Bluetongue.

Summary Bluetongue (BT) is an arthropod-transmitted viral disease of non-African ungulates, principally sheep. The disease results from vascular injury analogous to that of human haemorrhagic viral fevers, with characteristic tissue infarction, haemorrhage, vascular leakage, oedema, and hypovolaemic shock. Importantly, BT is not zoonotic. Bluetongue virus (BTV) infection of ruminants and vector Culicoides midges is endemic throughout many tropical and temperate regions of the world; however, within this global range the virus exists within relatively discrete ecosystems (syn. episystems) where

The pathology and pathogenesis of bluetongue.

Bluetongue (BT) is an insect-transmitted viral disease of wild and domestic ruminants and, occasionally, other species. Amongst domestic livestock, BT is most common in certain breeds of sheep whereas asymptomatic BT virus (BTV) infection of cattle is typical in enzootic regions. BT in cattle can be a feature of specific outbreaks, notably the current epizootic in Europe caused by BTV serotype 8. BTV replicates within mononuclear phagocytic and endothelial cells, lymphocytes and possibly other cell types in lymphoid tissues, the lungs, skin and other tissues. Infected ruminants may exhibit a p

Experimental bluetongue virus superinfection in calves previously immunized with bluetongue virus serotype 8.

The effect of a superinfection with bluetongue virus serotype 1 (BTV1) was evaluated on two groups of four calves. One group received a commercial inactivated BTV serotype 8 (BTV8) vaccine. This group and the non-vaccinated group of calves were challenged twice (4 months apart) with the European BTV8 strain isolated during the 2006-2007 epidemics. Calves were then infected with a BTV1 inoculum which was found to be unexpectedly contaminated by BTV serotype 15 (BTV15). BTV1 and BTV15 single infections were performed on two other groups of three BTV naïve calves. A severe clinical picture was ob

Investigation of a bluetongue disease epizootic caused by bluetongue virus serotype 17 in sheep in Wyoming.

OBJECTIVE: To characterize a 2007 bluetongue disease (BT) epizootic caused by bluetongue virus (BTV) serotype 17 in sheep in the Big Horn Basin of Wyoming. DESIGN: Cross-sectional study. ANIMALS: 1,359 sheep from ranches in Wyoming and Montana. PROCEDURES: Information on clinical signs and history of BT in sheep was obtained from ranchers and attending veterinarians. At 3 to 6 months after the 2007 BT epizootic, blood samples were collected from rams, ewes, and lambs within and outside the Big Horn Basin; blood samples were also collected from lambs born in the spring of 2008. Sera were tes

An outbreak of bluetongue in sheep in the Sudan.

An outbreak of bluetongue and the first isolation of the virus in the Sudan are reported. The disease occurred in sheep stressed by walking for five days when biting arthropods were prevalent. Estimates of the morbidity and mortality rates ranged from about 30 per cent and 2 per cent respectively in adult sheep to around 80 per cent and 100 per cent respectively in lambs. The virus was isolated by the inoculation of suckling mice and embryonated eggs with whole blood from febrile sheep. In a gel precipitation test it reacted with specific antiserum to type 10 BT8 strain. No other agent was iso

Scientific Opinion on bluetongue serotype 8.

To answer a question from the European Commission on the potential special characteristics of bluetongue virus (BTV) serotype 8 (BTV-8) compared to other serotypes and their possible impact on the epidemiology of the disease, a systematic literature review was carried out by a working group established by the Animal Health and Welfare Panel. Currently, three special features can be assigned to BTV-8, which are the ability to cause serious disease in cattle and goats, the ability to be transmitted transplacentally, and the ability to contaminate semen. The transplacental transmission and the co

Implications of a conserved region of bluetongue virus protein VP2 in cross-neutralisation of bluetongue virus serotypes.

Bluetongue (BT) is a vector-borne disease of ruminants caused by Bluetongue virus (BTV). Twenty-nine different serotypes of BTV are currently reported throughout the world. The main objective of this study is the development of a subunit vaccine model that could potentially be adapted to provide broad spectrum protection against multiple BTV serotypes, which the conventional vaccines fail to address. To this end, three different BTV proteins (conserved region of viral protein [VP]2, VP5 and NS1) were expressed and purified in an Escherichia coli expression system. The immunogenicity of these p

Characterization of bluetongue virus serotype 28.

Bluetongue virus (Reoviridae; Orbivirus, BTV), which is usually transmitted by biting midges, affects wild and domestic ruminants worldwide, thereby causing an economically important disease. Recently, a putative new BTV strain was isolated from contaminated vaccine batches. In this study, we investigated the genomic and clinical characteristics of this isolate, provisionally designated BTV-28. Phylogenetic analysis of BTV-28 segment 2 (Seg-2) showed that it is related to Seg-2 from BTV serotypes 4, 10, 11, 17, 20 and 24, sharing 64%-66% identity in nucleotide sequences (nt) and 59%-62% in ami

Bluetongue virus-induced hydranencephaly in cattle.

