Forensic Research Progress on Bongkrekic Acid Poisoning.

Bongkrekic acid (BA) is a toxin with stable properties and no distinctive smell. It exists in common foods such as fermented edible grain products, potato products, spoiled tremella fuciformis and auricularia polytricha, as well as auricularia polytricha that has been soaked too long. It can easily cause food poisoning. At present, there is still a lack of complete method to detect BA, and no specific antidote of BA has been found. Therefore, BA poisoning is easy to be misdiagnosed or missed diagnosed, and its mortality rate remains high. In recent years, studies have revealed the toxic mechan

Assessing the toxicity of bongkrekic acid in zebrafish embryos.

Bongkrekic acid (BA) is a potent bacterial toxin found in certain fermented foods, which poses a serious risk to human health through rapid multi-organ failure. Its toxic effects on the early stage of vertebrates remain poorly understood. In this study, we addressed this gap using zebrafish embryos as a model to evaluate BA's developmental toxicity. Following 96-h exposure to BA at concentrations of 0.01, 0.05, 0.1, and 1 mg/L, mortality rates increased sharply between 0.05 mg/L (25 %) and 0.1 mg/L (79 %). Morphological assessments at BA concentrations ≥0.1 mg/L revealed significant reductions

Bongkrekic Acid-a Review of a Lesser-Known Mitochondrial Toxin.

INTRODUCTION: Bongkrekic acid (BA) has a unique mechanism of toxicity among the mitochondrial toxins: it inhibits adenine nucleotide translocase (ANT) rather than the electron transport chain. Bongkrekic acid is produced by the bacterium Burkholderia gladioli pathovar cocovenenans (B. cocovenenans) which has been implicated in outbreaks of food-borne illness involving coconut- and corn-based products in Indonesia and China. Our objective was to summarize what is known about the epidemiology, exposure sources, toxicokinetics, pathophysiology, clinical presentation, and diagnosis and treatment o

Bongkrekic acid reprograms hepatic lipid metabolism to promote systemic organ injury.

Bongkrekic acid (BK), a highly lethal mitochondrial toxin produced by Burkholderia gladioli pathovar cocovenenans, poses a significant threat to food safety. Although clinical reports have documented its harmful effects, the underlying metabolic mechanisms driving BK toxicity remain fullly understood. In this study, we utilized multi-omics techniques to explore the impact of BK on metabolic processes across multiple organs using a mouse model. Our results demonstrated that BK treatment significantly altered the liver's cellular composition, with hepatocytes becoming the dominant cell type. Not

Bongkrekic acid poisoning: a neglected toxin at the edge of global food safety.

Bongkrekic acid (BA) poisoning is a rare but highly lethal foodborne intoxication caused by Burkholderia gladioli pathovar cocovenenans. This heat-stable mitochondrial toxin irreversibly inhibits the adenine nucleotide translocator, leading to abrupt energy failure, metabolic collapse, and high case fatality rates. Although historically confined to specific regions and traditional fermented foods, recent outbreaks, particularly in Asia, suggest an expanding and underrecognized global risk. The first documented outbreak in Taiwan in 2024 highlights a shift from household fermentation to modern

Menadione induces a low conductance state of the mitochondrial inner membrane sensitive to bongkrekic acid.

When rat liver mitochondria are allowed to cycle Ca(2+) and are incubated in the presence of the pro-oxidant menadione, they undergo swelling, membrane potential (DeltaPsi) collapse, and ion release. These effects, which are inhibited by cyclosporin A (CsA), are fully consistent with the opening of the so-called permeability transition pore. However, when Ca(2+) cycling is abolished by EGTA, the mitochondria remain energized (DeltaPsi collapse and swelling are avoided), but Ca(2+) efflux, promoted by the chelating agent, is stimulated by menadione. This stimulation goes together with the relea

Physiologically based toxicokinetic-toxicodynamic (PBTK-TD) modeling of oral exposure to bongkrekic acid in rats.

