Cellular and molecular characteristics of blastoderm adhesion of the chick embryo.

OBJECTIVE: During early avian development, the blastoderm border must adhere to the vitelline membrane (VM) in order to spread tissue evenly. Classical studies have demonstrated that blastoderm edge tissue possesses intrinsic adhesive properties to the VM; however, the spatial extent, developmental changes, and molecular characteristics of this adherence remain poorly understood. This study aimed to characterize the molecular and functional properties of blastoderm adhesion to the VM. METHODS: Explant assays were performed using chick embryos at different developmental stages. Tissues were is

Distribution of ring-X chromosomes in the blastoderm of gynandromorphic D. melanogaster.

Embryos of the D. melanogaster strain producing gynandromorphs by loss of the ring-X chromosome were treated with vinblastine to obtain blastoderms with all mitoses arrested in metaphase, and with tetracaine to improve the resolution of chromosomes. Ring-X and ringless mitoses were recorded in the major part of the blastoderm in 18 eggs. Limits between females and male areas were very irregular and some embryos had several isolated areas of one type or the other. The proportion of male nuclei varied from 80.8 to 0.4%, indicating that there must have been more than one loss of the ring-X in mos

Wound healing in the primitive deep layer of the young chick blastoderm.

Wound healing in the primitive deep layer of stage 4 chick blastoderms was studied in vitro by cinemicrophotography of living cultures and by photomicroscopy, scanning- and transmission electron microscopy after fixation. Experimental wounds with an average diameter of 0.3 mm healed completely within 2 to 4 h through migration of the cells at their rims. Healing occurred in mesenchyme-free areas, providing us with a purely epithelial reaction. The rim cells of the primitive deep layer formed extensions at their free flank, described as fila, filopodia, lamellae and lamellipodia. They were alre

Early embryonic development in the tick Ixodes scapularis suggests syncytial organization and cellularization before blastoderm formation.

Ixodes ticks are the most important vectors of arthropod-borne diseases in the United States, Canada, and Europe. Ixodes scapularis is the major vector that transmits the causative agent of Lyme disease in the eastern United States and can transmit up to six additional pathogens. In recent years, many advances have been made in building the toolkit for I. scapularis research, including genomic resources, transcriptomes, and forward and reverse genetics techniques. However, an understanding of the early embryonic development of this species is still lacking. In this study, we attempted to fill

Direct delivery of adenoviral CRISPR/Cas9 vector into the blastoderm for generation of targeted gene knockout in quail.

Zygotes at the 1-cell stage have been genetically modified by microinjecting the CRISPR/Cas9 components for the generation of targeted gene knockout in mammals. In the avian species, genetic modification of the zygote is difficult because its unique reproductive system limits the accessibility of the zygote at the 1-cell stage. To date, only a few CRISPR/Cas9-mediated gene knockouts have been reported using the chicken as a model among avian species, which requires 3 major processes: isolation and culture of primordial germ cells (PGCs), modification of the genome of PGCs in vitro, and injecti

Adult deficiencies and duplications of head and thoracic structures resulting from microcautery of blastoderm stage Drosophila embryos.

Drosophila embryos were damaged by microcautery at the cellular blastoderm stage at the sites of presumptive imaginal cells, identified from fate maps. The resulting adults were analyzed for abnormal structures. Cautery of any of the presumptive imaginal regions can lead to defects in the adult cuticle, though the majority of adults which hatch are morphologically normal. The abnormal adults had one or more discs either deleted, incompletely formed, or misarranged. Several of the structures which were incomplete had duplicated regions. The results suggest that, from the time fo their initial f

Origin, fate, and function of the components of the avian germ disc region and early blastoderm: role of ooplasmic determinants.

In the avian oocytal germ disc region, at the end of oogenesis, we discerned four ooplasms (alpha, beta, gamma, delta) presenting an onion-peel distribution (from peripheral and superficial to central and deep. Their fate was followed during early embryonic development. The most superficial and peripheral alpha ooplasm plays a fundamental role during cleavage. The beta ooplasm, originally localized in the peripheral region of the blastodisc, becomes mainly concentrated in the primitive streak. At the moment of bilateral symmetrization, a spatially oblique, sickle-shaped uptake of gamma and del

Studies on cell differentiation: inducing capacity of sulfhydryl-containing amino acids on post-nodal pieces of chick blastoderms.

