmium, zinc and copper ions has been investigated. The complexes have been fully characterized by techniques including (113)Cd and (207)Pb NMR, as well as (13)C and (113)Cd CP/MAS NMR and by single crystal X-ray diffraction. All the complexes have the general formula [ML](n) (n = 1-3 depending on the metal ion), with the ligand doubly deprotonated. The nickel complex [NiL] is a monomeric compound, the lead complex [PbL](2) shows a binuclear structure, whereas zinc [ZnL](3) and co

Parametric studies on phencyclidine enhancement of 3H quinuclidinyl benzilate accumulation in vivo.

The purpose of these experiments was to define the temporal parameters involved in the phencyclidine (PCP) enhancement of 3H quinuclidinyl benzilate (QNB) accumulation in mouse brain. PCP enhanced QNB accumulation in brain if given intraperitoneally (IP) 1 and 4, but not 16 hours before intravenous (IV) administration of QNB. This effect was found in hypothalamus, striatum, cortex and hippocampus, but not cerebellum. PCP given after QNB did not alter QNB accumulation. The PCP enhancement persisted for at least 72 hours after QNB administration. These results confirm previous studies demonstrat

In vitro and in vivo metabolism of 3-quinuclidinyl benzilate by high-resolution mass spectrometry.

3-Quinuclidinyl benzilate (QNB) is an anticholinergic compound that affects the nervous system. Its hallucinogenic action has led to its potential utility as an incapacitating warfare agent, and it is listed in Schedule 2 by the Organization for the Prohibition of Chemical Weapons. Although this compound has been known for a long time, limited information is available regarding its metabolism and mass spectrometric data of the metabolites, the information that could facilitate the identification of QNB in case of suspected intoxication. To the best of our knowledge, the analytical methods prev

The benzil-cyanide reaction and its application to the development of a selective cyanide anion indicator.

The benzil-cyanide reaction is a cyanide-specific reaction that has been exploited to produce a colorimetric indicator for this toxic anion. This was done by producing a pi-extended analogue of benzil, 7, which is soluble in a 70:30 (v/v) mixture of methanol-water. In this medium, dilute solutions of 7 are yellow but produce colorless products when exposed to low concentrations of cyanide anion (> or = 1.7 microM; added as an aqueous NaCN solution), but no other common anions (e.g., OH(-), F(-), N3(-), benzoate(-), and H2PO4(-)). On the basis of these observations and supporting mechanistic an

Morphological Study on the Gelation-Induced Phosphorescence of All-Organic Phosphors Based on Substituted Benzil.

We report gelation-induced phosphorescence at room temperature based on 4,4'-disubstituted benzils with alkyloxy and ester groups. The title compounds include five 4,4'-dialkyloxybenzils with varying alkyl chain lengths (from octyloxy to dodecyloxy) and two 4,4'-diesterbenzil (nonylester and phenyl ester). All of the title compounds showed excellent gelation properties in hydrocarbon, alcohol, and acetonitrile solvents. The room-temperature phosphorescence was turned on upon gelation, whereas no emission was observed in solution. The investigation of the photophysical properties of the dried g

Decreased (3H) quinuclidinyl benzilate binding to lymphocytes in Gilles de la Tourette syndrome.

In spite of an unknown pathophysiology, it has been suggested that central dopaminergic hyperactivity exists in Gilles de la Tourette syndrome (TS). Cholinergic influences have also been postulated as a dopaminergic-cholinergic balance seems to be important in other movement disorders. If TS is due to alterations of cholinergic activity, this may also be expressed at postsynaptic levels. Recently, we showed that circulating lymphocytes may serve as useful peripheral markers reflecting induced alterations or inherent changes in muscarinic receptors in the central nervous system (CNS). In the pr

Anesthesia increases in vivo N-([18F]fluoroethyl)piperidinyl benzilate binding to the muscarinic cholinergic receptor.

