Spectroscopy integration to miniature bioreactors and large scale production bioreactors-Increasing current capabilities and model transfer.

Spectroscopy techniques are being implemented within the biopharmaceutical industry due to their non-destructive ability to measure multiple analytes simultaneously, however, minimal work has been applied focussing on their application at small scale. Miniature bioreactor systems are being applied across the industry for cell line development as they offer a high-throughput solution for screening and process optimization. The application of small volume, high-throughput, automated analyses to miniature bioreactors has the potential to significantly augment the type and quality of data from the

In-situ product recovery in microfluidic bioreactors.

Microfluidic reaction formats are of increasing importance to produce low molecular weight chemicals. In-situ product recovery in such systems benefits from intrinsic physical advantages of microfluidics, including increased mass and energy transfer and continuous processing. This opinion highlights the design space for microfluidic in-situ recovery of gaseous products with respect to rates of production and product recovery, as well as physical constraints limiting product titers. In a theoretical case study, O2 produced by cyanobacteria in a microfluidic reactor is used to benchmark producti

Cultivation of Mycobacterium bovis BCG in bioreactors.

The Mycobacterium bovis BCG vaccine for commercial use is classically produced as surface pellicles by culture on synthetic medium. Under these conditions, reproducibility of the cultures and quality assessment are hampered by slow growth of the bacilli, the formation of bacterial aggregates and a high proportion of dead bacilli after processing and final formulation of the vaccine. Here, we established dispersed cultures of M. bovis BCG in synthetic media in small-scale bioreactors. These cultures allow recording and adjusting of culture parameters and give rise to single bacilli with a high

Development of fundamental technologies for micro bioreactors.

This chapter reviews the development of fundamental technologies required for microchip-based bioreactors utilizing living mammalian cells and pressure driven flow. The most important factor in the bioreactor is the cell culture. For proper cell culturing, continuous medium supply from a microfluidic channel and appropriate modification of the channel surface to accommodate cell attachment is required. Moreover, the medium flow rate should be chosen carefully, because shear stress affects cell activity. The techniques presented here could be applied to the development of micro bioreactors such

Studies of craniofacial development in rotating bioreactors.

Several studies in our laboratory assessed the effect of 3-D culture in various rotating bioreactors on craniofacial development. Initially, mouse first branchial arches were cultured. Molar and incisor development occurred in both upper and lower jaws, but maxilla development was deficient because no brain was present. In a second study using excised whole heads, the oral epithelia fused and teeth did not develop. External structure of the face was obliterated, although internally, eye development was excellent. To preserve both internal spaces and external face structure, subsequent experime

Additively Manufactured Bioreactors for Scaffold-Coupled Electrical Stimulation.

Electrical stimulation (ES) modulates diverse cellular processes, yet in vitro platforms often lack precision, scalability, and compatibility with 3D bioscaffolds. We present a modular bioreactor system that delivers low-voltage, bioscaffold-coupled ES via conductive graphene foam, enabling real-time waveform monitoring and live cell imaging. Components were fabricated using low-cost stereolithography with BioMed Clear resin and validated through mechanical, chemical, and cytocompatibility analyses. Printed parts exhibited high dimensional accuracy, retained mechanical integrity post-steriliza

Temporary Immersion Bioreactors for Sugarcane Multiplication and Rooting.

Sugarcane is used to produce sugar, ethanol, and other by-products, so it is considered one of the most important crops worldwide. Using temporary immersion systems for sugarcane micropropagation represents an alternative to reduce the labor force, increase plant development, and improve plant quality. Temporary immersion systems are semi-automated bioreactors designed for the large-scale propagation of tissues, embryos, and organs. These are temporarily exposed in a liquid culture medium under a specific time and immersion frequency. Using this protocol and a temporary immersion bioreactor, i

Modeling immunotherapies in live 3D human cancer tissue bioreactors.

Background: Cancer immunotherapies have shown remarkable efficacy in advanced malignancies, yet many patients remain unresponsive. This variability, along with concerns about adverse effects and healthcare costs, highlights the need for predictive biomarkers and physiologically relevant cancer models to forecast individual treatment responses. Existing systems inadequately recapitulate the human tumor microenvironment (TME), which is essential for understanding immune-tumor interactions and treatment efficacy. Here, we developed an ex vivo 3D human tissue culture model that preserves the nativ

Bioreactors for tissue engineering--a new role for perfusionists?

Tissue engineering is an exciting new area of medicine with rapid growth and expansion over the last decade. It has the potential to have a profound impact on the practice of medicine and influence the economic development in the industry of biotechnology. In almost every specialty of medicine, the ability to generate replacement cells and develop tissues will change the focus from artificial organs and transplantation to growing replacement organs from the patient's own stem cells. Once these organs are at a size that requires perfusion to maintain oxygen and nutrient delivery, then automated

Gas supply across membranes in bioreactors for hepatocyte culture.