Direct inoculation of bluetongue virus into 125-day bovine fetuses resulted in development of hydranencephaly. The earliest lesions after virus inoculation were a severe necrotizing encephalitis, which was most prominent in the cerebrum, and an associated nonsuppurative meningitis. At birth, the brains of infected fetuses had thin-walled cerebral hemispheres, dilated lateral ventricles, and cerebral cysts. No gross lesions were observed in the brain stem or cerebellum. Two morphologically different lesions were present in the brain of a fetus sacrificed 20 days after virus inoculation. There w

[Bluetongue, an emerging disease in Northern Europe].

Bluetongue is a non contagious viral disease of sheep transmitted by bites of haematophagous midges. The disease is caused by an orbivirus belonging to the Reoviridae family. The genome is segmented in 10 double-strand RNA encapsidated in a non-enveloped spherical particle with a icosaedral symetry. Twenty distinct serotypes have been identified so far, each of them inducing limited cross-protection against the others. Sheep are usually the only ones showing clinical signs like pyrexia, congestion of mucosa and cyanosis of the tongue. However, cattle, goat and wild ruminants can be asymptomati

Bluetongue virus as a cause of hydranencephaly in cattle.

Hydranencephaly was produced in a foetus and a calf by intra-uterine infection with an attenuated Type 10 bluetongue virus. Laparotomy was performed on the respective dams and the foetuses, respectively 126 days and 138 days old, were inoculated intramuscularly through the uterine wall with 1 ml of a virus suspension containing 5 x 103 tissue culture infective doese. The younger feotus was aborted on Day 262, while the other one was born alive on Day 273. Both foetuses showed marked hydranencephaly.

A preliminary survey of the epidemiology of bluetongue in Kenya.

A survey of the species composition and distribution of the Culicoides midge populations at a range of sites where bluetongue is enzootic isolated a group of dominant species: C. cornutus, C. grahamii, C. magnus, C. milnei, C. pallidipennis and C. 23. dagger Monthly light-trap sampling of Culicoides showed that the population densities of the dominant species greatly increased after the rain seasons and that these species concentrated around flocks of sheep and cattle. The larval habitats of C. cornutus and C. pallidipennis were found associated with stock pens. Precipitin tests on blood-fed C

Bluetongue virus vaccine: conventional to modern approach.

Bluetongue (BT) is an economically important, infectious and non-contagious disease of ruminant animals. BT disease is caused by bluetongue virus (BTV) of the genus Orbivirus (the family Reoviridae). BTV is transmitted by certain species of biting midges of the genus Culicoides. Although originally BT was restricted to African continent, now it is present in all the continents except Antarctica. Conventional BT vaccines such as live attenuated and inactivated vaccines showed different degree of success in BT control. However, conventional vaccines have certain disadvantages of reversion to vir

Bluetongue virus in pregnant elk and their calves.

Two pregnant North American elk (Cervus canadensis), in the 3rd and 4th months of gestation, were inoculated with bluetongue (BT) virus (BTV) serotype 11. The virus was not isolated from the blood of the cows beyond postinoculation day (PID) 8, but was isolated from bone marrow and spleen samples obtained at necropsy on PID 190. Although neither cow had overt clinical signs of BT infection, fluctuations in specific neutralizing BTV antibody titers indicated viral replication. However, in 2 attempts, BTV was not recovered biologically via bites of colonized Culicoides variipennis (biting gnats)

Studies on bluetongue. VI. Animal transmission studies.

The Cyprus strain of bluetongue virus was successfully transmitted through six passages and the Station strain through one passage in calves. Although the animals developed no visible evidence of infection, viremia as shown by both passage and fluorescent antibody examination of infected foetal bovine kidney culture, and by serological conversion was nevertheless demonstrated. No enhancement of virulence for calves or sheep was shown following bovine passage. A ewe inoculated in late pregnancy with blood drawn from a calf 59 days after its infection, gave birth to a lamb from whose blood the v

Observations on transplacental infection with bluetongue virus in sheep.

Twenty-four ewes were inoculated with 1 of 2 strains of bluetongue virus type 4 at 40, 60, or 80 days of gestation. Two ewes aborted, 2 ewes died, and 1 was killed during the experiment, but their fetuses were recovered. At term, 2 mummified fetuses, 4 dead lambs, and 17 clinically healthy lambs were produced by 12 sheep, and the remaining 7 sheep were barren. Porencephaly and cerebellar dysgenesis were found in term lambs born to sheep inoculated at 40 and 60 days of gestation. Bluetongue Virus: From BTV-1 to BTV-27.

Bluetongue virus (BTV) is the type species of genus Orbivirus within family Reoviridae. Bluetongue virus is transmitted between its ruminant hosts by the bite of Culicoides spp. midges. Severe BT cases are characterized by symptoms including hemorrhagic fever, particularly in sheep, loss of productivity, and death. To date, 27 BTV serotypes have been documented. These include novel isolates of atypical BTV, which have been almost fully characterized using deep sequencing technologies and do not rely on Culicoides vectors for their transmission among hosts. Due to its high economic impact, BT i
