Onychomycosis due to Ulocladium botrytis.

A case of disto-lateral onychomycosis of the third toe of the right foot of a 45-year-old man is reported. The agent of infection was Ulocladium botrytis, a mycete not hitherto reported to infect humans. The patient underwent mycological examination consisting of direct microscope observation and culture of pathological material. Diagnosis was based on the macro- and microscopic characteristics of the colonies. Fragments of the latter were also examined by scanning electron microscope. Clinical and mycological recovery was achieved after 3 months of topical therapy with ciclopiroxolamine.

[Properties of the laccase of Botrytis cinerea].

Laccase from Botrytis cinerea, secreted by the fungus into the grape berry, is responsible for the oxidation phynomena in musts and wines (oxidasic casse). From a fungus culture, the enzyme was extracted for a study of its properties: specificity; influence of inhibitors, pH and temperature; The results confirm that the enzyme from Botrytis cinerea belongs to the laccase family, though it has some characteristic differences from the other laccases previously described.

Botrytis cinerea Protoplasts Production and Inoculation.

The absence of a cell wall turns fungal protoplasts very receptive to the uptake of exogenous genetic material and, consequently, they can be used for genetic transformation or transfection experiments. Additionally, isolated monosporic fungal strains, naturally infected or previously transfected with a mycovirus, can be used in assays of fungal protoplasts fusion. This technology will overcome the barriers of vegetative incompatibility with other Botrytis cinerea strains, or even with other fungi, and successfully allow the transference of mycoviruses from an infected to a non-infected fungal

Resveratrol Oxidation in Botrytis cinerea Conidia.

ABSTRACT Observations using light microscopy showed that approximately 30% of Botrytis cinerea conidia treated with semi-lethal concentrations (i.e., 60 mug/ml) of the grapevine phytoalexin resveratrol possessed intracellular brown coloration. This coloration was never observed in the absence of resveratrol or in conidia treated with resveratrol together with sulfur dioxide (antioxidant compound) or sodium diethyldithiocarbamate (inhibitor of laccase action), suggesting that discoloration resulted from the laccase-mediated oxidation of resveratrol. Further studies using transmission electron m

Analysis of plant cell death-inducing proteins of the necrotrophic fungal pathogens Botrytis squamosa and Botrytis elliptica.

Fungal plant pathogens secrete proteins that manipulate the host in order to facilitate colonization. Necrotrophs have evolved specialized proteins that actively induce plant cell death by co-opting the programmed cell death machinery of the host. Besides the broad host range pathogen Botrytis cinerea, most other species within the genus Botrytis are restricted to a single host species or a group of closely related hosts. Here, we focused on Botrytis squamosa and B. elliptica, host specific pathogens of onion (Allium cepa) and lily (Lilium spp.), respectively. Despite their occurrence on diffe

Procedures for the Study of Botrytis cinerea Proteome.

Proteomics has been revealed as a key set of technologies that provide a detailed description of the molecular processes involved in the development of a specific phenotype. "Omics" technologies can collect an incredible amount of information. Among them, proteomics is an invaluable tool for defining specific biological information by studying the complete set of proteins under specific conditions, the proteome; or specific subsets of proteins, the subproteome. It is a crucial instrument for describing protein post-translational modifications, the functional annotation of the genome, and the d

Fluorescence and Ultrastructural Microscopy for Botrytis cinerea.

Botrytis cinerea, a notorious pathogenic fungus, leads to postharvest decay in horticultural crops and substantial economic losses, while the pathogenicity of B. cinerea is closely related to its structural characteristics and cellular homeostasis. Among the currently available techniques, fluorescence and ultrastructural observations serve as crucial tools for investigating subcellular structures and cellular events during B. cinerea growth. These techniques enable the monitoring of cell structural integrity, such as variations in the cell membrane and cell wall, as well as intracellular chan

Viable Storage of Botrytis Conidia and Sclerotia.

Long-term storage of microorganisms is essential for maintaining a consistent laboratory strain collection. To prevent a reduction in viability of Botrytis stocks, we can make use of the naturally formed survival structures of this fungus, conidiospores and sclerotia. Conidiospore stocks are the preferred method of storage and can be stored in 20% glycerol at -80°C for up to 30 years. When conidiospore storage is not feasible, sclerotia can be stored at 0°C, and agar plugs containing mycelium can be stored at 4°C. Sclerotia and mycelium stocks will only last several years and thus require more

Antifungal activity of tetrasulfanes against Botrytis cinerea.

