The blood-retinal barriers.

The Blood-Retinal Barrier (BRB) is a situation of restricted permeability which is present between the blood and the retina. This barrier has a well defined anatomic substrate, particular permeability characteristics and appears to play a role of major importance in the pathophysiology and therapeutics of retinal disease. The BRB phenomenon operates fundamentally at two levels, retinal vessels and chorioepithelial interface, forming which may be better called an inner BRB and an outer BRB. The main structures involved are, for the inner BRB, the endothelial membrane of the retinal vessels, and

The blood-retinal barrier in angioid streaks.

The blood retinal barrier was investigated in a group of patients with angioid streaks and pseudoxanthoma elasticum, using the technique of vitreous fluorophotometry. Angioid streaks are breaks in the elastic layer of Bruch's membrane with atrophy of the adjacent pigment epithelium. The retinal pigment epithelial cells and their intercellular junctions form part of the blood retinal barrier. In this study, it was found that despite the presence of angioid streaks and extensive areas of pigment epithelial atrophy the blood retinal barrier was intact. In contrast, when disciform degeneration had

The Role of Blood-Retinal Barrier in Retinal Neurodegenerative Diseases.

Disruption of the blood-brain or blood-retinal barrier (BBB or BRB, respectively) is a common phenomenon in neurodegenerative diseases of the central nervous system. Although many existing reviews focus on the link between BBB disruption and neuronal degeneration in the brain, a similar analysis of the BRB integrity in retinal degeneration is currently unavailable. Like the BBB, the inner BRB is established by retinal blood vessels encapsulated in a neurovascular unit. The retinal neurovascular unit and BRB are affected not only in retina-specific neurodegenerative diseases (eg, diabetic retin

Comparative transcriptome analysis of inner blood-retinal barrier and blood-brain barrier in rats.

Although retinal microvessels (RMVs) and brain microvessels (BMVs) are closely related in their developmental and share similar blood-neural barriers, studies have reported markedly different responses to stressors such as diabetes. Therefore, we hypothesized that RMVs and BMVs will display substantial differences in gene expression levels even though they are of the same embryological origin. In this study, both RMVs and BMVs were mechanically isolated from rats. Full retinal and brain tissue samples (RT, BT) were collected for comparisons. Total RNA extracted from these four groups were proc

Roles of Drug Transporters in Blood-Retinal Barrier.

Blood-retinal barrier (BRB) includes inner BRB (iBRB) and outer BRB (oBRB), which are formed by retinal capillary endothelial (RCEC) cells and by retinal pigment epithelial (RPE) cells in collaboration with Bruch's membrane and the choriocapillaris, respectively. Functions of the BRB are to regulate fluids and molecular movement between the ocular vascular beds and retinal tissues and to prevent leakage of macromolecules and other potentially harmful agents into the retina, keeping the microenvironment of the retina and retinal neurons. These functions are mainly attributed to absent fenestrat

Implication of Hyperhomocysteinemia in Blood Retinal Barrier (BRB) Dysfunction.

Elevated plasma homocysteine (Hcy) level, known as hyperhomocysteinemia (HHcy) has been linked to different systemic and neurological diseases, well-known as a risk factor for systemic atherosclerosis and cardiovascular disease (CVD) and has been identified as a risk factor for several ocular disorders, such as diabetic retinopathy (DR) and age-related macular degeneration (AMD). Different mechanisms have been proposed to explain HHcy-induced visual dysfunction, including oxidative stress, upregulation of inflammatory mediators, retinal ganglion cell apoptosis, and extracellular matrix remodel

Blood-retina barrier in acute retinal branch vein occlusion.

BACKGROUND: Extensive clinical studies on retinal branch vein occlusion have not yet been able to clarify its pathogenesis. A study designed to look at the associated blood-retina barrier changes may contribute to a better understanding of the different forms of evolution of this pathology. METHODS: A prospective study was done in seven patients with recent large temporal branch vein occlusion. Vitreous fluorophotometry, fluorescein angiography and retinal colour photography were performed within the 1st week after the onset of symptoms, 1 week later, and at 12 and 24 weeks. RESULTS: A more

WITHDRAWN: Bioengineering approaches for modelling retinal pathologies of the outer blood-retinal barrier.

Источник пометил публикацию как отозванную

Alterations of the junctional complex of the outer blood-retinal barrier (oBRB), which is integrated by the close interaction of the retinal pigment epithelium, the Bruch's membrane, and the choriocapillaris, contribute to the loss of neuronal signalling and subsequent vision impairment in several retinal inflammatory disorders such as age-related macular degeneration and diabetic retinopathy. Reductionist approaches into the mechanisms that underlie such diseases have been hindered by the absence of adequate in vitro models using human cells to provide the 3D dynamic architecture that enables

[Blood-Retinal Barrier Drug Transport: Aiming to Develop Oral Administration for Retinal Diseases].

The blood-retinal barrier (BRB) consists of the inner and outer blood-retinal barrier (abbreviated as inner BRB and outer BRB). The inner and outer BRB's together regulate the transport of substances between the blood and neural retina via various membrane transporters. To utilize the oral administration for the pharmacotherapy of retinal diseases, it is necessary and important to understand many drug transport mechanisms in the BRB's. This review discusses the BRB transport research reported over more than a quarter of a century, particularly the development of in vitro and in vivo transport

The Blood-Retinal Barrier in the Management of Retinal Disease: EURETINA Award Lecture.

