Atroposelective biocatalysis employing alcohol dehydrogenases.

The biocatalytic synthesis of axially chiral heterobiaryl compounds is an area of great interest. A particularly valuable approach involves the atroposelective transformation of achiral, configurationally labile, or racemic heterobiaryl molecules using biocatalysis. By judicious designing the structure of the starting material, Dynamic Kinetic Resolutions (DKR) have been developed to produce higablehly enantioenriched atropisomers with excellent yields and selectivities. Thus, indole- and quinoline-based aldehydes with a precise structure have been prepared. The non-covalent Lewis interaction

[Biocatalysis of formaldehyde to L-xylose].

It is of great significance to use biosynthesis to transform the inorganic substance formaldehyde into organic sugars. Most important in this process was to find a suitable catalyst combination to achieve the dimerization of formaldehyde. In a recent report, an engineered glycolaldehyde synthase was reported to catalyze this reaction. It could be combined with engineered D-fructose-6-phosphate aldolase, a "one-pot enzyme" method, to synthesize L-xylose using formaldehyde and the conversion rate could reach up to 64%. This process also provides a reference for the synthesis of other sugars. Wit

Repurposing thiamine-dependent enzymes for radical biocatalysis.

Thiamine diphosphate (ThDP)-dependent enzymes are ubiquitous and versatile biocatalysts in living systems, catalyzing diverse C-C bond formation or cleavage reactions. Inspired by ThDP-dependent enzymes, chemists have developed biomimetic N-heterocyclic carbenes (NHCs) for organocatalysis, ligand design, as well as material synthesis. Inspired by the recent development in chemo-NHC-enabled radical catalysis, and based on the structural plasticity of ThDP enzymes-conserved cofactor-binding motifs coupled with highly evolvable active sites, our group has repurposed ThDP-dependent enzymes into ef

Eutectozymes as Soft Hybrid Materials for Advanced Biocatalysis.

The development of soft hybrid materials that combine structural integrity, biocompatibility, and catalytic function is a central challenge in advanced biocatalysis. Here, we introduce eutectozymes, enzyme-loaded eutectogels built from natural hydrophobic deep eutectic solvents (HES) and stabilized through a dual supramolecular polymeric network. This unique architecture not only preserves the conformational integrity of enzymes but also creates confined microcavities acting as protective microreactors, thereby enhancing their stability and substrate affinity. Eutectozymes exhibit remarkable r

A direct electrode-driven P450 cycle for biocatalysis.

The large potential of redox enzymes to carry out formation of high value organic compounds motivates the search for innovative strategies to regenerate the cofactors needed by their biocatalytic cycles. Here, we describe a bioreactor where the reducing power to the cycle is supplied directly to purified cytochrome CYP101 (P450cam; EC 1.14.15.1) through its natural redox partner (putidaredoxin) using an antimony-doped tin oxide working electrode. Required oxygen was produced at a Pt counter electrode by water electrolysis. A continuous catalytic cycle was sustained for more than 5 h and 2,600

Customizable colloidosomes confining enzymes for continuous-flow biocatalysis.

Enzyme confinement at the molecular scale reveals catalytic performance that is previously unseen at the ensemble level, but all such delicate enzyme hosts now available encounter their intrinsic issues. Here, we present a mild, one-pot emulsification-directing approach to customize colloidosomes (CSs) with pores matching enzyme size as enzyme hosts for optimizing continuous-flow biocatalysis. By means of the dissolution-induced inside-out self-assembly, uniform and enzyme-matched pores can be readily achieved. Typically, 5.8-nm pores obtained using 22-nm SiO2 nanoparticles (SiO2 NPs) as build

Toward Sustainability: Intensification of Light-Driven Whole Cell Biocatalysis.

Photobiocatalysis with photoautotrophic whole cells has demonstrated strong potential for producing chiral molecules and platform chemicals using sustainable inputs such as light, water and CO2 under mild reaction conditions. Coupling enzymatic transformations directly to natural photosynthesis enables higher atom efficiency compared with heterotrophic systems. However, large-scale application remains challenging, particularly due to light attenuation in photobioreactors. In this review, we summarize recent advances in whole-cell photobiotransformations with emphasis on process conditions. We

A Biochar-Based Composite Hydrogel Microenvironment for Enhanced Biocatalysis.

Addressing water pollution with sustainable and eco-friendly strategies is a significant global challenge. Traditional methods of decontaminating water via enzyme catalysis have limitations, particularly due to the mass transfer resistance resulting from enzyme immobilization. In this study, we report a novel approach in which biochar (BC), a biomass-derived porous material rich in surface functional groups, is integrated with polyacrylamide hydrogels to encapsulate the enzyme horseradish peroxidase (HRP). The resulting Gel/BC-HRP composite hydrogel exhibits superior biocatalytic activity for

Enzyme engineering aspects of biocatalysis: cofactor regeneration as example.

Reaction engineering is an important tool in the case of cofactor depending enzyme-catalyzed reactions. It allows the establishment of conditions resulting in lower product specific cofactor costs as compared with product-specific enzyme costs. This is shown for the stereospecific reduction of carbonyl compounds yielding chiral amino acids and alcohols. In continuous processes, cofactor costs can be reduced if the cofactor can be retained within the bioreactor or recycled into it after separation of the product. In case of readily water-soluble substrates it is even possible to recycle the cof

Production of Biobased Ethylbenzene by Cascade Biocatalysis with an Engineered Photodecarboxylase.

Production of commodity chemicals, such as benzene, toluene, ethylbenzene, and xylenes (BTEX), from renewable resources is key for a sustainable society. Biocatalysis enables one-pot multistep transformation of bioresources under mild conditions, yet it is often limited to biochemicals. Herein, we developed a non-natural three-enzyme cascade for one-pot conversion of biobased l-phenylalanine into ethylbenzene. The key rate-limiting photodecarboxylase was subjected to structure-guided semirational engineering, and a triple mutant CvFAP(Y466T/P460A/G462I) was obtained with a 6.3-fold higher prod

As a major class of petroleum hydrocarbons, n-alkanes are important resources. Advances in biotechnology have led to the discovery of microorganisms capable of metabolizing n-alkanes. Liquid n-alkanes have moderate carbon chain length and good biocompatibility, allowing their complete mineralization into carbon dioxide and water for bioremediation of petroleum pollution. Moreover, they can also be used for the biosynthesis of value-added chemicals via fermentation of engineered microorganisms. Focusing on the microbial transformation pathway of n-alkanes, this review systematically summarizes
