In-silico analysis of Bifidobacterium bifidum strain 900791 genome in the context of the B. bifidum pangenome.

INTRODUCTION: Bifidobacterium bifidum is a key member of the human gut microbiota with well-recognised roles in intestinal homeostasis, glycan metabolism, and immunomodulation. Strain 900791, isolated from the meconium of a Siberian infant, has been used as a commercial probiotic ingredient for decades and has demonstrated, in previously published clinical trials, an ability to improve lactose tolerance and reduce gastrointestinal symptoms in both children and adults; however, the genomic basis for these properties has not been characterised. METHODS: We present the complete genome sequence a

Safety evaluation of a heat-treated Bifidobacterium bifidum OLB6378 concentrate.

Several bacterial strains, including probiotic strains, have undergone evaluations for their safety and potential beneficial health effects. Some of these strains have been introduced into various markets, including that for infant products. However, certain probiotic strains have been linked to serious infections in infants, such as septicemia and meningitis. Given this, it is crucial to assess the safety of each probiotic strain, including those of Bifidobacterium, which is a common genus of probiotics. One such strain, Bifidobacterium bifidum OLB6378 (NITE BP-31), referred to as OLB6378 her

Bifidobacterium bifidum Shows More Diversified Ways of Relieving Non-Alcoholic Fatty Liver Compared with Bifidobacterium adolescentis.

The occurrence of non-alcoholic fatty liver disease (NAFLD) is closely related to intestinal microbiota disturbance, and probiotics has become a new strategy to assist in alleviating NAFLD. In order to investigate the effect of Bifidobacterium on NAFLD and the possible pathway, a NAFLD model was established by using a high-fat diet (HFD) for 18 weeks. Fourteen strains of Bifidobacterium were selected (seven Bifidobacterium adolescentis and seven Bifidobacterium bifidum) for intervention. The effects of different bifidobacteria on NAFLD were evaluated from liver cell injury, liver fat depositio

Human milk oligosaccharide mediates mutualism between Escherichia coli and Bifidobacterium bifidum.

Infant gut microbiota development involves frequent colonization by Enterobacteriaceae, particularly Escherichia coli, yet their ecological role in healthy infants is unclear. Here, we analyse longitudinal stool samples from healthy, term-born, breastfed infants (n = 41) and related mothers (n = 30) using shotgun metagenomics and novel computational approaches. Strain-resolved profiling indicates that Bifidobacterium species are frequently shared within families, whereas E. coli derive from external sources, but often persist within individuals. Despite differing ecological strategies, these g

Protective effect of Bifidobacterium bifidum FSDJN7O5 and Bifidobacterium breve FHNFQ23M3 on diarrhea caused by enterotoxigenic Escherichia coli.

Enterotoxigenic Escherichia coli (ETEC) is the main bacterial cause of diarrhea among children in developing countries and of traveler's diarrhea. In this study, a mouse model was used to evaluate the effect of Bifidobacterium on alleviating diarrhea caused by ETEC. The results showed that B. breve FHNFQ23M3 and B. bifidum FSDJN7O5 could relieve the symptoms of diarrhea. Both strains significantly reduced the stool water content, restored the villi structure in the jejunum and ameliorated the fecal short-chain fatty acid (SCFA) content. In addition, B. breve FHNFQ23M3 restored body weight to t

Differential activation of eosinophils by 'probiotic' Bifidobacterium bifidum and 'pathogenic' Clostridium difficile.

BACKGROUND: Recent studies suggest that probiotics alleviate pathophysiological processes of allergic diseases and inflammatory bowel diseases, whereas 'non-probiotic' microflora has negative effects. However, the underlying mechanisms are not well known, especially in relation to eosinophils, the major effector cells of these inflammatory diseases. OBJECTIVE: To investigate the effects of probiotic Bifidobacterium bifidum (BB) on human eosinophil functions compared with pathogenic Clostridium difficile (CD). METHODS: Peripheral human eosinophils were cultured with heat-killed BB or CD. FISH

Effects of growth conditions on the ion composition of Bifidobacterium bifidum subsp. pennsylvanicum.

The cation content of Bifidobacterium bifidum subsp. pennsylvanicum was markedly influenced by the washing procedure of the cells, by the growth phase and the temperature, and by the composition of the culture medium. Optimal retention of cations was achieved by washing with 0.25 M MgCl2 at 20C. The intracellular Na+ concentration rose during growth in normal medium to a constant value in the stationary phase, the K+ concentration rose in the exponential phase, but fell in the stationary phase. Cells from 29-C cultures contained more Na+ and less K+ in the stationary phase than did cells from

Bifidobacterium bifidum improves intestinal integrity in a rat model of necrotizing enterocolitis.

Neonatal necrotizing enterocolitis (NEC) is a major cause of morbidity and mortality in premature infants. Oral administration of probiotics has been suggested as a promising strategy for prevention of NEC. However, little is known about the mechanism(s) of probiotic-mediated protection against NEC. The aim of this study was to evaluate the effects of Bifidobacterium bifidum treatment on development of NEC, cytokine regulation, and intestinal integrity in a rat model of NEC. Premature rats were divided into three groups: dam fed (DF), hand fed with formula (NEC), or hand fed with formula suppl

Synbiotic intervention with Bifidobacterium bifidum and prebiotics reduces enterohemorrhagic Escherichia coli colonization.