Various natural polysulfanes (RS(x)R', x > or = 3, R (double dagger) H), such as diallyltrisulfide and diallyltetrasulfide from garlic, are mostly harmless to humans, higher animals and plants, yet highly active against diverse microbes, including several fungi. Such natural organic sulfur compounds (OSCs) possess considerable practical potential against a wide range of agricultural pests. Unfortunately, their use is often hampered due to the inherently offensive smell, chemical instability and low water solubility. However, since the biological activity of polysulfanes is primarily based on t

Methods for Inoculating Botrytis cinerea on Leaves and Fruit.

Being a generalist pathogen with a broad host range, Botrytis cinerea causes tremendous postharvest loss and food waste, while the incidence of gray mold is contingent upon the pathogenicity of B. cinerea and the resistance of host plants. It can thus be anticipated that the goals for controlling gray mold can be achieved by either inhibiting the pathogenicity of B. cinerea or enhancing the resistance of host plants. To assess the pathogenicity of pathogens or the defense responses of hosts, a standardized inoculation protocol is indispensable. Notably, specific controlled conditions are also

Hormetic Effects of Carbendazim on the Virulence of Botrytis cinerea.

The ascomycete plant-pathogenic fungus Botrytis cinerea infects more than 1,400 plant species worldwide. Stimulatory effects of sublethal doses of fungicides on plant pathogens are of close relevance to disease management. In the present study, stimulatory effects of carbendazim on the virulence of B. cinerea to cucumber plants were investigated. Spraying carbendazim on cucumber plants at 3 to 200 μg/ml had stimulatory effects on the virulence of carbendazim-resistant isolates of B. cinerea and the maximum percent stimulations were 16.7 and 13.5% for isolates HBtom451 and HBstr491, respectivel

Ectopic expression of BOTRYTIS SUSCEPTIBLE1 reveals its function as a positive regulator of wound-induced cell death and plant susceptibility to Botrytis.

Programmed cell death (PCD) is integral to plant life and required for stress responses, immunity, and development. Our understanding of the regulation of PCD is incomplete, especially concerning regulators involved in multiple divergent processes. The botrytis-susceptible (bos1) mutant of Arabidopsis is highly susceptible to fungal infection by Botrytis cinerea (Botrytis). BOS1 (also known as MYB108) regulates cell death propagation during plant responses to wounding. The bos1-1 allele contains a T-DNA insertion in the 5'-untranslated region upstream of the start codon. This insertion results

Evaluation of the Antifungal Activity of Z24 Against Botrytis cinerea.

The gray mold disease of plants caused by Botrytis cinerea leads to severe economic losses. Our previous studies showed that the neocryptolepine derivative Z24 exhibits excellent antifungal activity, particularly against B. cinerea. To assess the application value of Z24, this study further evaluated its antifungal effects. The results of this study show that N-methyl-2-pyrrolidone, dimethyl sulfoxide, and cinnamon oil can all serve as solvents for Z24. The EC50 value of Z24 against B. cinerea was 0.8466 μg/ml, which was significantly lower than that of neocryptolepine, and there was no cross-

Effects of Azole Fungicides on Secreted Metabolomes of Botrytis cinerea.

Botrytis cinerea, gray mold, is one of the most notorious phytopathgens, causing serious economic loss in the agricultural industry. The phytotoxic effects are mainly derived from secreted virulence proteins and terpenoid-type secondary metabolites. Azole fungicides are commonly used to manage the disease. However, their biochemical effects other than sterol biosynthesis were not documented, especially toxic secreted metabolites. In this study, six azole fungicides were treated with in vitro and in vivo conditions. Comprehensive profiles of primary and secondary metabolites in culture media we

[Mechanism of thymol inhibiting Botrytis cinerea: PAO-H2O2 system].

Gray mold disease caused by Botrytis cinerea infection is one of the major crop diseases. The application of environmental-friendly fungicides to control gray mold disease has been drawing great attention. Thymol, a natural compound, showed strong antifungal activity against Botrytis cinerea. We investigated the role of polyamine oxidase (PAO)-dependent hydrogen peroxide (H2O2) production in thymol-inhibited B. cinerea growth by using physiological and biochemical approaches. The results showed that: 1) Thymol significantly inhibited the growth of B. cinerea, with remarkable increases in H2O2
