Pharmaceutical applications of microbial biosurfactants.

Biosurfactants are becoming more and more popular in biomedical and pharmaceutical applications due to their intriguing biological and physicochemical characteristics. The amphiphilic nature of molecules imparts antimicrobial and antiviral properties, which are largely exploited for their application as therapeutic agents in wound healing, dental health, and infection control, especially against antimicrobial-resistant, multidrug-resistant microbes and viruses such as HIV and herpes simplex virus. The ring-shaped peptide and hydrophobic chain interact with bacterial membranes, inserting into l

Biosurfactants and aqueous two-phase fermentation.

The partition of surfactants and a biosurfactant-producing microorganism was studied in polyethylene glycol and dextran aqueous two-phase systems. In the presence of sodium phosphate, surfactants distributed themselves according to charge. Cationic surfactants preferred the bottom phase, while anionic surfactants were attracted to the top phase. Increasing the phosphate molarity or the pH resulted in a more 1-sided surfactant partitioning. Biosurfactant partitioning was weaker than synthetic surfactant partitioning due to the weaker effective charge and lack to strong specific affinity for any

Microbial biosurfactants: Multifarious applications in sustainable agriculture.

Agriculture in the 21st century faces grave challenges to meet the unprecedented food demand of the burgeoning population as well as reduce the ecological footprint for achieving sustainable development goals. The extensive use of harsh synthetic surfactants in pesticides and the agrochemical industry has substantial adverse impacts on the soil and environment due to their toxic and non-biodegradable nature. Biosurfactants derived from plant, animal, and microbial sources can be an eco-friendly alternative to chemical surfactants. Microbes producing biosurfactants play a noteworthy role in bio

Replacement of hexachlorocyclohexane to environmentally friendly biosurfactant as precursor for the production of biosurfactant from Pseudomonas.

Production of biosurfactant can be substantially increased by the addition of precursors like vegetable oils, petroleum products, and other water-insoluble substances. Pseudomonas Ptm+ strain produces biosurfactant in the presence of hexachlorocyclohexane (HCH), which specifically emulsifies HCH, a recalcitrant organochlorine pesticide. Addition of previously produced crude biosurfactant by the same organism as a precursor instead of HCH increased production of biosurfactants with a decrease in the total fermentation time from 32 to 24 h. The main objective of this paper was to find alternativ

Biosurfactants Contribute to Distiller's Corn Oil Recovery during Ethanol Production.

Distiller's corn oil (DCO) is an important coproduct of the ethanol industry. However, this oil is partially emulsified in the fermentation medium and adheres to solid particles during ethanol production. The addition of surfactants can facilitate the DCO removal. This study is the first to investigate the use of biosurfactants, biodegradable molecules with demulsifying properties, for DCO recovery. For this, two rhamnolipid (RML)-type biosurfactants, one mono-RML-rich mixture and the other, di-RML, both produced using DCO as a carbon source, were added into ethanol production at a concentrati

Amide Modification of Glycolipid Biosurfactants as Promising Biocompatible Antibacterial Agents.

Discovering new antibacterial agents is crucial to addressing the increasing risk of bacterial infections induced by antimicrobial resistance in food and agricultural industries. Here, biocompatible acidic-type sophorolipids (ASLs) and glucolipids (GLs) prepared via chemical modification of natural sophorolipids from fermentation were functionalized via amide modification for use as potential antibacterial agents. It was found that the arginine methyl ester derivative of GLs (GLs-d-Arg-OMe) showed excellent antibacterial activity, killing more than 99.99% of Escherichia coli at 200 mg/L. The s

Optimization of liquid-liquid extraction of biosurfactants from corn steep liquor.

In this work, the optimization of the operational conditions for the chloroform-based extraction of surface-active compounds from corn steep liquor (CSL) was carried out and the nutritional properties of the remnant aqueous phase (CSL-less biosurfactant) was evaluated as microbial fermentation medium. The optimal conditions to obtain biosurfactants from CSL were as follows: chloroform/CSL ratio 2 (v/v), 56 °C at extraction times >30 min. At the optima conditions, 100 % of biosurfactant extract can be obtained from CSL, obtaining 12.0 ± 0.5 g of biosurfactant extract/Kg of CSL. The critical mic

Biosurfactants direct the evolutionary dynamics of surface-associated microbial systems.

Surface-associated microbial systems experience spatial constraints that can intensify stochastic lineage loss through genetic drift and alter how natural selection acts on competing lineages. A key feature of these systems is that steep resource gradients typically confine growth to a narrow band of cells located at the biomass periphery. This restricts the effective population size and constrains lineage interactions, thereby accelerating stochastic lineage exclusion. Here, we demonstrate that biosurfactants, which are secreted by many bacteria, can modulate the evolutionary dynamics of thes

The paradigm of prophylactic viral outbreaks measures by microbial biosurfactants.