Retinal diseases are the main causes of blindness in the Western world. Diabetic retinopathy and age-related macular degeneration continue to increase in prevalence and as main causes of vision loss. Intravitreal anti-VEGF and steroid injections have raised new expectations for their successful treatment. These agents act by stabilizing the blood-retinal barrier (BRB). Our group defined the BRB by identifying for the first time the tight junctions that unite retinal endothelial cells and are the basis for the inner BRB, an observation later confirmed in retinal pigment epithelial cells and in

Ocular Manifestations of Emerging Flaviviruses and the Blood-Retinal Barrier.

Despite flaviviruses remaining the leading cause of systemic human infections worldwide, ocular manifestations of these mosquito-transmitted viruses are considered relatively uncommon in part due to under-reporting. However, recent outbreaks of Zika virus (ZIKV) implicated in causing multiple ocular abnormalities, such as conjunctivitis, retinal hemorrhages, chorioretinal atrophy, posterior uveitis, optic neuritis, and maculopathies, has rejuvenated a significant interest in understanding the pathogenesis of flaviviruses, including ZIKV, in the eye. In this review, first, we summarize the curr

Crystalline retinopathy: evaluation of blood-retinal barrier by vitreous fluorophotometry.

A 58-year-old man with typical crystalline retinopathy had started complaining of night blindness in both eyes at the age of 56 years. The visual acuity was 0.8 in the right eye and 0.7 in the left eye. Ophthalmoscopic examination showed crystal deposits scattered throughout the posterior pole of the fundus in both eyes. Electroretinogram showed subnormal amplitude and the light-peak/dark-trough ratio of the electrooculogram was reduced. Fluorescein angiography demonstrated atrophy of the pigment epithelium and partial loss of the choriocapillaris in the posterior pole. The crystal deposits we

Ferroptosis in autoimmune uveitis: potential links to blood-retinal barrier dysfunction and retinal inflammation.

Autoimmune uveitis (AU) is a vision-threatening intraocular inflammatory disease characterized by disruption of ocular immune privilege, blood-retinal barrier (BRB) dysfunction, and progressive immune-mediated retinal injury. Although corticosteroids and conventional immunosuppressive therapies remain clinically effective, their long-term use is limited by adverse effects, relapse, and incomplete disease control. Ferroptosis is an iron-dependent form of regulated cell death driven by phospholipid peroxidation. It may provide a mechanistic link between oxidative stress, retinal cell injury, and

Retinal capillary degeneration and blood-retinal barrier disruption in murine models of Alzheimer's disease.

Extensive effort has been made studying retinal pathology in Alzheimer's disease (AD) to improve early noninvasive diagnosis and treatment. Particularly relevant are vascular changes, which appear prominent in early brain pathogenesis and could predict cognitive decline. Recently, we identified platelet-derived growth factor receptor beta (PDGFRβ) deficiency and pericyte loss associated with vascular Aβ deposition in the neurosensory retina of mild cognitively impaired (MCI) and AD patients. However, the pathological mechanisms of retinal vascular changes and their possible relationships with

Regulation of blood-retinal barrier cell-junctions in diabetic retinopathy.

Loss of the blood-retinal barrier (BRB) integrity and subsequent damage to the neurovascular unit in the retina are the underlying reasons for diabetic retinopathy (DR). Damage to BRB eventually leads to severe visual impairment in the absence of prompt intervention. Diabetic macular edema and proliferative DR are the advanced stages of the disease where BRB integrity is altered. Primary mechanisms contributing to BRB dysfunction include loss of cell-cell barrier junctions, vascular endothelial growth factor, advanced glycation end products-induced damage, and oxidative stress. Although much i

Hypoglycemia promotes inner blood-retinal barrier breakdown and retinal vascular leakage in diabetic mice.

The blood-retinal barrier (BRB) serves as a physiological boundary regulating the passage of nutrients, waste, ions, proteins, and water to and from the retina. In patients with diabetic retinopathy, breakdown of the inner BRB (iBRB) results in damage to the neurovascular unit and is a principal cause of vision loss in the diabetic population. Here, we demonstrate that hypoglycemia, a common consequence of tight glycemic control and high glycemic variability, results in accumulation of the transcription factors hypoxia-inducible factor-1α (HIF-1α) and HIF-2α and the expression of dozens of HIF

Blood-retinal barrier (BRB) and blood-aqueous barrier (BAB) permeability were assessed by various methods to clarify conflicting reports on whether blood-retinal barrier permeability changes occur in diabetic and galactose-fed rats and to assess the potential role of aldose reductase in this process. Different molecular weight probes were utilized in rats fed for 7-11 months either a normal diet or a diet containing 50% galactose with/without the aldose reductase inhibitor AI1576 or Ponalrestat. The role of Müller cells in the formation of the blood-retinal barrier.

We have compared the ability of Müller cells and astrocytes to induce the formation of barrier properties in blood vessels. Müller cells cultured from the rabbit retina, and astrocytes and meningeal cells cultured from the rat cerebral cortex, were injected into the anterior chamber of the rat eye, where they formed aggregates on the iris. We have examined the barrier properties of the vessels in those aggregates and, for comparison, the barrier properties of vessels in the retina, ciliary processes and iris. Two tracers were perfused intravascularly to test barrier properties. The movement of
