Azithromycin and Ciprofloxacin Can Promote Antibiotic Resistance in Biosolids and Biosolids-Amended Soils.

Spread of biosolids-borne antibiotic resistance is a growing public and environmental health concern. Herein, we conducted incubation experiments involving biosolids, which are byproducts of sewage treatment processes, and biosolids-amended soil. Quantitative reverse transcription-PCR (RT-qPCR) was employed to assess responses of select antibiotic resistance genes (ARGs) and mobile elements to environmentally relevant concentrations of two biosolids-borne antibiotics, azithromycin (AZ) and ciprofloxacin (CIP). Additionally, we examined sequence distribution of gyrA (encoding DNA gyrase; site o

Biosolids-amended soils: Part I. Effect of biosolids application on soil quality and ecotoxicity.

Investigations of potential risk from biosolids generally indicate that land application does not threaten human or ecosystem health, but questions continue to arise concerning the environmental effects of this practice. This research project was initiated to evaluate ecotoxicity resulting from the amendment of soils with biosolids from municipal wastewater treatment plants. Toxicity was evaluated using standard tests, including earthworm mortality, growth, and reproduction; seedling germination and root elongation; microbial respiration; and nematode mortality and reproduction. Nineteen munic

Aging of biosolids in fields prolongs the release of biosolid-borne PFAS and promotes the release of select PFCA in water.

Land application of biosolids can introduce PFAS into croplands. Biosolids transported to surface waters through runoff may subsequently release PFAS into aquatic environments. The objective was to evaluate the effects of the aging of biosolids and the presence of sediment on the release of biosolid-borne PFAS to water at the water-sediment interface under anaerobic conditions. Through two independent rounds of microcosm experiments, we demonstrate that by day 136, most of the PFAS compounds (e.g., perfluorocarboxylic acids, or PFCAs) released <10 ng per g biosolids dry weight, except for 5:3

Characterizing pyrethroid and fipronil concentrations in biosolids.

Pesticides are prevalent in wastewater, yet few studies have measured pesticides in biosolids and aqueous media from samples collected concurrently. Seventeen California wastewater treatment plants (WWTPs) were sampled in May 2020. Biosolids samples were analyzed for 27 analytes, and paired aqueous samples (influent and effluent) were analyzed for 23 analytes. Analytes included fipronil and its transformation products (fiproles), pyrethroids, novaluron, and several other pesticides with down-the-drain transport potential. Of the 27 compounds analyzed in biosolids samples, 16 were detected in a

Biosolid-borne tetracyclines and sulfonamides in plants.

Tetracyclines and sulfonamides used in human and animal medicine are released to terrestrial ecosystems from wastewater treatment plants or by direct manure application. The interactions between plants and these antibiotics are numerous and complex, including uptake and accumulation, phytometabolism, toxicity responses, and degradation in the rhizosphere. Uptake and accumulation of antibiotics have been studied in plants such as wheat, maize, potato, vegetables, and ornamentals. Once accumulated in plant tissue, organic contaminants can be metabolized through a sequential process of transforma

Chemical fractionation of phosphorus in stabilized biosolids.

Three chemicals-ferrous sulfate (FeSul), calcium oxide (CaO), and aluminum sulfate (alum)-were applied at different rates to stabilize P in fresh, anaerobically digested biosolids (FBS) obtained from an activated sewage treatment plant. A modified Hedley fractionation procedure was used to assess P forms in these sludge-borne materials and in a biosolids compost (BSC) prepared from the same FBS. Each biosolids material exhibited a unique pattern of P distribution among fractions. The most available P forms, namely: (i) water-soluble P (WSP); (ii) membrane-P; and (iii) NaHCO(3)-P, were stabiliz

Risk assessment of biosolids-borne ciprofloxacin and azithromycin.

Ciprofloxacin (CIP) and azithromycin (AZ) are commonly prescribed antibiotics for various infections in humans and are frequently detected in biosolids. Ecological and human health risks from biosolids-borne CIP and AZ are not well understood, but necessary for formulating policies on safe use and management of biosolids. A tiered integrated risk assessment (IRA), based on the World Health Organization (WHO) framework and the USEPA Part 503 Biosolids Rule, was conducted to assess human and ecological health risks from biosolids-borne CIP and AZ. The IRA utilized the hazard quotient (HQ) approa

Evaluation of PFAS extraction and analysis methods for biosolids.

Per- and polyfluoroalkyl substances (PFAS) in the environment is a growing concern leading to a focus on PFAS occurrence in biosolids, a byproduct of wastewater treatment processes, often applied to improve soil health. This led to the need for analytical method development for assessing PFAS in biosolids. This study compares three methods for PFAS quantitation, evaluating solvent extraction, clean-up techniques, and final injection solvents. Three biosolids examined included not stabilized, anaerobically digested, and activated sludge with long-term lagoon-stabilized solids, resulting in diff

Biosolids affect soil barium in a dryland wheat agroecosystem.

In December 2003, the USEPA released an amended list of 15 "candidate pollutants for exposure and hazard screening" with regard to biosolids land application, including Ba. Therefore, we decided to monitor soil Ba concentrations from a dryland wheat (Triticum aestivum L.)-fallow agroecosystem experiment. This experiment received 10 biennial biosolids applications (1982-2003) at rates from 0 to 26.8 dry Mg ha(-1) per application year. The study was conducted on a Platner loam (Aridic Paleustoll), approximately 30 km east of Brighton, CO. Total soil Ba, as measured by 4 M HNO(3), increased with

Solid phosphorus phase in aluminum- and iron-treated biosolids.