This study evaluated the protective effect of combining Bifidobacterium bifidum with the prebiotics inulin and isomalto-oligosaccharides against enterohemorrhagic Escherichia coli infection and investigated the underlying mechanisms, using an integrated strategy of in vitro screening followed by in vivo validation. Among ten prebiotics screened, IMO and inulin were identified as optimal for promoting B. bifidum proliferation and acetate production (>2.5-fold), with IMO further enhancing bacterial adhesion to Caco-2 cells (four-fold). In a murine infection model, B. bifidum alone reduced total

Bifidobacterium bifidum CCFM1359 alleviates intestinal motility disorders through the BDNF-TrkB pathway.

Intestinal motility disorder is characterised by abnormal intestinal motility function, often resulting in symptoms such as diarrhoea and constipation. Probiotics are increasingly recognised as an effective treatment for gastrointestinal disorders, including intestinal motility disorders. In this study, we used senna extract to induce an animal model of intestinal dysfunction characterised by BDNF downregulation. By assessing relevant indicators of intestinal dyskinesia, we found that Bifidobacterium bifidum CCFM1359 effectively alleviated the dyskinesia. However, this alleviating effect was n

Coculture of Bifidobacterium bifidum G9-1 With Butyrate-Producing Bacteria Promotes Butyrate Production.

Supplementation with Bifidobacterium bifidum G9-1 (BBG9-1) has been established to enhance the production of butyrate, a short-chain fatty acid (SCFA) known for its beneficial effects in alleviating constipation. We hypothesized that BBG9-1 alters gut microbiota such that bacteria that produce butyric acid from lactate and acetate become more abundant. In this study, we sought to determine whether BBG9-1 promotes the growth of butyrate-producing bacteria and thereby enhances butyrate production. BBG9-1 was cocultured with different butyrate-producing bacteria to compare differences in the SCFA

Comparative genomics of the gut commensal Bifidobacterium bifidum reveals adaptation to carbohydrate utilization.

Bifidobacterium bifidum is one of the most abundant members of the gut microbiota at the early stage of life. The established association of the bacterium with the human gut confers health benefits. Such successful persistence of B. bifidum necessitates metabolic adaptation to the host-derived carbohydrates, a process which is poorly understood. The current study focuses on revealing the genomic-based phylogeny (phylogenomics) of B. bifidum and utilizing comparative genomics to decipher the glycolytic abilities of bifidobacterial strains isolated from different human body niches (feces, human

Bifidobacterium bifidum in a rat model of necrotizing enterocolitis: antimicrobial peptide and protein responses.

INTRODUCTION: Necrotizing enterocolitis (NEC) is a devastating disease of premature infants. Probiotics decrease the risk of NEC in clinical and experimental studies. Antimicrobial peptides protect the gut against noxious microbes and shape the commensal microbiota, but their role in NEC remains unclear. METHODS: To investigate the expression of antimicrobial peptides in experimental NEC and the impact of probiotics on their expression, premature rats were divided into three groups: dam fed (DF), hand fed with formula (FF), or hand fed with formula containing Bifidobacterium bifidum (FF + BIF

Influence of Bifidobacterium bifidum on release of minerals from bread with differing bran content.

Bread is considered an important source of minerals; however, the presence of fiber and phytic acid reduces bioavailability of minerals from cereal products. It is well established that activity of microorganisms in human gut increases the amount of nutrients released during digestion. The aim of this study was to determine the influence of Bifidobacterium bifidum on release of some minerals from bread using an in vitro process of enzymatic digestion. White bread and with addition of 15, 30, or 45% of bran was baked in a bakery by traditional methods, with addition of yeasts and rye leaven, fr

Effects of antibiotics on metabolism of peptidoglycan, protein, and lipids in Bifidobacterium bifidum subsp. pennsylvanicus.

The formation of cell envelope components of Bifidobacterium bifidum subsp. pennsylvanicus was studied by measuring the incorporation of [(3)H]glycine, (14)C-labeled fatty acids, and N-benzoyl-[(14)C]glucosamine into the membrane protein, membrane lipids, and cell wall peptidoglycan, respectively. Inhibition of peptidoglycan synthesis by antibiotics (penicillin G, vancomycin, d-cycloserine, and bacitracin) and by the omission of glucosamine-containing growth factors caused a marked decrease in glycine incorporation into cellular as well as membrane protein, which was accompanied by a considera

Immunomodulatory and prophylactic effects of Bifidobacterium bifidum probiotic strain on influenza infection in mice.

The limited efficacy of available influenza vaccines against rapidly emerging new viral strains stresses the need for the development of new antigen-independent prophylactic treatment for enhancing immunity against influenza infection. Recent studies suggest that probiotics possess immunomodulatory properties and can reduce the severity of respiratory infections. Here, we investigated the potential of prophylactic Bifidobacterium bifidum in improving anti-influenza immune responses in an experimental lethal mouse-adapted influenza A (H1N1) infection in a BALB/c mouse model.