The in vivo binding of N-[18F]fluoroethyl-piperidinyl benzilate ([18F]FEPB) to the muscarinic cholinergic receptor was measured in awake and anesthetized rats. Studies were done using an equilibrium infusion technique to provide estimates of specific binding as distribution volume ratios. Anesthesia with either isoflurane or sodium pentobarbital produced a significant (65-90%) increase of radiotracer binding in receptor-rich brain regions (striatum, cortex, hippocampus) relative to awake controls. Pretreatment of anesthetized animals with the acetylcholinesterase inhibitor phenserine produced

Binding of [3H]quinuclidinyl benzilate to intestinal mucus. An artifact in identification of epithelial cell muscarinic receptors.

The widely used muscarinic receptor ligand [3H]quinuclidinyl benzilate ([3H]QNB) was found to bind in a site-specific but artifactual manner to rat intestinal mucus, obscuring specific binding to muscarinic receptors on intestinal epithelial cells. Atropine inhibited [3H]QNB binding to mucus with an apparent IC50 of 2.1 x 10(-7) M, compared to an IC50 of 1.4 x 10(-8) M obtained with a homogenate of intestinal epithelial cells. Unlabeled QNB also inhibited binding of [3H]QNB to mucus but the apparent IC50 (4 x 10(-7) M) was about 300-fold greater than the IC50 determined with a control tissue,

Side group dependent room temperature crystallization-induced phosphorescence of benzil based all organic phosphors.

In this work, we report alkoxy substituted benzil based all organic room temperature phosphors which showed crystallization induced phosphorescence (CIP). Nine title compounds were prepared with various alkyl lengths (OCnH2n+1: n = 8-16) and the effect of alkyl side group length on the phosphorescence performance was investigated, as compared to p-anisil. It was found that both phosphorescence quantum yield and lifetime increased concomitantly as the alkyl length increased up to nonyloxy (BZL-OC9). Further increase in the carbon number caused the phosphorescence performance to deteriorate due

The benzil rearrangement reaction: trapping of a hitherto minor product and its application to the development of a selective cyanide anion indicator.

The isolation and characterization of an intermediate from the benzil-cyanide reaction is reported. The use of this trapping chemistry to produce a chemical indicator for the cyanide anion is described. It relies on the synthesis and reaction of a pi-extended analogue of benzil. Addition of tetrabutylammonium cyanide to organic solutions of this species, referred to as compound 3 in the text, gives rise to a dramatic change in both color and fluorescence properties.

Isolation of Paraphaeoketones: A Plausible Biosynthetic Explanation for Paraphaeolactones via a Benzilic Acid Rearrangement Rather than a Favorskii Rearrangement.

Paraphaeoketones A-C (1-3) were isolated from the culture broth of Paraphaeosphaeria sp. KT4192. Their structures and relative configurations were determined using spectroscopic analysis and verified through density functional theory (DFT)-based chemical shift calculations. The absolute configurations of these compounds were determined by comparing the experimental electronic circular dichroism (ECD) spectra with those based on DFT calculations. We also propose a plausible biosynthetic route to 1-3. While our prior studies on the isolation and structural elucidation of paraphaeolactones (e.g.,

Muscarinic cholinergic receptor binding: in vivo depiction using single photon emission computed tomography and radioiodinated quinuclidinyl benzilate.

An attempt was made to characterize, in vivo, specific binding to the muscarinic cholinergic receptor in the calf using the radioiodinated ligand quinuclidinyl benzilate (123I-OH-QNB) and single photon detection emission computed tomography (SPECT). The supratentorial brain activity was significantly increased after the intravenous infusion of 123I-OH-QNB as compared to free 123I. Scopolamine, a muscarinic cholinergic receptor antagonist, decreased the measured brain activity when infused prior to 123I-OH-QNB consistent with pharmacologic blockade of specific receptor binding. Specific binding of the muscarinic antagonist [3H]quinuclidinyl benzilate is not associated with preganglionic motor neurons in the dorsal motor nucleus of the vagus.

The present study evaluates the binding of [3H]quinuclidinyl benzilate, [3H]QNB, as a measure of cholinergic muscarinic binding in six areas of the rat medulla oblongata associated with the cranial nerves. In an experimental group, the right vagus nerve was severed in the neck in order to determine whether the specific muscarinic binding sites might be located on cells that contribute efferent fibers to the vagus nerve. The level of activity of choline acetyltransferase (ChAT) also was determined in the same six areas. Additional experiments utilizing the retrograde transport of toxic ricin, a