The inducing capacity of sulfhydryl (SH)-containing amino acids (cysteine and glutathione) on post-nodal pieces (PNPs) of stage 4 chick blastoderms was investigated. PNPs were treated for different lengths of time with chick Ringer's solution (control group) or chick Ringer's solution containing cysteine or glutathione, followed by culturing for 2-10 days on Spratt-Haas agar medium or on the chorioallantoic membrane of 8-day chick embryos. Control PNPs rarely showed differentiation, but those treated with the amino acids for six hours or longer developed structures such as neural tissue, notoc

Collimated Microbeam Reveals that the Proportion of Non-Damaged Cells in Irradiated Blastoderm Determines the Success of Development in Medaka (Oryzias latipes) Embryos.

It has been widely accepted that prenatal exposure to ionizing radiation (IR) can affect embryonic and fetal development in mammals, depending on dose and gestational age of the exposure, however, the precise machinery underlying the IR-induced disturbance of embryonic development is still remained elusive. In this study, we examined the effects of gamma-ray irradiation on blastula embryos of medaka and found transient delay of brain development even when they hatched normally with low dose irradiation (2 and 5 Gy). In contrast, irradiation of higher dose of gamma-rays (10 Gy) killed the embry

Formation of ectopic neurepithelium in chick blastoderms: age-related capacities for induction and self-differentiation following transplantation of quail Hensen's nodes.

Hensen's node, regarded as the avian and mammalian homologue of Spemann's neural inducer (i.e., the amphibian dorsal blastoporal lip), has been transplanted in many previous studies to the germinal crescent of avian blastoderms to examine ectopic neural induction. All these studies have suffered from one or more major shortcomings, the most significant of which has been the lack of a reliable cell marker to determine the contributions of graft cells to ectopic embryos. In the absence of such marker, induced (i.e., derived from the host) and self-differentiated (i.e., derived from the graft) ne

The role of ion transport in the formation of sub-embryonic fluid by the embryo of the Japanese quail.

1. Using a modification of the method of New (1955) explanted blastoderms of the Japanese quail were cultured for up to 72 h, and the role of ions in the formation of sub-embryonic fluid (SEF) investigated. 2. Culture media deficient in either sodium or chloride ions reduced the volume of SEF secreted by up to a quarter. Ionic composition of the fluid was little altered, and sodium was transported against a concentration gradient. 3. Amiloride, an inhibitor of Na+/H+ exchange, reduced SEF production by half, whereas ionic composition and osmolality of the fluid was unchanged. 4. Likewise, acet

Observations on ploidy of cells and on reproductive performance in parthenogenetic turkeys.

Using flow cytometry, the ploidy levels of parthenogenetic turkeys were quantified from blastodisc stage to adulthood. Eggs were collected from noninseminated hens of the Beltsville Small White flock, known for their high degree of parthenogenesis, and the blastodermal cells from developing embryos were compared with those of embryos produced by hens inseminated with semen from males of the same flock. Erythrocytes of parthenogens from Day 10 of incubation to 27 mo of age were also used for ploidy determination. Sperm and erythrocyte preparations from normal males of the above flock served as

Possible role of calcium on oocyte development after intracytoplasmic sperm injection in quail (Coturnix japonica).

Although a rise in intracellular calcium concentration of vertebrate oocytes plays a pivotal role for the initiation of fertilization or oocyte activation, no study on this subject has been reported in birds. This study was conducted to study the role of intracellular calcium in relation to fertilization in avian oocytes. First, immediately after a quail oocyte was injected with a sperm, it was treated with strontium chloride as an inducer for intracellular calcium rise at doses of 0, 2.5, 5, 7.5, 10 mM for 4 hr in the culture medium and was followed by 20-hr culture. Organization and function of microfilaments during late epiboly in zebrafish embryos.