Stabilization of phosphorus (P) in sewage sludge (biosolids) to reduce water-soluble P concentrations is essential for minimizing P loss from amended soils and maximizing the capacity of the soil to safely serve as an outlet for this waste material. The chemical form at which P is retained in biosolids stabilized by Al(2)(SO(4))(3) x 18H(2)O (alum) or FeSO(4) x 7H(2)O (FeSul) was investigated by scanning electron microscopy (SEM) equipped with energy-dispersive X-ray elemental spectrometry (EDXS) and by X-ray diffraction (XRD). Both treatments resulted in the formation of a Ca-P phase, probabl

Concentrations of per- and polyfluoroalkyl substances (PFAS) in private well drinking water and serum of individuals exposed to PFAS through biosolids: The Maine Biosolids Study.

Per- and polyfluoroalkyl substances (PFAS) are synthetic and persistent chemicals that can contaminate food and water systems and have lasting negative implications for human health. We described paired residential well water and serum PFAS concentrations among adults residing where PFAS-contaminated biosolids were historically applied to farmland as fertilizer. In 2023, we measured 7 PFAS in serum from 145 participants who had up to 28 PFAS measured in their well water within the 3 prior years due to proximity to a biosolids application site. Participants were median(interquartile range) 63(1

Co-contaminant risks in water reuse and biosolids application for agriculture.

Agriculture made the shift toward resource reuse years ago, incorporating materials such as treated wastewater and biosolids. Since then, research has documented the widespread presence of contaminants of emerging concern in agricultural systems. Chemicals such as pesticides, pharmaceuticals and poly- and -perfluoroalkyl substances (PFASs); particulate matter such as nanomaterials and microplastics; and biological agents such as antibiotic resistance genes (ARGs) and bacteria (ARB) are inadvertently introduced into arable soils where they can be taken up by crops and introduced to the food-web

Characterization of phosphorus species in biosolids and manures using XANES spectroscopy.

Identification of the chemical P species in biosolids or manures will improve our understanding of the long-term potential for P loss when these materials are land applied. The objectives of this study were to determine the P species in dairy manures, poultry litters, and biosolids using X-ray absorption near-edge structure (XANES) spectroscopy and to determine if chemical fractionation techniques can provide useful information when interpreted based on the results of more definitive P speciation studies. Our XANES fitting results indicated that the predominant forms of P in organic P sources

Seven years of biosolids versus inorganic nitrogen applications to tall fescue.

Repeated applications of municipal wastewater biosolids is cost effective for biosolids managers, but may lead to undesirable accumulations of nutrients or contaminants. We evaluated the effects of seven years of biosolids applications on tall fescue (Festuca arundinacea Schreb.) production and nutrient availability. We compared two types of Class A biosolids applied to tall fescue on a sandy loam in western Washington. Mean annual biosolids rates of 290, 580, and 870 kg total N ha(-1) yr(-1) were compared with inorganic N and zero-N controls using a randomized complete block design. We measur

Levels of antibiotic resistance genes in manure, biosolids, and fertilized soil.

Increasing antibiotic resistance genes in the environment may pose a threat to public health. In this study, tetracycline and sulfonamide resistance genes (Tet-W, Tet-O, and Sul-I) were quantified in 24 manure samples from three farms and 18 biosolids samples from seven different wastewater treatment plants using quantitative polymerase chain reaction methods. Concentrations of Tet-W and Tet-O genes were observed to be significantly higher (p < 0.05) in manure than in biosolids samples. The background soil samples showed significantly lower concentration of the above genes compared with manure

Biosolid and alum effects on runoff losses during turfgrass establishment.

Large, volume-based rates of composted biosolids (CB) enhance turfgrass establishment and soil properties, but nonpoint-source runoff losses could occur during production and after transplanting of sod. The objective was to evaluate runoff losses of N, P, sediment, and organic C during establishment of sprigs or transplanted sod of Tifway bermudagrass (Cynodon dactylon L. Pers. X C. transvaalensis Burtt-Davey) with and without CB and aluminum sulfate (Alum). Four treatments comprised Tifway sprigged in a sandy loam soil with and without incorporation of 0.25 m(3) CB m(-3) soil and Alum. In fou

Subsurface transport of Cd, Cr, and Mo mediated by biosolid colloids.

The potential of biosolid colloids to transport metals associated with organic-waste amendments through subsurface soil environments was investigated with leaching experiments involving undisturbed soil monoliths. The monoliths (25 cm in height and 18 cm in diameter) were carved from the upper solum of an Alfisol, a Mollisol, and an Entisol in the Bluegrass region of Kentucky. Biosolid colloids were fractionated from two municipal wastes (lime-stabilized/LSB, and aerobically-digested/ADB), and a poultry-manure (PMB) organic waste and applied onto the monoliths at a rate of approximately 0.7 cm

Irrigation water quality influences heavy metal uptake by willows in biosolids.

Phytoextraction is an effective method to remediate heavy metal contaminated landscapes but is often applied for single metal contaminants. Plants used for phytoextraction may not always be able to grow in drier environments without irrigation. This study investigated if willows (Salix x reichardtii A. Kerner) can be used for phytoextraction of multiple metals in biosolids, an end-product of the wastewater treatment process, and if irrigation with reclaimed and freshwater influences the extraction process. A plantation of willows was established directly onto a tilled stockpile of metal-contam