The conditions required for hepatocyte cultures is a main topic in the development of bioreactors for hybrid liver support systems. The detoxification of ammonia and the synthesis of urea due to primary isolated hepatocytes was measured in order to compare two different models of gas supply in bioreactors: (a) indirect medium oxygenation and (b) direct membrane-contact oxygenation of the hepatocytes using polypropylene membranes. Increasing oxygen pressure promoted cell function. At day 6 of culture, urea synthesis was 0.8 +/- 0.3 mM in 21% of O2 cultures and 1.5 +/- 0.1 mM in oxygenated cultu

Biodegradation of trichloroethylene in continuous-recycle expanded-bed bioreactors.

Experimental bioreactors operated as recirculated closed systems were inoculated with bacterial cultures that utilized methane, propane, and tryptone-yeast extract as aerobic carbon and energy sources and degraded trichloroethylene (TCE). Up to 95% removal of TCE was observed after 5 days of incubation. Uninoculated bioreactors inhibited with 0.5% Formalin and 0.2% sodium azide retained greater than 95% of their TCE after 20 days. Each bioreactor consisted of an expanded-bed column through which the liquid phase was recirculated and a gas recharge column which allowed direct headspace sampling

Bone tissue engineering bioreactors: a role in the clinic?

Tissue engineered bone grafts have the potential to be used to treat large bone defects due to congenital abnormalities, cancer resections, or traumatic incidents. Recent studies have shown that perfusion bioreactors can be used to generate grafts of clinically relevant sizes and shapes. Despite these scientific and technological successes, there is uncertainty regarding the translational utility of bioreactor-based approaches due to the perceived high costs associated with these procedures. In fact, experiences over the past two decades have demonstrated that the widespread application of cel

Compression bioreactor for cultured meat.

Cultured meat is emerging as a sustainable and ethical alternative source of dietary protein. However, the commercial viability of cultured meat is fundamentally constrained by prolonged tissue maturation timelines and the high energy demands of conventional, motor-driven electronic bioreactors. To address these bottlenecks, this study introduces a novel, scalable, magnetic-valve controlled pneumatic compression bioreactor, which utilizes a mechanically tunable magnetic circuit to deliver precise cyclic compression without internal electronic sensors or digital feedback loops. We evaluated the

Scale-up of a cyclone bioreactor.

The operation of a cyclone bioreactor differs from conventional stirred tanks since the agitation is accomplished by means of a pumped recirculation loop. Oxygen transfer can occur across the swirling gas-liquid interface in the cyclone or from bubbles entrained in the recirculation loop. A cyclone bioreactor was scaled-up from a 1 dm3 bench top unit to a 75 dm3 Process Development Unit (PDU). A reduction in the aspect ratio was compensated for by extending the length of the recirculation loop and providing additional aeration. Performance of the two reactors for the production of microbial po

The oral cavity as a bioreactor in halitosis.

Halitosis is a prevalent yet often underestimated clinical condition with multifactorial etiology, predominantly arising from intra-oral sources such as periodontal disease, tongue coating and xerostomia. Central to its pathogenesis is the microbial degradation of sulfur-containing substrates, which lead to the production of volatile sulfur compounds and other volatile organic compounds responsible for the characteristic odor. These metabolites are produced by proteolytic anaerobes inhabiting the tongue dorsum, periodontal pockets as well as the dental plaque and predominantly exist as structu

Pesticides in denitrifying woodchip bioreactors: Current knowledge and research needs.

Extensive research has demonstrated that excessive fertilizer and pesticide application leads to contamination of soil, surface water, and groundwater. Consequently, there is an increasing need for mitigation strategies to reduce multiple agricultural contaminants before they reach receiving water bodies. Denitrifying woodchip bioreactors (DWBs) are designed primarily to remove nitrate from agricultural surface runoff. However, evidence also indicates that they can facilitate the removal of pesticides. To date, studies investigating the fate of pesticides representing several chemical classes

Genomic potential for complete denitrification by Desulfobacillus in anammox bioreactors.

Desulfobacillus are frequently observed as abundant flanking denitrifying microorganisms in anammox bioreactors and wastewater treatment systems. Previous studies suggested that Desulfobacillus lacked canonical nitric oxide reductase, restricting them to incomplete denitrification. As a result, they were thought to provide NO to anammox bacteria and act as an N2O sink. In this study, 48 Desulfobacillus genomes from GTDB were analyzed, including 21 from anammox-related bioreactors. Multiple potential nitric oxide reductases (eNOR, nNOR, and sNOR) were identified, and all 21 anammox-associated g

Determination of nitrate removal kinetics model parameters in woodchip bioreactors.

Woodchip bioreactors have emerged as a viable water management tool to reduce nitrate contamination from agricultural subsurface drainage, wastewater, and stormwater. Understanding of denitrification kinetics is critical to the design and application of field woodchip bioreactors. The denitrification process in woodchip bioreactors generally obeys a model of Michaelis-Menten type enzyme kinetics. The objective of this study was to determine Michaelis-Menten model parameters for nitrate removal in laboratory bioreactors using the fresh, composted and aged woodchips. The results showed that the