We report that, during epiboly in zebrafish, three F-actin--based structures appear only after the blastoderm migrates past the embryonic equator. They are composed of two ring-like F-actin structures that form at the deep cell and enveloping layer margins of the blastoderm and a punctate actin band that develops in the external yolk syncytial layer. Treatment with cytochalasin B or the calcium chelator dibromo-BAPTA results in the disruption of all three of these actin-based structures, leading to the slowing or immediate arrest of epiboly, respectively, followed by a failure of yolk cell occ

Functional analysis of the promoter of an early zygotic gene KLC2 in Aedes aegypti.

BACKGROUND: Aedes aegypti is an important mosquito vector that transmits arboviruses that cause devastating diseases including Zika, dengue fever, yellow fever and chikungunya. Improved understanding of gene regulation in the early development of Ae. aegypti will facilitate genetic studies and help the development of novel control strategies of this important disease vector. RESULTS: In this study, we demonstrated through transgenic assays that the promoter of an endogenous early zygotic gene KLC2 could drive gene expression in the syncytial blastoderm and early cellular blastoderm, which is

SCAR is a primary regulator of Arp2/3-dependent morphological events in Drosophila.

The Arp2/3 complex and its activators, Scar/WAVE and Wiskott-Aldrich Syndrome protein (WASp), promote actin polymerization in vitro and have been proposed to influence cell shape and motility in vivo. We demonstrate that the Drosophila Scar homologue, SCAR, localizes to actin-rich structures and is required for normal cell morphology in multiple cell types throughout development. In particular, SCAR function is essential for cytoplasmic organization in the blastoderm, axon development in the central nervous system, egg chamber structure during oogenesis, and adult eye morphology. Impact of 2% carbon dioxide during storage on egg quality and early embryo development.

Storing fertilized eggs for several weeks prior to incubation is a hatchery practice to meet fluctuating demands for chicks. However, prolonged storage leads to a progressive deterioration of the egg internal structures, which ultimately reduces hatchability. This decrease in egg quality results from an increase in albumen pH following the diffusion of CO2 naturally present in the egg, through the eggshell pores. In this study, we explored whether a CO2-enriched atmosphere (2%) could limit CO2 diffusion from eggs stored for 6, 13, 20, or 27 days at 12°C and 80% relative humidity, followed by a

A direct approach to the study of the effect of gravity on axis formation in birds.

A system has been developed to enable the normal development of aborted very early uterine avian embryos, outside the female's uterus. The shell-less aborted egg was put into a foster shell of a sister egg, previously laid by the same female. The empty space between the shell and aborted egg was filled with artificial uterine fluid. The reconstructed eggs were incubated at 42 degrees C for 30 hours in a vertical position. The atmosphere contained a high concentration of CO2 (8-10%). At the termination of the 30 h the eggs were transferred to incubation at 37 degrees C in normal atmospheric con

A pupal lethal mutation with a paternally influenced maternal effect on embryonic development in Drosophila melanogaster.

The maternal effect and zygotic phenotype of l(1)pole hole (l(1)ph) is described. l(1)ph is a zygotic lethal mutation which affects cell division of adult precursor cells in Drosophila larvae. The locus is located in 2F6 on the salivary gland chromosome map and four alleles have been characterized. Germ-line clonal analysis of amorphic alleles indicates that l(1)ph has a maternal effect lethal phenotype. Two lethal phenotypes are observed among embryos derived from female germ-line clones homozygous for amorphic alleles dependent upon the zygotic activity of l(1)ph+ introduced via the sperm. C

[On the method of injecting into the yolk-sac in teratological studies in rabbits (author's transl)].

A method is described by which the embryotoxic and teratogenic effects of substances can be examined through their application into the yolk-sac. On the ninth day of gestation rabbits (white New Zealand) were laparatomised and the uterus bicornis together with the blastoderms were treated in 3 different experiments. The results show that after application of physiological saline into the yolk-sac the high rate of malformations and resorptions are caused mainly by an avoidable venous obstruction and by postoperative inflammations and infections.