Bongkrekic acid (BA), a secondary metabolite produced by Burkholderia gladioli pathovar cocovenenans (BGC) in food, represents a major etiological agent of severe foodborne intoxication. The physiologically based toxicokinetic-toxicodynamic (PBTK-TD) model serves as a valuable tool for dose prediction, enabling the estimation of toxicant exposure in biological systems. This study established a multi-compartment PBTK-TD model based on a time-course dataset obtained following single oral administration of BA in rats. Through optimization of kinetic parameters, the PBTK component effectively char

Controlling the Storage Time Is an Effective Measure to Reduce the Risk of Bongkrekic Acid Poisoning Caused by Wet Rice Noodles.

In October 2018, bongkrekic acid (BKA) poisoning cases caused by consuming wet rice noodles contaminated with Burkholderia gladioli were first reported in Guangdong Province, resulting in three cases, and two of them died. Due to the high fatality rate, the Guangdong Health Commission issued a code. The core risk-control elements of the code were storage temperature and storage time management, and the storage time is controlled through adopting a small sealed package with a clearly labeled production date and shelf life. In this study, we followed up on the code's implementation and compared

ANT-Mediated Inhibition of the Permeability Transition Pore Alleviates Palmitate-Induced Mitochondrial Dysfunction and Lipotoxicity.

Hyperlipidemia is a major risk factor for vascular lesions in diabetes mellitus and other metabolic disorders, although its basis remains poorly understood. One of the key pathogenetic events in this condition is mitochondrial dysfunction associated with the opening of the mitochondrial permeability transition (MPT) pore, a drop in the membrane potential, and ROS overproduction. Here, we investigated the effects of bongkrekic acid and carboxyatractyloside, a potent blocker and activator of the MPT pore opening, respectively, acting through direct interaction with the adenine nucleotide translo

Palmitoyl-carnitine increases RyR2 oxidation and sarcoplasmic reticulum Ca2+ leak in cardiomyocytes: Role of adenine nucleotide translocase.

Long chain fatty acids bind to carnitine and form long chain acyl carnitine (LCAC), to enter into the mitochondria. They are oxidized in the mitochondrial matrix. LCAC accumulates rapidly under metabolic disorders, such as acute cardiac ischemia, chronic heart failure or diabetic cardiomyopathy. LCAC accumulation is associated with severe cardiac arrhythmia including ventricular tachycardia or fibrillation. We thus hypothesized that palmitoyl-carnitine (PC), alters mitochondrial function leading to Ca(2+) dependent-arrhythmia. In isolated cardiac mitochondria from C57Bl/6 mice, application of

Dissociated ROS production and ceramide generation in sulfasalazine-induced cell death in Raw 264.7 cells.

Sulfasalazine (SSZ) is a drug used in inflammatory bowel disease, whose precise mechanism of action remains to be clarified. Here, we report that incubation of Raw 264.7 cells with SSZ but not salicylates [acetylsalicylic acid (ASA), 4-aminosalicylic acid (4-ASA), and 5-ASA] causes a mixed apoptotic and necrotic form of cell death. In contrast to its metabolites, sulfapyridine and 5-ASA, SSZ exposure in Raw 264.7 cells resulted in a threefold increase in ceramide generation, as well as a robust production of reactive oxygen species (ROS). However, inhibition of ceramide production by fumonisin

Spinosad induces programmed cell death involves mitochondrial dysfunction and cytochrome C release in Spodoptera frugiperda Sf9 cells.

Spinosad, a reduced-risk insecticide, acts on the nicotinic acetylcholine receptors and the gamma-aminobutyric acid receptor in the nervous system of target insects. However, its mechanism of action in non-neural insect cells is unclear. This study aimed to evaluate mitochondrial functional changes associated with spinosad in Spodoptera frugiperda (Sf9) insect cells. Our results indicate that in Sf9 cells, spinosad induces programmed cell death and mitochondrial dysfunction through enhanced reactive oxygen species production, mitochondrial permeability transition pore (mPTP) opening, and mitoc