The recent emergence and outbreak of the COVID-19 pandemic confirmed the incompetence of countries across the world to deal with a global public health emergency. Although the recent advent of vaccines is an important prophylactic measure, effective clinical therapy for SARS-Cov-2 is yet to be discovered. With the increasing mortality rate, research has been focused on understanding the pathogenic mechanism and clinical parameters to comprehend COVID-19 infection and propose new avenues for naturally occurring molecules with novel therapeutic properties to alleviate the current situation. Recovery of Rhodococcus biosurfactants using methyl tertiary-butyl ether extraction.

In the present study, we proposed methyl tertiary-butyl ether (MTBE) as a solvent for extraction of biosurfactants from Rhodococcus bacterial cultures. After comparison with other well known solvent systems used for biosurfactant extraction, it was found that MTBE was able to extract crude surfactant material with high product recovery (10 g/l), efficiency (critical micelle concentration (CMC), 130-170 mg/l) and good functional surfactant characteristics (surface and interfacial tensions, 29 and 0.9 mN/m, respectively). The isolated surfactant complex contained 10% polar lipids, mostly glycoli

Effectiveness of biosurfactant for the removal of trihalomethanes by biotrickling filter.

In this study, the biodegradation of a mixture of two trihalomethane (THM) compounds, chloroform (CF) and dichlorobromomethane (DCBM), was evaluated using two laboratory-scale biotrickling filters (BTFs). The two BTFs, hereby designated as "BTF-A" and "BTF-B," were run parallel and used ethanol as co-metabolite at different loading rates (LRs), and a lipopeptide-type biosurfactant that was generated by the gram-positive bacteria, Surfactin, respectively. The results using BTF-A showed that adding ethanol at a higher rate of 4.59 g/(m3 h) resulted in removal efficiencies of 85% and 87% for CF a

Rhamnolipid biosurfactant complexation of rare earth elements.

Rare earth elements (REE) are vital for modern technologies and considered critical materials. This study investigated monorhamnolipid biosurfactant interactions with REE as the basis for REE recovery technology. Conditional stability constants (log β), measured using a resin-based ion exchange method, are reported for 16 REE and metals. These results were combined with existing data for 10 other metals to assess comparative strength and determinants of binding. The stability constants could be divided into three groups: weakly, moderately, and strongly bound. The REE were all in the strongly

Enhanced aqueous solubilization of tetrachloroethylene by a rhamnolipid biosurfactant.

A rhamnolipid biosurfactant produced by Pseudomonas aeruginosa ATCC 9027 was isolated, purified and characterized in terms of its ability to mobilize and solubilize tetrachloroethylene (PCE) for potential use in surfactant-enhanced aquifer remediation (SEAR) applications. Using a drop volume method, the PCE-biosurfactant steady-state interfacial tension was determined and found to be ca. 10 mN/m which is not low enough to cause significant PCE nonaqueous phase liquid (NAPL) mobilization. It was observed that the biosurfactant partitioned significantly into PCE at aqueous concentrations higher

Sophorolipid biosurfactants application as novel approach to lipid extraction from Chlorella cultures.

Algal lipids and high-value biomaterials have diverse applications in industries such as biofuels, nutraceuticals, pharmaceuticals, and cosmetics. They represent a promising alternative to traditional plant oils and animal fats. However, the high costs of extraction and recovery still pose significant challenges. In the present study, a novel method involving treatment of Chlorella sorokiniana cultures with sophorolipid (SL) biosurfactants, obtained from Starmerella bombicola yeast culture, was initially developed to facilitate the release of lipids prior to extraction with ethyl acetate (EA),

A novel family of fungal protein biosurfactants: Discovery and sustainable production.

Protein-based biosurfactants remain underexplored compared to glycolipids and lipopeptides, despite their unique interfacial properties and self-assembly behavior. PAC3, a surface-active protein produced by the marine fungus Acremonium sclerotigenum, exhibits dual behavior as both a biosurfactant and bioemulsifier. For this reason, it can be seen as a high molecular weight proteinaceous compound, able to efficiently reduce surface tension. Here, we identify PAC3 as the first member of a previously unrecognized family of fungal protein biosurfactants. The complete amino acid sequence of PAC3 wa

Novel biosurfactants produced from food waste: a variability and validation study.

One-third of global food production is wasted, much of which is composted, gasified or landfilled. This food waste (FW) represents an untapped resource, particularly towards the production of biosurfactants. Genetically engineered Starmerella bombicola yeast strains offer a route to produce novel biosurfactants on FW with enhanced structural diversity, such as bola sophorosides. However, FW streams are variable, and often contain inhibitory compounds (e.g. lactic acid (LA), ethanol, etc.), posing bioprocessing challenges. Here we show that hydrolyzed FW consistently supports S. bombicola growt
